Potassium (K) fertilization is crucial for plant resistance to pathogens, but the underlying mechanisms remain unclear. Here, we investigate the molecular mechanism by which the addition of K promotes resistance inNicotiana benthamianatoPhytophthora parasitica. We found thatN. benthamianawith high K content (HK, 52.3 g/kg) produced more ethylene in response toP. parasiticainfection, compared toN. benthamianawith low-K content (LK, 22.4 g/kg). An exogenous ethylene application effectively increased resistance in LKN. benthamianato the level under HK status, demonstrating the involvement of ethylene in the HK-associated resistance inN. benthamiana. Further, transcriptome analysis showed thatNbSAMS1, encoding ethylene biosynthesis, was induced to upregulateP. parasiticaabout five times higher in HK than in LKN. benthamiana. NbSAMS1overexpression enhanced resistance in LK plants, whereasNbSAMS1silencing reduced resistance in HK plants, confirming its importance in conferring resistance. Furthermore, we identified a calcium-binding protein, NbCBP1, which interacted with NbSAMS1, promoting its expression in HKN. benthamiana.SilencingNbCBP1compromised resistance in HKN. benthamiana, whereas its overexpression improved resistance in LKN. benthamiana. Notably, NbCBP1 protected NbSAMS1 from degradation by the 26S proteasome, thereby sustaining ethylene accumulation in HKN. benthamianain response toP. parasiticainfection. Thus, our research elucidated some mechanisms of the NbCBP1-NbSAMS1 module associated with disease resistance in HKN. benthamiana.

The principal disease impacting pepper production is Phytophthora blight , which nano‑selenium (nano-Se) controls well but has high storage requirements and low heat resistance. Varying pectin amounts yielded pectin-modified nano-Se (nano-Se@PT) with 0.1 %, 0.5 %, 1 %, and 2 % concentration. As pectin content grew, nano-Se@PT particle size reduced, zeta potential value rose, and thermogravimetric temperature climbed. The incorporation of nano-Se@PT can improve its stability and thermal resistance at 40 °C and 70 °C. Nano-Se@PT outperformed nano-Se in fungicidal and Phytophthora capsici inhibition. In vitro antifungal experiments showed that nano-Se@PT could significantly inhibit mycelium growth with an EC 50 of about 4 mg/L. The particles adsorbed on the surface of mycelium, inducing death by destroying cell membranes, mitochondria, and nuclei. Non-targeted metabolomics analysis showed that nano-Se@PT treatment disrupted the mycelium growth and metabolism and changed amino acids, nucleotides, and sugar levels. In vivo experiments, 2 % nano-Se@PT had the lowest disease index and the best preventive therapy. Rhizosphere soil microbial analysis showed that nano-Se@PT recruited beneficial microorganisms such as Bacteroides, Lactobacillus, and Bifidobacterium in pepper plants, improving their Phytophthora capsici resistance and growth. The findings suggest that nano-Se@PT might be a unique pepper disease control technique and shed light on plant fungal disease management.

Interspecies interactions play a key role in soil-borne disease suppression in intercropping systems. However, there are limited data on the underlying mechanisms of soil-borne Phytophthora disease suppression. Here, a field experiment confirmed the effects of maize and soybean intercropping on Phytophthora blight of soybean caused by Phytophthora sojae. Experimentally, the roots and root exudates of maize were found to attract P. sojae zoospores and inhibit their motility and the germination of cystospores. Furthermore, five phenolic acids (p-coumaric acid, cinnamic acid, p-hydroxybenzoic acid, vanillic acid, and ferulic acid) that were consistently identified in the root exudates and rhizosphere soil of maize were found to interfere with the infection behavior of P. sojae. Among them, cinnamic acid was associated with significant chemotaxis in zoospores, and p-coumaric acid and cinnamic acid showed strong antimicrobial activity against P. sojae. However, in the rhizosphere soil of soybean, only p-hydroxybenzoic acid, low concentrations of vanillic acid, and ferulic acid were identified. Importantly, the coexistence of five phenolic acids in the maize rhizosphere compared with three phenolic acids in the soybean rhizosphere showed strong synergistic antimicrobial activity against the infection behavior of P. sojae. In summary, the types and concentrations of phenolic acids in maize and soybean rhizosphere soils were found to be crucial factors for Phytophthora disease suppression in this intercropping system.

Phytophthora capsici causes devastating losses in global pepper production. This study deciphers a sophisticated plant-microbe-metabolite collaboration initiated by rhizosphere-colonizing Streptomyces. We demonstrate that Streptomyces specifically primes pepper root biosynthesis of 6-nitrocoumarin, a nitrated coumarin derivative exhibiting potent broad-spectrum antioomycete activity (EC50: P. capsici 44.39 mg/L, Phytophthora palmivora 47.06 mg/L, Peronophythora litchii 34.92 mg/L). Crucially, this Streptomyces-recruited metabolite functions as a keystone rhizosphere signal, selectively enriching beneficial bacterial consortia while directly disrupting the ABC transporters of pathogens. The restructured microbiome synergistically amplifies plant systemic resistance through upregulating defense genes and enhancing root immunity. Our work establishes 6-nitrocoumarin as a dual-functional inoculant, a direct actinomycete compound and microbiome modulator, revealing a novel plant-microbe codefense paradigm driven by Streptomyces-induced metabolic rewiring for sustainable crop protection.

The fermentation of endophytic Nigrospora chinensis GGY-3 resulted in the isolation of tropolone stipitaldehyde (1), which exhibited broad-spectrum inhibition activity against fungi and bacteria, especially against Phytophthora capsici, with an EC50 value of 0.83 μg/mL and Xanthomonas oryzae pv. oryzicola, with a minimum inhibitory concentration value of 4.0 μg/mL. The in vitro and in vivo assays demonstrated that 1 had a significant protective effect on P. capsici. Furthermore, 1 inhibited the spore germination of P. capsici and damaged the plasma membrane structure. As observed by SEM and TEM, after exposure to 1, mycelia exhibited swelling, shrunken, branch-increasing phenomena, cell wall and membrane damage, and disordered content. Transcriptome analysis revealed that 1 might affect starch and sucrose metabolism and fatty acid biosynthesis by suppressing the expression of genes relevant to cell wall synthetases and cell membrane-associated genes. These findings indicate that 1 may be a potential agrochemical fungicide for controlling phytophthora blight.
