Optimization of hydrogel containing toluidine blue O for photodynamic therapy in treating acne

Zheng Yixin,Yu Endian,Weng Qi,Zhou Leilei,Li Qingyong

Antibiotics and photodynamic therapy (PDT) are widely employed in curing acne. However, antibiotics as an effective treatment would lead to bacterial resistance and severe side effects. In this study, we aimed to develop a novel TBO hydrogel, which could prolong the retention time of photosensitizer (TBO) at the lesion site and improve therapeutic effect. In vitro antibacterial experiments (against Staphylococcus aureus and Escherichia coli ), the response surface methodology was used to optimize the formulation of TBO hydrogel. The results indicated that the optimal formulation was 0.5% ( v / v ) carbomer, 0.01 mg/mL TBO, 0.5% ( v / v ) ethanol concentration, 0.5% ( v / v ) Tween 80, the mass ratio of NaOH to carbomer of 0.4 ( w / w ). The TBO hydrogel formulation showed the strong antibacterial activity for Propionibacterium acnes . The stability, pH, and antibacterial activity of TBO hydrogel did not significantly change under 4 °C, 25 °C, and 40 °C during 6-week storage. Furthermore, TBO combined with carbomer hydrogel showed the 51.28% (4 h) and 69.80% (24 h) release. In summary, the hydrogel TBO might be a vital therapeutic strategy to promote the PDT applied in the topical therapy of acne. A TBO hydrogel for photodynamic therapy in the treatment of acne

Sophorolipid-toluidine blue conjugates for improved antibacterial photodynamic therapy through high accumulation

Xiaoxiao Gu,Lixian Xu,Haoyang Yuan,Cailing Li,Juan Zhao,Shuang Li,Dinghua Yu

Anti-bacterial photodynamic therapy is the most promising treatment protocol for bacterial infection, but low accumulation of photosensitizers has seriously hindered their development in clinical application. Here, with inherent outstanding affinity to bacterial cell envelope, sophorolipid produced from Candida bombicola has been conjugated to toluidine blue (SL-TB) through amidation reaction. The structure of SL-TB conjugates was identified by 1H-NMR, FT-IR and ESI-HRMS. The interfacial assembly and photophysical properties of SL-TB conjugates have been disclosed through surface tension, micro-polarity, electronic and fluorescence spectra. After light irradiation, the log10 (reduced CFU) of free toluidine blue to P. aeruginosa and S. aureus were 4.5 and 7.9, respectively. In contrast, SL-TB conjugates showed a higher bactericidal activity, with a reduction of 6.3 and 9.7 log10 units of CFU against P. aeruginosa and S. aureus, respectively. The fluorescence quantitative results showed that SL-TB could accumulate 2850 nmol/1011 cells and 4360 nmol/1011 cells by P. aeruginosa and S. aureus, which was much higher than the accumulation of 462 nmol/1011 cells and 827 nmol/1011 cells of free toluidine blue. Through the cooperation of triple factors, including sophorose affinity to bacterial cells, hydrophobic association with plasma membrane, and electrostatic attraction, higher SL-TB accumulation was acquired, which has enhanced antibacterial photodynamic efficiencies.

Ratiometric electrochemical immunosensor for the detection of procalcitonin based on the ratios of SiO2-Fc–COOH–Au and UiO-66-TB complexes

Juncong Miao,Kang Du,Xuan Li,Xiaoting Xu,Xue Dong,Jinglong Fang,Wei Cao,Qin Wei

Procalcitonin (PCT) as a disease marker is of great significance in the early diagnosis of septicemia and pyemia. Biosensor have good prospects for analysis and detection of disease markers, but developing highly sensitive detection methods for detecting PCT remains a daunting task. In this paper, we develop a ratiometric electrochemical immunosensor with Au NPs modified SiO 2 -Fc-COOH complex as matrix and UiO-66 loaded with Toluidine blue (TB) as a marker for quantitative detection of procalcitonin (PCT). The SiO 2 modified by APTES not only has a large specific surface area but also contains abundant amino groups, which can be connected to the electrochemical probe of ferrocenecarboxylic acid (Fc-COOH). UiO-66 has the advantages of large specific surface area, high porosity and adjustable structure, which can adsorb toluidine blue electrochemical probe through electrostatic attraction . Measurement and analysis of the prepared immunosensor by Differential Pulse Voltammetry (DPV) show that the oxidation peak currents of Fc-COOH and TB appear at potentials of 0.30 V and −0.30 V respectively. As the PCT concentration increases, the oxidation peak current of TB increases and the oxidation peak current of Fc-COOH decreases. The ratios ( ΔI = ΔI TB /ΔI Fc-COOH ) between the double signals could show a certain linear relationship with the concentration of the PCT within a certain range. Under optimal conditions, the linear range of detection obtained by the immunosensor was 1 pg/mL-100 ng/mL and the detection limit was 0.3 pg/mL. In this work, the developed ratiometric electrochemical immunosensor not only provides a simple, reliable and sensitive strategy for quantitative detection of PCT but also provides a useful method for clinical detection of other disease markers.

A resonance Rayleigh scattering and colorimetric dual-channel sensor for sensitive detection of perfluorooctane sulfonate based on toluidine blue

He Jincan,Qiu Peipei,Song Jiayi,Zhang Shiyun,Bai Yan

This work described the use of a basic phenothiazine dye (toluidine blue, TB) as a resonance Rayleigh scattering (RRS) and colorimetric probe for the detection of perfluorooctane sulfonate (PFOS). Owing to the electrostatic interactions between TB and PFOS, TB in the presence of PFOS caused great enhancement of RRS signal at dual-wavelength ( I 345 nm and I 506 nm ) and the ratio changes of absorbance ( A 502 nm / A 630 nm ). The RRS enhancement was attributed to the absorption-rescattering resonance effect, the increase of the molecular size, and the enhancement of hydrophobicity. The analytical procedure was implemented by physically mixing TB, Britton-Robinson buffer solution, and PFOS solution (or sample solution) all-in-one, avoiding the tedious pre-derivatization or the preparation of nanoparticles. The whole approach was less than 8 min. Under the optimal conditions, the analytical performance was acquired. The linear ranges for RRS and colorimetry were 0.04–8.0 and 1.0–20 μmol/L, with detection limits of 4.2 nmol/L and 112 nmol/L, respectively. The RRS method was applied to the determination of PFOS in environmental water with recoveries of 93.2–106%. The dual-channel sensor is convenient, rapidly responsive, sensitive, and cost-effective, integrating the advantages of RRS and colorimetry. Graphical abstract

Luyao Zhang,Weishan Tang,Lu Yang,Zeyi Wang,Yu Jiang,Xingyue Wang,Rong Su,Feng Xiao,Lichuan Chen,Ping He,Yali Zeng,Yun Zhou,Ying Wan,Bin Tang

In this work, a series of multiwalled carbon nanotubes/polytoluidine blue (MWCNTs/PTB) with network structure are synthesized by in-situ oxidative polymerization. Electrochemical performance of MWCNTs/PTB are optimized through precise control of mass ratio between two components. Field emission scanning electron microscope analysis reveals a more homogeneous dispersion of network structure under the mass ratio of 1:3. Prepared MWCNTs/PTB-3 electrode exhibits the highest specific capacity of 232.7C g −1 and satisfactory specific capacity retention of 91.9 % after 5000 charge/discharge cycles in 1.0 M H 2 SO 4 electrolyte. Additionally, the assembled asymmetric supercapacitor exhibits energy density of 13.4 Wh kg −1 at power density of 600 W kg −1 and maintains exceptional cyclic stability. In conclusion, constructing carbon nanotube-supported conductive polymer composite proves to be an effective approach for enhancing the energy storage performance.

Multifunctional Nanosystem for Synergistic Tumor Therapy Delivered by Two-Dimensional MoS2

Meng-Yun Peng,Di-Wei Zheng,Shi-Bo Wang,Si-Xue Cheng,Xian-Zheng Zhang

A multifunctional nanosystem based on two-dimensional molybdenum disulfide (MoS2) was developed for synergistic tumor therapy. MoS2 was stabilized with lipoic acid (LA)-modified poly(ethylene glycol) and modified with a pH-responsive charge-convertible peptide (LA-K11(DMA)). Then, a positively charged photosensitizer, toluidine blue O (TBO), was loaded on MoS2 via physical absorption. The negatively charged LA-K11(DMA) peptide was converted into a positively charged one under acidic conditions. Charge conversion of the peptide could reduce the binding force between positively charged TBO and MoS2, leading to TBO release. Furthermore, the positively charged nanosystem was easily endocytosed by cells. Photo-induced hyperthermia of MoS2 in the tumor areas could promote TBO release and exhibited photothermal therapy. In vitro and in vivo results demonstrated that fluorescence and photo-induced reactive oxygen species (ROS) generation of TBO were severely decreased by MoS2 under normal conditions. While in the acidic condition, the pH-responsive nanosystem exhibited a highly specific and efficient antitumor effect with TBO release and photo-induced ROS generation, suggesting to be a promising accessory for synergistic tumor therapy.

Light-Activated Antimicrobial Polymers with Citronellol-Enhanced Bacterial Accumulation for On-Demand Disinfection

Jiahao Ling,Chu-Ming Pang,Yingxin Tan,Liangzhi Hong

Antibacterial photodynamic therapy offers a promising approach to combating both susceptible and multidrug-resistant pathogens. However, conventional photosensitizers have limitations in terms of poor binding specificity and weak penetration for pathogens. In this study, we developed synergistic photobactericidal polymers that integrate hydrophilic toluidine blue O (TBO) with the lipophilic penetration enhancer citronellol (CT). The CT-containing monomer was copolymerized with glycidyl methacrylate, and then TBO was introduced via the epoxy group's ring-opening reaction. By combining the cationic charge and singlet oxygen (1O2) generation abilities of TBO with citronellol's membrane disrupting ability, the resulting copolymers have enhanced antimicrobial efficacy against Gram-positive, Gram-negative, and multidrug-resistant bacteria. CT not only enables the polymer to self-assemble and enhances the interaction between the polymer and bacteria, but also promotes the accumulation of TBO within cells by improving membrane permeability. 1O2 generated in specific subcellular regions maximize photodynamic therapy's therapeutic potential. Furthermore, these copolymers can be used to develop light-activated antimicrobial polydimethylsiloxane that prevents bacterial adhesion and enables on-demand disinfection.

A novel method for detection of ochratoxin A in foods—Co-MOFs based dual signal ratiometric electrochemical aptamer sensor coupled with DNA walker

Yan Guan,Peng-Bin Si,Tong Yang,Yuan Wu,Yun-Hui Yang,Rong Hu

Ochratoxin A (OTA) is a naturally occurring mycotoxin that poses serious threats, such as kidney damage, to human health. Therefore, we developed a DNA walker-based dual-signal electrochemical ratiometric platform for OTA detection, which could overlook the variations in environmental and instrumental factors and DNA load densities. Cobalt metal–organic frameworks (Co-MOFs) and toluidine blue were used as the electrochemical signal tag and internal reference probe, respectively. In the presence of OTA, this developed machine resulted in the DNA labelled-Co-MOFs far away from the electrode. Thus, Co-MOFs signal at −1.18 V decreased, while toluidine blue at −0.28 V increased. This proposed strategy has displayed superior sensitivity (limit of detection = 0.31 fg/mL, linear range = 1–50 ng/mL) and high reproducibility. The sensor was also applied for determining OTA content in red wine samples and the results were comparable to those of commercial enzyme-linked immunoassay kits with satisfactory results.

A multifunctional DNA tetrahedron for imaging, gene therapy, and chemotherapy-phototherapy combination: Binding affinity and anticancer activity

Jie Liu,Xinyu Li,Yushu Wu,Xinpeng Zhang,Xiangtai Wang,Lixia Yuan,Yanna Zhao,Min Liu

Imaging, silencing cancer-related microRNA, and chemotherapy-phototherapy (CTPT) combination therapy are crucial for cancer diagnosis and drug resistance overcoming. In this study, we designed a multifunctional DNA tetrahedron (MB-MUC1-TD) for the targeted delivery of combined daunorubicin (DAU) + toluidine blue O (TBO). The detection limit of miRNA-21 was determined to be 0.91 nM. The intercalation of DAU and TBO into MB-MUC1-TD was proved by spectroscopic and calorimetric methods. The thermodynamic parameters for the interactions of DAU and/or TBO with MB-MUC1-TD confirmed high drug loading. The first addition of TBO in the ternary system achieved a higher loading of both drugs and a more stable complex structure. Deoxyribonuclease I (DNase I) accelerated the release of DAU and/or TBO loaded in MB-MUC1-TD. Confocal laser scanning microscope demonstrated that MB-MUC1-TD exhibited good imaging ability for miRNA-21 to accurately identify cancer cells, and DAU/TBO was predominantly distributed within the nucleus of cancer cells. In vitro cytotoxicity showed better gene therapy efficacy of MB on MCF-7 cells, better biocompatibility of loaded DAU and TBO on LO2 cells, and stronger synergistic cytotoxicity of DAU + TBO on MCF-7/ADR cells. This study may establish a theoretical foundation for co-loading CTPT combination drugs based on multifunctional DNA nanostructures.

Enhanced amperometric response of a glucose oxidase and horseradish peroxidase based bienzyme glucose biosensor modified with a film of polymerized toluidine blue containing reduced graphene oxide

Wang Fang,Gong Wencheng,Wang Lili,Chen Zilin

Reduced graphene oxide (RGO) was used to construct a bienzyme biosensor containing horseradish peroxidase (HRP) and glucose oxidase (GOx). A poly(toluidine blue) (pTB) film containing RGO acted as both enzyme immobilization matrix and electron transfer mediator. The bienzyme biosensor was characterized by electrochemical techniques and displays a highly sensitive amperometric response to glucose and hydrogen peroxide (H 2 O 2 ) at a potential as low as −0.1 V (vs. SCE). It is shown that use of RGO causes a strong enhancement on the amperometric responses. H 2 O 2 formed by the action of GOx in the presence of oxygen can be further reduced by HRP in the pTB film contacting the RGO modified electrode. In the absence of oxygen, glucose oxidation proceeds by another mechanism in which electron transfer occurs from GOx to the electrode and with pTB acting as the mediator. Amperometric responses to glucose and H 2 O 2 follow Michaelis-Menten kinetics. The experimental conditions were optimized, and under these conditions glucose can be determined in the 80 μM to 3.0 mM range with a detection limit of 50 μM. H 2 O 2 , in turn, can be quantified in up to 30.0 μM concentration with a detection limit of 0.2 μM. The bienzyme biosensor is reproducible, repeatable and stable. Finally, it has been successfully applied to the determination of glucose in plasma samples. Schematic representation of glocuse detection at GCE/RGO/pTB-HRP-GOx.

A Raman immunosensor based on SERS and microfluidic chip for all-fiber detection of brain natriuretic peptide

Dongqiao Zheng,Zhiqian Wang,Jingjing Wu,Shiyu Li,Wei Li,Houjing Zhang,Li Xia

Early diagnosis of chronic heart failure (CHF) relies on the ultra-sensitive detection assay of its main biomarker, Brain natriuretic peptide (BNP). To meet such demand, we designed a near-infrared immunoassay biosensor, utilizing a fiber-embedded microfluidic chip for surface-enhanced Raman scattering (SERS) detection. A SERS probe, consisting of metal–organic framework and Au-HEPES coupling nanoparticles (AuHPs), was used to enhance the Raman signal of the toluidine blue (TB) dye. The probe anchored with TB is used as a SERS tag, which is linked to BNP through sandwich method. To enhance the Raman signal further, we embedded a SERS substrate at the bottom of the microfluidic channel, which serves an enhancer when the SERS tagged BNP passes through the microfluidic chip. The amplified Raman signal was collected by an optical fiber probe of a simple and portable Raman detection system with an all-fiber connected configuration. Such design resulted in the lowest detection limit of pg/mL, indicating that the proposed platform can be used for rapid and highly sensitive detection of BNP. In addition, our platform also has the advantages of low sample consumption and high throughput rate, providing an effective scheme for the early diagnosis of CHF.

Based on PI3K/Akt Signaling Pathway, the Effects of Gubi Decoction on Chondrocyte Proliferation, Apoptosis, and Expression of Inflammatory Factors in a Rat Model of Osteoarthritis.

Guo Z,Qin Y,Li Y,Tang P

Objective Given the crucial role of the PI3K/Akt signaling pathway in chondrocyte survival and inflammation in osteoarthritis (OA), this study aimed to investigate the effects of Gu Bi Tang on chondrocyte proliferation, apoptosis, and inflammatory factor expression in a rat model of OA, with a focus on the PI3K/Akt signaling pathway. Methods Forty-five specific pathogen-free (SPF) C57 mice were randomly divided into three groups: the model group (Group A), the Gu Bi Tang group (Group B), and the Amfenac group (Group C), with 15 rats in each group. All 45 rats underwent anterior cruciate ligament transection (ACLT) surgery to establish an OA model. The ACLT procedure is a well-established method for inducing OA in rodents, as it leads to the destabilization of the knee joint and the development of degenerative changes characteristic of human OA. After 8 weeks of modeling, Group A rats received an equivalent volume of normal saline by gastric lavage, Group B rats were administered 13 mL/kg of Gu Bi Tang, and Group C rats received 19.5 mg/kg of Amfenac solution by gastric lavage. The dosages of Gu Bi Tang and Amfenac were selected based on previous studies examining the therapeutic effects of these interventions in rodent OA models. The gastric lavage frequency for all three groups was maintained at twice daily. The researchers analyzed cartilage morphological changes using toluidine blue staining, chondrocyte proliferation and apoptosis using the TUNEL method, and the expression of PI3K/AKT/mTOR proteins in chondrocytes using Western blotting. Additionally, the expression of inflammatory factors (TNF-α and IL-1β) in serum was measured using ELISA. Results Staining: Compared to the model group (Group A), both the Gu Bi Tang group (Group B) and the Amfenac group (Group C) showed significant improvement in cartilage tissue, with deeper toluidine blue staining. Toluidine blue staining is a marker of cartilage integrity and glycosaminoglycan content, indicating improved cartilage structure and composition in the treatment groups. Chondrocyte proliferation and apoptosis: Compared to the model group (Group A), both the Gu Bi Tang group (Group B) and the Amfenac group (Group C) significantly reduced chondrocyte apoptosis (P < .05). This reduction in chondrocyte death contributes to a healthier cartilage environment and helps prevent further cartilage degradation. Protein expression: In comparison to the model group (Group A), the expression of PI3K, AKT, and mTOR proteins in the joint cartilage of the Gu Bi Tang group (Group B) and the Amfenac group (Group C) significantly decreased, with the Amfenac group showing a greater reduction than the Gu Bi Tang group (P < .05). The inhibition of this detrimental PI3K/AKT/mTOR signaling pathway promotes chondrocyte survival and a more favorable cartilage homeostasis. Inflammatory factor expression: Prior to treatment, there was no significant difference in the expression levels of the inflammatory factors TNF-α and IL-1β in serum among the three groups (P > .05). However, after treatment, both the Gu Bi Tang group (Group B) and the Amfenac group (Group C) showed a significant reduction in the serum expression of TNF-α and IL-1β compared to the model group (Group A), with the Amfenac group showing a greater reduction than the Gu Bi Tang group (P < .05). This is important, as TNF-α and IL-1β are key pro-inflammatory cytokines that drive the destructive processes in osteoarthritis. Conclusion In summary, this study demonstrates that Gu Bi Tang exerts protective effects on chondrocytes in a rat model of osteoarthritis. Specifically, Gu Bi Tang was shown to inhibit chondrocyte apoptosis, reduce the expression of key proteins in the PI3K/AKT/mTOR signaling pathway, and decrease the levels of the pro-inflammatory cytokines TNF-α and IL-1β. These findings suggest that Gu Bi Tang could offer a novel therapeutic approach for osteoarthritis by modulating key signaling pathways and inflammatory responses. The ability of Gu Bi Tang to preserve chondrocyte viability and maintain a more favorable cartilage homeostasis makes it a promising candidate for further investigation as a potential treatment for osteoarthritis. Future studies should explore the precise mechanisms by which Gu Bi Tang exerts its beneficial effects and evaluate its efficacy in additional animal models and clinical settings.
