The endogenous electrical potential generated by native bone and periosteum plays a key role in maintaining bone mass and quality. Inspired by the electrical properties of bone, different negative surface potentials are built on microspheres to restore electric microenvironment for powerful bone regeneration, which was prepared by the combination of strontium-doped barium titanate (Sr-BTO) nanoparticles and poly (lactic-co-glycolic acid) (PLGA) with high electrostatic voltage field (HEV). The surface potential was modulated through regulating the phase composition of nanoparticles in microspheres by the doping amount of strontium ion (Sr2+). As a result, the 0.1Sr-BTO/PLGA group shows the lowest surface potential and its relative permittivity is closer to natural bone. As expected, the 0.1Sr-BTO/PLGA microspheres performed cytocompatibility, osteogenic activity in vitro and enhance bone regeneration in vivo. Furthermore, the potential mechanism of Sr-BTO/PLGA microspheres to promote osteogenic differentiation was further explored. The lower surface potential generated on Sr-BTO/PLGA microspheres regulates cell membrane potential and leads to an increase in the intracellular calcium ion (Ca2+) concentration, which could activate the Calcineurin (CaN)/Nuclear factor of activated T-cells (NFAT) signaling pathway to promote osteogenic differentiation. This study established an effective method to modulate the surface potential, which provides a prospective exploration for electrical stimulation therapy. The 0.1Sr-BTO/PLGA microsphere with lower surface potential and bone-matched dielectric constant is expected to have great potential in the field of bone regeneration.

Conventional methods to stimulate the metabolism of bone marrow mesenchymal stem cells (BMSCs) for osteogenic differentiation typically involve systemic mobilization, which faces challenges including limited in vivo half-life, lack of selectivity, and potential side-effects. Therefore, localized modulation of BMSCs represents a more efficient and safer alternative. However, few studies have explored the regulation of a localized stimuli-responsive microenvironment to activate osteogenic differentiation via mitochondrial pathways and clarified its underlying mechanisms. Herein, a novel strategy to accelerate the metabolic switch of BMSCs in tissue defects through targeted modulation using built-in magnetoelectric biomaterials is proposed. BMSCs cultured in the magnetoelectric microenvironment exhibited an increased mitochondrial membrane potential, the highest oxygen consumption rate and enhanced adenosine triphosphate production. Furthermore, BMSCs in the magnetoelectric microenvironment demonstrated a successful metabolic switch of energy resource from glycolysis to oxidative phosphorylation, indicating a strong tendency toward osteogenic differentiation. The highest multiclass metabolite profile, indicating the most active metabolic state, was shown in rats cranial defect model treated with magnetoelectric microenvironment. This research introduces a novel approach to accelerate bone defect repair by targeted modulation of BMSC mitochondria with magnetoelectric microenvironment and provides a promising direction for exploring the intrinsic mechanisms through which the magnetoelectric microenvironment promotes bone regeneration.

The synergetic integration of living microorganisms and nonliving matrices into a new artificial micro-ecological system is a crucial challenge in bio-manufacturing especially towards the development of sustainable energy technology. Here we develop a way of creating a Shewanella oneidensis based conductive micro-niche to boost hydrogen production by integrating graphene, polydopamine and alginate. We reveal that both the respiratory metabolism induces localized hypoxic conditions inside the micro-niche and the wired-up extracellular electron reverses transfer pathway from the outer environment to the periplasmic hydrogenases, contributing to the enhanced hydrogen production rate which is about 12.7 folds enhancement compared with that of the free Shewanella oneidensis in solution. Significantly, the whole assembly process shows good biocompatibility, ensuring that the Shewanella oneidensis inside the micro-niche could maintain hydrogen production for 30 days. Overall, it is anticipated the hybrid integration of microorganisms and abiotic material will provide an effective strategy to modulate green biomanufacturer as well as enhance biological hydrogen production.

C-C chemokine receptor type 2 (CCR2 − ) cardiac-resident macrophages (CCR2 − cRMs) are known to promote cardiac repair after myocardial infarction (MI). However, the substantial depletion and slow recovery of CCR2 − cRMs pose significant barriers in cardiac recovery. Here, we construct a functional conductive cardiac patch (CCP) that can provide exogenously elastic conductive microenvironment and induce endogenously reparative microenvironment mediated by CCR2 − cRMs for MI repair. This CCP exhibits suitable mechanical properties, conductivity, and high water retention, reminiscent of natural myocardium, which can actively engage in modulating CCR2 − cRM renewal and their cell crosstalk. The functional CCP can promote the expression of Connexin43 between CCR2 − cRMs and cardiomyocytes (CMs) and regulate paracrine signaling to activate epicardial cell epithelial-to-mesenchymal transition (EMT) toward endothelial cells using rat and Wt1 CreERT2 transgenic lineage tracing mice. Overall, this study provides a promising strategy to construct a synergistic reparative microenvironment for MI repair.
