Electrophysiological and behavioral responses of Tamarixia radiata (Hymenoptera: Eulophidae) to volatiles of nymphal Diaphorina citri (Hemiptera: Liviidae)

Liu Yan-Mei,Huang Yuan-Yuan,Wang Fei-Feng,Hu Yu-Wei,Zhang Zhi-Lin,Cuthbertson Andrew G S,Qiu Bao-Li,Sang Wen

Huanglongbing (HLB), a devastating citrus disease caused by Candidatus Liberibacter asiaticus, is efficiently vectored by the Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Liviidae). Tamarixia radiata (Waterston) plays a crucial role as an ectoparasitoid, preying on D. citri nymphs. By collecting and identifying headspace volatiles from fifth instar nymphs of D. citri using a gas chromatograph–mass spectrometer (GC–MS), we obtained a collection of 9 volatile compounds. These compounds were subsequently chosen to investigate the electrophysiological and behavioral responses of female T. radiata. At a concentration of 10 μg/μl, 9 compounds were compared with cis-3-hexen-1-ol (control), resulting in trans-2-nonenal inducing the highest relative electroantennogram (EAG) value, followed by hexanal, heptanal, n-heptadecane, tetradecanal, n-tetradecane, n-pentadecane, 1-tetradecanol, and 1-dodecanol. The top 5 EAG responses of female T. radiata to these compounds were further investigated through EAG dose–response experiments. The results showed positive dose–responses as concentrations increased from 0.01 to 10 μg/μl. In Y-tube olfactometer bioassays, female T. radiata exhibited a preference for specific compounds. They were significantly attracted to tetradecanal at a concentration of 10 µg/µl and trans-2-nonenal at 0.01 µg/µl, while no significant attraction was observed toward hexanal, heptanal, or n-heptadecane. Our report is the first to demonstrate that volatiles produced by D. citri nymphs attract T. radiata, which suggests that this parasitoid may utilize nymph volatiles to locate its host.

Electrophysiological Mechanism and Identification of Effective Compounds of Ginger (Zingiber officinale Roscoe) Shoot Volatiles Against Aphis gossypii Glover (Hemiptera: Aphididae)

Jiawei Ma,Ye Tian,Xuli Liu,Shengyou Fang,Chong Sun,Junliang Yin,Yongxing Zhu,Yiqing Liu

Aphis gossypii Glover (Homoptera: Aphidinae), a major pest of Chinese pepper (Zanthoxylum bungeanum Maxim), causes significant agricultural damage. Ginger (Zingiber officinale Roscoe) has shown potential as a source for developing botanical pesticides due to its strong bacteriostatic and insecticidal properties; however, the underlying mechanisms remain poorly understood. This study evaluated the repellent activity of ginger shoot extract (GSE) across four solvent phases—petroleum ether, trichloromethane, ethyl acetate, and methanol—against A. gossypii. The results demonstrated that GSE exhibited significant repellent effects, with the methanol phase showing the most pronounced activity. Twelve fractions were chromatographically separated from the methanol phase, and electroantennography (EAG) analysis revealed that fraction 4 induced strong EAG responses in both winged and wingless aphids. Further identification of active compounds in fraction 4 by gas chromatography–mass spectrometry (GC–MS) indicated the presence of terpenes, aromatics, alkanes, esters, and phenols as major constituents. Subsequent EAG analysis identified several key compounds—octahydro-pentalene (C1), (Z)-cyclooctene (C2), dimethylstyrene (C3), tetramethyl-heptadecane (C5), tetrahydro-naphthalene (C6), and heptacosane (C9)—as responsible for eliciting EAG responses in both aphid forms. Additionally, results from Y-tube olfactometer assays showed that (Z)-cyclooctene and heptacosane were significantly attractive, while octahydro-pentalene acted as a strong repellent to both winged and wingless aphids. These findings offer valuable insights for the development of synthetic attractants and repellents for A. gossypii and provide a theoretical foundation for utilizing ginger in the creation of botanical pesticides targeting this pest.

Electrophysiological and behavioural responses of Plutella xylostella L. (Lepidoptera: Plutellidae) to thirteen non-host plant essential oils

Chengfei Song,Xinyao Ma,Jiao Liu,Li Ma,Xingtao Qie,Xizhong Yan,Chi Hao

The diamondback moth (DBM), Plutella xylostella (L.), is a cosmopolitan obligate pest of cruciferous crops. Currently, DBMs management is largely dependent on broad-spectrum synthetic pesticides, which has led to the adverse effects on the environment, human health, and nontargeted organisms. Thus, alternative control methods are needed. The essential oils (EOs) derived from bioactive species may be developed as effective alternatives for the management of DBMs that is safe and eco-sustainable than conventional pesticides. In this study, we investigated electrophysiological (EAG) and behavioral responses of DBM adults to the thirteen non-host EOs of Acorus calamus , Ageratum conyzoides , Artemisia argyi , Chenopodium ambrosioides , Cymbopogon citratus , Foeniculum vulgare , Ocimum basilicum , Pelargonium hortorum , Perilla frutescens , Pogostemon cablin , Polygonum hydropiper , Rosmarinus officinalis , and Ruta graveolens in order to contribute to the development of low environmental impact management alternatives. EAG recordings revealed all the tested EOs significantly elicited EAG responses of DBM females, and the males were sensitive to the eleven EOs except A. conyzoides and O. basilicum . Meanwhile, EAG recordings of DBMs exhibited dose-dependent responses to these tested EOs in most cases. In Y-tube olfactometer bioassays, the EOs of A. calamus , C. citratus , P. hortorum , P. hydropiper , and R. graveolen demonstrated prominent repellent effects on DBM females at concentrations of 10 μg/μL or higher. The results of oviposition bioassays found the number of eggs on the two plants with the five tested EOs were less than that on the two plants with paraffin oil. And the oviposition indexes of the plants that fixed with the EOs of P. hortorum , R. graveolen , A. calamus , P. hydropiper , C. citratus , and paraffin oil were –71.81%, –59.43%, –57.46%, –51.11%, –35.41%, and 0.09%, respectively. These findings indicate that the five EOs, especially P. hydropiper EO, provide active and safe tools for natural repellents to prevent and control DBMs infestations of cruciferous vegetables.

The EAG response and behavior of the Saperda populnea L. to volatiles from poplar branches

Defu Chi,Xiaocan Li,Jia Yu,Xing Xie,Guangli Wang

In order to investigate the effect of organic volatiles from poplar species on the host plants orientation of Saperda populnea , the essential oil in the 2–3   years old branches from the sapling or mature trees of Populus simonii Carr. and P. simonii Carr.   ×   P. nigra L. were extracted using the steam distillation method. The chemical composition and relative content in essential oil from those branches was analyzed by GC–MS. The main components contained in those volatiles from the four kinds of branches are mainly aromatic compounds such as 1,2 - cyclohexanedione, 2 - cyclohexene - 1 - one, p - xylene and 1,2,3 - trimethylbenzene and so on. The relative content of aromatic compounds contained in branches from saplings was higher than that contained in branches from old tress. EAG response and behavioral reaction of S. populnea L. to those four kinds of distillated essential oil were tested. The results shown that male and female adults of S. populnea L. shown significant choice behavior and strong olfactory response to those essential oil distillated from branches of P. simonii Carr. and P. simonii Carr.   ×   P. nigra L.. The EAG bioassay of S. populnea L. to 10 kinds of monomers compounds which had relative higher contents was tested. The results shown that the EAG response of female adults of S. populnea L. to 1   mol   L −1 p-xylene was intensest and reached 1.027   mV, the strength of their EAG response to 0.1   mol   L −1 phenol took the second place. The EAG response of male adult of S. populnea L. to 1   mol   L −1 p-xylene was strongest and reached to 0.824   mV, the strength of their EAG response to 1   mol   L −1 salicylaldehyde took the second place. In the choice behavior test of S. populnea L. adult to monomeric compound carried out in ‘Y’ type olfactometer have found that female and male adult of S. populnea L. all appeared obvious positive taxis to 0.1   mol   L −1 p-xylene. The selection rate of female and male adults ware (76.7   ±   4.1)% and (71.7   ±   2.6)%, respectively. The selective reaction to 0.1   mol   L −1 1,2-cyclohexanedione and 2-cyclohexene-1-one were not obviously than that to 0.1   mol   L −1 p-xylene. The male and female adult of S. populnea L. appeared repelling behavior to 0.1   mol   L −1 salicylaldehyde.

Phytoremediation of cadmium-trichlorfon co-contaminated water by Indian mustard (Brassica juncea): growth and physiological responses

Chao Zhang,Feng He,Lanzhou Chen

In this study, the morphological and physiological responses of Brassica juncea to the stresses of Cadmium (Cd) and trichlorfon (TCF), and the phytoremediation potential of B. juncea to Cd and TCF were investigated under hydroponics. Results showed that Cd exhibited strong inhibition on biomass and root morphology of B. juncea as Cd concentration increased. The chlorophyll a fluorescence intensity and chlorophyll content of B. juncea decreased with the increased Cd concentration, whereas the malondialdehyde and soluble protein contents and superoxide dismutase activity increased. TCF with different concentrations showed no significant influence on these morphological and physiological features of B. juncea. The biomass and physiological status of B. juncea were predominantly regulated by Cd level under the co-exposure of Cd and TCF. B. juncea could accumulate Cd in different plant parts, as well as showed efficient TCF degradation performance. A mutual inhibitory removal of Cd and TCF was observed under their co-system. The present study clearly signified the physiological responses and phytoremediation potential of B. juncea toward Cd and TCF, and these results suggest that B. juncea can be used as an effective phytoremediation agent for the Cd-TCF co-contamination in water.

Electrophysiological and laboratory behavioral responses of a leaf beetle pest of elm, Ambrostoma quadriimpressum, to selected plant volatiles and essential oils

Yan-Chen Wang,Peng Li,De-Fu Chi

The leaf beetle Ambrostoma quadriimpressum Motschulsky (Coleoptera: Chrysomelidae) is distributed in China, Siberia, and elsewhere in northeastern Asia. This pest feeds only on buds and leaves of elm trees (Ulmaceae) and inhibits elm trees sprouting, increasing the risk of damage and destruction by other pests. We investigated selected plant compounds that could be used for the development of semiochemical-based push-pull methods for the control of this elm pest. Electroantennogram (EAG), Y-tube olfactometer, and wind tunnel bioassays were conducted to test the electrophysiological and behavioral response of A. quadriimpressum to nine individual volatiles and five essential oils. Individual volatiles and essential oils were selected based on their activity against insects in previous studies. The individual volatiles and essential oils were dissolved in dichloromethane and adsorbed by filter paper. Using a pipette, odor stimuli were delivered as 0.5-s puffs of air into a continuously humidified air stream in the odor delivery tube at 400 ml per min. Ambrostoma quadriimpressum adults display strong EAG responses to l mol l −1 isoeugenol, (1 R )-(+)-α-pinene, 1% pepper oil (derived from the pericarp of Piper chinense Miq.), and peppermint oil [derived from the aerial parts of Mentha haplocalyx (Briq.)] compared to the control. In behavioral assays conducted using a Y-tube olfactometer and a wind tunnel, adult beetles were attracted by (1 R )-(+)-α-pinene but repelled by 0.1% pepper oil. The results provide a basis for the development of a ‘push-pull’ strategy to manage A. quadriimpressum .

Treatments of orange-spotted grouper (Epinephelus coioides) against Cryptocaryon irritans with •OH, ClO2 or HCHO: Survival, physiological and histological response

Ziqing Zhong,Xiping Wu,Mindong Bai,Xiaodian Huang,Qilin Zheng,Chunxiang Ai

Cryptocaryon irritans causes one of the most serious diseases in various wild and cultured marine fish, leading to mass mortality and economic loss. In this study, hydroxyl radical (•OH) solution produced by strong ionization discharge combined with water jet cavitation effect was injected into orange-spotted grouper ( Epinephelus coioides ) aquaculture tanks for C. irritans control. The results showed that all C. irritans theronts were inactivated by •OH solution at concentrations of 0.5 mg/L within 2 min. •OH could induce alteration of shape, the absence of motility and macronucleus dispersion in theronts. A possible explanation was that the macronucleus of C. irritans might be damaged by •OH; as a result, its metabolism and life activities were disturbed. The •OH treatment increased the survival rate of E. coioides challenged with C. irritans from 64.7 ± 8.0% (mean ± SD) to 100% and reduced their infection intensity significantly. Stress response biomarkers such as malonaldehyde, glutathione, glutathione peroxidase, superoxide dismutase (SOD) and catalase levels in the gills of E. coioides at different time points were analysed. The SOD activity in the •OH group first decreased and then recovered to the initial level at the end of the experiment. The other stress response biomarkers had no significant difference from that in the uninfected control group after •OH treatment. Additionally, the gill of E. coioides in the •OH group exhibited slight and reversible transformation compared with the uninfected control group. Compared with •OH treatment, chlorine dioxide and formalin treatment reduced the survival rate, induced oxidative damage and changed the histological gill structure in E. coioides . In conclusion, •OH could be applied effectively to control C. irritans infection without affecting the normal physiological condition of E. coioides .

Identification and Field Bioassay of the Sex Pheromone of Trichophysetis cretacea (Lepidoptera: Crambidae)

Peng Cheng-Lin,Gu Ping,Li Juan,Chen Qi-Yu,Feng Chuang-Hong,Luo Huai-Hai,Du Yong-Jun

The jasmine bud borer Trichophysetis cretacea (Butler) (Lepidoptera: Crambidae) is an important agricultural pest of jasmine flowers Jasminum sambac in China. The extract from the pheromone gland of the female moth was analyzed by gas chromatography coupled to mass spectroscopy (GC-MS). (Z)-11-hexadecenyl acetate (Z11–16:Ac), (Z)-11-hexadecenal (Z11–16:Ald), and (Z)-11-hexadecenol (Z11–16:OH) were identified as the probable sex pheromone components. The electroantennogram (EAG) bioassay confirmed the results from the chemical analysis. Field bioassays conducted in the late summer and fall in Quanwei, Sichuan, China, showed that the synthetic chemical blend of Z11–16:Ac, Z11–16:Ald, and Z11–16:OH was highly attractive to male T. cretacea moths, and none of the three components was attractive by itself. All three components were necessary and the ratio of the three was critical. The highest number of male moths was captured when the ratio of Z11–16:Ac: Z11–16:Ald:Z11–16:OH was 10:10:1 at a dosage per lure of 200 μg Z11–16:Ac. The binary mixture of 200 μg Z11–16:Ac and 25 μg Z11–16:OH captured a number of Nymphicula mesorphna (10.3 ± 4.4). The results indicate that traps with synthetic pheromone lures can be used to monitor jasmine bud borer populations in the field and potentially to control this pest.

Electrophysiological and behavioral responses of Asian and European honeybees to pear flower volatiles

Weihua Ma,Denglong Long,Yi Wang,Xinyu Li,Jiaxing Huang,Jinshan Shen,Wenting Su,Yusuo Jiang,Jie Li

Honeybee pollination behavior is influenced by flower volatiles, which honeybees sense via olfactory receptors . Honeybees are only weakly attracted to pear flowers. To investigate the potential reasons, we extracted and determined the floral volatile compounds from three pear cultivars (Su, Ya, and Xuehua) using headspace solid-phase micro-extraction (HS-SPME) and gas chromatography-mass spectrometry (GC–MS). The effects of pear flower volatiles on the Asian honeybee ( Apis cerana cerana Fabricius) and the European honeybee ( Apis mellifera ligustica Spinola) were determined by electroantennogram (EAG) assays and behavioral tests in a three-arm olfactometer . Among the 76 flower volatiles detected with GC–MS, 21 were found in all three pear cultivars, accounting for approximately 70% of the total volatile content. 3-Methyl-1-butanol and (+)-limonene volatiles had the highest relative content. Five compounds elicited strong EAG responses in both bee species: 2-methylbutyraldehyde, 1-nonanal, 6-methyl-5-hepten-2-one, 3-methyl-1-butanol, and (+)-limonene. Neither bee species showed positive taxis to these volatiles. In behavioral tests, A. mellifera ligustica showed a low preference for 6-methyl-5-hepten-2-one (20%, 400 µg/µL) and 2-phenethyl alcohol (16.7%, 400 µg/µL). Apis cerana cerana showed a low preference for 6-methyl-5-hepten-2-one (6.7%, 400 µg/µL) and 1-nonanal (10%, 400 µg/µL), whereas its preferences for 3-methyl-1-butanol (43.3%, 400 µg/µL) and α-farnesene (40%, 400 µg/µL) were similar to that for the control. Therefore, a lack of attractive volatile compounds could explain why honeybees are only weakly attracted to pear flowers. Therefore, to achieve acceptable pollination in pear orchards, we suggest using flower-scent sugar syrup feeding and a saturation pollination strategy.

Effects of plant volatiles on electrophysiological and behavioral responses of Cryptorrhynchus lapathi

Qing-Jie Cao,Jia Yu,Ya-Li Ran,De-Fu Chi

Electroantennogram (EAG), Y-tube olfactometer, and wind tunnel bioassays were conducted to test the electrophysiological and behavioral responses of Cryptorrhynchus lapathi L. (Coleoptera: Curculionidae) to six individual volatiles and seven essential oils (compounded volatiles). The aim of this study was to select effective plant compounds that can be used in the development of semiochemical-based push–pull methods for the control of this harmful insect. Male and female C. lapathi displayed strong EAG responses to linoleic acid, α-pinene, ( Z )-3-hexen-1-ol, geraniol, turpentine oil, and salicylaldehyde. Y-tube olfactometer assays indicated that salicylaldehyde and α-pinene elicited strong repellent effects on female C. lapathi . Linoleic acid and ( Z )-3-hexen-1-ol showed greater luring effects on male and female C. lapathi compared with the control. Turpentine oil was stronger repellent, and geraniol showed stronger luring effects on male C. lapathi than the control. Wind tunnel assays with both male and female C. lapathi indicated that salicylaldehyde, α-pinene, and turpentine oil elicited repellent effects compared with the control. Linoleic acid, ( Z )-3-hexen-1-ol, and geraniol were stronger lures of both male and female C. lapathi than the control. These results provide a basis for the further development of C. lapathi luring and repellent agents.

Female-Biased Odorant Receptor LstiOR3 Mediates Oviposition Preference via Linalool Recognition in Loxostege sticticalis (Lepidoptera: Pyralidae)

Huanhuan Dong,Yafei Qu,Ertao Li,Jing Luo,Ibrahim Osman,Zhimin Wang,Hongnian Li,Kebin Li,Jiao Yin

The beet webworm, Loxostege sticticalis, whose oviposition behavior significantly affects population quantity and causes crop losses, has poorly understood olfactory mechanisms governing this behavior. Odorant receptors (ORs) play a critical role in the insect olfactory system. Here, three ORs (LstiOR1, LstiOR2, and LstiOR3) were cloned, with all highly expressed in female antennae. The two-electrode voltage clamp recording revealed that only LstiOR3 displayed broad tuning to nine host plant volatiles, especially linalool. Electroantennogram recordings confirmed that the nine ligands induced electrophysiological responses, with particularly strong responses observed for linalool in female antennae. Subsequently, dual-fragment RNA interference (RNAi) effectively silenced LstiOR3, significantly reducing the electrophysiological and behavioral responses of L. sticticalis to linalool. Additionally, molecular docking showed that LstiOR3 binds strongly to linalool through hydrogen bonds at VAL-199 and GLY-203. In summary, our studies elucidate the specific biological function of LstiOR3 via linalool detection in oviposition site selection.
