Bio-adhesives used clinically, commonly have the ability to fill surgical voids and support wound healing, but which are devoid of antibacterial activity, and thus, could not meet the particular needs of the infected wound site. Herein, a series of natural polyphenolic antibacterial bio-adhesives were prepared via simple mixing and heating of polyphenols and acid anhydrides without any solvent or catalyst. Upon the acid anhydride ring opening and acylation reactions, various natural polyphenolic bio-adhesives could adhere to various substrates (i.e., tissue, wood, glass, rubber, paper, plastic, and metal) based on multi-interactions. Moreover, these bio-adhesives showed excellent antibacterial and anti-infection activity, rapid hemostatic performance and appropriate biodegradability, which could be widely used in promoting bacterial infection wound healing and hot burn infection wound repair. This work could provide a new strategy for strong adhesives using naturally occurring molecules, and provide a method for the preparation of novel multifunctional wound dressings for infected wound healing.

In addition to the robust adhesive properties, there is a pressing demand for ideal adhesives in tumor surgery that possess anti-tumor therapeutic effects. In this study, we introduce BSA-MnO 2 -GP@Ca-Y (BMGY) bio-glue by integrating bovine serum albumin (BSA)-MnO 2 , genipin (GP), and Ca-Y zeolite. Ca-Y zeolite exhibits the thrombin activity for hemostasis, while the cross-linking of BSA, GP, and skin tissue induces wound adherence upon laser irradiation for normalized skin structure within nine days. The heat generated during the “photothermal suture” process ablates residual tumor cells and produces antigen fragments, which are internalized by antigen presenting cells. The released Mn ions subsequently activate the cGAS-STING pathway, enhancing immunogenicity. Consequently, tumor-infiltrating p-TBK1 and interferon-β levels are significantly increased, ensuring robust anti-tumor immunity following BMGY treatment. Thus, BMGY bio-glue achieves hemostasis, wound bonding, ablation of residual tumor cells, and tumor recurrence inhibition, simultaneously. Beyond Ca-Y zeolite, BSA-MnO 2 -GP serves as a versatile platform for loading other drugs or active species to boost therapeutic efficacy. Therefore, we present a successful bio-glue paradigm with significant translational potential for various postsurgical management applications.

Bacterial infection and wound hemorrhage remain critical determinants of mortality in traumatic injuries, necessitating the development of bioglues capable of rapid hemostasis and efficient bacterial elimination. While numerous bioglues have been reported, challenges including insufficient adhesive strength, prolonged curing time, cytotoxicity, and persistent microbial contamination limit their clinical utility. Herein, a series of multifunctional bioglues are prepared via a combination of o-nitrobenzyl alcohol-modified carboxymethyl cellulose (CNC-NB), dopamine-modified carboxymethyl chitosan (CMC-DA), and antibacterial drug tobramycin (TOB). Upon irradiation by UV light, these bioglues exhibited rapid gelation behavior, outstanding bioadhesion, and hemostatic performances. Moreover, the bioglues demonstrated exceptional biocompatibility and robust antibacterial efficacy, rendering them suitable for promoting infectious wound healing. This study establishes a versatile platform for next-generation bioglues combining immediate hemostasis, antibacterial efficacy, and tissue regeneration capabilities, offering a promising alternative to conventional surgical sutures.

Colon leakage is one of the most severe complications in abdominal trauma or surgery cases. It can lead to severe abdominal infection and abdominal adhesions, resulting in prolonged hospital stays and increased mortality. In this study, a photosensitive hydrogel is proposed, which can swiftly form a strong adhesion coating on the damaged colon after UV irradiation, to realize quick cure and suture-free repair of colon leakage. The newly developed biological gel consists of hyaluronic acid methacryloyl (HAMA) and hyaluronic acid o-nitroso benzaldehyde (HANB) in the optimal ratio of 3: 1, which exerts both the rapid photocuring properties of HAMA and the strong tissue adhesion properties of HANB. HAMA/HANB shows excellent adhesion stability on wet surfaces, presenting controllable mechanical properties, ductility, adhesion stability, and chemical stability; it also evades foreign body response, which relieves the degree of abdominal adhesion. The underlying mechanism for HAMA/HANB promoting wound healing in colon leakage involves the reconstruction of the colon barrier, as well as the regulation of the immune reaction and neovascularization. In all, HAMA/HANB is a promising alternative suture-free approach for repairing colon leakage; it has a reliable healing effect and is expected to be extended to clinical application for other organ injuries.

Postoperative anastomotic leakage (AL) and abdominal adhesions are two major complications after intestinal surgery, with an incidence of 2–25% for AL and 93% for adhesion. Until now, there is no method addressing AL and abdominal adhesions simultaneously. In this work, Bi-PEG–succinimidyl succinate (PEG-NHS), amino-gelatin (Agel) is combined with cefoperazone-sulbactam (SCF) to prepare a multifunctional bioadhesive (SCF/SEAgel) for the postoperative leakage and adhesion prevention. SCF/SEAgel possesses a great sealing capability for tissue, with a bursting pressure of 54 kPa. The loaded SCF endows the systems with good antibacterial properties. The in vivo antiadhesion experiments show that SCF/SEAgel possesses better anti-adhesion properties than the commercially used sodium hyaluronate gel. In the cecum leakage model, the SCF/SEAgel effectively seals the leakage with a survival rate of 100%, superior to commercial products (Sainaoning). Meanwhile, it significantly reduces tissue adhesion. Finally, the laparoscopic surgery with dogs shows that the SCF/SEAgel can be injected through minimally invasive surgery, demonstrating its ease of use. Combined with its great biocompatibility, SCF/SEAgel is very promising in intestinal surgery.

Nanocomposite membranes with specific recognition, durability and regeneration ability that can rapidly adsorb and separate target compounds have remarkable technological applications for areas ranging from solid-phase extraction devices to architecture. In this work, inspired by the highly bioadhesive performance of mussel protein, urgently desired molecularly imprinted nanocomposite membranes (MINCMs) were prepared by developing a simple “bio-glue” imprinted strategy. By simply immersing the “bio-glue” m-cresol-imprinted PDA@SiO2 into casting solution accompanied by persistently mechanically stirring, a highly bio-adhered and homo-dispersedly distributed structure could be generated into MINCMs during a phase inversion process, which directed the higher perm-selectivity and reusability. Additionally, due to the unique properties of PDA modified layers and SiO2 nanoparticles (high surface-to-volume ratio and large surface area), the as-prepared MINCMs not only exhibited rapid adsorption dynamics, but also possessed an excellent separation performance of template molecule (m-cresol in this work). The excellent separation (perm-selectivity factor is 3.477) and recognition behavior (imprinted factor is more than 3.0) along with the low preparation consumed and green, quick, facile synthesis conditions make the as-prepared MINCMs attractive in broad technological applications for areas ranging from drug delivery to bioseparation.

Although bio-adhesives have achieved many progresses, creating robust bio-glues that could withstand the complex physiological microenvironment remains a significant challenge such as limited bioadhesion strength and stability, as well as slow adhesion speed caused by pH, enzymes, salt, and bacteria. Herein, a series of strong bio-glues are successfully fabricated using a polyphenol–acrylamide–ruthenium crosslinking (PARC) reaction under visible light irradiation, involving acrylamides radical polymerization, polyphenols (extracts) coupling reaction, and ruthenium-mediated photochemistry. The resulting bio-glues underwent rapid sol–gel transition and exhibited adhesive properties within a short of time (≈5 s), which could quickly bond to various bio-tissues. Moreover, these bioadhesives demonstrate high adhesion strength and stability with mucosal tissue in the oral environment, effectively promoting rat and pig oral mucosal wound healing. It is anticipated that this class of visible light-responsive polyphenolic bio-glues could serve as a robust tool for addressing oral mucosal defects in the clinic, offering a promising alternative to traditional treatments for oral mucosal disorders.

In this study, a Paenibacillus polymyxa ZF129 microcapsule suspension (CS-ZF129) was prepared. P. polymyxa ZF129 was microencapsulated by gum arabic via the spray-drying method. Coating seeds with the CS-ZF129 not only increased the control efficacy against cucumber root rot to 46.30 ± 0.02%, but also promoted the growth of the cucumber roots and other aspects, effectively improving the colonization ability of polymyxa ZF129 in the cucumber root system, which was analyzed to clarify the biological control mechanism of the cucumber Fusarium root rot disease through the enzyme activity. This study provides an efficient means based on seed coating for the biological control of cucumber Fusarium root rot. Fusarium root rot, a destructive soil-borne fungal disease, necessitates eco-friendly biocontrol strategies. This study developed a microbial seed-coating approach using the antagonistic strain Paenibacillus polymyxa ZF129, formulated into a microencapsulated powder (10 8 CFU/g) and a suspension (CS-ZF129). CS-ZF129 application enhanced cucumber resistance, achieving 46.30 ± 0.02% disease suppression while promoting root growth. The maximum increase in the fresh weight of the root in the promotion of rectangular growth was 47.16%. The colonization dynamics of ZF129 in the rhizosphere were systematically tracked, revealing its antagonistic correlation with Fusarium proliferation. An enzymatic activity analysis further uncovered the underlying regulatory mechanisms, demonstrating induced defense responses through pathogenesis-related protein activation. These findings highlight ZF129’s dual functionality as a biocontrol agent and a plant growth promoter, offering a sustainable strategy against soil-borne pathogens.

Although smart bio-glues have been well documented, the development of internal bio-glues for non-invasive or minimally invasive surgery is still met with profound challenges such as safety risk and the lack of deep tissue penetration stimuli for internal usage. Herein, a series of smart internal bio-glues are developed via the integration of o-nitrobenzene modified biopolymers with up-conversion nanoparticles (UCNPs). Upon irradiation by near-infrared (NIR) light, the prepared smart bio-glues can undergo a gelation process, which may further induce strong adhesion between tissues under both dry and wet conditions based on multi-interactions. Moreover, those NIR light-responsive bio-glues with deeper tissue penetration ability demonstrate good biocompatibility, excellent hemostatic performance, and the potent ability to accelerate wound healing for both external and internal wounds. This work provides new opportunities for minimally invasive surgery, especially in internal wound healing using smart and robust bio-glues.

Bioadhesives crosslinked with dynamic bonds exhibit shear-thinning, self-healing, and on-demand detachment properties, but generally show a weak bonding performance due to their poor bulk strength. Obtaining a strong bioadhesive with reversible crosslinking remains a challenge. To address this issue, herein we engineered a dynamic thiol–aldehyde crosslinked solvent-free adhesive based on hyperbranched polymer. The adhesive was obtained by directly mixing a liquid hyperbranched polymer with thiol end groups (HBPTE) and benzaldehyde-terminated polyethylene glycol (PEGCHO) without any additional catalyst or solvent. The solvent-free strategy yielded a dense crosslinking structure with many aldehyde groups, so this HBPTE–PEGCHO adhesive can strongly bond to tissue and various non-biological substrates. In addition, the HBPTE–PEGCHO adhesive has self-healing and thermo-reversible bonding properties due to the dynamic thiol–aldehyde crosslinking matrix. In vivo wound healing experiments show that this HBPTE–PEGCHO adhesive is tissue-benign, suggesting it can be applied in clinical practice. Combining the hyperbranched polymer-based solvent-free strategy and dynamic thiol–aldehyde crosslinking chemistry provides a simple but effective way to engineer a multifunctional bioadhesive with the desired bonding performance.

Pancreatic leakage is severe and life-threatening when happening pancreatitis or conducting pancreatectomy on patients, which easily induces bleeding, infection and even death. Commercial tissue adhesives cannot meet the demands for pancreatic leakage sealing due to the weak adhesiveness, potential toxicity and inability to degrade. Few reports have been feasibly paid attention to pancreas sealants due to the complicated conditions such as enzymatic microenvironments, mechanical dynamics and pre-infections. Herein, we prepared a rapid high-adhesion, antibacterial, biodegradable and injectable bioadhesive based on the FDA-approved polyethylene glycol to seal the pancreatic leakage for clinical translations and commercial applications. The tetra-armed poly(ethylene glycol) succinimidyl glutarate (PEG-SG), tetra-armed poly(ethylene glycol) amine (PEG-NH 2 ) and tannic acid (TA) were integrated to rapidly form the adhesive hydrogel (gelling time < 5 seconds) for convenient operations, while this bioadhesive achieved tough tissue adhesion (∼ 30 kPa), robust antibacterial activity (∼ 95% efficiency) and biocompatibility, revealing the excellent performances for pancreatic wound closure. In the established pre-clinical pancreatectomy using Bama miniature pig model, the bioadhesive presented efficient outcomes on pancreatic leakage, and this indicated the promising feasibility of the bioadhesive for clinical trials and translations.

Effective wound management is critical in post-operative recovery, particularly in sensitive areas such as circumcision. The aim of this study was to design and assess the efficacy of a novel two-component polyurethane (PU) bioadhesive, designated as PU1000S, for its application in advanced wound care within the context of clinical circumcision procedures. The glue-type bioadhesive was fine-tuned to conformally adhere to the moist tissue surfaces. It rapidly absorbed interfacial water and cured within 160 s, ensuring remarkable surface adaptability to wet tissue surfaces. The PU1000S demonstrated superior lap-shear strength, peaking at 55.12 ± 6.88 kPa, along with exceptional durability. These attributes underscored its strong wet adhesion and remarkable resilience at the interface with moist tissues. Owing to its mechanical adaptability to wet skin tissue, the adhesive layer of PU1000S maintained stability under complex loading associated with twisting, folding and significant volumetric deformation, resulting in minimal debonding. In addition, PU1000S was found to significantly accelerate wound healing by promoting re-epithelialization and collagen deposition, confirming its excellent bioadaptability for initial closure and subsequent tissue repair and regeneration following circumcision. The comprehensive results position PU1000S as a promising candidate for advanced wound care in circumcision, offering superior performance in terms of wet adhesion, durability, and bioadaptability. Its application could potentially enhance clinical outcomes and elevate patient satisfaction.
