Impacts of Non-Lethal High-Temperature Stress on the Development and Reproductive Organs of Bradysia odoriphaga

Jingrong Hu,Rudoviko Galileya Medison,Seng Zhang,Peifang Ma,Caihua Shi

Simple SummaryBradysia odoriphagais a soil-dwelling insect native to China, and its preferred host is Chinese chives. In this study, non-lethal high-temperature as an important limiting factor to affect the population and development ofB. odoriphagawas assessed. Meanwhile, the physiological mechanism on non-lethal high-temperature to reduce the population was also identified. These would lay a key theoretical foundation for the future development of high-temperature products for environment friendly pest control strategies.AbstractBradysia odoriphagais an agricultural pest in China’s vegetable industry. In this study, pupae and adults were exposed to various non-lethal high-temperatures. The results demonstrated a decreased rate of eclosion once the pupae were exposed to temperatures exceeding 37 °C for 1 h. No effect on the lifespan of unmated female adults was observed after exposure to temperature stress, while unmated male adult lifespan decreased (>37 °C for 2 h). The size of the testis and ovaries for unmated male and female adults decreased, as did the fecundity and egg hatching rate for mated females. Compared with the control group (25 °C), the testis size of unmated male adults decreased after high-temperature stress followed by recovery at 25 °C for 1 h, though the size of the ovaries of female adults did not change. Additionally, the size of the testis and ovaries for unmated male and female adults decreased following high-temperature stress and 24 h of recovery at 25 °C. High temperatures affected males more than females; 37 °C is the critical temperature to control the population ofB. odoriphaga. These results lay the foundation for the future development of environmentally friendly high-temperature prevention and pest-control strategies. Keywords:Allium tuberosum;chive gnats;emergence;fertility;sublethal

Lipid metabolism-mediated fertility decline in male mice exposed to Fluorene-9-bisphenol: An integrated DNA methylation and transcriptomics analysis

Danni Jiang,Yang Yang,Xiaojuan Han,Qianni Li,Yingxiu Ma,Jun Jiao,Lan Chao

Fluorene-9-bisphenol (BHPF), a widely recognized alternative to bisphenol A (BPA), has been increasingly used in a wide range of industries. However, despite numerous studies, the direct mechanism underlying its male reproductive toxicity remains poorly understood. Here, a BHPF-exposed mouse model was established to evaluate the effect of BHPF on male fertility. The results showed that BHPF exposure induced reproductive dysfunction, including testis damage, spermatogenesis impairment, steroid hormone disruption and sperm quality degradation. Mechanistically, over 90,000 differentially methylated regions were identified and increased global testicular methylation levels indicated that DNA methylation may be associated with BHPF-induced testicular damage. Transcriptome analysis revealed 221 up-regulated genes and 227 down-regulated genes in BHPF-exposed mice, mainly enriched in the steroid biosynthetic pathway. Further combined methylome and transcriptome analysis revealed the critical methylated genes potentially involved in lipid metabolism. The differentially expressed genes (Cyp4a10 and Rdh1) were further confirmed to correlate with impaired male mouse fertility. Taken together, the results of this study provide a better understanding of the molecular mechanisms of toxicity effects induced by BHPF exposure from the perspective of methylome and transcriptome.

The role of N6-methyladenosine modification in benzene-induced testicular damage and the protective effect of melatonin

Lin Han,Jingyu Wang,Lei Zhang,Jiaru Jing,Wei Zhang,Ziyan Liu,Ai Gao

Benzene is a universal ambient pollutant. Population-based studies have shown that benzene exposure affects male fertility. However, the mechanism of benzene-induced reproductive toxicity is unknown. Here, we established a dynamic inhalation model and exposed C57BL/6J mice to 0, 10, and 50 ppm benzene (6 h/day, 6 days/week, 7 weeks). Our study revealed that benzene exposure caused testicular injury, including structural damage to spermatogenic tubules, reduced semen quality, and decreased testosterone levels. In addition, the decrease in the global level of N 6 -Methyladenosine (m 6 A) and the change of m 6 A important regulatory enzymes in mice testes suggested that m 6 A was involved in the benzene-induced testicular injury. Further genome-wide m 6 A methylation analysis showed that 1469 differential m 6 A peaks were present in the testes of control and benzene groups, indicating that benzene exposure modulated m 6 A methylation in testes. Furthermore, the comprehensive analysis of m 6 A-sequencing and transcriptome revealed that hypermethylated Rara and its consequent reduced expression impaired the sperm production process. In particular, melatonin alleviated benzene-induced testicular injury by modulating m 6 A-related genes. Overall, our research provides a new idea and fundamental knowledge into the possible mechanisms of m 6 A modifications in benzene-induced testicular impairment, as well as a new experimental basis for benzene-induced male fertility therapy.

Chlorpyrifos inhibits sperm maturation and induces a decrease in mouse male fertility

Xuelian Zhang,Wenbo Cui,Ke Wang,Rui Chen,Mingyue Chen,Kangshu Lan,Yanpei Wei,Chuanying Pan,Xianyong Lan

Background Pesticides, especially organophosphorus pesticides such as chlorpyrifos (CPF), play an important role in modern agriculture. Studies have shown that pesticide residues are an important cause of male reproductive injury in mammal. Aim The aim of this study was to evaluate the reproductive damage caused by CPF in male mice and investigate the underlying mechanisms. Methods In vivo , C57BL/6 mice (6–8 weeks old) were treated with CPF for 14, 70, and 80 days by intraperitoneal injection, intragastric administration, and dietary supplementation, respectively. Then, sperm from the cauda epididymidis was cultured in vitro to confirm the deleterious effects of CPF. Results The in vivo results indicated that, after treatment with CPF by dietary supplementation and intraperitoneal injection, the expression of reproduction-related genes in the mouse testes was altered, although the mice were fertile and the testes presented no morphological abnormalities. Notably, mating experiments revealed that the fertility of male mice was decreased following CPF administration by gavage. Sperm motility within the cauda epididymidis declined significantly after CPF treatment, which was accompanied by a decrease in sperm density, upregulation of relative reactive oxygen species (ROS) levels, and downregulation of glutathione reductase activity. In vitro incubation experiments showed that sperm rapidly lost their capacity for linear movement; the relative ROS levels also increased significantly, while the mitochondrial membrane potential (MMP) showed a significant decrease. However, the integrity of the plasma membrane was not affected by CPF administration. Conclusions The above data indicated that exposure to CPF reduces sperm motility by disrupting mitochondrial function and increasing the level of oxidative stress during sperm maturation, thereby reducing the fecundity of male mice.

Comparison of the Evaluation of Combination of Ultrasonography of the Reproductive Tract With Hormone Administration on Dairy Cow Fertility

Bingke Wang,Jinbang Xiao,Yongjie Ma,Chuxi Gao,Hanbing Li,Yonghong Jia,Yaping Jin,Pengfei Lin

Further optimization of reproduction management programs in dairy cows is a contemporary research topic. In this context, our study aimed to compare a hormone program, named “uterus-ovary monitoring and classified use of hormone program” (M+C), with the Pre-OvSynch program. The M+C was based on regular application of B-mode ultrasonography during a voluntary waiting period to monitor the uterus and ovaries, while using various treatments under different conditions. Results of the 30–33-day and 60-day pregnancy/artificial insemination after the first AI of M+C were significantly better than the Pre-OvSynch (p < 0.05). The pregnancy rates within 180 days in milk after M+C was significantly higher than that after Pre-OvSynch (p < 0.05). The total number of inseminations used for M+C was significantly lower than that for Pre-OvSynch (p < 0.01). The number of open days was fewer after M+C than after the Pre–OvSynch throughout the experimental period with highly significant differences (p < 0.01). In summary, the use of M+C enhances reproductive benefits and reduces the need for hormone drugs among cows.

Gestational exposure to arsenic reduces female offspring fertility by impairing the repair of DNA double-strand breaks and synapsis formation in oocytes

Cong Ma,Kaiqin Hu,Xueke Zhang,Yaru Yu,Zuying Xu,Tian Ma,Hongzhen Ruan,Yingying Zhang,Jiajia Wang,Xinyu Yue,Chunmei Liang,Huifen Xiang,Rui Guo,Yunxia Cao,Zhiming Ding

Arsenic is a pollutant that can cross the placenta; however, research on the effects of arsenic exposure during pregnancy on the fertility of female offspring is limited. To address this gap, we developed a mouse model to investigate the relationship between arsenic exposure during pregnancy and fertility in female offspring. Our fertility assessment revealed that gestational exposure to 1 mg/kg arsenic or higher (10 mg/kg) resulted in reduction in litter size, ovarian volume, and multistage-follicle number in female offspring. By assessing the in vitro developmental capacity of oocytes and zygotes, we confirmed that the reduced fertility was due not to impaired oocyte quality but rather to a reduction in oocyte quantity. Arsenic exposure impedes synapsis formation in MPI and compromises homologous recombination-mediated repair of double-strand breaks, resulting in fewer crossovers. This disruption activates the pachytene-checkpoint, hindering the progression of the MPI and resulting in the elimination of defective oocytes through p-Chk2 activation. Our study reveals for the first time the detrimental effects of arsenic exposure during pregnancy on the fertility of female offspring, underscoring the urgent need to prevent such exposure to safeguard reproductive health.

Agomelatine alleviates palmitic acid-induced mouse oocyte meiosis defects by restoring mitochondrial function

Zhao Ruolin,Tang Yujie,Zhang Yingbing,Liu Jinwang,Zhong Changsheng,Ji Bozhen,Song Weijia,Zhang Chengtu,Su Jianmin

Background/objective Palmitic acid (PA) is known to be elevated in the follicular fluid of women with obesity, negatively affecting female fertility. However, the mechanism by which PA exposure reduces female fertility is not fully understood, and how it can be treated requires further investigation. Methods We first established in vivo and in vitro models of mouse oocyte maturation at high concentrations of PA and determined the effects of treatment with agomelatine (Ago) which is a melatonin receptor agonist with antioxidant properties. We assessed oocyte maturation rates, spindle morphology and chromosome morphology, oxidative stress and apoptosis levels. Lastly, we examined energy levels, mitochondrial function, and mitochondrial synthesis-related protein expression levels. Results Our results showed that PA exposure disrupted spindle assembly and chromosome alignment, reduced microtubule stability, and impaired the meiotic maturation of oocytes. PA also disrupted mitochondrial function, leading to decreased ATP production, elevated Reactive Oxygen Species(ROS) levels, oxidative stress, and apoptosis. Remarkably, Ago supplementation promoted oocyte quality by restoring spindle/chromosome conformation, maintaining mitochondrial function, lowering ROS levels, and inhibiting apoptosis. Conclusions This study establishes that Ago ameliorates metabolic stress-induced oocyte deterioration through mitochondrial functional restoration, providing mechanistic insights into obesity-associated infertility. Importantly, our study identifies a potentially favorable drug for combating obesity-induced female infertility.

Epigallocatechin Gallate Cooperated with Hydrogel Encapsulation Enable High-performance Cryopreservation of Mouse Ovaries

Qi Feng,Jianping Wang,Dengyao Yi,Suhuan Li,Yue Cheng,Zhongrong Chen,Gang Zhao

Cryopreservation and transplantation of ovarian tissue have emerged as promising techniques for preserving fertility in prepubescent girls and cancer patients who cannot postpone treatment. However, this technology faces several challenges. In particular, conventional methods often fail to mitigate serious oxidative stress issues and cytotoxic damage caused by cryoprotectants, which can affect transplant outcomes. To overcome this bottleneck, a new strategy was proposed here, a new cryoprotectant formula (12% ethylene glycol+12% propylene glycol +0.5 M sucrose+75 μM epigallocatechin gallate) was developed and combined with hydrogel encapsulation to optimize ovarian cryopreservation. This approach avoids the utilization of DMSO and reduces the concentration of penetrating cryoprotectant by 20%. The findings indicate that this strategy significantly reduced oxidative damage, fibrosis degree, and tissue apoptosis during cryopreservation and enhanced the quality of cryopreserved mouse ovaries. Furthermore, in vivo and in vitro tests assessing ovarian and oocyte quality further validate the safety and reliability of this research protocol. This study is anticipated to provide a new strategy for female fertility preservation through ovarian cryopreservation.

Potential role of 4-hydroxyisoleucine in enhancing fertility in male mice with diet-induced obesity

Emmanuel Osei Nkansah,Yunzhu Lan,Hui Zhang,Binbin Xu,Qiaodan Li,Mohammad Ishraq Zafar,Jian Xu

Background: Obesity is associated with hormonal imbalance, increased oxidative stress, and inflammation in the testis. These conditions adversely affect sperm quality, leading to impaired male fertility. Therefore, therapeutic interventions to counteract the adverse effects of obesity are crucial. This study explored the therapeutic effects of 4-hydroxyisoleucine (4-HIL) on fertility in male mice with diet-induced obesity. Methods: C57BL6 male mice (n=45) were randomly divided into normal diet (ND, n=15) and high-fat diet (HFD, n=30) groups for 10 weeks. The HFD group was then randomized into untreated (HFD, n=15) and 4-HIL-treated (200 mg/kg/day, intraperitoneal injection, HFD + 4-HIL group, n=15) for 6 weeks. ND and HFD controls received saline (0.3 mL/30 g body weight) throughout the intervention period. Comprehensive evaluations included (1) metabolic assessments (body weight, glucose, insulin and pyruvate tolerance tests, and blood serum lipids), (2) sperm analysis (count, concentration, and morphology), (3) fertility testing (mating trials and in vitro fertilization), (4) testicular histopathology (fat deposition and apoptosis), (5) biochemical assays (reproductive hormones, oxidative stress markers, and inflammatory cytokines), and (6) molecular analyses (mRNA sequencing and qPCR validation of differentially expressed genes). Results: 4-HIL treatment improved metabolic parameters, including reduced weight gain, enhanced glucose tolerance, and optimized blood serum lipids, compared to HFD controls. Treated mice exhibited superior sperm quality with increased count and concentration, reduced histomorphological abnormalities in the testis, and attenuated oxidative stress and inflammation. Furthermore, the key spermatogenic gene expressions, including spem1 and spata24, were significantly optimized in the testes of mice treated with 4-HIL compared to those of untreated mice (HFD group). Conclusion: This study demonstrates that 4-HIL therapy ameliorates obesity-induced testicular dysfunction and improves fertility markers in mice. The beneficial effects of this compound on metabolic parameters, sperm quality, and spermatogenic gene expression suggest its potential as a therapeutic agent for obesity-related male infertility. Further research is warranted to elucidate the underlying mechanisms and assess the clinical translatability of these findings.

Endometrial ECM Modified Hemispherical Hydrogels Delivering MenSCs Promote the Regeneration of Thin Endometrium

Ming Chen,Xuemin Liu,Xiaoxia Song,Xiaofeng Ye,Zeshen Liang,Guoqing Chen,Peixian Cheng,Huihua Yang,Ling Shuai,Dongdong Li,Liping Wang

Thin endometrium (TE), a major cause of embryo implantation failure, increases the risk of early miscarriage, ectopic pregnancy, and perinatal complications. Owing to the poor utilization and short-term effectiveness of current stem cell therapies, their efficacy in improving reproductive outcomes in patients with TE is limited. Endometrial extracellular matrix (EMECM) modified hemispherical hydrogels (ECMHPs) are developed to deliver menstrual blood-derived stem cells (MenSCs) to promote TE repair. The irregular porous structure on one side of the hemispherical hydrogel microspheres significantly enhances their retention capability on the uterine wall, whereas the bioactive decellularized EMECM promotes MenSCs proliferation and paracrine function. In vitro experiments demonstrate that ECMHPs@MenSCs promote the proliferation of damaged human endometrial stromal cells (HESCs) while inhibiting their fibrosis and apoptosis. In TE rat models, intrauterine transplantation of the ECMHPs delivering MenSCs system (ECMHPs@MenSCs) extends retention time, promotes endometrial thickness by 2.3-fold, increases glandular number by 3.7-fold, inhibits endometrial fibrosis. Additionally, this system improves endometrial regeneration markers and uterine receptivity, and restores fertility, with a mean gestational sac number of 11.0 ± 2.8 compared with only 2.3 ± 2.1 in the untreated group. This study provides an efficient and convenient therapeutic strategy for TE repair and fertility restoration.

Minimally invasive delivery of human umbilical cord-derived mesenchymal stem cells by an injectable hydrogel via Diels–Alder click reaction for the treatment of intrauterine adhesions

Sentao Hu,Yangyang Dai,Liaobing Xin,Xiaowen Zheng,Zi Ye,Songying Zhang,Lie Ma

Intrauterine adhesions (IUA) are the most common cause of uterine infertility, and conventional treatments have not consistently achieved satisfactory pregnancy rates. Stem cell therapy shows promising potential for the clinical treatment of IUA. Although various advanced biomaterials have been designed for delivering stem cells to the uterine cavity, there remain significant challenges, particularly in devising therapeutic strategies for clinical application that minimize surgical incisions and conform to the intricate structure of uterine cavity. Herein, an injectable hydrogel loaded with human umbilical cord-derived mesenchymal stem cells (UCMSCs) was synthesized via the Diels-Alder click reaction for endometrial regeneration and fertility restoration, exhibiting suitable mechanical properties, good biocompatibility, and desirable degradation properties. Notably, this hydrogel permitted minimally invasive administration and integrated seamlessly with surrounding tissue. Our study revealed that the UCMSCs-laden injectable hydrogel enhanced cell proliferation, migration, angiogenesis, and exhibited anti-fibrotic effects in vitro. The implantation of this hydrogel significantly facilitated endometrium regeneration and restored fertility in a rat endometrial damage model. Mechanistically, in vivo results indicated that the UCMSCs-laden injectable hydrogel effectively promoted macrophage recruitment and facilitated M2 phenotype polarization. Collectively, this hydrogel demonstrated efficacy in regenerating damaged endometrium, leading to the restoration of fertility. Consequently, it holds promise as a potential therapeutic strategy for endometrial damage and fertility decline arising from intrauterine adhesions. Statement of significance Severe endometrial traumas frequently lead to intrauterine adhesions and subsequent infertility. Stem cell therapy shows promising potential for the clinical treatment of IUA; however, challenges remain, including low delivery efficiency and compromised stem cell activity during the delivery process. In this study, we fabricated an injectable hydrogel loaded with UCMSCs via the Diels-Alder click reaction, which exhibited unique bioorthogonality. The in situ-gelling hydrogels could be introduced through a minimally invasive procedure and adapt to the intricate anatomy of the uterus. The UCMSCs-laden injectable hydrogel promoted endometrial regeneration and fertility restoration in a rat endometrial damage model, efficaciously augmenting macrophage recruitment and promoting their polarization to the M2 phenotype. The administration of UCMSCs-laden injectable hydrogel presents a promising therapeutic strategy for patients with severe intrauterine adhesion.

Injectable, stable, and biodegradable hydrogel with platelet-rich plasma induced by l-serine and sodium alginate for effective treatment of intrauterine adhesions

Xiangyan Xie,Xue Ao,Ruijuan Xu,Hongyi Lv,Shiqiao Tan,Jinrong Wu,Lijuan Zhao,Yi Wang

The combination of pharmacological and physical barrier therapy is a highly promising strategy for treating intrauterine adhesions (IUAs), but there lacks a suitable scaffold that integrates good injectability, proper mechanical stability and degradability, excellent biocompatibility, and non-toxic, non-rejection therapeutic agents. To address this, a novel injectable, degradable hydrogel composed of poly(ethylene glycol) diacrylate (PEGDA), sodium alginate (SA), and l -serine, and loaded with platelet-rich plasma (PRP) (referred to as PSL-PRP) is developed for treating IUAs. l -Serine induces rapid gelation within 1 min and enhances the mechanical properties of the hydrogel, while degradable SA provides the hydrogel with strength, toughness, and appropriate degradation capabilities. As a result, the hydrogel exhibits an excellent scaffold for sustained release of growth factors in PRP and serves as an effective physical barrier. In vivo testing using a rat model of IUAs demonstrates that in situ injection of the PSL-PRP hydrogel significantly reduces fibrosis and promotes endometrial regeneration, ultimately leading to fertility restoration. The combined advantages make the PSL-PRP hydrogel very promising in IUAs therapy and in preventing adhesions in other internal tissue wounds.
