E-eye and FT-NIR combined with multivariate algorithms to rapidly evaluate the dynamic changes in the quality of Gastrodia elata during steaming process

Jiu-ba Zhang,Bin Wang,Yun-fei Zhang,Yi Wu,Ming-xuan Li,Ting Gao,Tu-lin Lu,Zhen-hua Bian,Lian-lin Su

Gastrodia elata (GE) is traditionally subjected to steaming, and steaming duration plays a crucially important role in determining GE quality. This study examined the variations in bioactive components during the steaming process and proposed the utilization of electronic eye and Fourier Transform near-infrared (FT-NIR) spectroscopy for quality assessment. The findings revealed that the levels of parishin E parishin B, parishin A, and gastrodin initially rose and subsequently declined, while 4-Hydroxybenzyl alcohol exhibited a rapid decrease followed by stabilization. With prolonged steaming, the brightness of GE decreased, while the red and yellow tones became more pronounced and the color saturation increased. FT-NIR divided the steaming process into three stages: 0 min (raw GE), 0–9 min (partially steamed GE), and 9–30 min (fully steamed GE), and the partial least squares regression models effectively predicted the levels of five components. Overall, this study provided valuable insights into quality control in food processing.

On-site monitoring of thiram via aggregation-induced emission enhancement of gold nanoclusters based on electronic-eye platform

Xu Zhao,Deshuai Kong,Rui Jin,Hongxia Li,Xu Yan,Fangmeng Liu,Peng Sun,Yuan Gao,Geyu Lu

The rapid and on-site monitoring of thiram is of great significance because the residue of thiram in agriculture has caused great contamination in environment and food, as well as threaten to the human health. Herein, we constructed a fluorometric strategy for on-site monitoring thiram via the aggregation-induced emission enhancement (AIEE) effect of glutathione-capped gold nanoclusters (GSH-AuNCs). The dissociated Ag + ions can effectively enhance the fluorescent emission intensity of GSH-AuNCs at 570 nm owing to the AIEE phenomenon. Upon the addition of thiram, the fluorescence enhancement effect was weakened due to the strong coordinative interaction between Ag + and thiram. By monitoring the signal change, the thiram residues were quantitatively analyzed with the detection limit of 0.05 μg mL −1 . Notably, a home-made electronic-eye (E-eye) system, containing of a test-strip and a smartphone, was constructed as a portable platform to recognize and quantitatively analyze thiram in 10 min on the basis of captured fluorescence signals from a test strip with immobilized probe by direct photographing, which provides a direct quantitative tool to identify pesticide. Taking advantage of user-friendliness, portability and simplicity, the E-eye system are expected to fulfill the demand for quantitative on-site testing of pesticide especially in resource-limited areas.

A novel portable biosensor based on aptamer functionalized gold nanoparticles for adenosine detection

Shuqi Zhou,Ying Gan,Liubing Kong,Jiadi Sun,Tao Liang,Xinyi Wang,Hao Wan,Ping Wang

Adenosine has received great attentions acting as a potential biomarker for monitoring lung cancer. Most of the reported studies for adenosine detection require large instruments and complicated procedures. Herein, a sensitive, rapid and in-situ colorimetric aptasensor was developed for adenosine detection. Moreover, a homemade biomimetic electronic-eye (E-eye) was established and utilized as a portable in-time detection equipment. The entire measurement can be completed within 20 min, including the combination of aptamer with adenosine or AuNPs and the detection of adenosine. Four different kinds of aptamer were compared and the results showed that the AuNPs-aptamer-biotin system was the most stable and with the widest detection range of 5.0 μM–60.0 μM and the lowest LOD of 0.17 μM. Moreover, the artificial urine samples were also tested with a linear range from 5.0 to 50.0 μM and a LOD of 0.48 μM. The results validated that the aptasensor together with the E-eye can be a promising platform for adenosine detection.

Efficiency of four different dietary preparation methods in extracting functional compounds from dried tangerine peel

Huijuan Zhang,Jiefen Cui,Guifang Tian,Christina DiMarco-Crook,Wei Gao,Chengying Zhao,Genyuan Li,Yunhe Lian,Hang Xiao,Jinkai Zheng

Dried tangerine peel (DTP) is an excellent plant resource that has been used as ingredients for both food and traditional Chinese medicine. In this study, the efficiency of four different dietary preparation methods (i.e. soaking, boiling, steaming, and ethanol extraction) in extraction of functional compounds (i.e. flavonoids and essential oil constituents) from DTP was evaluated systematically for the first time. To conduct a comprehensive evaluation of the extraction of the functional compounds, a synthetic evaluation model based on a weighting method was established. The optimum conditions of each dietary preparation method (e.g., time, temperature, solid-liquid ratio, etc.) were determined by response surface methodology . Ethanol extraction showed the best extraction efficiency, followed by soaking, boiling, and steaming. Additionally, different DTP extracts were shown to be clearly distinguished by electronic eye and electronic tongue. This research provides essential findings for the effective dietary instruction of DTP consumption.

MnO2 nanosheets as the biomimetic oxidase for rapid and sensitive oxalate detection combining with bionic E-eye

Ying Gan,Ning Hu,Chuanjiang He,Shuqi Zhou,Jiawei Tu,Tao Liang,Yuxiang Pan,Dmitry Kirsanov,Andrey Legin,Hao Wan,Ping Wang

Urolithiasis commonly occurs in kidney and ureteral, and may cause local organ/tissue damage, even kidney failure. The incidence of this disease is increasing worldwide, in which calcium oxalate is the major composition forming the urinary calculus. Therefore, to monitor this disease for the prevention and treatment, measuring the oxalate in the urine is of great significance. Here, a rapid and sensitive colorimetric method was developed based on 3,3',5,5'-tetramethylbenzidine-manganese dioxide (TMB-MnO 2 ) nanosheets for oxalate detection. MnO 2 nanosheets acted as an efficient biomimetic oxidase to catalyze the reaction with TMB and oxalate. Pale yellow TMB can be oxidized to blue oxide TMB catalyzed by BSA-stabilized MnO 2 nanosheets, and oxalate can selectively inhibit this reaction by consuming and reacting with MnO 2 nanosheets, thus achieving the quantitative detection of oxalate. Moreover, a home-made bionic electronic-eye (E-eye) system was developed as a portable in-situ detection platform to efficiently measure the oxalate concentrations in 10 s by direct photographing. By optimizing experimental conditions, this method shows a wide linear range (7.8 μM to 250 μM) and a low detection limit (0.91 μM) for oxalate detection. Besides, this method exhibits high selectivity even with 80-fold interfering chemicals. Furthermore, the performance of the method was validated by testing the artificial urine samples, indicating its great potential for monitoring and diagnosis of urolithiasis in point-of-care applications.

A method combining a kit with the Bionic e-Eye for rapid on site detection of diarrhetic shellfish poisoning

Xianxin Qiu,Longjie Zhong,Ying Gan,Kaiqi Su,Shihui Hu,Ping Wang

Okadaic acid is a kind of fat-soluble diarrhetic shellfish poison existing extensively in shellfish. The rapid on site detection of shellfish contaminated by the toxin is a serious problem. Herein, a new method combining a kit with the Bionic e-Eye was developed for the rapid detection of okadaic acid on site. After optimization the kit reached a very low limit of detection, 0.19 μg L−1, and the linear detection range was 0.2–10 μg L−1. The cross reaction rate with dinophysistoxins 1 was 23.6%. The kit had a small cross reaction with other kinds of toxin like saxitoxin, gynotoxin 2 & 3 and pectenotoxins 2-b. The recovery range was about 75.78–102.56%, and the CVs were lower than 20%. A correlation analysis was conducted between the Bionic e-Eye and a microplate reader in PBS and shellfish backgrounds. The detection results of the real shellfish samples by the Bionic e-Eye exhibited great consistency with high performance liquid chromatography. The total time taken to perform the detection was less than 90 minutes. Above all, the method integrated kit with the Bionic e-Eye was very suitable for the rapid detection of okadaic acid on site.

Microfluidic-based fluorescent electronic eye with CdTe/CdS core-shell quantum dots for trace detection of cadmium ions

Xinyi Wang,Liubing Kong,Ying Gan,Tao Liang,Shuqi Zhou,Jiadi Sun,Hao Wan,Ping Wang

Cadmium ions (Cd 2+ ) greatly threat human health and the environment due to its extremely severe toxicity. Therefore, it is of paramount importance to establish a sensitive and portable platform for monitoring Cd 2+ on site. In this work, a novel microfluidic-based fluorescent electronic eye (E-eye) combined with tetrasodium iminodisuccinate (IDS)-etched CdTe/CdS quantum dots (QDs) was developed for portable and sensitive detection of trace Cd 2+ in water environment. The fluorescent E-eye consists of a microfluidic chip for miniaturized flow analysis, an ultraviolet light excitation module for fluorescent excitation, an optical lens for device miniaturization and a smartphone for portable photographing and analysis. The IDS was added in the CdTe/CdS QDs to cause fluorescence quenching due to the chemical etching. Subsequently added Cd 2+ will be recognized by etched QDs, thus inducing the fluorescence changes that can be directly captured by the E-eye for quantitative detection of Cd 2+ . With the optimization of all parameters including pH, reaction time and the concentration of IDS, the proposed platform could detect Cd 2+ with a low detection limit of 0.26 μg/L in the range of 1–250 μg/L. It is worth noting that the performance of the developed fluorescent E-eye is quite comparable to a commercial microplate reader with a detailed comparison in linearity, sensitivity and detection limit. In summary, the proposed microfluidic-based fluorescent E-eye provides a promising platform for portable and high sensitive detection of trace cadmium in water environment.
