Foodborne pathogenic bacteria always threaten human health. Flavonoids are commonly used in antibacterial applications. Studying the antibacterial effect of flavonoids on bacteria is significant. Capillary electrophoresis is a versatile separation means. It has different separation modes, so, it can be applied in the analysis of both bacteria and flavonoids. In this study, an efficient method of indirect separation and detection of three bacteria Vibrio parahaemolyticus , Escherichia coli , and Staphylococcus aureus was developed. The determination of bacteria was realized by separating the aptamers corresponding to the bacteria by capillary sieving electrophoresis. This method avoided the disadvantages of low resolution, channel adsorption and bacterial aggregation, encountered in the direct separation of bacteria. The separation of the flavonoid kaempferide was realized by capillary zone electrophoresis, and kaempferide in Kaempferia galanga was determined. Kaempferide and the extract of Kaempferia galanga were applied in the antibacterial tests separately. Using the developed method, the bacterial growth curves and the curves of the kaempferide concentration were studied respectively. The results showed that both kaempferide and Kaempferia galanga could effectively inhibit the bacteria. The inhibitory effect on the terrestrial bacteria Escherichia coli , and Staphylococcus aureus was better than that on the marine bacterium Vibrio parahaemolyticus .

In this study, we synthesized polyelectrolyte complexed nanoparticles using an ion exchange reaction between poly(hexamethylene guanidine hydrochloride) and sodium caffeate. The morphology of the obtained antiparticle was observed by scanning electron microscopy, and FT-IR and XPS were employed for the structural characterization. The antimicrobial properties of E. coli and S. aureus were characterized through minimum inhibitory concentration (MIC), growth curve analysis, plate colony counting method, and crystal violet method. Notably, the sample showed a 100% bactericidal rate against E. coli at 0.095 μg/mL and against S. aureus at 0.375 μg/mL within 1 h, demonstrating excellent antimicrobial performance against E. coli and S. aureus. The CA-PHMG-containing acrylic resin coatings exhibited exceptional antimicrobial and antiadhesive properties when examined under an inverted fluorescence microscope, particularly at a 4% weight concentration of the antibacterial agent. This study holds vast potential for development in the field of antimicrobial coatings.

Bacterial infections caused by the surface of material are one of the world's major public health issues and the drug–resistance to the current antibiotics brings a serious concern to global healthcare. Although there have been significant efforts devoted to develop novel alternative antimicrobial materials, most of the developed organic antibacterial agents are derived from petrochemicals, which are suffered from long–term biosafety due to their non–degradability and potential long–term toxicity. In this study, amino acids (DL–Methionine, L–Tryptophan and L–Serine) and o–Vanillin were used as starting material to prepare fully bio–based Schiff bases and their Cerium coordinated metallic complexes. The structural characterization of ligands was performed by nuclear magnetic resonance spectrum (NMR), Fourier transform infrared spectroscopy (FT–IR), ultraviolet–visible spectrum (UV) and complexes were analyzed by X–ray electron diffraction (XPS) and FT–IR. The antibacterial performance of as prepared material was tested using minimum inhibitory concentration (MIC), growth curve, plate colony count, inhibition test (ZOI), staining of live/dead bacteria method. The results revealed that MIC of ligands was 0.5 mg/mL and 0.75 mg/mL against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) respectively. For plant colony count, compared to ligands, the bactericidal performance of complexes was increased by 20 times at 6 h, up to 32 μg/mL. Noteworthily, the complex of DL–Methionine Schiff base ligand with cerium (MSL–Ce) exhibited the best antibacterial activities, showing broad–spectrum and long–lasting antibacterial behavior in the coating against Gram–positive bacterial and Gram–negative bacterial. The fully bio–based antibacterial agents indicate tremendous potential for antimicrobial coatings on different substrates for a variety of applications such as marine antifouling and biomedical fields.

In this paper, the effect of different concentrations of sucrose stress on color formation of the Staphylococcus xylosus was investigated. The results showed that the highest a* value and the best coloring effect similar to those of nitrite were observed after the addition of 0.05 g/mL sucrose to stress the S. xylosus. UV-Vis and electron spin resonance spectra analysis showed that production of coloring product Mb-NO was significantly enhanced after 0.05 g/mL sucrose stress. The growth curve, reactive oxygen content, cell cycle, nitric oxide synthase (NOS) activity, zeta potential, cell size, and protein composition of S. xylosus were investigated to reveal the mechanism of sucrose stress to enhance the coloring effect of the strain. The result showed that sucrose inhibited the growth of S. xylosus, which changed the physiological state by activating the oxidative stress response. The stress altered the rate of intracellular metabolism of S. xylosus by delaying the cell cycle and increasing cell surface zeta potential and cell particle size. These changes altered the protein composition of the cells and significantly enhanced the activity of intracellular NOS, which could improve the chromogenic ability of S. xylosus. This study will provide theoretical support for sucrose stress on S. xylosus to enhance its coloring effect, and sucrose stress for S. xylosus might be a promising biological alternative to nitrite in meat products.

Ferulic acid (FA) is a natural antibacterial agent rich in plants, FA has excellent antioxidant and antibacterial properties. However, because of its short alkane chain and large polarity, FA is difficult to penetrate the soluble lipid bilayer in the biofilm to enter the cell to play an inhibitory role, limiting its biological activity. To improve the antibacterial activity of FA, with the catalytic condition of Novozym 435, four alkyl ferulic acid esters (FCs) with different alkyl chain lengths were obtained by fatty alcohols (including 1-propanol (C3), 1-hexanol (C6), nonanol (C9), and lauryl alcohol (C12)) modification. The effect of FCs on P. aeruginosa was determined by Minimum inhibitory concentrations (MIC), minimum bactericidal concentrations (MBC), Growth curves, alkaline phosphatase (AKP) activity, crystal violet method, scanning electron microscopy (SEM), membrane potential, PI, cell contents leakage. Results showed that the antibacterial activity of FCs increased after esterification, and the antibacterial activity significantly increased and then decreased with the extension of the alkyl chain of the FCs. Hexyl ferulate (FC6) showed the best antibacterial activities against E. coli and P. aeruginosa (MIC for E. coli was 0.5 mg/ml, MIC for P. aeruginosa was 0.4 mg/ml). And Propyl ferulate (FC3) and FC6 showed the best antibacterial activities S. aureus and B. subtilis (MIC for S. aureus was 0.4 mg/ml, The MIC of B. subtilis was 1.1 mg/ml). In addition, the growth, AKP activity, bacterial biofilm, bacterial cell morphology, membrane potential and cell contents leakage of P. aeruginosa after different FCs were investigated, which found that FCs could damage the cell wall of P. aeruginosa and showed different effects on the P. aeruginosa cell biofilm. FC6 showed the best inhibition on the biofilm formation of P. aeruginosa cells, which caused the surface of P. aeruginosa cells to be rough and wrinkled. Some P. aeruginosa cells showed aggregation and adhesion, even rupture. The membrane hyperpolarization was obvious, which appeared as holes, leading to cell contents leakage (protein and nucleic acid). All these results concluded that the antibacterial activities FCs against foodborne pathogens depended on different fatty alcohol esterification of FA. FC6 showed the best inhibition on P. aeruginosa due to its effect on P. aeruginosa cell walls and biofilms and the leak of the cell contents. This study provides more practical methods and a theoretical basis for giving full play to the bacteriostatic effect of plant FA.

Ethnopharmacology relevance Cananga oil (CO) is derived from the flowers of the traditional medicinal plant, the ylang-ylang tree. As a traditional antidepressant, CO is commonly utilized in the treatment of various mental disorders including depression, anxiety, and autism. It is also recognized as an efficient antibacterial insecticide, and has been traditionally utilized to combat malaria and acute inflammatory responses resulting from bacterial infections both in vitro and in vivo . Aim of the study The objective of this study is to comprehensively investigate the anti- Salmonella activity and mechanism of CO both in vitro and in vivo , with the expectation of providing feasible strategies for exploring new antimicrobial strategies and developing novel drugs. Methods The in vitro antibacterial activity of CO was comprehensively analyzed by measuring MIC, MBC, growth curve, time-killing curve, surface motility, biofilm, and Live/dead bacterial staining. The analysis of the chemistry and active ingredients of CO was conducted using GC-MS. To examine the influence of CO on the membrane homeostasis of Salmonella , we conducted utilizing diverse techniques, including ANS, PI, NPN, ONPG, BCECF-AM, DiSC3(5), and scanning electron microscopy (SEM) analysis. In addition, the antibacterial mechanism of CO was analyzed and validated through metabolomics analysis. Finally, a mouse infection model of Salmonella typhimurium was established to evaluate the toxic side effects and therapeutic effects of CO. Results The antibacterial effect of CO is the result of the combined action of the main chemical components within its six (palmitic acid, α-linolenic acid, stearic acid, benzyl benzoate, benzyl acetate, and myristic acid). Furthermore, CO disrupts the balance of purine metabolism and the tricarboxylic acid cycle (TCA cycle) in Salmonella , interfering with redox processes. This leads to energy metabolic disorders and oxidative stress damage within the bacteria, resulting in bacterial shock, enhanced membrane damage, and ultimately bacterial death. It is worth emphasizing that CO exerts an effective protective influence on Salmonella infection in vivo within a non-toxic concentration range. Conclusion The outcomes indicate that CO displays remarkable anti- Salmonella activity both in vitro and in vivo . It triggers bacterial death by disrupting the balance of purine metabolism and the TCA cycle, interfering with the redox process, making it a promising anti- Salmonella medication.
