A PHYB-PIF4-auxin module promotes tomato graft formation in response to elevated ambient temperature

Xianmin Meng,Feng Zhang,Wencai Yang,Qingmao Shang

Grafting is an effective technique for increasing the resistance of vegetables to biotic and abiotic stresses. It has been widely applied to produce solanaceous and melon vegetables. Temperature is an important external factor affecting graft formation. However, the molecular mechanism by which external ambient temperature affects tomato graft formation remains unclear. In this study, we demonstrated that elevating ambient temperature during grafting to 35 °C for more than 24 h after grafting accelerated vascular reconnection. We generated self- or heterografted combinations between phyB1B2 and pif4 loss-of-function mutant and wild-type plants, and were mutants unresponsive to graft formation at elevated ambient temperature. In addition, elevated ambient temperature induced SlPIF4 expression during grafting. SlPIF4 directly binds the promoters of auxin biosynthesis genes SlYUCCA s and activates their expression. Further investigation revealed auxin accumulation in the graft junction under elevated ambient temperature. The results illuminate the mechanism by which the PHYB-PIF4-auxin module promotes tomato graft formation in response to elevated ambient temperature.

AaEIL2 and AaERF059 are involved in fruit coloration and ripening by crossly regulating ethylene and auxin signal pathway in Actinidia arguta

Yukuo Li,Zhe Song,Xu Zhan,Miaomiao Lin,Xiaohan Li,Ran Wang,Leiming Sun,Hong Gu,Feng Wei,Jinbao Fang,Xiujuan Qi

In kiwifruit ( Actinidia arguta ), fruit coloration is typically accompanied by ripening; however, the intrinsic connection between these two processes remains unclear. In this study, we found that ethylene and auxin accelerated and suppressed fruit coloration and ripening, respectively, in A. arguta . ETHYLENE INSENSITIVE 3-LIKE 2 (AaEIL2) and ETHYLENE RESPONSIVE FACTOR (AaERF059) were mined and identified using yeast two-hybrid library screening and transcriptome analysis. AaEIL2, specifically induced to high expression by ethylene, was confirmed as a transcription factor that positively regulates coloration and ripening by targeting AaLDOX (leucoanthocyanidin dioxygenase involved in anthocyanin biosynthesis) and AaPG18 (polygalacturonase involved in cell wall degradation) based on its subcellular localization in Arabidopsis protoplasts, stable genetic transformation in transgenic tomato, and yeast one-hybrid and luciferase activity assays. AaERF059 also responds to ethylene and regulates ethylene-/auxin-mediated fruit coloration and ripening by targeting the downstream genes AaACS2 (ACC synthase, which is involved in ethylene biosynthesis) and AaGH3 (Gretchen-Hagen 3, which is involved in the auxin pathway). Overall, AaEIL2 and AaERF059 regulate ethylene-and auxin-mediated fruit coloration and ripening by maintaining a dynamic balance in a positive and negative regulatory manner. Our results not only identified key genes but also established an intrinsic connection between fruit coloration and ripening in A. arguta .

Evidence for the Involvement of Auxin, Ethylene and ROS Signaling During Primary Root Inhibition of Arabidopsis by the Allelochemical Benzoic Acid

Zhang Wei,Lu Li-Ying,Hu Li-Yan,Cao Wei,Sun Kai,Sun Qi-Biao,Siddikee Ashaduzzaman,Shi Run-Han,Dai Chuan-Chao

Allelopathy is mediated by plant-derived secondary metabolites (allelochemicals) which are released by donor plants and affect the growth and development of receptor plants. The plant root is the first organ which senses soil allelochemicals this results in the production of a shorter primary root. However, the mechanisms underlying this process remain elusive. Here, we report that a model allelochemical benzoic acid (BA) inhibited primary root elongation of Arabidopsis seedlings by reducing the sizes of both the meristem and elongation zones, and that auxin signaling affected this process. An increase in auxin level in the root tips was associated with increased expression of auxin biosynthesis genes and auxin polar transporter AUX1 and PIN2 genes under BA stress. Mutant analyses demonstrated that AUX1 and PIN2 rather than PIN1 were required for the inhibition of primary root elongation during BA exposure. Furthermore, BA stimulated ethylene evolution, whereas blocking BA-induced ethylene signaling with an ethylene biosynthesis inhibitor (Co2+), an ethylene perception antagonist (1-methylcyclopropene) or ethylene signaling mutant lines etr1-3 and ein3eil1 compromised BA-mediated inhibition of root elongation and up-regulation of auxin biosynthesis-related genes together with AUX1 and PIN2, indicating that ethylene signal was involved in auxin-mediated inhibition of primary root elongation during BA stress. Further analysis revealed that the BA-induced reactive oxygen species (ROS) burst contributed to BA-mediated root growth inhibition without affecting auxin and ethylene signals. Taken together, our results reveal that the allelochemical BA inhibits root elongation by increasing auxin accumulation via stimulation of auxin biosynthesis and AUX1/PIN2-mediated auxin transport via stimulation of ethylene production and an auxin/ethylene-independent ROS burst.

Detachment-accelerated ripening and senescence of strawberry (Fragaria × ananassa Duch. cv. Akihime) fruit and the regulation role of multiple phytohormones

Chen Jingxin,Mao Linchun,Mi Hongbo,Zhao Yuying,Ying Tiejin,Luo Zisheng

To study the detachment stress on the ripeness of strawberry fruit, physiological characteristics of strawberry fruit on and off plant during ripeness and senescence processes were investigated. The results indicated that the ripeness of strawberry fruit upon detachment was accelerated, in terms of firmness, soluble solid content and especially color development. The color of fruit off plant changed rapidly from white to full red in 1–2 days. The respiratory rate in fruit off plant was strengthened, higher than that on plant. Abscisic acid level and ethylene production in fruit off plant were also higher than those on plant and auxin degradation was exacerbated by detachment. Expression levels of FaMYB1 , FabHLH3 and FaTTG1 were generally reduced with phenotypes of redder color and more anthocyanin accumulation in fruit off plant. Results also suggested that the detachment initially stimulated ethylene and abscisic acid production and auxin degradation, which modulated ripening-related gene expression and at last enhanced fruit pigmentation.

Yucasin Alleviates Aluminum Toxicity Associated with Regulating Reactive Oxygen Species Homeostasis in Tomato Seedlings

Huabin Liu,Chuangyang Bai,Jiahui Cai,Yue Wu,Changwei Zhu

The phytotoxicity of aluminum (Al) to plants is well known. Auxin accumulation and reactive oxygen species (ROS) burst induced by Al toxicity are the key factors in root growth inhibition. Yucasin, an auxin synthesis inhibitor, effectively ameliorates Al phytotoxicity in tomato seedlings. However, the physiological mechanisms by which yucasin alleviates Al phytotoxicity in tomatoes remain elusive. Here, we examined the regulatory mechanisms of yucasin involved in tomato seedling growth under Al conditions through phenotypic, plant physiology analysis, and cellular experiments. Exogenous indole-3-acetic acid (IAA) application increased Al accumulation in tomato seedling roots, while yucasin decreased Al accumulation. Yucasin application reduced Al-induced ROS accumulation, lipid peroxidation, and cell death, enhanced root viability, and promoted tomato seedling root growth. Further, yucasin enhanced the antioxidant enzyme activities of superoxide dismutase, catalase, and peroxidase in plants under Al conditions. The results suggest that yucasin improves the scavenging capacity of ROS by maintaining the activities of antioxidative enzymes. This study elucidates the physiological mechanism by which yucasin alleviates Al phytotoxicity, highlighting its potential to enhance plant tolerance under acidic Al conditions.

Trichoderma-secreted anthranilic acid promotes lateral root development via auxin signaling and RBOHF-induced endodermal cell wall remodeling

Yu Chen,Yansong Fu,Yanwei Xia,Youzhi Miao,Jiahui Shao,Wei Xuan,Yunpeng Liu,Weibing Xun,Qiuyan Yan,Qirong Shen,Ruifu Zhang

Trichoderma spp. have evolved the capacity to communicate with plants by producing various secondary metabolites (SMs). Nonhormonal SMs play important roles in plant root development, while specific SMs from rhizosphere microbes and their underlying mechanisms to control plant root branching are still largely unknown. In this study, a compound, anthranilic acid (2-AA), is identified from T. guizhouense NJAU4742 to promote lateral root development. Further studies demonstrate that 2-AA positively regulates auxin signaling and transport in the canonical auxin pathway. 2-AA also partly rescues the lateral root numbers of CASP1 pro :shy2-2 , which regulates endodermal cell wall remodeling via an RBOHF -induced reactive oxygen species burst. In addition, our work reports another role for microbial 2-AA in the regulation of lateral root development, which is different from its better-known role in plant indole-3-acetic acid biosynthesis. In summary, this study identifies 2-AA from T. guizhouense NJAU4742, which plays versatile roles in regulating plant root development.

Transcriptomic and hormone analyses reveal mechanisms underlying petal elongation in Chrysanthemum morifolium ‘Jinba’

Wang Jingjing,Wang Haibin,Ding Lian,Song Aiping,Shen Feng,Jiang Jiafu,Chen Sumei,Chen Fadi

Key message Auxin regulates chrysanthemum petal elongation by promoting cell elongation. Transcriptomic analysis shows that auxin signal transduction may connect with other transcription factors by TCPs to regulate chrysanthemum petal elongation. Abstract As an ornamental species, Chrysanthemum morifolium has high ornamental and economic value. Petal size is the primary factor that influences the ornamental value of chrysanthemum, but the mechanism underlying the development of C. morifolium petals remains unclear. In our study, we tracked the growth of petals and found that the basal region of ‘Jinba’ petals showed a higher elongation rate, exhibiting rapid cell elongation during petal growth. During petal elongation growth, auxin was demonstrated to promote cell elongation and an increase in cell numbers in the petal basal region. To further study the molecular mechanisms underlying petal growth, the RNA-seq (high-throughput cDNA sequencing) technique was employed. Four cDNA libraries were assembled from petals in the budding, bud breaking, early blooming and full blooming stages of ‘Jinba’ flower development. Analysis of differentially expressed genes (DEGs) showed that auxin was the most important regulator in controlling petal growth. The TEOSINTEBRANCHED 1, CYCLOIDEA and PCF transcription factor genes ( TCPs ), basic helix-loop-helix- encoding gene ( bHLH ), glutaredoxin-C ( GRXC ) and other zinc finger protein genes exhibited obvious up-regulation and might have significant effects on the growth of ‘Jinba’ petals. Given the interaction between these genes in Arabidopsis thaliana , we speculated that auxin signal transduction might exhibit a close relationship with transcription factors through TCPs. In summary, we present the first comprehensive transcriptomic and hormone analyses of C. morifolium petals. The results offer direction in identifying the mechanism underlying the development of chrysanthemum petals in the elongated phase and have great significance in improving the ornamental characteristics of C. morifolium via molecular breeding.

Identification of Potential Auxin-Responsive Small Signaling Peptides through a Peptidomics Approach in Arabidopsis thaliana

Weigui Luo,Yuan Xiao,Qiwen Liang,Yi Su,Langtao Xiao

Small signaling peptides (SSPs) are a class of short peptides playing critical roles in plant growth and development. SSPs are also involved in the phytohormone signaling pathway. However, identification of mature SSPs is still a technical challenge because of their extremely low concentrations in plant tissue and complicated interference by many other metabolites. Here, we report an optimized protocol to extract SSPs based on protoplast extraction and to analyze SSPs based on tandem mass spectrometry peptidomics. Using plant protoplasts as the material, soluble peptides were directly extracted into phosphate buffer. The interference of non-signaling peptides was significantly decreased. Moreover, we applied the protocol to identify potential SSPs in auxin treated wild type and auxin biosynthesis defective mutantyuc2yuc6. Over 100 potential SSPs showed a response to auxin inArabidopsis thaliana. Keywords:small signaling peptides;plant protoplast;peptidomics;auxin

Daidzin from Pueraria thomsonii regulate root plasticity and cell proliferation

Shiyan Yue,Xiaoming Ye,Tiegui Nan,Chen Li,Luqi Huang,Yuan Yuan

Root plasticity is essential for optimizing crop yields and ensuring survival under stress. However, the regulatory roles of root-enriched secondary metabolites in developmental processes have remained largely unexplored. Pueraria thomsonii , a leguminous species distinguished by its rare tuberous roots, exhibits remarkable isoflavonoid accumulation. Chemical quantification and in vitro functional analyses have identified daidzin as a key regulator of root architectural plasticity in this species. Using an integrated approach combining Arabidopsis thaliana (L.) root phenotyping system, cellular thermal shift assays (CETSA) and thermal proteome profiling (TPP), daidzin was demonstrated to modulate root development via stem cell-related processes and auxin signaling pathways. Specifically, daidzin led to the reduced auxin levels, silenced DR5::GFP expression in the stele, and suppressed PIN protein expression accompanied by a lateral shift in PIN2 polarity. Furthermore, daidzin decreased SHR, SCR, PLT1, and PLT2 protein levels while increasing WOX5 and CYCD6;1 accumulation. In the wox5 mutant, daidzin inhibited root elongation, whereas in the p35S:WOX5-GR transgenic line, it enhanced root elongation and attenuated starch granule degradation. These findings reveal that WOX5 functionally counteracts daidzin-mediated root growth inhibition. Furthermore, daidzin stimulated cell proliferation in Pueraria thomsonii (Benth.) and induced root thickening in Panax quinquefolius (L.). Collectively, these results suggest that daidzin's coordinates root architectural plasticity by modulating auxin transport, polarity, and stem cell niche maintenance, highlighting its potential as a valuable tool for enhancing root development in economically important crops.

The function of BoTCP25 in the regulation of leaf development of Chinese kale

Zeng Jiajing,Yang Mengyu,Deng Jing,Zheng Dongyang,Lai Zhongxiong,Wang-Pruski Gefu,XuHan Xu,Guo Rongfang

XG Chinese kale (Brassica oleracea cv. ‘XiangGu’) is a variety of Chinese kale and has metamorphic leaves attached to the true leaves. Metamorphic leaves are secondary leaves emerging from the veins of true leaves. However, it remains unknown how the formation of metamorphic leaves is regulated and whether it differs from normal leaves. BoTCP25 is differentially expressed in different parts of XG leaves and respond to auxin signals. To clarify the function of BoTCP25 in XG Chinese kale leaves, we overexpressed BoTCP25 in XG and Arabidopsis, and interestingly, its overexpression caused Chinese kale leaves to curl and changed the location of metamorphic leaves, whereas heterologous expression of BoTCP25 in Arabidopsis did not show metamorphic leaves, but only an increase in leaf number and leaf area. Further analysis of the expression of related genes in Chinese kale and Arabidopsis overexpressing BoTCP25 revealed that BoTCP25 could directly bind the promoter of BoNGA3, a transcription factor related to leaf development, and induce a significant expression of BoNGA3 in transgenic Chinese kale plants, whereas this induction of NGA3 did not occur in transgenic Arabidopsis. This suggests that the regulation of Chinese kale metamorphic leaves by BoTCP25 is dependent on a regulatory pathway or elements specific to XG and that this regulatory element may be repressed or absent from Arabidopsis. In addition, the expression of miR319’s precursor, a negative regulator of BoTCP25, also differed in transgenic Chinese kale and Arabidopsis. miR319’s transcrips were significantly up-regulated in transgenic Chinese kale mature leaves, while in transgenic Arabidopsis, the expression of miR319 in mature leaves was kept low. In conclusion, the differential expression of BoNGA3 and miR319 in the two species may be related to the exertion of BoTCP25 function, thus partially contributing to the differences in leaf phenotypes between overexpressed BoTCP25 in Arabidopsis and Chinese kale.

Transcriptome profiling of postharvest strawberry fruit in response to exogenous auxin and abscisic acid

Chen Jingxin,Mao Linchun,Lu Wenjing,Ying Tiejin,Luo Zisheng

Main conclusion Auxin and abscisic acid regulate strawberry fruit ripening and senescence through cross-talk of their signal transduction pathways that further modulate the structural genes related to physico-chemical properties of fruit. The physiological and transcriptomic changes in harvested strawberry fruits in responses to IAA, ABA and their combination were analyzed. Exogenous IAA delayed the ripening process of strawberries after harvest while ABA promoted the postharvest ripening. However, treatment with a combination of IAA and ABA did not slow down nor accelerate the postharvest ripening in the strawberry fruits. At the molecular level, exogenous IAA up regulated the expressions of genes related to IAA signaling, including AUX/IAA , ARF , TOPLESS and genes encoding E3 ubiquitin protein ligase and annexin, and down regulated genes related to pectin depolymerization, cell wall degradation, sucrose and anthocyanin biosyntheses. In contrast, exogenous ABA induced genes related to fruit softening, and genes involved in signaling pathways including SKP1 , HSPs , CK2 , and SRG1 . Comparison of transcriptomes in responses to individual treatments with IAA or ABA or the combination revealed that there were cooperative and antagonistic actions between IAA and ABA in fruit. However, 17 % of the differentially expressed unigenes in response to the combination of IAA and ABA were unique and were not found in those unigenes responding to either IAA or ABA alone. The analyses also found that receptor-like kinases and ubiquitin ligases responded to both IAA and ABA, which seemed to play a pivotal role in both hormones’ signaling pathways and thus might be the cross-talk points of both hormones.

AUXIN RESPONSE FACTOR7 integrates gibberellin and auxin signaling via interactions between DELLA and AUX/IAA proteins to regulate cambial activity in poplar

Hu Jian,Su Huili,Cao Hui,Wei Hongbin,Fu Xiaokang,Jiang Xuemei,Song Qin,He Xinhua,Xu Changzheng,Luo Keming

Cambial development in the stems of perennial woody species is rigorously regulated by phytohormones. Auxin and gibberellin (GA) play crucial roles in stimulating cambial activity in poplar (Populus spp.). In this study, we show that the DELLA protein REPRESSOR of ga1-3 Like 1 (RGL1), AUXIN RESPONSE FACTOR 7 (ARF7), and Aux/INDOLE-3-ACETIC ACID 9 (IAA9) form a ternary complex that mediates crosstalk between the auxin and GA signaling pathways in poplar stems during cambial development. Biochemical analysis revealed that ARF7 physically interacts with RGL1 and IAA9 through distinct domains. The arf7 loss-of-function mutant showed markedly attenuated responses to auxin and GA, whereas transgenic poplar plants overexpressing ARF7 displayed strongly improved cambial activity. ARF7 directly binds to the promoter region of the cambial stem cell regulator WOX4 to modulate its expression, thus integrating auxin and GA signaling to regulate cambial activity. Furthermore, the direct activation of PIN-FORMED 1 expression by ARF7 in the RGL1–ARF7–IAA9 module increased GA-dependent cambial activity via polar auxin transport. Collectively, these findings reveal that the crosstalk between auxin and GA signaling mediated by the RGL1–ARF7–IAA9 module is crucial for the precise regulation of cambial development in poplar.
