Efficient degradation of tylosin by Klebsiella oxytoca TYL-T1

Tao Zhang,Shuang-yan Xu,Hui Lin,Jian Yang,Zhuo-qun Zhao,Damià Barceló,Hua-bao Zheng

Tylosin is widely used in livestock; however, the release of tylosin through animal manure can cause serious environmental problems. In this study, a new tylosin-degrading strain, TYL-T1, was isolated. Its phylogenetic similarity to Klebsiella oxytoca was found to be 99.17 %. TYL-T1 maintained good growth at 40 °C over a broad pH range (4.0–10). TYL-T1 degraded 99.34 % of tylosin in 36 h under optimal conditions (tylosin initial concentration: 25 mg/L, pH: 7.0, and temperature: 35 °C). After LC-MS-MS analysis, a new degradation pathway for tylosin was proposed, including ester bond breaking of the macrolide lactone ring, redox reaction, and loss of mycinose and mycarose. Based on a transcriptome analysis, 164 genes essential for degradation were upregulated through hydrolysis and redox of tylosin. Among various transferases, lipopolysaccharide methyltransferase, glycogen glucosyltransferase, and fructotransferase were responsible for tylosin degradation. The present study revealed the degradation mechanism of tylosin and highlighted the potential of Klebsiella oxytoca TYL-T1 to remove tylosin from the environment.

Enhancement of the solubility and oral bioavailability of altrenogest through complexation with hydroxypropyl-β-cyclodextrin

Wojun Chen,Xianwen Zheng,Wenxuan Lao,Hongxin Wang,Shengfeng Chen,Canying Liu,Zhisheng Chen,Yinshan Bai,Hui Zhang,Xiaoshu Zhan,Bingyun Wang

Altrenogest (ALT), a synthetic progestogen, serves a critical role in estrus synchronization among animals like gilts and mares. However, its practical application in animal husbandry is hampered due to its poor solubility and limited oral bioavailability. To address this challenge, a solvent evaporation method was employed to create an inclusion complex of ALT with hydroxypropyl-β-cyclodextrin (ALT/HP-β-CD). The formation of this inclusion complex was confirmed by scanning electron microscopy, power X-ray diffraction, differential scanning calorimetry, Fourier transform infrared spectroscopy, nuclear magnetic resonance spectroscopy, and docking calculations. In addition, we further conducted pharmacokinetic investigation involving gilts, comparing ALT/HP-β-CD inclusion complex to an ALT oral solution. The physicochemical characterization results unveiled a transformation of ALT's crystal morphology into an amorphous state, with ALT effectively entering the cavity of HP-β-CD. Compared with ALT, the solubility of ALT/HP-β-CD inclusion complex increased by 1026.51-fold, and its dissolution rate demonstrated significant improvement. Pharmacokinetic assessments further revealed that the oral bioavailability of ALT/HP-β-CD inclusion complex surpassed that of the ALT oral solution, with a relative bioavailability of 114.08 %. In conclusion, complexation with HP-β-CD represents a highly effective approach to improve both the solubility and oral bioavailability of ALT.

rGO-K-CD-MOFs based non-enzymatic electrochemical sensor toward sensitive hyodeoxycholic acid determination

Cancan Yang,Chongyue Bai,Xiaoting Chen,Hamdy Khamees Thabet,Saad Alshammari,Yunlong Sun,Jiongru Li,Juanna Ren,Hassan Algadi,Xiangzheng Hu,Faming Gao,Dapeng Cui,Zhanhu Guo,Huige Wei

The increasing utilization of hyodeoxycholic acid (HDCA) additives in animal feed has created a pressing need for reliable detection methods. This study presents a novel non-enzymatic electrochemical platform for specific detection of HDCA using liquid nanocomposites from potassium-coordinated cyclodextrin metal-organic frameworks (K-CD-MOFs) immobilized on reduced graphene oxide (rGO) substrates. Employing neutral red (NR) as the probe, rGO-K-CD-MOFs@NR exhibits outstanding sensing performance with a wide linear detection range (5.0–100.0 μM), a high sensitivity (0.10 μA·μM −1 ), and low detection limit (1.97 μM, S/N = 3). The sensor also demonstrates exceptional selectivity for HDCA and maintains stable performance for six months under low-temperature storage conditions. The excellent performance originates from the synergistic effect between rGO's excellent electron transfer capability and K-CD-MOFs' unique host-guest recognition properties. This work not only provides an effective solution for HDCA monitoring in feed products but also offers a promising strategy for developing advanced electrochemical sensors for food safety and quality control applications in the animal husbandry industry.
