Nanocarriers boost non-systemic fluazinam transportation in plants and microbial community enrichment in soil

Xiao Wenhui,Du Jiang,Zheng Yuxin,Chen Shanshan,Liu Zhanyun,Chen Fengping,Liu Feng,Li Beixing,Liu Xili,Zhang Can

Utilizing nanotechnology for pesticide delivery can enhance their uptake and distribution in plants, thereby boosting pesticide efficiency. Fluazinam, a highly effective broad-spectrum fungicide with unique mode of action, is hindered by its poor systemic conductivity, limiting its field control efficacy. In this study, fluazinam-loaded nanocapsules were developed, which exhibited significantly higher inhibitory activity against various Phytophthora species compared to the commercial suspension concentrate. Notably, the nanocapsules demonstrated pronounced effects on reducing ATP content in Phytophthora capsici and effectively controlled pepper blight and cucumber downy mildew. Tracking the distribution of fluorescein isothiocyanate (FITC) fluorescein-labeled fluazinam nanocapsules revealed their presence in roots seven days after treatment. High-performance liquid chromatography (HPLC) analysis confirmed the easy absorption and transport of fluazinam nanocapsules by pepper plants. Moreover, treatment with fluazinam nanocapsules significantly increased the expression of two exocytosis genes, FACP and RABF2a , in pepper tissues. Lastly, fluazinam nanocapsules were found to enhance the relative abundance of beneficial bacteria in the soil microbial community. This study presents a promising approach to enhance the upward transport capability of non-systemic pesticides, offering novel insights into their application and improved efficacy.

Plant secondary metabolite citral interferes with Phytophthora capsici virulence by manipulating the expression of effector genes

Wen Song,Zhiyuan Yin,Xinyu Lu,Danyu Shen,Daolong Dou

Phytophthora capsici is a notorious pathogen that infects various economically important plants and causes serious threats to agriculture worldwide. Plants deploy a variety of plant secondary metabolites to fend off pathogen attacks, but the molecular mechanisms are largely unknown. In this study, we screened 11 plant secondary metabolites to evaluate their biofumigation effects against P. capsici , and found that citral, carvacrol, and trans -2-decenal exhibited strong antimicrobial effects. Intriguingly, a low concentration of citral was effective in restricting P. capsici infection in Nicotiana benthamiana , but it was unable to inhibit the mycelial growth. A high concentration of citral affected the mycelial growth and morphology, zoospore germination, and cell membrane permeability of P. capsici. Further investigations showed that citral did not induce expression of tested plant immunity-related genes and reactive oxygen species (ROS) production, suggesting that a low concentration of citral could not trigger plant immunity. Moreover, RNA-Seq analysis showed that citral treatment regulated the expression of some P. capsici effector genes such as RxLR genes and P. cactorum - fragaria (PCF)/small cysteine-rich (SCR)74-like genes during the infection process, which was also verified by reverse transcription-quantitative PCR assay. Five candidate effector genes suppressed by citral significantly facilitated P. capsici infection in N. benthamiana or inhibited ROS triggered by flg22, suggesting that they were virulence factors of P. capsici . Together, our results revealed that plant-derived citral exhibited excellent inhibitory efficacy against P. capsici by suppressing vegetative growth and manipulating expression of effector genes, which provides a promising application of citral for controlling Phytophthora blight.

Identification of volatile organic compounds for the biocontrol of postharvest litchi fruit pathogen Peronophythora litchii

Li Zheng,Jun-jian Situ,Qing-feng Zhu,Ping-gen Xi,Yin Zheng,Hong-xia Liu,Xiaofan Zhou,Zi-de Jiang

Litchi is an important economic fruit in subtropical countries. The litchi downy blight (LDB) caused by the oomycete Peronophythora litchii severely affects the production and quality of litchi fruit, and is widespread in almost all litchi production regions of China. Therefore, there is an urgent need for effective and sustainable control strategies against LDB. Our previous study showed that Bacillus amyloliquefaciens LI24 and PP19, B. licheniformis HS10, B. pumilus PI26, and Exiguobacterium acetylicum SI17 are promising biocontrol agents (BCAs) in controlling LDB, and their volatile organic compounds (VOCs) could inhibit the growth of P. litchii in vitro . In this study, we found that pre-exposure of litchi fruit to VOCs produced by PP19, SI17 and PI26 can significantly reduce the severity of LDB during 36 h to 72 h post inoculation. We further analyzed VOCs produced from the three BCAs ( i.e. , PP19, SI17, PI26) by solid phase microextraction gas chromatography-mass spectrometry (SPME-GC–MS), and found that their chemical compositions varied substantially over incubation time and between BCAs. In total, 70, 98, 101 chemicals were detected in PP19, SI17, PI26 from 24 to 72 h of incubation, respectively; 17 of them were commonly produced at more than one time points by PP19, and 11 were selected for further study. Two of the compounds 1-(2-Aminophenyl)ethanone (EA) and Benzothiazole (BTH) showed inhibitory activity against both P. litchii on plates and LDB on litchi fruit when the compounds were directly applied, while another compound α-Farnesene (AF) was able to suppress LDB in vivo , but did not exhibit antagonistic activity against the pathogen in vitro , suggesting that it may act through induction of host defense mechanisms. Our results showed that the bacterial VOCs and compounds of BTH or AF could be promising for the control of LDB on harvested litchi fruit.

Antifungal activity of zedoary turmeric oil against Phytophthora capsici through damaging cell membrane

Bi Wang,Fei Liu,Qi Li,Shu Xu,Xingzeng Zhao,Peilin Xue,Xu Feng

Phytophthora capsici is a plant oomycete pathogen, which causes many devastating diseases on a broad range of hosts. Zedoary turmeric oil (ZTO) is a kind of natural plant essential oil that has been widely used in pharmaceutical applications . However, the antifungal activity of ZTO against phytopathogens remains unknown. In this study, we found ZTO could inhibit P. capsici growth and development in vitro and in detached cucumber and Nicotiana benthamiana leaves. Besides, ZTO treatment resulted in severe damage to the cell membrane of P. capsici , leading to the leakage of intracellular contents. ZTO also induced a significant increase in relative conductivity, malondialdehyde concentration and glycerol content. Furthermore, we identified 50 volatile organic compounds from ZTO, and uncovered Curcumol, β-elemene, curdione and curcumenol with strong inhibitory activities against mycelial growth of P. capsici . Overall, our results not only shed new light on the antifungal mechanism of ZTO, but also imply a promising alternative for the control of phytophthora blight caused by P. capsici .

Functionalized Carbon Dot-Delivered RNA Nano Fungicides as Superior Tools to Control Phytophthora Pathogens through Plant RdRP1 Mediated Spray-Induced Gene Silencing

Zhiwen Wang,Yu Li,Borui Zhang,Xiang Gao,Mengru Shi,Sicong Zhang,Shan Zhong,Yang Zheng,Xili Liu

Spray-induced gene silencing (SIGS) represents an attractive avenue for plant protection, but limited uptake efficiency of double-stranded RNA (dsRNA) has restricted its application against the notorious oomycetes, Phytophthora . To control Phytophthora , dsRNAs targeting two essential genes in Phytophthora are screened for their ability to partially decrease Phytophthora capsici ( P. capsici ) infection and fecundity. To further facilitate SIGS for Phytophthora control, functionalized carbon dots (CDs) are complexed with the screened dsRNAs (dsRNA-CDs) through electrostatic interaction. dsRNA-CDs significantly enhanced the control efficacy of dsRNA against Phytophthora infestans , Phytophthora sojae , and both the wild-type and fungicide-resistant P. capsici . The synergism is based on enhancing dsRNA stability and internalization. Dual treatment with dsRNA-CDs and the corresponding fungicide reduced the amount of fungicide required to achieve the same protective effect by 90%. Plant RdRP1 is the main effector in processing various lengths of small RNAs to induce translation inhibition in Phytophthora . Notably, here the first application of a nano-delivery system to improve the effect of SIGS against Phytophthora pathogens is reported. Moreover, the elucidation of how CD facilitates dsRNA internalization in recipient cell and the molecular mechanism of SIGS can benefit the application of dsRNA-CDs in other plant-pathogen pathosystems through improving dsRNA bioactivity and reducing chemical fungicides application.

Fungicidal activity of curcumol against Phytophthora capsici via inhibiting phosphatidylcholine biosynthesis and its systemic translocation in plants

Jingjing Yang,Hai Zhu,Wenhao Liang,Xingzeng Zhao,Min Yin,Xu Feng,Bi Wang

Phytophthora capsici is an infamously soil-borne pathogen that poses a serious threat to agricultural production. Curcumol is a natural plant-derived sesquiterpene lactone, whose antimicrobial effect against plant pathogens remains unclear. In this study, curcumol exhibited pronounced antifungal activity against a diverse range of plant pathogens, particularly against plant pathogenic oomycetes, which including P. capsici , Phytophthora infestans , Phytophthora parasitica , and Phytophthora sojae . The median effective concentration values of curcumol against P. capsici for spore germination and mycelial growth were 4.75 and 2.11 μg mL −1 , respectively. After treatment with curcumol, mycelia of P. capsici exhibited morphological and ultrastructual defects, which included swelling, hyperbranching, dissolution of plasma membrane, and loss of organelles. In addition, curcumol effectively inhibited the synthesis of phosphatidylcholine (PC), a primary component of cell membrane, by downregulating the expression levels of genes participated in PC synthesis such as Phospholipid N-methyltransferase and Cholinephosphotransferase . This inhibition decreased the accumulation of PC and phospholipids within the cell, thereby increasing the cell membrane permeability and damaging its integrity. In the in vivo antifungal tests, curcumol reduced the disease incidence of P. capsici on tomato leaves as well as pepper seedlings. The systemicity tests further validated the strong phloem and xylem mobility of curcumol in both upward and downward directions. Taken together, these results indicated that curcumol could effectively combat diseases caused by P. capsici and had the potential for development into a novel fungicide for P. capsici management.

Biocontrol Mechanisms of Three Plant Essential Oils Against Phytophthora infestans Causing Potato Late Blight

Yongqiang Tian,Jianglai Wang,Qingqing Lan,Yang Liu,Jinfeng Zhang,Lu Liu,Xu Su,Rehmat Islam

Late blight, caused by the notorious pathogen Phytophthora infestans , poses a significant threat to potato ( Solanum tuberosum ) crops worldwide, impacting their quality as well as yield. Here, we aimed to investigate the potential use of cinnamaldehyde, carvacrol, and eugenol as control agents against P. infestans and to elucidate their underlying mechanisms of action. To determine the pathogen-inhibiting concentrations of these three plant essential oils (PEOs), a comprehensive evaluation of their effects using gradient dilution, mycelial growth rate, and spore germination methods was carried out. Cinnamaldehyde, carvacrol, and eugenol were capable of significantly inhibiting P. infestans by hindering its mycelial radial growth, zoospore release, and sporangium germination; the median effective inhibitory concentration of the three PEOs was 23.87, 8.66, and 89.65 μl/liter, respectively. Scanning electron microscopy revealed that PEOs caused the irreversible deformation of P. infestans , resulting in hyphal shrinkage, distortion, and breakage. Moreover, propidium iodide staining and extracellular conductivity measurements demonstrated that all three PEOs significantly impaired the integrity and permeability of the pathogen's cell membrane in a time- and dose-dependent manner. In vivo experiments confirmed the dose-dependent efficacy of PEOs in reducing the lesion diameter of potato late blight. Altogether, these findings provide valuable insight into the antifungal mechanisms of PEOs vis-à-vis late blight-causing P. infestans . By utilizing the inherent capabilities of these natural compounds, we could effectively limit the harmful impacts of late blight on potato crops, thereby enhancing agricultural practices and ensuring the resilience of global potato food production.
