Involvement of ethylene in glutamate-mediated tomato fruit resistance to Alternaria alternata

Jiali Yang,Tengfei Wang,Jianbing Di,Yaping Liu,Yu Wang

Recent progress showed the potential of glutamate to regulate stress responses by affecting phytohormones homeostasis , but much remains to be explored. To decipher the role of ethylene in glutamate-induced resistance to Alternaria alternata , ethephon application, ethylene perception inhibitor (1-MCP) experiment and ethylene measurement were pursued. Concurrently, temporal alteration in ethylene biosynthesis and signaling pathways were transcriptionally analyzed in tomato fruit. Our data exhibited that ethylene conferred tomato fruit susceptibility rather than resistance to Alternaria alternata . The enhanced resistance in glutamate treatment was related to decreased ethylene production during the early stages of interaction, accompanied by lower expression levels of SlACS1 , SlACS2 and SlACO1 relative to the control. Interestingly, 1-MCP induced amounts of resistance similar to that observed in glutamate treatment, while ethylene receptors ( SlETR3 and SlETR4 ), mitogen-activated protein kinase SlMPK3 and ethylene response factor SlERF1 were down-regulated by glutamate during infection. Collectively, antagonizing the ethylene biosynthesis or signaling might play a pivotal role in glutamate-induced resistance to the necrotrophic pathogen Alternaria alternata .

Abscisic acid mediates wound-healing in harvested tomato fruit

Xiaoya Tao,Linchun Mao,Jiayin Li,Jingxin Chen,Wenjing Lu,Si Huang

The development of wound periderm is vital to prevent water vapor loss and pathogen invasion in wounded fruit. The effect of abscisic acid (ABA) on wound-healing in harvested tomato fruit was evaluated in this study. Wounded tomato fruit were treated with ABA or fluridone (FLD, an inhibitor of ABA biosynthesis), respectively, and allowed to wound-heal at 20   °C in darkness for 4   days. Weight loss of tomato fruit reflected the intuitionistic effects of different treatments during wound-healing. Autofluorescence and histochemical staining demonstrated the development of suberized wound periderm in harvested tomato fruit at 3–4   day after wounding. Alkanoic acids, n -alkanes, unsaturated fatty acids and pentacosane represented the major components in the suberin. ABA-treated fruit showed enhanced autofluorescence, histochemical intensity and suberin deposition along with increased activities of phenylalanine ammonia-lyase (PAL) and peroxidase (POD). FLD treatment, however, alleviated the features of suberization and enzyme activities during wound-healing. The results suggest that ABA is involved in the stimulation of wound-induced suberization in harvested tomato fruit.

Postharvest UV-C irradiation inhibits the production of ethylene and the activity of cell wall-degrading enzymes during softening of tomato (Lycopersicon esculentum L.) fruit

Jianwen Bu,Yucong Yu,Guzhanuer Aisikaer,Tiejin Ying

Mature green cherry tomato fruit were harvested and treated with ultraviolet-C (UV-C) irradiation at a predetermined dose of 4.2   kJ   m −2 and stored at 18   °C for 35 days. The effects of UV-C treatment on firmness, cell wall compositions (pectin and cellulose), cell wall ultra-structure, levels of ethylene production, and activities of cell wall degrading enzymes in the fruit were investigated during storage. Major genes involved in cell wall degradation ( PME 2.1, Cel 1, PG cat and Exp 1) were also checked. Fruit firmness was better maintained in UV-C treated fruit corresponding with higher contents of cellulose and acid-soluble pectin. Transmission electron microscopy indicated that UV-C irradiation retarded cell wall disassembly in the pericarp. Ethylene production was significantly inhibited by UV-C treatment. The UV-C treatment also suppressed the transcriptional expression of PME2.1 , Cel1 , PGcat and Exp1 genes, and inhibited pectin methylesterase (PME; EC 3.1.1.11), polygalacturonase (PG; EC3.2.1.15) and cellulase (EC3.2.1.4) activities during storage. These results suggested that the inhibition of ethylene production, which in turn down-regulated expression of genes encoding cell wall degrading enzymes, might be one of the possible mechanisms of UV-C delaying tomato fruit softening.

Overactivation of glutamate consuming pathways in l-glutamate treated tomato fruits lead to resistance against Alternaria Alternata

Jiali Yang,Xu Jing,Tengfei Wang,Jianbing Di,Li Chen,Yu Wang

Previous studies reported the ability of l -glutamate (glutamate) to induce host resistance against Alternaria alternata , but the underlying mechanism is still scarce. Here, temporal alteration in antioxidant enzymes and glutamate consuming pathways were transcriptionally and enzymatically monitored in tomato fruits. The results showed that exogenous glutamate application enhanced the activities of antioxidant enzymes, including peroxidase, superoxide dismutase and catalase, in tomatoes upon inoculation with Alternaria alternata , while lower concentrations of H 2 O 2 and malondialdehyde were observed. Interestingly, glutamate led to a decline in carbon:nitrogen ratio and induction of deaminating activity of glutamate dehydrogenase, resulting in replenishment of the tricarboxylic acid cycle. Concurrently, proline dehydrogenase mediated the proline catabolism was downregulated by glutamate, accompanied by the enhanced biosynthesis of proline due to increased expression of pyrroline-5-carboxylate synthase. Together our findings favored a model whereby overactivation of antioxidant enzymes and glutamate consuming pathways, deamination of glutamate and proline biosynthesis from glutamate, played a vital role in the mechanism of glutamate-induced resistance.

Suppression of the Jasmonate Signaling Pathway by EIN3-LIKE1 Increases Postharvest Susceptibility to Gray Mold in Ripening Tomato Fruits

Liyao Liu,Mengjia Xiang,Ruixi Shi,Dandan Sun,Liping Qiao,Laifeng Lu

Postharvest decay, primarily occurring after ripening, leads to substantial losses. Ripening increases tomato susceptibility to Botrytis cinerea, linked to desensitization of jasmonate (JA) biosynthesis and defense signaling. During ripening, key JA biosynthesis genes such as allene oxide synthase (AOS) and 12-oxophytodienoate reductase 3 (OPR3) were downregulated in response to wounding or fungal challenges. The Slmyc2 mutants with impaired JA responses showed heightened pathogen sensitivity. Conversely, EIN3-Like sleil1 mutants exhibited elevated SlMYC2 levels, which increased expression of JA biosynthesis genes such as lipoxygenase D (LOXD), OPR3, AOS, and allene oxide cyclase (AOC) alongside defense genes including proteinase inhibitor PI-6, PI-27, polyphenol oxidase, and pathogenesis-related proteins, thereby enhancing resistance. Treatment with DNA methyltransferase inhibitor 5-azacytidine increased JA content, suggesting epigenetic regulation of JA-mediated defense. Collectively, intensified ethylene signaling during ripening, mediated by SlEIL1, suppressed JA-dependent responses against B. cinerea, highlighting ethylene-JA antagonism as a key driver of postharvest decay.

Assessment of the role of brassinosteroid in regulating the disease resistance of postharvest tomato fruit by proteomic analysis

Yang Ding,Jiping Sheng,Fansheng Cheng

This study aimed to investigate the effects of brassinosteroid (BR) on disease development, disease resistance-related enzyme activities, gene expression, and proteomic change in tomato ( Solanum lycopersicum ) fruits infected by Alternaria solani . The results showed that BR pretreatment inhibited disease development and increased the activities of disease resistance-related enzymes. Fruits treated by brassinazole, the biosynthesis inhibitor of BR, are more sensitive to A . solani . BR could promote the gene expression of downstream and upstream in the systematic acquired resistance (SAR) pathways and the catalyzers in the biosynthetic pathways of phenolic and quinone compounds. The proteomic analysis showed that BR upregulated the expression of 2 proteins and downregulated the expression of 13 proteins. Furthermore, one upregulated protein was involved in the phenylpropanoid biosynthetic pathway. These results suggested that BR-mediated disease resistance in tomato fruits could share some SAR downstream genes and phenylalanine ammonia-lyase and also have distinct pathways. Practical applications Brassinosteroid treatment before harvest could be a useful strategy to improve the quality of tomato fruit and reduce postharvest losses caused by fungi disease.

Postharvest ultraviolet-C irradiation suppressed Psy 1 and Lcy-β expression and altered color phenotype in tomato (Solanum lycopersicum) fruit

Jianwen Bu,Zhendan Ni,Guzhanuer Aisikaer,Zhenhui Jiang,Zia Ullah Khan,Wangshu Mou,Tiejin Ying

Mature green cherry tomato fruit were harvested and treated with ultraviolet-C (UV-C) irradiation at a predetermined dose of 4.2   kJ   m −2 , and stored at 18   °C for 35 days. The effects of UV-C treatment on color change, pigment contents, and the expression of major genes involved in carotenoid metabolism, including Psy 1, Pds , Lcy - β , and Lcy - ɛ , encoding phytoene synthase, phytoene desaturase, lycopene β-cyclase and lycopene ɛ-cyclase, respectively, were examined. The UV-C treated fruit developed a pink red color in contrast to the normal orange red color of control fruit. Lycopene accumulation during ripening in UV-C treated fruit was significantly inhibited but its final content was not affected. However, both accumulation and final content of β-carotene were significantly suppressed in UV-C treated fruit. The lower content of β-carotene, leading to a higher lycopene to β-carotene ratio, is probably responsible for the altered color phenotype in UV-C treated fruit. Psy 1, a major gene involved in lycopene synthesis was inhibited by UV-C irradiation. Significantly suppressed expression of Lcy-β gene was also observed in UV-C treated fruit. Thus it is possible that the lower transformation from lycopene to carotenes contributed to the relatively stable content of lycopene.

Exogenous abscisic acid regulates primary metabolism in postharvest cherry tomato fruit during ripening

Xiaoya Tao,Qiong Wu,Suqing Huang,Beiwei Zhu,Feng Chen,Bin Liu,Luyun Cai,Linchun Mao,Zisheng Luo,Li Li,Tiejin Ying

Primary metabolites play crucial roles during fruit development and ripening. Phytohormone abscisic acid (ABA) has been reported to regulate postharvest tomato fruit ripening. However, knowledge about ABA in regulating primary metabolism in tomato fruit is obscure. In the present research, cherry tomato fruit were infiltrated with 1.0 mM ABA or deionized water under vacuum, and kept in darkness for 15 d. The changes of the relative contents of sugars, organic acids, and amino acids, as well as the expression levels of key genes involved in sugar and organic acid metabolisms, were analyzed. Results showed exogenous ABA accelerated tomato fruit ripening, accompanying with the enhanced changes of fruit color and firmness, the higher levels of respiration rate and ethylene production. Meanwhile, exogenous ABA enhanced the relative levels of some primary metabolites, such as glucose, lyxose and fructose; citric acid, citramalic acid, ascorbic acid and gluconic acid; also serine, pyroglutamic acid, GABA, glutamic acid, aspartic acid, asparagine and glutamine (1.69-3.25 folds). Furthermore, exogenous ABA stimulated the expression levels of most key genes (such as SPSs, SPP2, TIV1, Lins, SUSs, PPCs, MDH and CS ) (log 2 fold change ranged from 1.06 to 3.64). Results indicate ABA is systematically involved in the regulation of primary metabolism, which may provide valuable information for exploring technical means to improve tomato taste and flavor.

Transcription factor Constans-like 4 may regulate tomato fruit ripening by interacting with abscisic acid and ethylene signaling

Qiong Wu,Xiaoya Tao,Chunxiao Cui,Yanan He,Dongdong Zhang,Yurong Zhang,Li Li

Phytohormones are vital for tomato ripening, and abscisic acid (ABA) is speculated to induce ethylene biosynthesis at the initiate of tomato fruit ripening; however, its interaction sites remain unclear. In the present study, Constans-like 4 (SlCOL4), a CO/COL transcription factor, was cloned from tomato fruit. Amino acid sequence and subcellular location analysis proved that SlCOL4 was a typical transcription factor of the COL family. Gene expression analysis showed SlCOL4 expressed in different tissues of tomato plants and its transcription abundance declined at the onset of fruit ripening, coincidentally it was found to be down-regulated in ABA-treated tomato fruit and responded to exogenous ABA within 6 h after treatment. Meanwhile, yeast one-hybrid and dual-luciferase analysis showed that the ABA response factor SlAREB1 inhibited the transcription of SlCOL4 , indicating it was directly regulated by ABA at the mature green stage of tomato fruit . In addition, the pericarp color turned red faster than that of control fruit, and the key ethylene biosynthesis genes, ACS2, ACS4, and ACO1 , were up-regulated in SlCOL4 silenced tomato fruit, suggesting that SlCOL4 may be a negative regulator of ripening. Furthermore, several genes associated with the ethylene signaling pathway were identified downstream of SlCOL4 by chromatin immunoprecipitation sequencing (ChIP-Seq) analysis. Overall, the transcription factor SlCOL4 may interact with ABA and ethylene signals and negatively regulate tomato fruit ripening. The results in this study provide new insights for elucidating the molecular mechanism of ABA-ethylene interaction in regulating tomato fruit ripening, and provide references for further research on the role of COLs transcription factors in tomato fruit ripening as well.

Quantitative proteomic analysis of tomato fruit ripening behavior in response to exogenous abscisic acid

Qiong Wu,Yanan He,Chunxiao Cui,Xiaoya Tao,Dongdong Zhang,Yurong Zhang,Tiejin Ying,Li Li

BACKGROUND To determine how abscisic acid (ABA) affects tomato fruit ripening at the protein level, mature green cherry tomato fruit were treated with ABA, nordihydroguaiaretic acid (NDGA) or sterile water (control, CK). The proteomes of treated fruit were analyzed and quantified using tandem mass tags (TMTs) at 7 days after treatment, and the gene transcription abundances of differently expressed proteins (DEPs) were validated with quantitative real-time polymerase chain reaction. RESULTS Postharvest tomato fruit underwent faster color transformation and ripening than the CK when treated with ABA. In total, 6310 proteins were identified among the CK and treatment groups, of which 5359 were quantified. Using a change threshold of 1.2 or 0.83 times, 1081 DEPs were identified. Among them, 127 were upregulated and 127 were downregulated in the ABA versus CK comparison group. According to KEGG and protein–protein interaction network analyses, the ABA-regulated DEPs were primarily concentrated in the photosynthesis system and sugar metabolism pathways, and 102 DEPs associated with phytohormones biosynthesis and signal transduction, pigment synthesis and metabolism, cell wall metabolism, photosynthesis, redox reactions, allergens and defense responses were identified in the ABA versus CK and NDGA versus CK comparison groups. CONCLUSION ABA affects tomato fruit ripening at the protein level to some extent. The results of this study provided comprehensive insights and data for further research on the regulatory mechanism of ABA in tomato fruit ripening. © 2023 Society of Chemical Industry.

Dextran as an elicitor of phenylpropanoid and flavonoid biosynthesis in tomato fruit against gray mold infection

Laifeng Lu,Lifeng Ji,Ruixi Shi,Shuhua Li,Xi Zhang,Qingbin Guo,Changlu Wang,Liping Qiao

Alpha-1,3-glucan is often synthesized on the surface of pathogenic filamentous fungi cell walls to block pathogen-associated molecular patterns (PAMPs) generation by host plant enzymes and the subsequent immune system response of the plant. Here, Botrytis cinerea susceptibility was assessed in tomato fruit to determine whether the fruit could recognize this camouflage and mount an immune response to it. The results showed that local mechanical wounds treated with dextran and laminarin, except amylopectin, could locally and then systemically activate disease resistance against B. cinerea infection in tomato fruit. Dextran treatment effectively elicited fruit callose deposition and phenylpropanoid and flavonoid biosynthesis to a greater extent than α-glucanase activity relative to the mock group surface wounds. Enzymatic hydrolysis of this polysaccharide provided some help in improving host disease resistance. Taken together, these results demonstrate that tomato fruit can perceive α-1,3-glucan as a kind of PAMPs but have limited ability to degrade it.

Oligogalacturonide-accelerated healing of mechanical wounding in tomato fruit requires calcium-dependent systemic acquired resistance

Laifeng Lu,Ying Yang,Haoran Zhang,Dandan Sun,Zhenjing Li,Qingbin Guo,Changlu Wang,Liping Qiao

Mechanical wounding causes significant economic losses of fresh produce due to accelerated senescence and spoilage as well as loss of nutritional value. Here, pre-application of oligogalacturonides (OGs) enzymatically hydrolyzed from apple pectin effectively reduced the healing times of mechanical wounds from>24 h in mock groups to 12 h, and the Botrytis cinerea infection rate was reduced from 37.5% to 12.5%. OGs accordingly increased callose deposition; SlPR1, SlPAL and SlHCT gene expression; and phenylalanine ammonia-lyase (PAL) activity around the wounds. Inhibition of Ca 2+ signaling using the inhibitor Ruthenium Red markedly inhibited OG accelerated healing of mechanical wounding on fruit. SlPG2, SlEXP1 , and SlCEL2 mRNAs accumulation was reduced in OG-elicited tomato fruit compared to water-treated fruit with subsequent retardation of the fruit softening during ripening. These results indicated that apple pectin OGs accelerate wound healing and inhibit fruit softening by activating calcium signaling in tomato fruits during postharvest storage.
