Optimization of hydrogel containing toluidine blue O for photodynamic therapy in treating acne

Zheng Yixin,Yu Endian,Weng Qi,Zhou Leilei,Li Qingyong

Antibiotics and photodynamic therapy (PDT) are widely employed in curing acne. However, antibiotics as an effective treatment would lead to bacterial resistance and severe side effects. In this study, we aimed to develop a novel TBO hydrogel, which could prolong the retention time of photosensitizer (TBO) at the lesion site and improve therapeutic effect. In vitro antibacterial experiments (against Staphylococcus aureus and Escherichia coli ), the response surface methodology was used to optimize the formulation of TBO hydrogel. The results indicated that the optimal formulation was 0.5% ( v / v ) carbomer, 0.01 mg/mL TBO, 0.5% ( v / v ) ethanol concentration, 0.5% ( v / v ) Tween 80, the mass ratio of NaOH to carbomer of 0.4 ( w / w ). The TBO hydrogel formulation showed the strong antibacterial activity for Propionibacterium acnes . The stability, pH, and antibacterial activity of TBO hydrogel did not significantly change under 4 °C, 25 °C, and 40 °C during 6-week storage. Furthermore, TBO combined with carbomer hydrogel showed the 51.28% (4 h) and 69.80% (24 h) release. In summary, the hydrogel TBO might be a vital therapeutic strategy to promote the PDT applied in the topical therapy of acne. A TBO hydrogel for photodynamic therapy in the treatment of acne

Light-Activated Antimicrobial Polymers with Citronellol-Enhanced Bacterial Accumulation for On-Demand Disinfection

Jiahao Ling,Chu-Ming Pang,Yingxin Tan,Liangzhi Hong

Antibacterial photodynamic therapy offers a promising approach to combating both susceptible and multidrug-resistant pathogens. However, conventional photosensitizers have limitations in terms of poor binding specificity and weak penetration for pathogens. In this study, we developed synergistic photobactericidal polymers that integrate hydrophilic toluidine blue O (TBO) with the lipophilic penetration enhancer citronellol (CT). The CT-containing monomer was copolymerized with glycidyl methacrylate, and then TBO was introduced via the epoxy group's ring-opening reaction. By combining the cationic charge and singlet oxygen (1O2) generation abilities of TBO with citronellol's membrane disrupting ability, the resulting copolymers have enhanced antimicrobial efficacy against Gram-positive, Gram-negative, and multidrug-resistant bacteria. CT not only enables the polymer to self-assemble and enhances the interaction between the polymer and bacteria, but also promotes the accumulation of TBO within cells by improving membrane permeability. 1O2 generated in specific subcellular regions maximize photodynamic therapy's therapeutic potential. Furthermore, these copolymers can be used to develop light-activated antimicrobial polydimethylsiloxane that prevents bacterial adhesion and enables on-demand disinfection.

A multifunctional DNA tetrahedron for imaging, gene therapy, and chemotherapy-phototherapy combination: Binding affinity and anticancer activity

Jie Liu,Xinyu Li,Yushu Wu,Xinpeng Zhang,Xiangtai Wang,Lixia Yuan,Yanna Zhao,Min Liu

Imaging, silencing cancer-related microRNA, and chemotherapy-phototherapy (CTPT) combination therapy are crucial for cancer diagnosis and drug resistance overcoming. In this study, we designed a multifunctional DNA tetrahedron (MB-MUC1-TD) for the targeted delivery of combined daunorubicin (DAU) + toluidine blue O (TBO). The detection limit of miRNA-21 was determined to be 0.91 nM. The intercalation of DAU and TBO into MB-MUC1-TD was proved by spectroscopic and calorimetric methods. The thermodynamic parameters for the interactions of DAU and/or TBO with MB-MUC1-TD confirmed high drug loading. The first addition of TBO in the ternary system achieved a higher loading of both drugs and a more stable complex structure. Deoxyribonuclease I (DNase I) accelerated the release of DAU and/or TBO loaded in MB-MUC1-TD. Confocal laser scanning microscope demonstrated that MB-MUC1-TD exhibited good imaging ability for miRNA-21 to accurately identify cancer cells, and DAU/TBO was predominantly distributed within the nucleus of cancer cells. In vitro cytotoxicity showed better gene therapy efficacy of MB on MCF-7 cells, better biocompatibility of loaded DAU and TBO on LO2 cells, and stronger synergistic cytotoxicity of DAU + TBO on MCF-7/ADR cells. This study may establish a theoretical foundation for co-loading CTPT combination drugs based on multifunctional DNA nanostructures.

An electrochemical biosensor based on DNAzyme and TB-loading mesoporous silica nanoparticles for detection of TdT in serum

Minqiao Feng,Chengyan Yin,Hailiang Qin,Wei Liu,Yunqi Wang,Ruth Antwi-Baah,Yao Rao,Shuxia Xu

Terminal deoxynucleotidyl transferase (TdT) is actively expressed in leukemia patients and plays an important role in maintaining the genetic diversity of an organism. Therefore, developing simple and sensitive analytical methods for the accurate detection of TdT in serum is of significant importance. Herein, an electrochemical biosensor based on a toluidine blue (TB)-loading mesoporous silica nanoparticles (MSN) was established for the detection of TdT in serum. First, MSN with amino group was synthesized and subsequently was carbonylated to obtain carboxyl-functionalized MSN (MSN-COOH), which was used to load TB. Thereafter, MSN-COOH was sealed with the substrate strand S(rA)-NH 2 of the Mg 2+ -dependent DNAzyme to form S(rA)-NH 2 /TB/MSN-COOH. In the presence of TdT, it can add deoxyadenosine triphosphate (dATP) to the 3’-OH end of Enc-5 A, activating its cleavage activity on the substrate chain S(rA)-NH 2 , thus releasing TB encapsulated in MSN-COOH. By measuring the change in the electrochemical signal of TB, the TdT concentration can be determined. The linear range is from 5 to 400 U/mL and the limit of detection (LOD) is 0.369 U/mL (3 σ/K). Since DNAzyme can be extended by TdT without the need of a template, the proposed biosensor demonstrates excellent selectivity towards TdT in the presence of other DNAzyme, such as T4 DNA ligase, phi29 DNA polymerase, DreamTaq DNA polymerase, and Klenow fragment ( exo -). Furthermore, the proposed biosensor was successfully applied to the detection of TdT in serum with the recovery rates ranged from 91.0 % to 94.7 %, indicating its potential for early detection of leukemia and other related diseases.

Preparation and characterisation of novel quaternized chitosan-based hollow salt and its sodium-reducing effect in air-dried ducks

Zihang Shi,Yangyang Hu,Changyu Zhou,Qiang Xia,Yangying Sun,Hongbing Yan,Kaiyong Yao,Chen Chen,Weitao Zhao,Daodong Pan

In this study, hollow salt was fabricated using quaternized chitosan (Qac), with the effects of varying ultrasonic pretreatment conditions (100 W–500 W, 40 Hz, 60 °C, 30 min) on its physicochemical properties, saltiness-enhancing mechanisms, and salt-reduction efficacy in air-dried duck processing systematically investigated. The findings revealed that ultrasonic pretreatment increased the degree of cross-linking and Na + content in the hollow salts. It also altered the binding interactions, thereby influencing the thermal stability of the hollow salts. The ultrasonic pretreatment reduced the size of the hollow salt and increased its flowability and hygroscopicity, which promoted its rapid dissolution to release Na + and increased the retention time of Na + on the tongue, and ultimately achieved salt reduction. Using hollow salt in air-dried duck curing reduces the usage of NaCl by 22–26.75 % without affecting product quality. This research provides a theoretical foundation for the development of novel salt-reduction strategies in food processing.

The improvement of flocculation performance of organic polymer flocculants via regulation of their branched structure and block structure in removal of soluble dyes from wastewater

Yujie Wang,Jingxin Li,Honghua Wang,Shixin Zhang,Jie Zhang,Hailin Yang,Xiaomin Tang,Huaili Zheng

The efficient treatment of dyeing wastewater, a typical industrial wastewater, has aroused widespread attention. Flocculation was a vital pretreatment in dyeing wastewater treatment as it removed colloidal particles, reduced organic loading and improved the treatment efficiency of subsequent processes. However, the soluble dyes were rarely removed by traditional flocculation. In this study, the removal efficiencies of soluble dyes by flocculation were enhanced via synthesizing organic polymer flocculants with branched structure and block structure (PADS). The block length on PADS was regulated by the molecular weight of the template. PADS owned the high molecular weight, rich functional groups and outstanding flocculation performance. The largest removal rates of soluble Congo red, chrome black T and toluidine blue, 90.16 %, 88.41 % and 80.19 %, were achieved. The flocculation mechanisms for dyeing wastewater treatment were emphasized via Extended Derjaguin-Landau-Verwey-Overbeek (XDLVO) calculations and molecular dynamics simulations. Charge neutralization and the interaction between PADS and dyes mainly contributed to dye removal. The block structure of PADS strengthened these two processes. The branched structure of PADS facilitated bridging and promoted the formation of large and compact flocs, improving solid-liquid separation of dyes. PADS were potential candidates for improving dye removal in the pretreatment of dyeing wastewater.

Engineering solvent-controlled multi-color emission carbon dots in single carbon source and their application in multi-channel Ag+ detection

Yanan Yan,Youhong Tang,Dan Chang,Shengmei Song,Ayesha Aziz,Muhammad Asif,Shaomin Shuang,Chuan Dong

Multi-color carbon dots (CDs), as advanced carbon nanomaterials with exceptional features, have gained significant attention, yet the facile and rapid synthesis of CDs with tunable multi-color emission from a single carbon source remains a challenge. By simply varying the solvent (water, EtOH, or DMF), blue-, green-, and yellow-emitting CDs are obtained, with corresponding tunable emission peaks of 469, 485, and 528 nm and quantum yields of 31.2 %, 34.1 % and 35.0 %. This emission tunability attributed to variations in their carbon core and surface states. Furthermore, multi-color CDs are employed as fluorescent probes to assess their detection capabilities. The multi-color CDs enabled highly sensitive Ag + detection, achieving detection limits of 4 pM, 34 pM, and 42 pM, respectively. This strategy enabled a multichannel smartphone platform for on-site analysis, with the CDs also proving effective for bioimaging in HeLa cells and zebrafish, collectively establishing their dual potential for environmental and biomedical applications.

Conjugated cationic dye modulates the ion-confined partitioning of anions/cations in graphene membrane to enhance seawater desalination

Ruixue Meng,Yuan Li,Jiawei Peng,Huanhuan Guo,Yanhui Li,Long Chen

Obtaining low-salinity water from seawater via nanofiltration technology has emerged as a crucial solution to solve the freshwater scarcity. Graphene nanofiltration membrane exhibits revolutionary potential but faces serious challenges, primarily insufficient structural stability and poor rejection for small salt ions. Herein, reduced graphene oxide (rGO)/Dye composite membranes with positively charged surfaces by intercalating π-conjugated cationic dye molecules into rGO membranes through π-π interaction and electrostatic interactions were successfully fabricated. The introduction of dye molecules not only regulates the interlayer spacing but also achieves electrostatic-induced ion-confined partitioning, effectively inhibiting the co-transport of cation-anion pairs to enhance the salt rejection performances. The optimized membrane demonstrated remarkable rejection of 85.3 % for NaCl and 91.4 % for Na 2 SO 4 with high permeance due to the interlayer spacing modulation and ion-confined partitioning in nanochannels. Notably, those membranes exhibited exceptional chemical and mechanical stability during long-term nanofiltration operation involving high-salinity solution, mixed feed solution, and actual seawater. Transmembrane mass transfer experiments further confirmed that rGO/Dye composite membranes effectively suppress cation-anion pair co-transport, thereby increasing mass transfer resistance for salt ions. This study presents a novel strategy for designing high-performance desalination nanofiltration membranes through the synergistic regulation of nanochannels and surface charge characteristics.

Eco-friendly and cost-effective modification strategy for high-performance polyamide membranes: Synergistic regulation of interfacial tension reduction and diffusion inhibition

Yuxuan Chen,Xiaoming Xu,Zhiwei Wang,Sunxinyi Wang,Guoyan Hua,Fuqiang Liu

The eco-friendly and cost-effective regulation of interface polymerization (IP) reaction is an effective strategy to enhance the energy efficiency of nanofiltration (NF). In this work, a novel polyamide (PA) NF membrane was prepared by introducing glycerol-doped sugarcane cellulose nanofibers as a biomass composite interlayer. The optimized interlayer could modulate the water-organic interfacial tension and regulate piperazine (PIP) diffusion, thereby enabling a more controlled IP reaction. The optimized membrane (TFC-8) featured a more homogeneous and integrity PA separation layer with the thickness of only 25 nm. Compared to the control membrane, TFC-8 exhibited a lower zeta potential, reduced pore size, a narrower pore size distribution, and a high pure water permeance of 45.78 LMH/bar. Additionally, TFC-8 achieved a high monovalent/divalent anion selectivity of 160.38 in the treatment of dyeing wastewater and showed great potential in the removal of organic micropollutants. Molecular dynamics simulations and interfacial tension confirmed that the composite interlayer greatly improved the tension at the water-organic interface, promoting a more uniform trans -interfacial mass transfer process of PIP. Overall, this work represents a novel eco-friendly modification method for the preparation of homogeneous and structurally intact PA NF membranes for efficient separation of inorganic salts from wastewater.

A one-for-four redox probe strategy for multiplex electrochemical POCT of thrombotic biomarkers

Cai Minghui,Zhang Wenwen,Yao Haixu,Su Huiyu,Cao Yang,Li Jiaxin,Min Xinyu,Cui Feiyun,Zhu Hui

Thrombotic diseases remain a major global health concern with high morbidity and mortality. Four biomarkers (TAT, t-PAIC, TM, PIC) enable real-time monitoring of coagulation, addressing limitations of traditional diagnostics. Herein, we developed a multiplex electrochemical immunosensor for the simultaneous detection of these biomarkers. The sensing platform is constructed on a glassy carbon electrode modified with gold nanoparticles and laser-induced graphene (AuNPs-LIG), which enhances conductivity and signal output. A “One-for-Four” strategy was employed using four distinct redox probes (AQ, OAP, MB, PPD), each conjugated to a specific antibody via amidation and anchored to functionalized carbon nanotubes (Au@CNTs), forming a multichannel signal amplification system with high sensitivity. The immunosensor exhibited excellent linearity (10–100 ng/mL, R 2  > 0.98) and low detection limits: 3.99 ng/mL (TAT), 5.25 ng/mL (t-PAIC), 2.51 ng/mL (TM), and 4.20 ng/mL (PIC). It also demonstrated high selectivity, reproducibility and long-term stability. Recovery in human plasma ranged from 84.1% to 112.21%, indicating strong applicability to complex clinical samples. This work presents a robust, portable POCT platform with great potential for early diagnosis and dynamic monitoring of thrombotic diseases. Graphical

A Terminator-Inspired Liquid Metal Stent for Treating Atherosclerotic Vascular Stenosis with Precise Vascular Matching and Magnetothermally Regulated Active Degradation

Jianxiang Xiong,Li Huang,Jingzheng Huang,Zhibin Zhou,Shen Hu,Wenpeng Zhao,Guizhen Xu,Xingchun Lai,Guoyu Yang,Weimin Zhou,Xiaolei Wang

Atherosclerosis represents the primary pathological basis of cardiovascular diseases. Vascular stent implantation as a key intervention for treating atherosclerosis, currently faces challenges such as the lack of personalized customization, difficulty in precisely matching diseased vessels, and prolonged risks of thrombosis and restenosis. Inspired by the shape-shifting liquid-metal robot T-1000 from the film Terminator , this research pioneers the use of liquid metal (LM) in vascular interventional therapy. LM was cast into a mold, then drilled and ground to the Hep/PLL/PDA/LM stent. Its heparin coating confers effective anti-thrombogenic activity and mitigates restenosis. More importantly, after the process of vascular remodeling, the stent can be degraded non-invasively through the application of an alternating magnetic field. Furthermore, the stent material exhibits notable antioxidant and anti-inflammatory properties. The findings above suggest that the magnetothermal characteristics of LM make it possible to precisely match diseased vessels and achieve controllably degradable vascular interventional therapy. This capability can fundamentally address the safety risks associated with the prolonged presence of traditional vascular stents, potentially eliminating the necessity for follow-up surgical interventions.

Nanocolloidal Hydrogel Coatings With Multiple Functions for Antithrombogenic Biointerfaces

Tiantian Ding,Chunxia Ren,Liyuan Meng,Xu Zhang,Hengchong Shi,Bai Yang,Yunfeng Li

Hydrogel coatings offer a promising strategy for improving the biocompatibility and functionality of medical devices. However, conventional hydrogel coatings often suffer from swelling-induced delamination, poor durability, and limited resistance to thrombosis and biological adhesion. Here, we report a nanocolloidal hydrogel coating (NHC) that shows an important and highly impactful combination of properties, such as lubrication, antibioadhesion, and antithrombogenicity. The NHC is prepared by anchoring a layer of nanocolloidal hydrogel made from methacryloyl-modified hyaluronate nanoparticles on the surface of biomedical implants and medical devices. We show that the long-term nonswelling behavior of nanocolloidal hydrogels renders the good durability and the stable adhesion to diverse substrates under physiological conditions. NHCs with tunable mechanical properties significantly reduce the friction of biomedical devices such as catheters, stents, and contact lenses. NHCs effectively resist the adhesion of proteins, cells, and bacteria. Moreover, NHCs show good hemocompatibility and minimal in vivo inflammatory response. Notably, NHCs covalently modified by heparin further exhibit robust antithrombotic activity both in a dynamic in vitro circulation model and an arteriovenous shunt model in a rabbit. NHCs with greatly enhanced properties will offer a clinically translational platform for innovative hydrogel coatings on diverse medical devices and implantable bioelectronics.
