Studies on polysaccharides from leaf skin of Aloe barbadensis Miller: Part II. Structural characteristics and molecular properties of two lower molecular weight fractions

Xiao-Dan Shi,Jun-Yi Yin,Liu-Jing Zhang,Ou-Ye Li,Xiao-Jun Huang,Shao-Ping Nie

Aloe barbadensis Miller ( Aloe vera ) has long been used in food , cosmetic and pharmaceutical industries. The solid wastes such as the spikes and the outer green skins are usually separated from the inner gel and discarded during the process of Aloe vera leaves. Three kinds of polysaccharides (designated as ASP-4N, ASP-6N and ASP-8N) were successfully purified from the leaf skin of Aloe vera in our previous work. The present study aimed to analyze the structural and molecular features of ASP-6N and ASP-8N using methylation and GC-MS, NMR, high performance size-exclusion chromatography coupled with multi-angle laser light scattering (HPSEC-MALLS) and scanning electron microscopy (SEM) techniques. Results showed that ASP-6N and ASP-8N were β-(1→4)-glucomannans with acetyl groups, which may attach to O -2, O -3 or O -6 of mannopyranosyl residues in the backbone as mono-, di-, or tri-acetylated forms. HPSEC-MALLS analysis indicated that the polysaccharide fractions from Aloe leaf skin adopted various conformations in the aqueous solution depending on their molecular mass. The polysaccharides had fibrous, ribbon-like and spherical morphology by SEM observation.

Structure identification of α-glucans from Dictyophora echinovolvata by methylation and 1D/2D NMR spectroscopy

Xiao-dan Shi,Ou-ye Li,Jun-yi Yin,Shao-ping Nie

Dictyophora echinovolvata is a kind of edible mushroom in the Dictyophora genus, of which polysaccharide is an important chemical substance. Herein, three polysaccharide fractions (DEP-4P, DEP-6P and DEP-8P) were prepared from water extract of D. echinovolvata using gradient ethanol precipitation method. Their chemical structures were analyzed by methylation analysis and 1D/2D NMR spectroscopy. Molecular weights were determined by multi-angle laser light scattering combined with size-exclusion chromatography (HPSEC-MALLS). HPSEC data showed that DEP-4P had the highest values of molecular weight, intrinsic viscosity and hydrodynamic radius. DEP-6P and DEP-8P had lower molecular weights, which contributed to their easily distinguished 1D and 2D NMR spectra. Methylation and NMR analysis suggested that the three fractions were linear α-(1 → 4)-glucans with α-Glc p residues linked to the backbone at C -6. Differences among the three fractions were the molar ratios of the identified glycosidic bonds. Repeating units of polysaccharides from D. echinovolvata were proposed as follows: Download : Download high-res image (60KB) Download : Download full-size image

Structural studies of a new fraction obtained by gradient ethanol precipitation from Acacia seyal gum

Jingen Li,Qihuan Deng,Xiaoyun Yu,Wenjun Wang

AY60 was a fraction precipitated from Acacia seyal gum by 60% ethanol, and its detailed structure was elucidated by methylation analysis and 1D/2D NMR spectroscopy. AY60 (Mw~92.4 kDa) was found to be a highly branched polysaccharide , and the terminal residues accounted for about 29.76% of total residues, which were t-Ara f (26.59%), t-Rha p (2.25%) and t-Gal p (0.92%) respectively. The branching residues included →3,6-Gal p 1→ (15.81%), →3,4-Gal p 1→ (13.59%) and →3,4,6-Gal p 1→ (11.25%). NMR spectroscopy ( 1 H NMR, 13 C NMR, 1 H– 1 H COSY, TOCSY, HSQC and HMBC) confirmed that the backbone of AY60 was composed of 1,3-linked galactopyransyl residues substituted at O -4 and O -6 positions. The side chains were mainly composed of →4)-β-Gal p A-(1→ (14.43%) and →3)-α-Ara f -(1→ (2.25%), and terminated by arabinofuranosyl (Ara f ) and occasionally by rhamnopyranosyl (Rha p ) or glacuronopyranosyl (Gla p ) residues.

Structural characteristics and rheological properties of alkali-extracted arabinoxylan from dehulled barley kernel

Lin-Yan Li,Yu-Xiao Wang,Ting Zhang,Jian-Fang Zhang,Meng Pan,Xiao-Jun Huang,Jun-Yi Yin,Shao-Ping Nie

Arabinoxylan (BIF-60) was isolated from barley water-insoluble fiber (BIF) by ethanol precipitation at 60 % (v/v). BIF-60 was composed of xylose (48.5 %) and arabinose (30.3 %). Its average molecular weight was 1360 kDa. Methylation and 1D/2D NMR analysis showed that BIF-60 possessed β-(l→4)-xylan as backbone, comprised of un-substituted (1,4-linked β-Xyl p , 56.9 %), mono-substituted (1,2,4-linked and 1,3,4-linked β-Xyl p , 22.1 %) and di-substituted (1,2,3,4-lin4ked β-Xyl p , 18.4 %) xylose units, as well as other residues (T-Ara f -(1→, T-Xyl p -(1→, →5)-Ara f -(1→, →2)-Ara f -(1→, →3)-Ara f -(1→ and →4)-Glc p -(1→). BIF-60 exhibited shear-thinning behaviour, low gel stability and weak gelling ability at high concentrations. This work provides a theoretical and experimental basis for molecular structure and properties of the alkali-extracted arabinoxylan from barley kernel, which could guide further functional research and application of barley-derived arabinoxylan.

Structural characteristics of a highly branched and acetylated pectin from Portulaca oleracea L.

Wei Tang,Dan Liu,Yu Li,Ming-Yue Zou,Yu-Chen Shao,Jun-Yi Yin,Shao-Ping Nie

The water-extracted polysaccharide from Portulaca oleracea L. mainly consisted of two pectic polysaccharides designated as POWP-H and POWP-L. The POWP-L was rich in linear homogalacturonan with GalA content of 77.6%, while POWP-H might be a rhamnogalacturonan I (RG-I) pectin composed of Rha (15.7%), Ara (31.7%), Gal (42.5%) and GalA (10.1%). The structure of POWP-H was elucidated by partial degradation, methylation analysis combined with GC-MS method and NMR spectroscopy. It had a weight-average molecular weight of 1.2 × 10 6  g/mol and high degree of branch of 0.57. The presence of abundant arabinogalactan-II and certain galactan side chains in POWP-H was verified by the T-Ara f (17.4%), 1,3,6-Gal p (11.8%), 1,3-Gal p (8.1%), T-Gal p (13.4%) and 1,4-Gal p (14.3%). The evidence for the RG-I backbone of POWP-H was demonstrated by the structure analysis of its degraded product POWP-H-H, which had the backbone of →4)-α-Gal p A-(1 → 2)-α-Rha p -(1 → 4)-α-Gal p A-(1 → 2,4)-α-Rha p -(1→ substituted by short side chain of β-Gal p -(1 → 4)-β-Gal p -(1→ at O -4 position of →2,4)-α-Rha p -(1 → . Moreover, the POWP-H-H was mainly O -acetylated at O -3 position of α-Rha p with 24.8% acetylation degree. In conclusion, POWP-H was mainly the highly branched and acetylated RG-I pectin with relative short RG-I backbone and abundant arabinogalactan II and certain galactan side chains. Besides, it adopted a flexible chain conformation in 0.1 M NaNO 3 solution.

An alternative GC–MS library for methylation analysis of polysaccharides via partially methylated aldononitrile acetates

Ye Li,Jia-Ning Gao,Jun Liang,Xin-Hua Zhu,Hai-Xue Kuang,Yong-Gang Xia

The accurate characterization of glycosyl linkages in complex carbohydrates is always a challenging task due to the occurrence of isomer diversity and the unavailability of known standards. Because of the significant difference between head and tail in structures, this article built a complete methylation analysis library based on partially methylated aldononitrile acetates (PMANs) for facilitated identification of complex glycosidic linkages in polysaccharides. This library covered full EI-MS spectra and detailed fragmentation behaviors of corresponding PMANs ascribed to 46 aldohexoses, 32 aldopentoses and 16 deoxyhexoses. Furthermore, the elute behaviors of these PMANs are summarized on two capillary columns ascribed to complementary polarities. It is quite benefit to elucidate the glycosyl linkages of polysaccharides for inexperienced researchers in this field. The newly established PMAN library has been conformed to be an effective and alternative strategy for glycosyl linkage analysis of polysaccharides.

Structural characterization and immunomodulatory activity analysis of a novel pectic polysaccharide extracted from Tetrastigma hemsleyanum Diels et Gilg and its hydrolysis products

Shasha Tian,Zian Mao,Yuexia Wang,Kewei Li,Yafei Li,Bingqi Zhu,Fangmei Zhou,Jingchao Li,Yingzhi Shen,Zhishan Ding

Tetrastigma hemsleyanum Diels et Gilg ( T. hemsleyanum ) is a rare traditional Chinese medicine with high medicinal value and T. hemsleyanum polysaccharides (THPs) have attracted increasing attention in recent years. However, detailed structural and functional insights into THPs remain limited. Here, a novel pectic polysaccharide (STHP4) with an average molecular weight (Mw) of 119 kDa was extracted and purified from the aboveground part of T. hemsleyanum . Structurally, STHP4 was mainly composed of galacturonic acid, glucuronic acid, and galactose, with minor glucose, rhamnose, arabinose, and mannose, and its primary backbone was 1,4-GalA. Pectinase treatment of STHP4 yields two distinct fractions: STHP4-1 and STHP4-2. STHP4-1 shared the same types of monosaccharides with STHP4, except for the absence of glucose, but had a broad Mw distribution (9.9 to 251 kDa). Another product, STHP4-2, a mixture of oligosaccharides and free monosaccharides, has a Mw of 1.387 kDa. Their structures were further elucidated via methylation analysis and NMR spectra. Furthermore, STHP4-1 exhibited superior immune regulatory effects on macrophages, including increased nitric oxide release and elevated interleukin-6, interleukin-1β, and tumor necrosis factor α expression. These findings suggest that STHP4-1 might be the core active domain of STHP4, and both STHP4 and STHP4-1 are potential immune enhancers.

Preparation and structural characteristics of polysaccharides from loquat peel waste, and their preliminary bioactivities

Dan Liu,Lingju Zhang,Hongge Huang,Changchun Fu,Yunxiao Wei,Zuolong Yu,Chao Han,Wei Tang

BACKGROUND Loquat peel, often as food waste, is a valuable source of bioactive polysaccharides. However, study of such polysaccharides is insufficient, leaving a significant gap in understanding their preparation, structure and bioactivities. RESULTS In this study, three types of loquat peel polysaccharides (LPWP, LPHP and LPNP) were sequentially extracted using hot water, HCl and NaOH solutions, respectively. Among them, LPWP was the purest, with a yield of 3.4% and molecular weight of 470.6 kDa, and it differed from LPHP and LPNP in structure, as evidenced by Fourier transform infrared spectroscopy, X-ray diffraction and scanning electron microscopy, which demonstrated that LPWP consisted of more arabinose (Ara) but less galacturonic acid, rhamnose and galactose, with molar percentages of 71.3%, 23.3%, 3.5% and 1.9%, respectively. Besides, LPWP also exhibited superior antioxidant and antihyperglycemic activities in vitro , particularly in inhibiting α-amylase and α-glucosidase. Methylation and nuclear magnetic resonance analysis confirmed that LPWP was a methyl-esterified pectic polysaccharide rich in branched arabinan, as evidenced by the notable proportion of α-Ara residues, including T-α-Ara f , 1,5-α-Ara f and 1,2,3,5-α-Ara f , with molar percentages of 27.1%, 23.1% and 10.2%, respectively. AFM imaging further revealed its branched-chain morphology and aggregation behavior. CONCLUSION This study highlights the potential of loquat peel polysaccharides as a bioactive ingredient with significant antioxidant and antihyperglycemic properties, particularly LPWP, which was found as a methyl-esterified pectic polysaccharide with abundant-branched arabinan. Our work provides valuable insights into the application of loquat peel polysaccharides in functional foods. © 2024 Society of Chemical Industry.

Structural characteristic of pectin-glucuronoxylan complex from Dolichos lablab L. hull

Dan Liu,Wei Tang,Xiao-Jun Huang,Jie-Lun Hu,Jun-Qiao Wang,Jun-Yi Yin,Shao-Ping Nie,Ming-Yong Xie

Polysaccharides are important constituents in Dolichos lablab hull. Herein, pectin-glucuronoxylan complex from D. lablab hull designated as DLHP-3 ( D. lablab hull polysaccharide,) was prepared by ion exchange and gel permeation chromatography , and further characterized by acid degradation and enzymatic hydrolysis , methylation combined with GC–MS, NMR and MALDI-TOF-MS analysis. Both of pectin and glucuronoxylan regions were found in DLHP-3. The glucuronoxylan region consisted of a →4)-β-Xyl p -(1→ backbone with branches of α-Glc p A-(1→ substituted at O -2 site, and the ratio of xylose to glucuronic acid was about 5:1. Acetyl groups were mainly attached to O -3 site of →2,4)-β-Xyl p -(1→ residues. The main chain of pectin region could be represented by →4)-α-Gal p A-(1→4)-α-Gal p A-(1→ and →2)-α-Rha p -(1→4)-α-Gal p A-(1→ with partial methyl-esterification. The side chains were deduced to embrace arabinan and arabinogalactan linked to rhamnogalacturonan-I region. Pectin was probably covalently bound to glucuronoxylan. Our findings uncovered the molecular structure of pectin-glucuronoxylan complex from D. lablab hull.

Chemical-Oxidation Cleavage Triggered Isothermal Exponential Amplification Reaction for Attomole Gene-Specific Methylation Analysis

Yuzhi Xu,Chang Niu,Xiaofen Xiao,Wenyuan Zhu,Zong Dai,Xiaoyong Zou

Genomic 5-methylcytosine (5-mC) modification is known to extensively regulate gene expression. The sensitive and convenient analysis of gene-specific methylation is wishful but challenging due to the lack of means that can sensitively and sequence-selectively discriminate 5-mC from cytosine without the need for polymerase chain reaction. Here we report a chemical-oxidation cleavage triggered exponential amplification reaction (EXPAR) method named COEXPAR for gene-specific methylation analysis. EXPAR was proved to not only have rapid amplification kinetics under isothermal condition but also show excellent sequence-selectivity and linear-dependence on EXPAR trigger. Further initiation of EXPAR by chemical-cleavage of DNA at 5-mC, the COEXPAR showed high specificity for methylated and nonmethylated DNA, and ∼107 copies of triggers were replicated in 20 min, which were used to quantify the methylation level at the methylation loci. As a result, the gene-specific methylation level of a p53 gene fragment, as a target model, was analyzed in two linear ranges of 10 fM–1 pM and 1 pM–10 nM, and limits of detection of 411 aM (S/N = 3) by fluorescence, and 576 aM (S/N = 3) by electrochemistry. The method fulfilled the assay in an isothermal way in ∼5 h without the need for tedious sample preparation and accurate thermocycling equipment, which is likely to be a facile and ultrasensitive way for gene-specific methylation analysis.

Preparation, structural characterization and biological activities of Laetiporus sulphureus polysaccharide and its stabilized selenium nanoparticles

Qu Yunhe,Zhang Pingping,Zhao Boya,Xu Jing,Shi Dongfang

The total polysaccharides extracted from Laetiporus sulphureus fruiting bodies by ultrasound-assisted complex enzyme method were separated by freeze–thaw combined with fehling reagent to prepare purified polysaccharide (LSPS1). The results of monosaccharide composition and molecular weight distribution demonstrated that LSPS1 contained galactose (51.83%), mannose (26.89%), fucose (16.13%) and glucose (5.15%), and the molecular weight was around 17.3 kDa. Methylation analysis indicated that the backbone of LSPS1 consisted of 1,6-Gal p residues, branched at O-2 of Gal p with t-Man p , t-Fuc p and t-Glc p residues as side chains. In addition, LSPS1 was used for the synthesis of SeNPs based on the redox system of sodium selenite and ascorbic acid. The L. sulphureus polysaccharide selenium nanoparticles (LSPS1–SeNPs) were characterized by UV, FT-IR, ICP-MS, EDX, DLS and SEM, inferring that LSPS1–SeNPs were stabilized with spherical and granular surface structure as well as a mean particle size of 96.72 ± 1.12 nm. Both of LSPS1 and LSPS1–SeNPs possessed strong antioxidant and hypoglycemic activities while the effects of LSPS1–SeNPs were superior to LSPS1. The findings provided valuable structural information for L. sulphureus polysaccharide, and will promote the application of SeNPs by L. sulphureus polysaccharide in potential antioxidant agent and hypoglycemic candidate. Ultrasound-assisted extraction optimization of Ganoderma sinense polysaccharides and comparative structural analysis across six Ganoderma species

Zhou-Wei Wu,Xiao-Cui Liu,Xue-Fang Zhao,Chen-Xi Quan,Yu-Jie Li,Xing-Rong Peng,Ming-Hua Qiu

This study optimized ultrasound-assisted extraction (UAE) of Ganoderma sinense polysaccharides using response surface methodology with a Box-Behnken design. Optimal conditions (47–52 °C, solid–liquid ratio 1:35–1:40, ultrasound time 45–55 min) improved extraction yield at relatively low temperatures compared to conventional hot water extraction (80–100 °C). Comparative analysis of six Ganoderma species ( G. sinense, G. lucidum, G. applanatum, G. resinaceum, G. cochlear, and G. hainanesis ) revealed substantial interspecies variations in yields (3.86 %–12.15 %), total sugar contents (11.24 %–42.88 %), and molecular weights (1.76 kDa–1.44 × 10 5  kDa). Monosaccharide profiles were dominated by glucose and galactose (>70 %), with species-specific distribution. FT-IR spectroscopy revealed species-specific functional group variations, while methylation analysis of G. lucidum polysaccharides identified primarily (1 → 3)- and (1 → 4)-linked β-glucans with complex branching. Ethanol-precipitated fractions showed structural differentiation: GL-40 was rich in (1 → 6)-linked galactans, GL-60 had a balanced linkage profile, and GL-80 was enriched in branched (1 → 3,6)-β-glucans. These findings offer insights into Ganoderma polysaccharide diversity and structure–function relationships, supporting quality control and development of standardized functional foods.
