Exposure to pathogens induces hyperactivate inflammatory immune responses, oxidative stress and even death in fish. Therefore, suppressing excessive inflammatory responses is a means of treating fish diseases. Scutellariae Radix (SR), an herbal medicine, has anti-inflammatory and antioxidant properties. However, the active ingredients of SR that produce a marked effect and their mechanisms are poorly understood. In this study, 32 active ingredients of SR and their 124 target genes associated with anti-inflammatory effect were screened. These target genes in zebrafish were mainly enriched in biological processes associated with response to stress and stimulus. The KEGG enrichment analysis and pathway–pathway network analysis showed that the MAPK signalling pathway and nine other pathways were the key pathways associated with anti-inflammatory effect. A protein–protein interactions (PPI) network of target genes was constructed, and AKT2 and nine other genes were identified as the hub genes by cytoHubba. Combining the above results, we constructed an active ingredient–hub protein–key pathway network, showing that wogonin-AKT2 was the key regulated module. Molecular docking and SPR-based assays were used to investigate the possible interaction of wogonin with the AKT2 protein. The infected zebrafish and grass carp treated with wogonin displayed a higher survival rate and anti-inflammatory and antioxidant effects than the infected fish without wogonin treatment. Dietary wogonin also enhanced grass carp anti-inflammatory and antioxidant effects. These findings further reveal the molecular biological mechanism by which wogonin from SR exerts anti-inflammatory effects and provide a theoretical basis for the prevention of fish diseases.

A low-molecular-weight molecule (4-(2-(3-(dicyanomethyl)-5,5-dimethylcyclohex-1-en-1-yl)vinyl)phenyl-benzoate, DDPB) has been developed. The organic framework possesses very weak fluorescence . The feasibility of the signal transduction has been performed via fluorometric titrations in solution. DDPB gives rise to responses to carboxylesterase 2 (CES2) based on “off-on” responses. The red emission at 670 nm has been derived from the enzyme-induced hydrolysis of ester linkages, thus suppressing the intramolecular charge transfer (ICT) effect and thereby generating the fluorescent segment. The optical excitation window for this probe is extended to the visible light range ( λ ex = 516 nm), and it will induce less harmful influence on biological substances. The detection limit for the measurement of CES2 concentration is as low as 2.33 mU/mL. The conventional studies concerning the activation process are generally performed within only a single liveing cell system. In this study, it is the first time that expression of carboxylesterase 2 in five kinds of cell lines (HeLa > C1498 > active T cell > Jurkat > unactive T cell) has been clarified by flow cytometry, Western blotting, and confocal microscopy analysis. The elucidation of CES2 and its variability in a variety of cells will open new ways for drug metabolism and disease prevention.
We reported a new “substrate-mediated light-on” strategy based on an ester bond cleavage reaction. Most of prepared nanomaterials and organic fluorophores possessed short wavelength emissions in the blue or green region which will not be difficult for cellular imaging. In this study, a novel functional molecule (DDPB) was considered as the substrate for CES2 and the optical “off-on” response was realized. DDPB was cell permeable and possessed very low cytotoxicity. Moreover, the identification of CES2 and their subtle changes in five different cells afforded the sequence for carboxylesterase-2 as Hela > C1498 > Active T cell > Jurkat > Unactive T cell. Inhibition studies showed that the hydrolysis of DDPB was effectively suppressed by bis-p-nitrophenyl phosphate and the cellular tracking results firmly supported this point. To our knowledge, the inter-individual variability for the CES2 expressions in five different cell lines has never been reported via the substrate induced optical changes.

Relying on the specific coordination of Ag + and mismatched cytosine-cytosine (C–C), the high-efficiency inhibition of urease by Ag + ion, and the rapid and sensitive response of phenol red to pH, a sensitive ratiometric sensor has been designed for visual detection of human immunodeficiency virus gene (HIV DNA). This sensor utilizes the HIV DNA to initiate catalytic hairpin assembly (CHA) process, releasing Ag + to inhibit subsequent urease-catalyzed urea hydrolysis and prevent the pH of the solution from rising. The CHA process and the absorbance ratio of phenol red at different wavelengths (A 559 /A 432 ) amplify the signal, allowing the sensor to detect HIV DNA from 10 to 130 nM in a sensitive and highly selective manner with a low detection limit of 7.8 nM. In addition, this sensor can visually distinguish different concentrations of HIV DNA within a certain range and possesses a good recovery in 1% of serum samples, which will provide new ideas for biosensor design, dipstick test, blood test, and other clinical disease prevention.

Wearable, noninvasive sensors enable the continuous monitoring of metabolites in sweat and provide clinical information related to an individual’s health and disease states. Uric acid (UA) is a key indicator highly associated with gout, hyperuricaemia, hypertension, kidney disease, and Lesch–Nyhan syndrome. However, the detection of UA levels typically relies on invasive blood tests. Therefore, developing a wearable device for noninvasive monitoring of UA concentrations in sweat could facilitate real-time personalized disease prevention. Here, we introduce 1,3,6,8-pyrene tetrasulfonic acid sodium salt (PyTS) as a bifunctional molecule functionalized with Ti3C2Tx via π–π conjugation to design nonenzymatic wearable sensors for sensitive and selective detection of UA concentration in human sweat. PyTS@Ti3C2Tx provides many oxidation–reduction active groups to enhance the electrocatalytic ability of the UA oxidation reaction. The PyTS@Ti3C2Tx-based electrochemical sensor demonstrates highly sensitive detection of UA in the concentration range of 5 μM–100 μM, exhibiting a lower detection limit of 0.48 μM compared to the uricase-based sensor (0.84 μM). In volunteers, the PyTS@Ti3C2Tx-based wearable sensor is integrated with flexible microfluidic sweat sampling and wireless electronics to enable real-time monitoring of UA levels during aerobic exercise. Simultaneously, it allows for comparison of blood UA levels via a commercial UA analyzer. Herein, this study provides a promising electrocatalyst strategy for nonenzymatic electrochemical UA sensor, enabling noninvasive real-time monitoring of UA levels in human sweat and personalized disease prevention.

Field effect transistor (FET) has attracted high attention in biomolecules detection. However, the sensitivity of FET-based biosensors is often limited by the charge screening effect. In addition, the facile and ultra-sensitive detection for small biomolecules, which possess weak charge or electroneutral, remains to be studied. Here, the self-assembled monolayer AuNPs/three-dimensional (3D) crumpled graphene FET is structured for a biosensor by shrinking flexible polystyrene (PS) films through heat treatment method and realized label-free and ultra-sensitive detection of dopamine (DA) using DA aptamer as probes immobilized on the AuNPs/3D crumpled graphene surface. The nanoscale deformation caused by thermal expansion effect of flexible graphene/PS can effectively reduce charge screening and the synergistic application of crumpled graphene and AuNPs can promote electron transfer and the graphene carrier concentration, leading to conductivity increase and hydrophilicity enhancement of 3D graphene. In addition, the binding of DA aptamer to DA will cause conformational changes of the aptamer molecule, which affects the charge transport properties of the sensor, thus improving its selectivity and stability. The biosensor can also easily distinguish interfering substances for DA detection in complex components (PBS, human urine, and fetal calf serum) with the detection limits as low as 60, 240 and 316 zM (10 −19 –10 −11 M), respectively. DA released from exocytosis induced by K + stimulation was selectively detected. The results show that the biosensor can be used as an excellent tool for ultrasensitive molecular recognition and detecting the effects of exogenous reagents on living cells, which is promising for clinical diagnosis and early disease prevention.
