One-Dimensional Ferroelectric Nanoarrays with Wireless Switchable Static and Dynamic Electrical Stimulation for Selective Regulating Osteogenesis and Antiosteosarcoma

Cairong Xiao,Lei Fan,Shiqi Zhou,Xinchang Kang,Pengfei Guan,Rumin Fu,Changhao Li,Jian Ren,Zhengao Wang,Peng Yu,Yan Wang,Chunlin Deng,Lei Zhou,Chengyun Ning

Preventing local tumor recurrence and simultaneously improving bone-tissue regeneration are in great demand for osteosarcoma therapy. However, the current therapeutic implants fail to selectively suppress tumor growth and enhance osteogenesis, and antitumor therapy may compromise osseointegration of the bone implant. Here, based on the different responses of bone tumor cells and osteoblasts to different electric stimulations, we constructed ferroelectric BaTiO3 nanorod arrays (NBTO) on the surface of titanium implants with switchable dynamic and static electrical stimulation for selective bone-tumor therapy and bone tissue regeneration. Polarized NBTO (PNBTO) generated a sustained dynamic electrical stimulus in response to wireless ultrasonic irradiation (“switch-on”), which disrupted the orientation of the spindle filaments of the tumor cell, blocked the G2/M phase of mitosis, and ultimately led to tumor cell death, whereas it had almost no cytotoxic effect on normal bone cells. Under the switch-off state, PNBTO with a high surface potential provided static electrical stimulation, accelerating osteogenic differentiation of mesenchymal stem cells and enhancing the quality of bone regeneration both in vitro and in vivo. This study broadens the biomedical potential of electrical stimulation therapy and provides a comprehensive and clinically feasible strategy for the overall treatment and tissue regeneration in osteosarcoma.

Magnetic manipulation of Fe3O4@BaTiO3 nanochains to regulate extracellular topographical and electrical cues

Yusheng Zhang,Borui Su,Yuan Tian,Zhuoting Yu,Xiaoyang Wu,Jie Ding,Chengheng Wu,Dan Wei,Huabin Yin,Jin Sun,Hongsong Fan

Magnetic fields play an essential role in material science and biomedical engineering. Magnetic-responsive materials can be arranged orderly in matrix to realize the construction of an aligned scaffold under magnetic induction. However, a single topological cue is insufficient to activate neural tissue regeneration, demanding more cues to promote regeneration synergistically, such as electrical stimulation and a biomimetic matrix. Herein, we propose one-dimensional (1D) magnetoelectric Fe 3 O 4 @BaTiO 3 nanochains with controllable lengths under the regulation of a magnetic field. These nanochains can be oriented in the biomimetic hydrogel under magnetic guidance and induce the hydrogel microfiber to align along the direction of the nanochains, which is beneficial for cell-oriented outgrowth. This aligned hydrogel enabled wireless electrical stimulation mediated by magnetoelectric nanochains under magnetic stimulation, thereby activating the voltage-gated ion channel. Consequently, topological and electrical cues in this multifunctional biomimetic hydrogel synergistically enhanced the expression of neural functional proteins, facilitating synapse remodeling and neural regeneration. Predictably, the construction of multifunctional hydrogels based on low-cost and facile synthesis of magnetoelectric nanochains is an emerging patient-friendly and effective therapeutic strategy for neural or other tissue regeneration. Statement of significance A facile and controllable magnetic strategy is established to manipulate 1D nanomaterial growth, matrix topography, and wireless electrical stimulation of cells. First, the magnetic-assisted interface co-assembly was used to control the length of Fe 3 O 4 @BaTiO 3 nanochains with enhanced magnetoelectric effect. Then, the motion of the magnetic-induced nanochains guided the orientation of nanofibers in a 3D biomimetic hydrogel matrix. Finally, wireless electrical signals and topological cues in the biomimetic matrix synergistically promoted orderly aligned cell outgrowth and membrane depolarization by Ca 2+ influx, thus enhancing nerve cell synaptic plasticity and functional expression. Consequently, this work provides a conceptual strategy from material design to extracellular matrix signal manipulation and synergistic induction of tissue regeneration.

Transcutaneous electrotherapy of facial nerve injuries based on dissolvable conductive microneedles

Long Lin,Xiaojing Yuan,Lili Ma,Liang Xu,Wenyue Xie,Ying Jiang,Jidong Xiu,Hongwei Xu,Zhuoling Tian,Jianan Qu,Chunhui Zhao,Zuoying Yuan,Huilin Liu,Jianyong Huang,Xing Wang

Local electrical stimulation (LES) is a widely used clinical method to treat peripheral nerve injury. However, existing plate electrodes (PE) usually suffer from low electronic transport efficiency, whereas acupuncture electrical stimulation may cause damage to normal tissues. There is an urgent need to develop an innovative LES technique with efficient electronic transport, minimal tissue injury and enhanced biosafety. Here, we present a new stretchable transcutaneous electrical stimulation system (STESS) that integrates dissolvable conductive microneedles (DCMN) for optimal skin penetration and electronic transport, along with snake-like electrodes that offer superior flexibility, which can effectively penetrate the stratum corneum, enabling deep tissue electrostimulation with minimal injury. In vitro studies and 3D finite-element analysis demonstrate that the DCMN-based electrotherapy significantly enhances subcutaneous nerve stimulation compared to the conventional PE-based technique. By diminishing the electrical resistance of the stratum corneum, the DCMN-based electrotherapy has shown remarkable in vivo therapeutic efficacy in a rat facial nerve crush injury model. This will pave the way for innovative physiotherapeutic methodologies in the realm of neurological rehabilitation.

3D-printed Ti3C2/polycaprolactone composite scaffold with a DOPA-SDF1 surface modified for bone repair

Yu Han,Li-Hui Sun,Bo Cai,Ming Xia,Chun-Quan Zhu,Dong-Song Li

Large bone defects are a major clinical challenge in bone reconstructive surgery. 3D printing is a powerful technology that enables the manufacture of custom tissue-engineered scaffolds for bone regeneration. Electrical stimulation (ES) is a treatment method for external bone defects that compensates for damaged internal electrical signals and stimulates cell proliferation and differentiation. In this study, we propose a simple, reliable, and versatile strategy to prepare multifunctional 3D printed scaffold combined with ES for bone defect therapy. Firstly, scaffolds composed of polycaprolactone (PCL) and Ti 3 C 2 were prepared by 3D printing technology, and then a stromal cell derived factor 1 (SDF1) containing DOPA tag was loaded onto the scaffold surface. Ti 3 C 2 was selected as the electrode component because of its excellent electrical conductivity. The selection of DOPA-modified SDF-1(DOPA-SDF1) can improve the material binding ability and exert long-term stem cell recruitment function. The results show that prepared 3D printed scaffold (DOPA-SDF1@PCL#Ti 3 C 2 ) has good hydrophilicity, electrical conductivity, antibacterial property, biocompatibility and stem cell recruitment ability. Furthermore, the expression of osteogenic specific genes in scaffold surface cells was significantly increased when pulse ES (PES) treatment was applied. The results of tibial plateau defect repair experiment showed that DOPA-SDF1@PCL#Ti 3 C 2 scaffold can significantly promote the formation of new bone and collagen fibres. When the DOPA-SDF1@PCL#Ti 3 C 2 scaffold was used in combination with PES therapy, the bone defect regeneration rate was further improved. This kind of scaffold could provide a new strategy for promoting the healing of large bone injuries and could expand the application of adjuvant therapy such as PES.

Dual electrical stimulation at spinal-muscular interface reconstructs spinal sensorimotor circuits after spinal cord injury

Zhou Kai,Wei Wei,Yang Dan,Zhang Hui,Yang Wei,Zhang Yunpeng,Nie Yingnan,Hao Mingming,Wang Pengcheng,Ruan Hang,Zhang Ting,Wang Shouyan,Liu Yaobo

The neural signals produced by varying electrical stimulation parameters lead to characteristic neural circuit responses. However, the characteristics of neural circuits reconstructed by electrical signals remain poorly understood, which greatly limits the application of such electrical neuromodulation techniques for the treatment of spinal cord injury. Here, we develop a dual electrical stimulation system that combines epidural electrical and muscle stimulation to mimic feedforward and feedback electrical signals in spinal sensorimotor circuits. We demonstrate that a stimulus frequency of 10−20 Hz under dual stimulation conditions is required for structural and functional reconstruction of spinal sensorimotor circuits, which not only activates genes associated with axonal regeneration of motoneurons, but also improves the excitability of spinal neurons. Overall, the results provide insights into neural signal decoding during spinal sensorimotor circuit reconstruction, suggesting that the combination of epidural electrical and muscle stimulation is a promising method for the treatment of spinal cord injury.

A Safe, Stable, Simple, Serviceable, and Self-Powered Wound Dressing With Continuous Low-Voltage Direct Current Electrical Stimulation: an Efficient Approach to Accelerate Wound Healing

Chenxi Shi,Huan Wang,Xiaojing Wang,Kefeng Li,Pengbi Liu,Lihuan Wang,Hui Yu

Electrical stimulation (ES) dressings have garnered considerable attention owing to their profound impact on chronic wound care, while the existing ES dressings necessitate external power supply or intricate structures, posing potential safety risks and usage inconveniences. Herein, a safe, stable, simple, serviceable, and self-powered (5S) ES dressing composed of an electrospinning asymmetric nanofiber membrane with screen-printed electrodes and a polyurethane foam is developed. The asymmetric nanofiber membrane acts as the power due to its electrical double-layer effect during the unidirectional exudate transfer, and the polyurethane foam with excellent water absorption and retention properties effectively manages the exudate. This 5S dressing generates a sustained low-voltage direct current ES in situ at the wound site and creates a favorable microenvironment for wound re-epithelization. In vitro and in vivo studies indicate that this 5S dressing can significantly promote wound healing. Compared with the control group (on the 7th day), the collagen deposition of the 5S dressing treated group increased by 15.9%, the capillary density increased by 90.6%, and the epidermal thickness increased by 228.3%, resulting in a 23.6% enhancement in the wound healing rate. Consequently, the 5S wound dressing presents a highly sophisticated and effective therapeutic approach for accelerating wound healing.

Artificial Synapse Based on Organic–Inorganic Hybrid Perovskite with Electric and Optical Modulation

Jie Lao,Wen Xu,Chunli Jiang,Ni Zhong,Bobo Tian,Hechun Lin,Chunhua Luo,Jadranka Travas-Sejdic,Hui Peng,Chun-Gang Duan

Artificial synapse of neuromorphic computing has been attracting significant attention as an alternative concept for next-generation computing. Here, a two-terminal synaptic device based on potassium iodide (KI) and CH 3 NH 3 PbI 3 (MAPbI 3 ) hybrid film (KI–MAPbI 3 ) is reported. The use of KI as an additive in the film preparation process greatly reduces grain boundaries, facilitating the improvement of device performance. The prepared synaptic device exhibits dual response to electric and light stimuli due to the unique properties of MAPbI 3 . Besides the successful simulation of biological synaptic functionalities such as paired-pulse facilitation (PPF), plasticity in long-term potentiation (LTP), and long-term depression (LTD), the learning behavior of human under different moods and Pavlov's dog experiment are also simulated by combining electric and light stimuli. For image recognition, the simulation result shows that the device achieves an accuracy of 84.2% under light illumination after only 1500 learning phases.

Thermal and electro-induced shape-memory Eucommia ulmoides gum composites filled with carbon nanotubes

Lin Xia,Qi Wang,Jiafeng Meng,Zhenguo Ma,Ping Zhao

A series of thermal and electro double-stimulus shape memory composites were processed using natural Eucommia ulmoides gum (EUG) and carbon nanotubes (CNTs). Mechanical, cure, and thermal properties were studied by universal testing machine, differential scanning calorimetry (DSC), and dynamic mechanical analysis (DMA), respectively. The shape memory behavior of the composites was explored via heat and electrical stimulation. The cross-linked network in the composites acted as a fixed domain, and the crystalline regions acted as a reversible domain during shape memory process. We studied the shape memory behavior of composites at different voltage and monitored the temperature change of composites during the process of electro-induced deformation memory in real time. Experimental results showed that appropriate amount of carbon nanotubes could not only improve the strength of composites but also provide electrical shape memory properties to composites. So, it is underscored that these kinds of shape memory composites exhibit excellent electro-responsive properties and have the potential to be used for intelligent and multiresponse shape memory applications.

A capacitive polypyrrole-wrapped carbon cloth/bacterial cellulose antibacterial dressing with electrical stimulation for infected wound healing

Wang Hao,Zheng Ruizhu,He Pengyu,Li Xiaoming,Shi Zhijun,Yang Guang

The formulation of an antibiotic-free antibacterial approach is imperative in circumventing escalating bacterial drug resistance. Electrical stimulation presents a viable therapeutic modality for such an approach. Nonetheless, obstacles persist in achieving efficacious sterilization with biosafe low-voltage electrical fields (EFs) and enduring antibacterial capabilities. In this study, we have devised a novel capacitive antibacterial dressing comprising polypyrrole-wrapped carbon cloth (PPy-CC) electrodes and a bacterial cellulose (BC) hydrogel separator. Subjected to 1 V electrical stimulation for 10 min, the dressing attains high bactericidal efficiency (up to 99.97%) and enhanced activity against multidrug-resistant (MDR) bacteria (up to 99.99%). Its considerable electric capacity and rechargeability allow for repeated charging to achieve sustained sterilization. In vivo results demonstrate significant inhibition of wound infection and facilitated wound recovery in infected full-thickness defects in mouse models. This represents an antibiotic-free, physically-stimulated treatment modality for infected wounds with considerable potential for clinical application. TOC A capacitive antibacterial dressing consists of polypyrrole-wrapped carbon cloth as electrode layer and bacterial cellulose hydrogel as separator with efficiently accelerated infected-wound healing. The resultant hydrogel dressing exhibits excellent high capacitance characteristics, good biocompatibility, efficient and sustained antibacterial ability after low voltage charging. It is an antibiotic-free, physical stimulation-based strategy for infected wounds.

Multifunctional Organohydrogels for pH-Responsive Fluorescent and Electrostimulus Writing

Siyu Yang,Qiuyan Luo,Chuanluan Guo,Jia Jiang,Xiaohong Wang,Juguo Dai,Dongxu Li,Kaibin He,Yiting Xu,Conghui Yuan,Weiang Luo,Lizong Dai

Hydrogels have attracted widespread attention in anticounterfeiting due to their unique physical/chemical properties and designability. However, hydrogels’ poor mechanical properties and sluggish response to chemical stimuli pose challenges for their wide application. A fluorescent tough organohydrogel capable of freeform writing of information is reported in this work. By incorporation of the fluorescent monomer 7-methylacryloxy-4-methylcoumarin into the polyacrylamide network in a covalently cross-linked manner while intertwining with the carboxymethyl cellulose sodium network, a fluorescent tough organohydrogel with a dual-network structure is prepared. The organohydrogel shows acid–base-mediated adjustable fluorescence through the transformation of fluorescent monomers. Ion printing and electrical stimulation design achieved free information storage and encryption. In addition, the prepared organohydrogel has good antifreezing properties and can be encrypted and decrypted at subzero temperatures. The encrypted information in the organohydrogel can be read only after UV-light irradiation. These patterned fluorescent organohydrogels should find applications in protected message displays for improved information security.

Refining and in-situ growth of polyaniline endows the cellulose fibers with electrical stimulation sterilization

Jinxin Lan,Lvlv Xu,Yao Wu,Jiazhen Chen,Hui Chen,Jinfeng Huang,Xiaofeng Yong,Dongdong Lu,Xiaojuan Ma,Shilin Cao

Bacteria and virus infections have posed a great threat to public health and personnel safety. For realizing rapid sterilization of the bacteria and virus, electrical stimulation sterilization was adopted to endow cellulose fibers with instantaneous antibacterial and antiviral properties. In the proposed strategy, the fiber is fluffed by mechanical refining, and then by means of the hydrogen bond between hydroxyl and aniline, the polyaniline (PANI) directionally grows vertically along the fine fibers via in-situ oxidative polymerization. Benefiting from the conductive polyaniline nanorod arrays on the fiber stem, the paper made from PANI modified refined fibers (PANI/BCF/P) exhibited excellent antibacterial and antiviral activity, the inhibition rates against S. aureus , E. coli , and bacteriophage MS2 can up to 100 %, 100 %, and 99.89 %, respectively when a weak voltage (2.5 V) was applied within 20 min. This study provides a feasible path for plant fiber to achieve efficient antibacterial and antiviral activity with electrical stimulation, which is of great significance for the preparation of electroactive antibacterial and antiviral green health products.
