Mercury ions (Hg2+) pollution in the water environment can cause serious harm to human health. Trace Hg2+detection is of vital importance for environmental monitoring. Herein, we report a novel design of Ru-MOFs modified gold microelectrode for Hg2+determination. Ru-MOFs are synthesized directly by the cathodic method on gold microelectrode, with the covered area accurately controlled. Cathodic synthesized Ru-MOFs show good conductivity and are suitable to be used as the electrode surface material directly. The synergy of the pre-deposition process and the adsorption process of Ru-MOFs can effectively improves the performance of the sensor. The results show good linearity (R2= 0.996) from 0.1 ppb to 5 ppb, with a high sensitivity of 0.583μA ppb−1mm−2. The limit of detection is found to be 0.08 ppb and the test process is within 6 min. Most importantly, the senor has a good anti-interference ability and the recoveries are satisfactory. This miniature electrochemical sensor has the potential for on-site detection of trace mercury in the field. Keywords:mercury ions;metal-organic frameworks;electrochemical micro-sensor;cathodic synthesis

A novel electrochemical microsensor was developed for the ratiometric and simultaneous determination of hydrogen peroxide (H2O2) and ascorbic acid (AA) based on the borate-phenol “switch” recognition mechanism and carbon nanotube (CNT) catalytic characteristics. First of all, a carbon fiber microelectrode (CFME) was coated with CNTs. Then, a specific probe, 9-anthraceneboronic acid pinacol ester (9-AP), was screened and decorated on CNTs through π–π stacking for the recognition of H2O2 based on the transformation of boric acid ester into electroactive phenols. CNTs not only served as the amplifiers of current signals, but also as catalysts facilitating AA oxidation. Meanwhile, ferrocenecarboxylic acid (Fc), inert to H2O2 and AA, was modified on another amino-functionalized CNT microelectrode via an amide bond as an internal reference channel for avoiding errors caused by environmental discrepancies. The two-channel ratiometric microsensor enabled the sensitive and accurate detection of H2O2 and AA simultaneously, and the detection limits were estimated to be 0.09 μM and 4.12 μM, respectively. The developed microsensor with remarkable analytical performance was finally applied for the simultaneous detection of H2O2 and AA in the live rat brain.

A novel electrochemical microsensor is constructed on a traditional acupuncture needle (AN) and used to monitor a biomarker of the SARS-CoV-2-N protein. The reversible interaction of the borate bond between the cis-diol in this glycoprotein and the phenylboronic acid in 4-mercaptophenylboronic acid (4-MPBA) is effectively exerted. This interaction was applied to anchor the SARS-CoV-2-N protein onto 4-MPBA, which is covalently self-assemblied onto electrodeposited AuNPs by the S-Au bond. Meldola blue was then electropolymerized around the protein template. After the template was eluting, three-dimensional nanocavities complementary with the protein were generated within the polymelola blue (pMB) layer. Interestingly, nanocavities could play a channel role for the electron-transfer of outer [Fe(CN)6]3−/4−, and the signal of electrochemical probe could be hindered after recombining the SARS-CoV-2-N protein, which laid a platform for the detection of this biomarker. After optimizing the influencing factors, the prepared microsensor exhibits a linear range of 0.1 ~ 1000 ng mL−1 with a low detection limit of 0.01 ng mL−1 (S/N = 3). In particular, the sensing ability was dramatically affected by the thickness correlative factors for the polymer matrix. The suitable thickness is effective for the sensing signals, which corresponds to the behavior of surface-imprinted polymer. The microsensor showed comparatively high sensitivity and selectivity and practically detected the SARS-CoV-2-N protein in the serum sample, which is of scientific significance for the development of the electrochemical microsensor and acupuncture.

Osteopontin (OPN) is an important biomarker for reflecting osteoarthritic inflammation and endochondral ossification. In the field of electroanalysis, OPN belongs to non-electroactive protein, which is usually detected by means of an outer probe or biolabel. Here, a novel microsensor can directly electroanalyze OPN, which was constructed by coordinating surface molecularly imprinted polymer (SMIP) with a built-in electroactive probe of poly (methylene blue) (pMB) on an acupuncture needle microelectrode (ANME). The OPN template can be reversibly anchored using 4-mercaptophenylboric acid (4-MBPA) via a borate bond between phenylboronic acid and the external cis-diol of the glycoprotein. Methylene blue (MB) and dopamine (DA) were sequentially electropolymerized and grown around templates, which played pivotal roles in the detection signal from the built-in pMB through the imprinted nanocavities. After the recombination of OPN molecules with imprinted nanocavities, the current strength of built-in pMB could be impeded with a highly sensitive response. This microsensor shows a linear relationship to the concentration of OPN from 0.01 to 1000 ng mL-1 with a detection limit of 3 pg mL-1. The microsensor also exhibits high selectivity and stability, which is attributed to the recognizing ability of the imprinted nanocavities and the hindrance and anti-interference function of coated polydopamine (pDA). This strategy of preparing a sensor shows practical and scientific significance for functionalizing microelectrodes and constructing microsensors for non-electroactive glycoproteins. In the future, it will be fascinating to integrate this microsensor with the acupuncture technique.

Measurement of signal molecule is critically important for understanding living systems. Nitric oxide (NO) is a key redox signal molecule that shows diverse roles in virtually all life forms. However, probing into NO's activities is challenging as NO has restricted lifetime (<10 s) and limited diffusion distance (usually <200 μm). So, for the direct acupuncture of NO within the time-space resolution, an electrochemical microsensor has been designed and fabricated in this work. Fabrication of the microsensor is achieved by (1) selective assembly of an electrocatalytic transducer, (2) attaching the transducer on carbon fiber electrode, and (3) covered it with a screen layer to reduce signal interference. The fabricated microsensor exhibits high sensitivity (LOD, 13.5 pM), wide detection range (100 pM–5 μM), and good selectivity . Moreover, studies have revealed that the availability of the sensor for efficient detection of NO is due to the formation of a specific DNA/porphyrin hybrid structure that has synergetic effects on NO electrocatalysis . Therefore, NO release by cells and tissues can be directly and precisely traced, in which we have obtained the release pattern of NO by different cancer cell lines, and have known its dynamics in tumor microenvironment . The fabricated electrocatalytic microsensor may provide a unique and useful tool for the direct assay of NO with high time-space resolution, which promisingly gives a technical solution for the bioassay of NO in living systems.

Mycophenolic acid (MPA), which is a potent immunosuppressant, has been widely used to prevent organ rejection after organ transplantation. In clinical practice, it is essential to monitor the free MPA concentration. Herein, convenient and fast electrochemical microsensing technology is developed for directly detecting the MPA concentration. To implement this method, ultrathin copper-based metal–organic framework (Cu–MOF) nanosheets with a thickness of about 6.6 nm are used to detect MPA. An analysis of electrochemical behavior reveals the direct electrochemical catalytic oxidation mechanism of MPA molecules on a Cu(II) tetrakis(4-carboxyphenyl)porphyrin (Cu–TCPP)-modified electrode. Furthermore, electrochemical testing results show that the ultrathin Cu–TCPP nanosheet-based microsensor exhibits high sensitivity and specificity for MPA in the nano- to micromolar range, with a detection limit of 10 nM. X-ray absorption spectroscopy (XAS) characterization reveals that the unsaturated metal ions in ultrathin nanosheets are the active sites responsible for boosting the catalytic oxidation of MPA molecules. High-performance liquid chromatography–tandem mass spectrometry further shows the catalytic oxidation product of MPA and that the phenolic hydroxyl group in the MPA molecule is the active site where the catalytic reaction occurs. The considerable sensing performance of the Cu–TCPP nanosheet-based microsensor proves that it is possible to develop a fast and convenient method for the direct high-frequency monitoring of immunosuppressive drugs.

Electroactive bacteria (EAB) hold great promise for the development of electrochemical biosensors given their unique ability to transfer electrons extracellularly via specialized pathways, a process termed extracellular electron transfer (EET). Ongoing research aims to overcome current limitations and fully harness the potential of EABs for high-performance biosensing applications. Herein, we report the fabrication of an electrochemical microsensor based on biomineralized electroactive bacteria, specifically Shewanella oneidensis MR-1. We exploited the EET capability of S. oneidensis to mineralize gold nanoparticles (AuNPs) in situ, thereby enhancing the EET efficiency and improving the electrochemical performance. With lactate serving as the sole electron donor, S. oneidensis functions as a whole-cell electrical transducer, converting the metabolism of lactate into detectable electrical signals. The resulting microsensor, designated AuNPs@S. oneidensis@CFE, exhibited enhanced sensitivity, a broad linear detection range, and high specificity for lactate detection, along with relatively good stability and reproducibility. Furthermore, when integrated into a three-in-one microelectrode, it enabled online monitoring of lactate fermentation. Thus, this work highlights the potential of combining the EET reduction capacity and sensory ability of EABs with miniaturized devices, demonstrating the feasibility of integrating biomineralization with EET pathways of EABs to develop robust electrochemical microbial biosensors.
