Preparation of Cellulose Nanoparticles from Foliage by Bio-Enzyme Methods

Zhengjie Tang,Mingwei Yang,Mingli Qiang,Xiaoping Li,Jeffrey J. Morrell,Yao Yao,Yanwei Su

There are vast reserves of foliage in nature, which is an inexhaustible precious resource. In this study, the chemical components of five foliage types (pine needles, black locust tree leaves, bamboo leaves, elm leaves and poplar leaves) were analyzed, including cellulose content, hemicellulose content, and lignin content. The bio-enzymatic method was then used to prepare cellulose nanoparticles (CNPs) from these five kinds of leaves, and the prepared CNPs were analyzed using TEM, FTIR, FESEM, and XRD. The results showed that the content of hemicellulose in bamboo leaves was the highest, and the lignin content in the other four leaves was the highest. The cellulose content in the five kinds of foliage was arranged from large to small as pine needles (20.5%), bamboo leaves (19.5%), black locust leaves (18.0%), elm leaves (17.6%), and poplar leaves (15.5%). TEM images showed that the CNPs prepared by the five kinds of foliage reached the nanometer level in width and the micrometer level in length; therefore, the CNPs prepared in this study belonged to cellulose nanofibers (CNFs). The results of FTIR and XRD showed that CNFs prepared by the enzyme treatment exhibited a typical crystalline structure of cellulose II. The degree of crystallinity (DOC) of CNFs prepared from pine needle, poplar leaves, and bamboo leaves are 78.46%, 77.39%, and 81.51%, respectively. FESEM results showed that the CNFs prepared from pine needles, poplar leaves and bamboo leaves by enzymatic method presents a three-dimensional (3D) network structure, and their widths are 31 nm, 36 nm, and 37 nm, respectively. This study provides a meaningful reference for broadening the use of foliage types and improving their added value. Keywords:foliage;cellulose nanoparticles;bio-enzyme

Jiaxin Zhao,Xiaoxiao Wu,Xushuo Yuan,Xinjie Yang,Haiyang Guo,Wentao Yao,Decai Ji,Xiaoping Li,Lianpeng Zhang

Cellulose is one of the most abundant, widely distributed and abundant polysaccharides on earth, and is the most valuable natural renewable resource for human beings. In this study, three different particle sizes (250, 178, and 150 μm) of Neosinocalamus affinis cellulose were extracted from Neosinocalamus affinis powder using bio-enzyme digestion and prepared into nanocellulose (CNMs). The cellulose contents of 250, 178, and 150 μm particle sizes were 53.44%, 63.38%, and 74.08%, respectively; the crystallinity was 54.21%, 56.03% and 63.58%, respectively. The thermal stability of cellulose increased gradually with smaller particle sizes. The yields of CNMs for 250, 178, and 150 μm particle sizes were 14.27%, 15.44%, and 16.38%, respectively. The results showed that the Neosinocalamus affinis powder was successfully removed from lignin, hemicellulose, and impurities (pectin, resin, etc.) by the treatment of bio-enzyme A (ligninase:hemicellulose:pectinase = 1:1:1) combined with NH3·H2O and H2O2/CH3COOH. Extraction of cellulose from Neosinocalamus affinis using bio-enzyme A, the smaller the particle size of Neosinocalamus affinis powder, the more cellulose content extracted, the higher the crystallinity, the better the thermal stability, and the higher the purity. Subsequently, nanocellulose (CNMs) were prepared by using bio-enzyme B (cellulase:pectinase = 1:1). The CNMs prepared by bio-enzyme B showed a network structure and fibrous bundle shape. Therefore, the ones prepared in this study belong to cellulose nanofibrils (CNFs). This study provides a reference in the extraction of cellulose from bamboo using bio-enzymes and the preparation of nanocellulose. To a certain extent, the utilization of bamboo as a biomass material was improved. Keywords:Neosinocalamus affinis;cellulose;nanocellulose;bio-enzymes;particle size

Bio-inspired Fe/Ti-MOF photocatalysts for efficient nitrogen fixation under mild conditions

Can Sun,Runze Guo,Zhexiao Zhu,Shouxin Zhu,Jingyi Qu,Zijie Fang,Xiaolu Xu,Jiahui Lin,Yangben Chen,Shijie Zhang,Hui Zheng

Photocatalytic nitrogen fixation as a green, environmentally friendly, mild and low energy-consuming nitrogen fixation is considered as an ideal way to produce ammonia. Herein, inspired by nitrogen-fixing enzymes, the MOF materials Fe-sdc and Ti-sdc were synthesized by a simple solvothermal synthesis method using Fe and Ti as metal nodes and 4,4′-stilbenedicarboxylic acid (H 2 sdc) as a ligand. The ammonia production rates of Fe-sdc and Ti-sdc were 61.01 μmol g −1 h −1 and 46.84 μmol g −1 h −1 . The 15 N 2 isotope experiments were utilized to determine the nitrogen source for ammonia synthesis during the nitrogen fixation process of Fe-sdc. Various characterization analyses were also utilized to analyze the reasons for the better nitrogen fixation performance of the two catalysts.

The chemo-enzymatic modification and degumming of hemp fiber by the laccase-2,2,6,6-tetramethylpiperidine-1-oxyl radical-hemicellulase system and physico-chemical properties of the products

Xiang Yeping,Yu Jianyong,Liu Liu,Zhang Ruiyun,Qu Yongshuai,Jing Miaolei

In this study, we found an environmentally friendly system in the degumming of hemp fiber without using a strong acid and alkali. The chemo-enzymatic modification and degumming of hemp fiber used an acetic acid–sodium acetate buffer solution containing laccase, hemicellulase and 2,2,6,6-tetramethylpiperidine-1-oxyl radical (TEMPO) under a mild condition (pH = 5, T = 50℃). Through comparing and analyzing the physical and chemical properties of fiber products under different treatments, we found the method involving treatment with TEMPO/laccase/hemicellulase not only greatly removed most of the gum attached on the surface of the raw hemp fiber, but also introduced surface-active aldehyde and carboxyl groups into the fiber. The performances of the hemp fibers with different treatments were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy and X-ray diffraction. We also found the product was finer, with a lower content of non-cellulosic components and it had better water uptake properties (moisture sorption and water retention value), which has potential application values in textile industries.

Self-sacrificial bioenzyme-reinforced injectable hydrogel bone adhesives for enhancing diabetic fracture healing

Dongming Duan,Song Liu,Tianhua Xiao,Aiju Lou,Haiyan Huang,Yu Cai,Zunlei Gong,Lei Zhou,Le Wang

The increased risk of diabetic fractures, driven by high oxidative stress and a proinflammatory response at the site of bone injury, remains a treatment challenge. Traditional internal fixation materials pose numerous limitations, prompting the emergence of new biomaterials as superior alternatives due to advancements in bone tissue engineering research. This study develops an injectable magnesium-gallate (MgGA) bioenzyme-reinforced hydrogel bone adhesive to remodel the local inflammatory microenvironment and enhance diabetic fracture healing. The MgGA bioenzyme significantly improves toughness and bone adhesive strength owing to its interaction with polymer chains. The MgGA-containing, catalyst-like injectable bone adhesive facilitates the scavenging of overexpressed reactive oxygen species, promotes macrophage polarization to the M2 phenotype, and attenuates early-stage inflammation. The pathological tissue microenvironment-responsive MgGA enables the self-sacrificial and sustained release of Mg 2+ and gallic acid, regulating redox balance and promoting angiogenesis and osteogenesis. The developed MgGA hydrogel serves as a promising multifunctional bone adhesive, offering fracture fixation and accelerating diabetic bone regeneration in vivo, thereby representing a novel strategy for treating diabetic fractures.

Bioenzyme-responsive L-arginine-based carbon dots: the replenishment of nitric oxide for nonpharmaceutical therapy

Chuyi Han,Qinghua Yu,Junhao Jiang,Xianming Zhang,Fan Wang,Mingyue Jiang,Ruihong Yu,Tao Deng,Chao Yu

Nitric oxide (NO) is a short-lived, bioactive gas that has been found to have affinitive effects on cardiovascular diseases as well as cancer biology, while NO deficiency may cause serious pathological responses. The existing chemically-synthesized NO donors have inevitable systemic toxicity and cannot be released adaptively. Hence, L-arginine, an endogenous NO precursor, merits investigation as a natural efficient NO donor. Herein, we designed amino acid-doped L-arginine CDs-based bioenzyme-responsive NO donors, which could adaptively replenish NO/ONOO− in response to different microenvironments. Our results indicated the mechanism of the NO/ONOO− supplementation of L-arginine-based CDs and their potential for nonpharmaceutical gas-involving theranostics for the first time.

Fiber characterization of old corrugated container bleached pulp with laccase and glycine pretreatment

Chen Guozheng,Dong Junjing,Wan Jinquan,Ma Yongwen,Wang Yan

In response to deal with the increasingly serious environmental problems and shortage of fiber raw materials, biological enzyme pretreatment is an effective way to replace a large number of chemical additives to improve the properties of waste paper fibers. Fiber characterization of old corrugated container bleached pulp with laccase and glycine (Lac/Gly) pretreatment was investigated by means of Fourier transform infrared spectroscopy (FTIR), headspace gas chromatography (HSGC), fiber quality measurements (FQA), X-ray diffraction method (XRD), and atomic force microscope (AFM). Results showed that, compared with the control pulp, the whiteness and brightness of the Lac/Gly-treated pulp increased by 16.17% and 7.41%, respectively. And the FTIR showed that Lac/Gly pretreatment promotes the effective removal of lignin by hydrogen peroxide (H 2 O 2 ) bleaching. The content of carboxyl groups in pulp increased remarkably by 21.92%. The paper physical analysis showed that the paper strength properties have improved remarkably. The fiber quality analyses indicated that the fiber length, coarseness, and curl index changed a little. The XRD analysis showed that the crystallinity decreased by 5.83% due to Lac/Gly treatment. The AFM analysis showed that through Lac/Gly treatment, the lignin and extracts over the fiber surface are decreased significantly.

Biological enzyme treatment of starch-based lithium-ion battery silicon-carbon composite

Runhong Wei,Ruhui Xu,Keyu Zhang,Feng Liang,Yaochun Yao

Silicon/carbon composites have the disadvantages of large volume expansion and high cost, which limits their commercial application. In this study, green and economic starch was used to prepare porous starch (PS) under the action of enzymes, and then nano-silica was embedded in the PS. A PS based carbon/silicon/carbon composite was prepared by coating and carbonizing the starch slurry, which can alleviate the volume expansion of silicon. The results show that the anode composite material with 20% silicon content has a high initial capacity of 869 mAh g−1 and an initial Coulombic efficiency of 66% at 0.2 A g−1, and the specific capacity is maintained 450 mAh g−1 after 100 cycles. When the silicon content reaches 30%, the reversible capacity of the composite is 1490 mAh g−1 at a current density of 0.2 A g−1, and the capacity remains 850 mAh g−1 after 100 cycles. Its excellent properties and stability are attributed to the abundant porosity of the carbon in the starch derived layer, which improves the structural stability and electrochemical kinetics. This method provides a reference for the sustainable and environmental protection of lithium-ion battery anode materials.

Study on the effects of different pectinase/cellulase ratios and pretreatment times on the preparation of nanocellulose by ultrasound-assisted bio-enzyme heat treatment

Xiaoxiao Wu,Xushuo Yuan,Jiaxin Zhao,Decai Ji,Haiyang Guo,Wentao Yao,Xiaoping Li,Lianpeng Zhang

With the development of science and technology, efficient, fast and green methods are increasingly being pursued. The production of nanocellulose by green methods, such as bio-enzymes-assisted ultrasound treatment, has been the focus of many studies. However, the yield of cellulose nanocrystals prepared by this method is very low. In this paper, by pretreatment of microcrystalline cellulose (MCC), nanocellulose was prepared by heating and stirring + pectinase/cellulase + ultrasonic treatment (HSt – P/C – Ultr). The effects of the ratios of pectinase and cellulase and the hydrolysis time on the yield of nanocellulose were studied. FTIR, XRD, SEM, TEM and TG were used to determine the structure, crystallinity, morphology and thermal stability of nanocellulose. The results showed that optimal hydrolysis conditions were determined as a pectinase : cellulase ratio of 1 : 1, 90 min and 50 °C. The yield of nanocellulose was about 32.0%. The yield of pectinase cellulase = 1 : 1 was higher than that of microcrystalline cellulose (MCC) treated by a single bio-enzyme. This indicated that the synergistic effects of pectinase and cellulase have a certain effect on the formation of nanocellulose. During the preparation, the crystalline form of cellulose did not change. It was still cellulose I with a crystallinity of 73.5%, which is 9.50% higher than that of microcrystalline cellulose (MCC), a width of 20–50 nm, a high aspect ratio and a winding network structure. Therefore, nanocellulose prepared by this method is an ideal toughening material for manufacturing composite materials.

Preparation and characterization of nanocellulose fiber (CNF) by biological enzymatic method

Jing Wang,Yumin Song,Futong Cui,Wenbin Wu,Haodong Zhu,Xiaoping Li,Kunyong Kang

In this paper, using Styphnolobium japonicum, Cryptomeria fortunei and Pinus yunnanensis as raw materials, cellulose (CE) was first extracted by chemical method (CM1) and biological enzymatic method 2 (BM2), and then further prepared into nanocellulose fiber (CNF) by biological enzymatic method 1(CPM (CPM stands for cellulase and pectinase)). The CE and CNF were characterized by FTIR, XRD, SEM, DSC and TG. The results showed that the CNF prepared by CM1-CPM was cellulose type II, CNF prepared by BM2-CPM was cellulose type I. The best crystallinity of the CNF prepared by CM1-CPM reached 74.1% (C. fortunei CNF), the best yield was 24.0% (S. japonicum CNF) and the minimum average diameter was 62.6 nm (C. fortunei CNF). However, the best crystallinity of CNF prepared by BM2-CPM reached 80.1% (C. fortunei CNF), the best yield was 27.4% (C. fortunei CNF), and the minimum average diameter was 31.6 nm (C. fortunei CNF). And also, the thermal degradation analysis of the CNF prepared from the three raw materials revealed that the CNF prepared by BM2-CPM had better thermal degradability. Finally, the thermal stability analysis of the C. fortunei CNF was carried out, and it was found that the residual mass of CNF prepared by BM2-CPM was 48%, while the residual mass of C. fortunei CNF prepared by BM1-CPM was 22%, which showed the C. fortunei CNF prepared by BM2-CPM had better thermal stability. Therefore, the properties of nanocellulose prepared by BM2-CPM are optimal.

Effects of nanoplastic exposure routes on leaf decomposition in streams

Jingjing Du,Xilin Wang,Tianying Tao,Yan Su,Xueting Zhang,Jing Shao,Yufan Zhang,Luyao Yu,Baodan Jin,Wenrui Qv,Xia Cao,Lan Wang,Yanqin Yang

Polystyrene nanoparticles (PS NPs) released from plastic products have been demonstrated to pose a threat to leaf litter decomposition in streams. Given the multitrophic systems of species interactions, the effects of PS NPs through different exposure routes on ecosystem functioning remain unclear. Especially dietary exposure, a frequently overlooked pathway leading to toxicity, deserves more attention. A microcosm experiment was conducted in this study to assess the effects of waterborne and dietary exposure to PS NPs on the litter-based food chain involving leaves, microbial decomposers , and detritivores (river snails). Compared to waterborne contamination, dietary contamination resulted in lower microbial enzyme activities and a significantly higher decrease in the lipid content of leaves. For river snails, their antioxidant activity was significantly increased by 20.21%–69.93%, and their leaf consumption rate was significantly reduced by 16.60% through the dietary route due to the lower lipid content of leaves. Besides, the significantly decreased nutritional quality of river snails would negatively influence their palatability to predators. The findings of this study indicate that dietary exposure to PS NPs significantly impacts microbial and detritivore activities, thus affecting their functions in the detritus food chain as well as nutrient cycling.

Bio-enzymatic synthesis of nitrogen-doped porous carbon/SnO2/carbon microspheres as high-performance composite materials for lithium-ion and sodium-ion batteries

Yin Li,Li Wang,Wenhui Ma,Bin Yang,Runhong Wei,Yaochun Yao

In this study, a nitrogen-doped 3D porous starch-derived carbon/SnO 2 /carbon (PSC/SnO 2 /C) composite is synthesized with porous starch as a carbon source by biological enzymatic hydrolysis . Compared with the traditional complex acid-base reagent method, the biological enzymatic method is more environmentally friendly and economical, and it can also naturally introduce nitrogen sources and dope the carbon layer . Many mesoporous nanostructures provide enough buffer space and promote the ions' and electrons’ transmission rate. The formation of the Sn– O –C bond between SnO 2 and carbon ensures the stability of the structure. As a result, the PSC/SnO 2 /C composite exhibits a high initial discharge capacity (1802 mAhg −1 at 0.2 A g −1 for LIBs and 549 mAh g −1 at 0.1 A g −1 for SIBs) and good cycle stability (701 mAh g −1 at 0.2 A g −1 after 100 cycles for LIBs and 271 mAh g −1 at 0.1 A g −1 after 100 cycles for SIBs). This synthesis method can prepare other energy storage systems such as fuel cells, supercapacitors , and metal ion batteries .
