Screening fluorescent dyes for anti-counterfeit labelling in rape seed (Brassica napus)

Tian Y.X.,Guan Y.J.,Hu J.,Wang Q.T.,Hu Q.J.,Zhu S.J.

Five fluorescent dyes – rhodamine B (RB), rhodamine 6G (R6G), lissamine rhodamine B (LRB), 3, 6-diamincacridine hulmisulfate (DH) and safranine T (ST) – were used to label seeds as an anti-counterfeiting technology. The effects of the d yes on physiological parameters of rape and the patterns of fluorescence of rape seedlings were investigated. The results showed that s oaking seeds for 24 hours in 0.1 mg RB ml−1, 0.1 mg LRB ml−1 or 0.1 mg ST ml−1 had no significant effects on seed germination, seedling growth, seedling protective enzyme activities or chlorophyll content. RB and LRB emitted bright red and bright orange fluorescence under a fluorescence microscope, when excited by green light (546 nm) and blue light (495 nm), respectively, whereas ST showed little difference compared with the control. Moreover, the vascular bundles of stem, seedling roots and aerial parts of seedlings treated with RB all emitted brilliant fluorescence for a long time, which could be used as a marker in seedlings. Our results suggest that labelling rape seeds with RB dye at a suitable concentration can be used as an anti-counterfeiting technique.

Xiao-Chun Yang,Xiao-Xue Wang,Chen-Yu Wang,Meng Yin,Ke Ma,Yan Li,Ke-Zheng Chen,Sheng-Lin Qiao

Meningitis is a critical condition, particularly prevalent among young children, necessitating prompt diagnosis and intervention to prevent severe complications and mortality. This study presents the design and application of Janus-adhesive ionogels for real-time monitoring of physiological parameters with a focus on the early detection and management of infantile meningitis. The ionogel features a dual-layer structure, with an inner adhesive layer that strongly adheres to the skin and an outer anti-adhesive layer that minimizes external interference, ensuring accurate and reliable monitoring. These ionogels demonstrate superior properties such as high tensile strength, breathability, anti-freezing capabilities, and excellent ionic conductivity, making them ideal for continuous, long-term monitoring. The integrated monitoring system effectively tracks joint activity, body temperature, and ECG signals, providing real-time data critical for early warning and intervention in meningitis cases. This research represents a significant advancement in the field of wearable health monitoring technologies, showcasing the potential of Janus-adhesive ionogels to enhance the performance of wearable devices and contribute to personalized medicine.

Altered life history traits and transcripts of molting- and reproduction-related genes by cadmium in Daphnia magna

Wei Xinrong,Li Xiao,Liu Hongsong,Lei Haojun,Sun Weijun,Li Dan,Dong Wu,Chen Hongxing,Xie Lingtian

Cadmium (Cd) is a non-essential element and can be toxic to aquatic organisms at low concentrations. Despite its well-known toxicity to Daphnia magna , the effects of Cd on physiological parameters (heart rate and thoracic limb activity) and molting- and reproduction-related genes are relatively understudied. In this study, D. magna were exposed to 0 (control), 25, 50 and 75 μg L −1 of Cd for 7 d and 21 d to determine the toxicity of Cd. The results showed that the Cd body burden in D. magna was significantly increased with elevated Cd concentrations, up to 13.4 μg Cd/g dry weight (dw) after exposure to 75 μg L −1 for 21 d. After 21 d of exposure, the body length and body weight of D. magna were significantly decreased in all Cd treatments compared to the control. The heart rate and thoracic limb activity were reduced by 4.3–11.7 and 5.0–10.3%, respectively. The levels of malondialdehyde (MDA) were increased by ~24–37% and the activity of catalase (CAT) was inhibited by ~50% compared to the control. The reproductive parameters (i.e., size of the first brood, the total number of offspring per female and the number of offspring per brood) were remarkably reduced, causing adverse effects on the population dynamics. In addition, the transcripts of genes ( cyp314 , cyp18a1 , ecra , usp , hr3 , cut , cht and cht3 ) related to the molting of D. magna were altered, whereas the transcripts of genes ( vtg1 , vtg2 and vmo1 ) related to reproduction were down-regulated. This study helps better understand the effects of Cd at different biological levels.

Integrated physiological and transcriptomic analyzes reveal the duality of TiO2 nanoparticles on alfalfa (Medicago sativa L.)

Zhao Chen,Zhipeng Guo,Mengli Han,Yuxi Feng,Jin Ma

Alfalfa ( Medicago sativa L.) is a feed crop due to its rich nutrition and high productivity. The utilization of titanium oxide nanoparticles (TiO 2 NPs) brings benefits to agricultural production but also has potential hazards. To investigate the duality and related mechanism of TiO 2 NPs on alfalfa, its different doses including 0, 50, 100, 200, 500, and 1000 mg L − 1 (CK, Ti-50, Ti-100, Ti-200, Ti-500, and Ti-1000) were sprayed on leaves. The results showed that greater doses of TiO 2 NPs (500 and 1000 mg L −1 ) negatively affected the physiological parameters, including morphology, biomass, leaf ultrastructure, stomata, photosynthesis, pigments, and antioxidant ability. However, 100 mg L −1 TiO 2 NPs revealed an optimal positive effect; compared with the CK, it dramatically increased plant height, fresh weight, and dry weight by 22%, 21%, and 41%, respectively. Additionally, TiO 2 NPs at low doses significantly protected leaf tissue, promoted stomatal opening, and enhanced the antioxidant system; while higher doses had phytotoxicity. Hence, TiO 2 NPs are dose-dependent on alfalfa. The transcriptomic analysis identified 4625 and 2121 differentially expressed genes (DEGs) in the comparison of CK vs. Ti-100 and CK vs. Ti-500, respectively. They were mainly enriched in photosynthesis, chlorophyll metabolism, and energy metabolism. Notably, TiO 2 NPs-induced phytotoxicity on photosynthetic parameters happened concurrently with the alterations of the genes involved in the porphyrin and chlorophyll metabolism and carbon fixation in photosynthetic organisms in the KEGG analysis. Similarly, it affected the efficiency of alfalfa energy transformation processes, including pyruvate metabolism and chlorophyll synthesis. Several key related genes in these pathways were validated. Therefore, TiO 2 NPs have positive and toxic effects by regulating morphology, leaf ultrastructure, stomata, photosynthesis, redox homeostasis, and genes related to key pathways. It is significant to understand the duality of TiO 2 NPs and cultivate varieties resistant to nanomaterial pollution.

Highly Sensitive Fabric Sensors Based on Scale-like Wool Fiber for Multifunctional Health Monitoring

Wenjing Fan,Cheng Li,Xiaoshi Li,He Tian

Highly sensitive, multifunctional, and comfortable fabric sensors with splendid electrical properties for precise detection of human physiological health parameters have attractive prospects in next-generation wearable flexible devices. However, it remains a non-ignorable challenge to construct a multifunctional fabric sensor to meet the requirements of compact structure, high sensitivity, fast response, excellent stability, and air permeability. Here, a wool felt@MXene fabric sensor (WF@MFS) prepared by felting large quantities of wool coated with MXene is reported for measuring multiple physiological parameters in a noninvasive manner. With the high conductivity and outstanding mechanical properties of MXene and the special scale-like surface structure of the wool fiber, the sensor exhibits remarkable sensing performance such as high pressure sensitivity (80.79 kPa–1), fast response (40 ms), low detection limit (12 Pa), and strong stability (>12,500 cycles). Furthermore, to avoid direct contact between MXene and the human body, the WF@MFS is encapsulated in pure wool without MXene, thereby enabling the fabricated sensor to be tightly integrated into a variety of clothing for monitoring different physiological signals and information about human activities. More importantly, we develop an intelligent cushion with a square and panda pattern and an intelligent neckerchief in the form of arrays based on the WF@MFS, which can intuitively observe the real-time force distribution of the thigh and cervical spine by means of machine learning when a human body sits in different postures. The sensor proposed in this work demonstrates the great ability to prevent cardiovascular disease and the related diseases caused by improper sitting postures in advance, paving a promising path for future wearable smart fabric electronics.

A novel anti-counterfeiting method: Application and decomposition of RB for broad bean seeds (Vicia faba L.)

Yixin Tian,Zhan Li,Fei He,Yajing Guan,Shuijin Zhu,Jin Hu

Combining with seed coating method, rhodamine B (RB), a kind of fluorescent compounds had been used in seed labeling as an anti-counterfeiting technology. Effects of broad bean seed coating with RB on physiological parameters and fluorescence performance in broad bean seedlings were performed. The prolonged fluorescence in vascular bundles of seedlings treated with RB could be used as a marker of seed anti-counterfeiting. The experiment showed that RB had no negative effect on seed germination, seedling growth, seedling protective enzyme activities, and the contents of malondialdehyde (MDA) and chlorophyll, after coating seeds with ratio 1   kg RB to 10–30   kg seeds. In addition, the RB adsorption by soil was accorded with the isothermal adsorption equations of Langmuir, which exhibited the linear correlation as 0.9959, the maximum adsorption as 1.716   mg/g, and the adsorption equilibrium constant as 0.042   L/mg. The soil adsorption of RB could be effectively degraded by UV/Fenton. Present research suggested that coating broad bean seeds with appropriate concentration RB were safe to seed germination and this technology was suitable for anti-counterfeiting application.

An ultrasensitive multimodal intracranial pressure biotelemetric system enabled by exceptional point and iontronics

Li Jie,Zhang Fan,Xia Xiaobin,Zhang Kaihang,Wu Jianhui,Liu Yulu,Zhang Chi,Cai Xinyu,Lu Jiaqi,Xu Liangquan,Wan Rui,Hazarika Dinku,Xuan Weipeng,Chen Jinkai,Cao Zhen,Li Yubo,Jin Hao,Dong Shurong,Zhang Shaomin,Ye Zhilu,Yang Minye,Chen Pai-Yen,Luo Jikui

The accurate monitoring of vital physiological parameters, exemplified by heart rate, respiratory rate, and intracranial pressure (ICP), is of paramount importance, particularly for managing severe cranial injuries. Despite the rapid development of implantable ICP sensing systems over the past decades, they still suffer from, for example, wire connection, low sensitivity, poor resolution, and the inability to monitor multiple variables simultaneously. Here, we propose an ultrasensitive multimodal biotelemetric system that amalgamates an iontronic pressure transducer with exceptional point (EP) operation for the monitoring of ICP signals. The proposed system can exhibit extraordinary performance regarding the detection of minuscule ICP fluctuation, demonstrated by the sensitivity of 115.95 kHz/mmHg and the sensing resolution down to 0.003 mmHg. Our system excels not only in the accurate quantification of ICP levels but also in distinguishing respiration and cardiac activities from ICP signals, thereby achieving the multimodal monitoring of ICP, respiratory, and heart rates within a single system. Our work may provide a pragmatic avenue for the real-time wireless monitoring of ICP and thus hold great potential to be extended to the monitoring of other vital physiological indicators.

Effects of selenium and sulfur on antioxidants and physiological parameters of garlic plants during senescence

Bo CHENG,Hai-feng LIAN,Ying-ying LIU,Xin-hui YU,Ya-li SUN,Xiu-dong SUN,Qing-hua SHI,Shi-qi LIU

A hydroponic study was conducted to determine the effects of selenium (Se: 0, 3, 6 μmol L –1 ) on senescence-related oxidative stress in garlic plants grown under two sulfur (S) levels. We evaluated the yields of plants harvested at 160 and 200 days after sowing. Plants grown under a low Se dose (0.3 μmol L –1 ) at low S level showed higher yields (12.0% increase in fresh weight yield, 13.7% increase in dry weight yield) than the controls, despite a decrease in chlorophyll concentration. Compared with control plants, the Se-treated plants showed lower levels of lipid peroxidation. The Se-treated plants also showed higher activities of glutathione peroxidase and catalase, but lower superoxide dismutase activities. Changes in F v / F m values and proline contents were affected more strongly by S than by Se. On the basis of our results, we can conclude that Se plays a key role in the antioxidant systems in garlic seedlings. It delays senescence by alleviating the peroxide stress, but it can be toxic at high levels. A high S level may increase tolerance to high Se concentrations through reducing Se accumulation in plants.
