Improving anaerobic digestion of lignocellulosic hydrolysate derived from hydrothermal pretreatment via applied voltages

Chao Wang,Xiangrong Wu,Ao Xia,Abdul-Sattar Nizami,Yun Huang,Xianqing Zhu,Xun Zhu,Qiang Liao

Anaerobic digestion (AD) is an effective means to convert biomass hydrolysate to biomethane for bioenergy utilization. However, byproducts such as furfural and 5-hydroxymethylfurfural may be generated from soluble sugars during the hydrothermal pretreatment of lignocellulosic biomass, potentially inhibiting subsequent AD. Emerging electro-anaerobic digestion technology may be effective in releasing the inhibition. In this study, various voltages (0.3−1.2 V) were applied for the first time in the AD of lignocellulose hydrolysate. The results revealed that maximum chemical oxygen demand (COD) removal efficiency of 95.5 % occurred at 0.9 V, outperforming the voltage absence control by 22.5 %. Meanwhile, the most significant methane yield (336.9 NmL/g COD) and energy yield (12.1 kJ/g COD) were also achieved at 0.9 V, exceeding the control by 60.1 %. At 0.3, 0.6, and 1.2 V, AD performance outperformed that of the voltage absence, although it was not optimal due to either inadequate potential drive or excess-voltage suppression. The imposed voltage stimulated the growth of biofilm and tryptophan and humic acid secretion, enhancing the electrochemical activity. The more functionally selective electroactive bacteria Petrimonas and Sedimentobacter and archaebacteria Methanosarcina and Methanomassiliicoccus were significantly enriched under applied voltages. Efficient reciprocal metabolism among electroactive microorganisms was promoted to diversify methane production pathways.

Degradation efficiency of antibiotics by the sewage-fed microbial fuel cells depends on gram-staining property of exoelectrogens

Peng Cheng,Cong Yang,Shang Zhou,Junyu Huang,Rongrong Liu,Bing Yan

Microbial fuel cells (MFCs) are green technologies for bioenergy capture. Gram-negative bacteria have been shown as the dominant electrogenic functional microbes on the anode of MFC. However, how gram-positive bacteria perform in the mixed colonies remains unclear. In this study, specific classes of antibiotics are employed to characterize the properties of electroactive microorganisms in the sewage-fed MFC. Our results reveal that MFCs exhibit high efficiencies in degrading some widely used bacterial antibiotics and generating electricity, while being significantly influenced by gram-staining properties of microbial communities . The MFC is effective against the broad-spectrum bacterial antibiotic cefepime with a degradation efficiency of 92.5% (5 mg/L) and a maximum power density of 263.6 mW/cm 2 (25 mg/L). Electrochemical analyses demonstrate that the gram-positive bacteria on anode are more resistant to their specific antibiotic vancomycin and the maximum power output of the system is enhanced by 13.7%. Furthermore, the exoelectrogens in the mixed colonies show insensitivity to fungi-specific antibiotics such as amphotericin with extremely low degradation rate (<4.5%). Our findings provide a new perspective on exploring the characteristics of functional exoelectrogens based on the microbial responses to antibiotics with specific antibacterial spectrum. It is also crucial to guide the application of bioelectrochemical systems in medical wastewater treatment for urban sustainable development .

Validation of effective roles of non-electroactive microbes on recalcitrant contaminant degradation in bioelectrochemical systems

Yingfeng Xu,Zhipeng Ge,Xueqin Zhang,Huajun Feng,Xianbin Ying,Baocheng Huang,Dongsheng Shen,Meizhen Wang,Yuyang Zhou,Yanfeng Wang,Hanqing Yu

Bioelectrochemical systems (BESs) have been widely investigated for recalcitrant waste treatment mainly because of their waste removal effectiveness. Electroactive microbes (EMs) have long been thought to contribute to the high effectiveness by interacting with electrodes via electron chains. However, this work demonstrated the dispensable role of EMs for enhanced recalcitrant contamination degradation in BESs. We revealed enhanced p -fluoronitrobenzene ( p -FNB) degradation in a BES by observing a defluorination efficiency that was three times higher than that in biodegradation or electrochemical processes. Such an improvement was achieved by the collaborative roles of electrode biofilms and planktonic microbes, as their individual contributions to p -FNB degradation were found to be similarly stimulated by electricity. However, no bioelectrochemical activity was found in either the electrode biofilms or the planktonic microbes during stimulated p -FNB degradation; because no biocatalytically reductive or oxidative turnovers were observed on cyclic voltammetry curves. The non-involvement of EMs was further proven by the similar microbial community evolution for biofilms and planktonic microbes. In summary, we proposed a mechanism for indirect electrical stimulation of microbial metabolism by electrochemically generating the active mediator p -fluoroaniline ( p -FA) and further degradation by a sequential combination of electrochemical p -FNB reduction and biological p -FA oxidation by non-EMs.

Boosting the electricity generation of nonclassical electroactive microorganisms enabled by carbon dots

Siyu Zhang,Xinpeng Zhao,Xinqi Guo,Gaoqi Zhang,Tianchong Li,Shiyu Xu,Yufu Zhao,Jian Li,Shouxin Liu,Zhijun Chen,Shujun Li,Chenhui Yang

Producing electricity via microorganisms is promising as a sustainable and inexpensive energy source. However, most of the reported classical electroactive microorganisms are unusual in the environment, hindering their practical application. Here, we used carbon dots (CDs) to boost the power generation of common but nonclassical electroactive microorganisms, such as Escherichia coli and Staphylococcus aureus , in the natural environment. The CDs, which have good biocompatibility and high electron transfer efficiency, are taken up by bacterial cells and enable electrons to migrate rapidly to the ambient electrode because of the formation of robust electron transfer networks. The CDs promote electrochemical redox activity and increase the internal charge generation, and reduce the electrochemical impedance of the bacteria. As a result, the CDs facilitate rapid and high power generation in the microbial chemical cells and microbial fuel cells, with a nearly 94-fold and 172-fold improvement in current and power output compared with unmodified Escherichia coli . This work offers an important strategy for enhancing the generation of bioelectricity and facilitates the functional optimization of technologies that depend on microbial electron transfer.
