From waste to value: Multi-omics reveal pomegranate peel addition improves corn silage antioxidant activity and reduces methane and nitrogen losses

Xia Zhang,Huixian Zhang,Dongcai Wang,Yuanqing Zhang

Fermentation technology presents promising opportunities for food waste valorization. Pomegranate peel ( PP ), a food by-product, has potential applications in fermented feed. This study examined the effects of a 6% dry PP additive on the ensiling characteristics, antioxidant activity, metabolites, bacterial community, and in vitro ruminal fermentation, methane (CH 4 ) emission of corn silage ensiled for 7 days and 60 days using microbiome and metabolome analyses. PP-treated silage inhibited ( P  < 0.05) protein degradation by reducing ammonia nitrogen and non-protein nitrogen concentrations during ensiling. The PP additive increased ( P  < 0.05) water-soluble carbohydrate content and reduced ethanol production in corn silage. Lactiplantibacillus dominated PP-treated silage at the initial ensiling stage, while Levilactobacillus prevailed at the final stage . Notably, the PP additive exhibited strong antioxidant activity by modulating antioxidant enzymes and flavonoid biosynthesis mediated by key metabolites (epigallocatechin and catechin). Correlation analysis identified Lactiplantibacillus , Citrobacter , Phytobacter and Burkholderia as key microbes in the production of antioxidant metabolites and enzymes in PP-treated silage. Additionally, PP supplementation reduced ( P < 0.05) in vitro ruminal CH 4 and nitrogen losses, while decreasing dry matter (DM) digestibility in corn silage. In summary, PP-treated corn silage enhanced antioxidant properties and reduced the nitrogen losses and in vitro ruminal CH 4 emissions, but lowered DM digestibility. Thus, PP can be recommended as a silage additive, though the dry PP level should be lower than that used in this study.

Rice root Fe plaque increases paddy soil CH4 emissions via the promotion of electron transfer for syntrophic methanogenesis

Jinzhi Yao,Minghui Xie,Linpeng Yu,Ting Liu,Tim J. Clough,Nicole Wrage-Mönnig,Jiafa Luo,Chunsheng Hu,Tida Ge,Shungui Zhou,Shuping Qin

Iron (Fe) plaque is a concentrated form of microbially available Fe oxide that coats rice plant root surfaces, representing a high density of Fe oxide and which potentially mediates paddy soil CH 4 emissions. Using a combination of methods including Fe plaque induction, isotopic labeling , pure microbial strains, and Fe oxide addition experiments, we investigated the impact of Fe plaque on methane (CH 4 ) emissions from paddy soils and explored the associated mechanisms underlying the influence of Fe plaque on CH 4 emissions. A 13 C–CH 4 isotopic labeling experiment showed that Fe plaque did not significantly affect CH 4 oxidation and associated gene expression, whereas Fe plaque significantly enriched methanogenic archaea and their expression of genes associated with methanogenesis. Pure microbial strain and Fe oxide addition experiments showed that the enhancement of CH 4 production in the presence of Fe plaque was caused by the (semi) conductive minerals within the Fe plaque, specifically, hematite , which promoted the extracellular electron transfer between the methanogenic archaea and their syntrophic bacteria, resulting in enhancement of methanogenesis. Our results imply that the presence of Fe plaque will accelerate CH 4 emissions from paddy soils and suppressing Fe plaque has the potential to mitigate CH 4 emissions.

Effects of fumaric acid supplementation on methane production and rumen fermentation in goats fed diets varying in forage and concentrate particle size

Li Zongjun,Liu Nannan,Cao Yangchun,Jin Chunjia,Li Fei,Cai Chuanjiang,Yao Junhu

Background In rumen fermentation, fumaric acid (FA) could competitively utilize hydrogen with methanogenesis to enhance propionate production and suppress methane emission, but both effects were diet-dependent. This study aimed to explore the effects of FA supplementation on methanogenesis and rumen fermentation in goats fed diets varying in forage and concentrate particle size. Methods Four rumen-cannulated goats were used in a 4 × 4 Latin square design with a 2 × 2 factorial arrangement of treatments: low or high ratio of forage particle size: concentrate particle size (Fps:Cps), without or with FA supplementation (24 g/d). Fps:Cps was higher in the diet with chopped alfalfa hay plus ground corn than in that with ground alfalfa hay plus crushed corn. Results Both increasing dietary Fps:Cps and FA supplementation shifted ruminal volatile fatty acid (VFA) patterns toward more propionate and less acetate in goats. An interaction between dietary Fps:Cps and FA supplementation was observed for the ratio of acetate to propionate (A:P), which was more predominant when FA was supplemented in the low-Fps:Cps diet. Methane production was reduced by FA, and the reduction was larger in the low-Fps:Cps diet (31.72%) than in the high-Fps:Cps diet (17.91%). Fumaric acid decreased ruminal total VFA concentration and increased ruminal pH. No difference was found in ruminal DM degradation of concentrate or alfalfa hay by dietary Fps:Cps or FA. Goats presented a lower ruminal methanogen abundance with FA supplementation and a higher B. fibrisolvens abundance with high dietary Fps:Cps. Conclusions Adjusting dietary Fps:Cps is an alternative dietary model for studying diet-dependent effects without changing dietary chemical composition. Fumaric acid supplementation in the low-Fps:Cps diet showed greater responses in methane mitigation and propionate increase.

Effects of melatonin on rumen microorganisms and methane production in dairy cow: results from in vitro and in vivo studies

Fu Yao,Yao Songyang,Wang Tiankun,Lu Yongqiang,Han Huigang,Liu Xuening,Lv Dongying,Ma Xiao,Guan Shengyu,Yao Yujun,Liu Yunjie,Yu Haiying,Li Shengli,Yang Ning,Liu Guoshi

Background Methane (CH 4 ) is a major greenhouse gas, and ruminants are one of the sources of CH 4 which is produced by the rumen microbiota. Modification of the rumen microbiota compositions will impact the CH 4 production. In this study, the effects of melatonin on methane production in cows were investigated both in the in vitro and in vivo studies. Results Melatonin treatment significantly reduced methane production in both studies. The cows treated with melatonin reduced methane emission from their respiration by approximately 50%. The potential mechanisms are multiple. First, melatonin lowers the volatile fatty acids (VFAs) production in rumen and reduces the raw material for CH 4 synthesis. Second, melatonin not only reduces the abundance of Methanobacterium which are responsible for generating methane but also inhibits the populations of protozoa to break the symbiotic relationship between Methanobacterium and protozoa in rumen to further lowers the CH 4 production. The reduced VFA production is not associated with food intake, and it seems also not to jeopardize the nutritional status of the cows. This was reflected by the increased milk lipid and protein contents in melatonin treated compared to the control cows. It is likely that the energy used to synthesize methane is saved to compensate the reduced VFA production. Conclusion This study enlightens the potential mechanisms by which melatonin reduces rumen methane production in dairy cows. Considering the greenhouse effects of methane on global warming, these findings provide valuable information using different approaches to achieve low carbon dairy farming to reduce the methane emission. Simultaneous mitigation of cadmium contamination and greenhouse gas emissions in paddy soil by iron-modified biochar

Xin Wang,Tong Zou,Jiapan Lian,Yonglong Chen,Liping Cheng,Yasir Hamid,Zhenli He,Paramsothy Jeyakumar,Xiaoe Yang,Hailong Wang

Cadmium (Cd) contamination in agricultural soils is one of the major environmental challenges globally. Biochar is a promising material for mitigating Cd pollution, but it carries the risk of increasing greenhouse gas emissions . Herein, we incorporate iron-based materials into biochar to simultaneously enhance soil nutrients, mitigate heavy metal contamination, and reduce greenhouse gas emissions. The results showed that the iron-modified biochar (FeBC) increased soil available potassium , alkali-hydrolyzable nitrogen and soil organic carbon . All materials promoted the formation of strongly bound Cd (FMO-Cd), with FeBC outperforming standalone iron or biochar by reducing soil Cd bioavailability by 17.0–44.9 %. And the goethite-modified biochar (GBC) further enhanced iron plaque [FeO(OH)] formation, achieving the highest Cd reduction (80.4 %) in rice grains . In addition, except for biochar and zero-valent iron, the other treatments significantly suppressed CH 4 emission and stabilized CO 2 and N 2 O. Among them, GBC treatment reduced the relative abundance of the mcrA gene, a CH 4 emission-related gene, by 22.7 %, ultimately leading to the highest reduction in CH 4 emissions (26.3 %). These findings suggest the potential of FeBC as soil amendments to improve soil nutrients and food safety, while reducing greenhouse gas emissions.

Synergistic Effects of 3-Nitrooxypropanol with Fumarate in the Regulation of Propionate Formation and Methanogenesis in Dairy Cows In Vitro

Zihao Liu,Kun Wang,Xuemei Nan,Meng Cai,Liang Yang,Benhai Xiong,Yiguang Zhao

3-Nitrooxypropanol (3-NOP) is effective at reducing ruminal methane emissions in ruminants. But it also causes a drastic increase in hydrogen accumulation, resulting in feed energy waste. Fumarate is a key precursor for propionate formation and plays an important role in rumen hydrogen metabolism. Therefore, this study examined the effects of 3-NOP combined with fumarate on volatile fatty acids, methanogenesis, and microbial community structures in dairy cows in vitro. The in vitro culture experiment was performed using a 2-by-2 factorial design, two 3-NOP levels (0 or 2 mg/g dry matter [DM]) and two fumarate levels (0 or 100 mg/g DM), including 3 runs with 4 treatments, 4 replicates, and 4 blanks containing only the inoculum. Rumen fluid was collected from three lactating Holstein cows with permanent ruminal fistulas. The combination of 3-NOP and fumarate reduced methane emissions by 11.48% without affecting dry matter degradability. The propionate concentration increased and the acetate/propionate ratio decreased significantly. In terms of bacteria, the combination of 3-NOP and fumarate reduced the abundances of Ruminococcus and Lachnospiraceae_NK3A20_group and increased the abundances of Prevotella and Succiniclasticum. For archaea, the combination of 3-NOP and fumarate significantly increased the abundances of Methanobrevibacter_sp._AbM4, while the abundance of operational taxonomic unit 581 (OTU581) (belonging to an uncultured_rumen_methanogen_g__Methanobrevibacter strain) was significantly decreased. These results indicated that the combination of 3-NOP and fumarate could alleviate the accumulation of hydrogen and enhance the inhibition of methanogenesis compared with 3-NOP only in dairy cows.IMPORTANCE The global problem of climate change and the greenhouse effect has become increasingly severe, and the abatement of greenhouse gases has received great attention from the international community. Methane produced by ruminants during digestion not only aggravates the greenhouse effect but also causes a waste of feed energy. As a methane inhibitor, 3-nitrooxypropanol can effectively reduce methane emissions from ruminants. However, when it inhibits methane emissions, the emission of hydrogen increases sharply, resulting in the waste of feed resources. Fumarate is a propionic acid precursor that can promote the metabolism of hydrogen to propionic acid in animals. Therefore, we studied the effects of the combined addition of 3-nitrooxypropanol and fumarate on methanogenesis, rumen fermentation, and rumen flora. It is of great significance to inhibit methane emission from ruminants and slow down the greenhouse effect.

Integrating anodic membrane diffusion/biodegradation with UV photolysis, Adsorptive oxidation by activation of peroxymonosulfate over activated carbon fiber based photo cathode in one reactor system for removing toluene gas

Qiyuan Chen,Lifen Liu

Emissions of volatile organic compounds (VOCs) air pollutants adversely affected human health and air quality. A novel reactor is designed by a bio-anode chamber with a PVDF/CFC diffusion membrane module and a catalytic air cathode chamber with persulfate and activated carbon fiber (ACF) based photocathode . Peroxysulfate is activated to generate oxidizing species that promote toluene removal and oxidation together with UV photolysis and photo-electrochemical catalysis. This new reactor as air pollution control technique is able to continuously remove toluene gas from an air stream that sequentially flows through firstly the cathode chamber where UV photolysis, photo-electrochemical (PEC) oxidation are integrated with activation of peroxymonosulfate (PMS), then flows through the bio-anode chamber where adsorption/diffusion and biodegradation take place in microbial fuel cell (MFC). This integrated system exhibits high efficiency in toluene removal (∼95 %, ∼100 ppm toluene) with stable power output (cell voltage: 0.40 V) under UV light illumination. In addition, the existence of reactive species such as OH, O 2 − and SO 4 - radicals are confirmed by EPR tests. The results show that both ·OH and SO 4 - are involved in the toluene oxidative degradation with the addition of PMS, where SO 4 - plays a predominant role, indicating that combination of photolysis, cathodic adsorption/oxidation with anodic adsorption/diffusion and biodegradation provides a promising way for VOCs removal and simultaneous electricity generation at room temperature.

Effects of combined addition of 3-nitrooxypropanol and vitamin B12 on methane and propionate production in dairy cows by in vitro-simulated fermentation

Zihao Liu,Kun Wang,Xuemei Nan,Liang Yang,Yue Wang,Fan Zhang,Meng Cai,Yiguang Zhao,Benhai Xiong

ABSTRACT The compound 3-nitrooxypropanol (3-NOP) is a promising methane inhibitor, which performs well in inhibiting methane emission and does not affect animal feed intake and digestibility. However, it causes a significant increase in hydrogen production while suppressing methane emission, resulting in a waste of feed energy. Vitamin B 12 is a key factor in the propionate production pathway and thus plays an important role in regulating the hydrogen utilization pathway. In this study, the effects of 3-NOP combined with vitamin B 12 supplementation on rumen fermentation and microbial compositional structure in dairy cattle were investigated by simulating rumen fermentation in vitro. Experiments were performed using a 2 × 2-factorial design: two 3-NOP levels (0 or 2 mg/g dry matter) and 2 vitamin B 12 levels (0 or 2 mg/g dry matter). Three experiments were performed, each consisting of 4 treatments, 4 replicates, and 4 blanks containing only inoculum. The combined supplementation of 3-NOP and vitamin B 12 reduced methane emission by 12% without affecting dry matter digestibility. The combined addition of 3-NOP and vitamin B 12 significantly increased the concentration of propionate and reduced the concentration of acetate and the acetate to propionate ratio. At the bacterial level, 3-NOP increased the relative abundances of Christensenellaceae_R-7_group and Lachnospiraceae_NK3A20_group . Vitamin B 12 increased the relative abundances of unclassified_f__Prevotellaceae and Prevotellaceae_UCG-003 and decreased the relative abundance of Lachnospiraceae_NK3A20_group . At the archaeal level, the combination of 3-NOP and vitamin B 12 increased the relative abundances of Methanobrevibacter_ sp._ Abm4 , OTU1125, and OTU95 and decreased the relative abundances of uncultured_methanogenic_archaeon_g__Methanobrevibacter , OTU1147, OTU1056, and OTU55. The results indicated that 3-NOP combined with vitamin B 12 could alleviate rumen hydrogen emission and enhance the inhibition of methane emission compared with 3-NOP alone.

Effect of fumarate and live yeast on ruminal fermentation, methane emissions, and blood metabolites in dairy goats

Huixin Dong,Xintong Zhang,Shuaishuai Li,Jingbo Ma,Jun Zhang,Hongyang Chen,Yudith lamothe Crespo,Junhu Yao,Zongjun Li

The objective of the current study was to evaluate the short-term effects of fumarate and active dry yeast (ADY) on ruminal fermentation, methane (CH 4 ) emissions, microbiota (composition and function), apparent total-tract digestibility, production performance, and blood metabolites in dairy goats using a 2 × 2 factorial design. Twenty-eight primiparous dairy goats were blocked and randomly assigned to 1 of 4 treatments: (1) control; (2) fumarate supplied at 34 g/d; (3) ADY supplied at 1.5 g/d; and (4) fumarate and ADY supplied in combination. The treatments lasted for 38 d. The results showed that fumarate supplementation reduced CH 4 production (L/d), yield (L/kg DMI), and intensity (L/kg milk yield) by 11.3%, 15.5%, and 21.9%, respectively, without affecting DMI, milk yield, or apparent nutrient digestibility. However, the CH 4 -depressing effect of fumarate began to decline 3 h postprandially and gradually diminished thereafter. Additionally, fumarate supplementation increased FCM with a higher NE L while reducing the ADG of goats with a lower NE G , resulting in no overall change in the net energy available for production. Fumarate supplementation also increased ruminal pH but reduced total VFA concentrations and microbial α diversity. Although fumarate did not alter the ruminal microbial structure at the genome or gene level, it stimulated the growth of fumarate- and lactate-utilizing bacteria, which may contribute to CH 4 mitigation and raise rumen pH. Supplementation with ADY increased the NE L and the molar proportion of ruminal butyrate and tended to increase CH 4 production and FCM without affecting CH 4 yield or intensity. Most of the blood metabolites and enzymes influenced by fumarate were involved in the tricarboxylic acid (TCA) and the urea cycles. Fumarate reduced the substrate concentrations of the TCA cycle (e.g., glucose, malondialdehyde) and the urea cycle (e.g., ammonia) in blood, but it increased the end products of the urea cycle (BUN, MUN) and enhanced the activity of malate dehydrogenase, a key enzyme in the TCA cycle. These findings indicate that fumarate can reduce enteric CH 4 emissions and enhance the TCA and the urea cycles in host animals.

Soil water content, carbon, and nitrogen determine the abundances of methanogens, methanotrophs, and methane emission in the Zoige alpine wetland

Zhang Wantong,Kang Xiaoming,Kang Enze,Audet Joachim,Davidson Thomas A.,Zhang Xiaodong,Yan Liang,Li Yong,Yan Zhongqing,Zhang Kerou,Wang Jinzhi,Hu Zhengyi

Purpose Alpine wetland ecosystems can contribute large amounts of methane (CH 4 ) to the atmosphere; however, their emissions vary with environmental conditions. Microbial activity is known to drive CH 4 emissions, but how environmental conditions determine microbial activity is still uncertain. Here, we seek to quantify the variability of the CH 4 flux, to detect the effects of CH 4 -related microbes on CH 4 emissions, and to study the dependency of these effects on environmental conditions. Materials and methods We measured the CH 4 flux, environmental conditions, and CH 4 -related microbial communities ( mcrA and pmoA gene abundances for methanogens and methanotrophs, respectively) under three hydrological conditions (submerged, soil–water interface, and emerged) from seven sampling sites in the Zoige alpine wetland, China. Results and discussion The CH 4 flux varied greatly from 0 to 41 mg m −2  h −1 in the Zoige alpine wetland. The methanogenic and methanotrophic abundances both showed positive correlations with CH 4 flux, while CH 4 flux increased linearly with the increase of soil water content (SWC) when SWC was above 60%. CH 4 flux and methanogenic and methanotrophic abundances maintained the high levels when soil C:N ratio was in the range of 11–24 and decreased exponentially with the increase of soil DOC:TN ratio in Zoige alpine wetland, which might result from its influence on nutrient supply for microbial decomposition process. Conclusion The results provided new insight into the effects of CH 4 -related microbes on CH 4 emission and its response to different environmental conditions and helped us to comprehend the risks of high CH 4 emissions from alpine wetlands under climatic change and anthropogenic disturbance.

Electron shuttles enhance anaerobic oxidation of methane coupled to iron(III) reduction

Qiuxiang He,Linpeng Yu,Jibing Li,Dan He,Xixi Cai,Shungui Zhou

Anaerobic oxidation of methane (AOM) has recently been coupled with the reduction of insoluble electron acceptors such as iron minerals. However, effects of electron shuttles (ESs) on this process and the underlying coupling mechanisms remain not well understood. Here, we evaluated AOM-coupled ferrihydrite reduction by a mixed culture in the absence and presence of ESs. The results showed that ESs (AQS, flavin, HA and AQDS) significantly enhanced the rate (up to 7.4 times) of AOM-dependent ferrihydrite reduction compared with the control. The enhancements were linearly related with the electron transfer capacity of ESs. Illumina high-throughput sequencing and DNA-based stable isotope probing revealed that the AOM-coupled iron reduction depended on the syntrophic interaction of Methanobacterium and the partner bacteria. Methanobacterium as the dominant microorganism , did not assimilate methane into its biomasses. However, it played a crucial role in the partial oxidation of methane into an intermediate (i.e. propionate), which was then assimilated by the partner bacteria (e.g. Cellulomonas , Desulfovibrio , Actinotalea , etc.) for ferrihydrite reduction. This work suggests that ESs in natural environments can mitigate the methane emissions by facilitating the AOM process and biogeochemical cycles of iron.
