Isolation, identification, and pathogenic effects of Trichoderma spp. from Auricularia auricula

Dang Hui,Kong Qianqian,Winchester William,Wan Xin,Lei Yu,Zhang Haisheng,Zhao Yu,Liu Xinyu,Xu Ben Bin,Zhang Baoshan,Wang Zhe

Auricularia auricula , one of the most important edible mushrooms, is affected heavily by Trichoderma . We collected the diseased samples from the main A. auricula cultivation regions to characterize the pathogen and study the effect of Trichoderma spp. on A. auricula species. We identified one Trichoderma species, T. pleuroticola , based on the internal transcribed spacer and morphology characteristics, and two types of A. auricula strains, Heiwei 15 (HW 15) and Hei 29 (H 29), were tested in this work. The growth rate of T. pleuroticola was 3.26–3.52 times higher than that of A. auricula and advantageously competed for living space and nutrients. In confrontation culture, T. pleuroticola completely inhibited the mycelium growth of A. auricula and grew on it, resulting in a diverse impact on HW 15 and H 29. In addition, T. pleuroticola can produce metabolites with antibacterial activity. The inhibition rate of volatile metabolites to H-29 and HW 15 was 13.46% and 10.44%, and the inhibition rate of nonvolatile metabolites to H-29 and HW 15 was 36.04% and 31.49%, respectively. Further analysis showed that these antifungal activities inhibiting Auricularia auricula growth were mainly attributed to the organic compounds from T. pleuroticola , nonanal, tyrosine, beta-sitosterol, and wortmannin. In short, T. pleuroticola was a highly pathogenic fungi in the production of A. auricula . Graphical Abstract T. pleuroticola can affect the normal growth of A. auricula hypha, and its metabolites also have inhibitory effects on A. auricula hypha.

Identification of the ADH gene family in Trichosporon asahii and the role of TaADH_like in pathogenicity and fluconazole resistance

Liu Zhen,Ma Xiaoping,Zeng Xiangwen,Li Zhiguo,Liu Ruiguo,Luo Rongyan,Wang Weichen,Tahir Muhammad Salman,Wang Chengdong,Gu Yu

Alcohol dehydrogenase has been studied in regulation of fungal growth and development, stress response and pathogenesis, but its function in T. asahii remains unexplored. In this study, we analyzed the ADH gene family in T. asahii for the first time, identifying six ADH genes and containing conserved ADH_N and ADH_Zinc_N domains. We constructed an overexpression strain of the most significantly differentially expressed gene TaADH_like and compared its phenotypes with those of the wild-type strain, focusing on colony morphology, biofilm biomass, stress response, drug resistance, and pathogenicity. The results showed that TaADH_like overexpression reduced sensitivity to hypoxic conditions, altered the hyphae-to-yeast transition, and led to slower growth, decreased colonization ability, reduced tissue damage, and lower lethality. Increased osmotic stress sensitivity and the involvement of the HOG MAPK pathway in the hyphae-to-yeast conversion contributed to the reduced colonization capacity of T. asahii . Furthermore, the overexpression of TaADH_like promoted biofilm formation and led to a slight enhancement in fluconazole resistance in T. asahii . This study is the first to elucidate the function of the alcohol dehydrogenase gene in T. asahii , providing a foundation for future genetic research on this pathogen.

Influence of Metal Oxides Nanoparticles on Pathogenicity of Steinernema carpocapsae Nematodes Against Lepidopteran Galleria mellonella

Makirita Winisia E.,Zhang Fengqin,Mbega Ernest R.,He Nongyue,Li Xiaolong,Chacha Musa,Liu Tonghua

Entomopathogenic nematodes (EPNs) have been cited as a safe and effective method for pest management. Their virulence against lepidopterans and other foliar pest has been demonstrated in the laboratory, but achieving field efficacy is challenged with environmental conditions such as desiccation and ultraviolet radiations. Nanoparticles (NPs) as UV-blocking agents have been reported to provide an alternative approach to enhance the EPNs efficacy. We screened the effect of ZnO, TiO2 and Fe3O4 NPs on survival and pathogenicity of EPNs, and determined whether the nanoparticles could provide protection to the EPNs at low concentrations when applied in direct sunlight. All nanoparticles had less influence on the survival of infective juveniles and did not deprive their pathogenic properties following prolonged exposure. The survival rate of nematodes decreased with increased concentrations, with no measurable difference between NPs. Moreover, the survival rate of nematodes exposed to UV radiation in different formulation decreased significantly with the increase in exposure time (p < 0.001). On the other side when EPNs in nanoparticles formulations were exposed to sunlight, their efficacy significantly improved by protecting nematodes from ultraviolet radiation when compared to the water-based formulation. Pathogenic efficacy after exposure to sunlight was in the order of ZnO > TiO2 > Fe3O4 > H2O, indicating the compatibility of nematodes and NPs, and the benefit of different NPs in EPNs formulations. General nanoparticles are novel ingredients that provide suitable protection of EPNs for management of foliar pests. However, the EPNs nanoparticles formulation under a specific agricultural system and climatic condition need to be established.

Spider-Web-Inspired SiO2/Ag nanofibrous aerogels with superelastic and conductive networks for electroporation water disinfection

Dingming Yu,Lifang Liu,Bin Ding,Jianyong Yu,Yang Si

Water-borne pathogenic diseases significantly threaten human health and the global economy, and traditional water disinfection strategies such as chlorination or membrane filtration to remove pathogens still suffer from harmful byproducts, biofouling, and limited handling capacity. Here, a bottom-up strategy is reported to develop biomimetic nanofibrous aerogels with superelastic and conductive networks by assembling Ag nanowires in a 3D SiO 2 nanofibrous skeleton via freeze-drying method for safe and efficient water disinfection. Inspired by the prey capture behavior of spiders using their webs, the core of this design is constructing spanning and interconnected networks with millions of conductive nanotips, which could inactivate microbes instantaneously based on the synergistic effect of interception and electroporation. Benefitting from the high porosity (99.81 %), superelasticity, and robust structural stability of the resultant aerogels, an electroporation-disinfection device (EDD) equipped with aerogels exhibits superior bactericidal (>99.9999 %) and virucidal (>99.9 %) performances at low derive voltage (1 V) and high flux (9900 L m −2 h −1 ) with energy consumption of 0.83 Wh m −3 . This novel disinfection method provides new insights into practical water treatment in an efficient, environmental, and sustainable way.

Exosomes derived from bladder epithelial cells infected with uropathogenic Escherichia coli increase the severity of urinary tract infections (UTIs) by impairing macrophage function

Zihao Wang,Ziming Jiang,Yu Zhang,Congwei Wang,Zhaoyang Liu,Zhankui Jia,Sudhanshu Bhushan,Jinjian Yang,Zhengguo Zhang

Uropathogenic Escherichia coli (UPEC) is the primary causative agent of urinary tract infections (UTIs) in humans. Moreover, as one of the most common bacterial pathogens, UPEC imposes a substantial burden on healthcare systems worldwide. Epithelial cells and macrophages are two major components of the innate immune system, which play critical roles in defending the bladder against UPEC invasion. Yet, the routes of communication between these cells during UTI pathogenesis are still not fully understood. In the present study, we investigated the role of membrane-bound nanovesicles (exosomes) in the communication between bladder epithelial cells and macrophages during UPEC infection, using an array of techniques such as flow cytometry, miRNA profiling, RNA sequencing, and western blotting. Moreover, our in vitro findings were validated in a mouse model of UPEC-induced cystitis. We found that UPEC infection induced the bladder epithelial MB49 cell line to secrete large numbers of exosomes (MB49-U-Exo), which were efficiently absorbed by macrophages both in vivo and in vitro. Assimilation of MB49-U-Exo induced macrophages to produce proinflammatory cytokines, including tumor necrosis factor (TNF)α. Exposure of macrophages to MB49-U-Exo reduced their phagocytic activity (by downregulating the expression of phagocytosis-related genes) and increased their rate of apoptosis. Mechanistically, we showed that MB49-U-Exo were enriched in miR-18a-5p, which induced TNFα expression in macrophages by targeting PTEN and activating the MAPK/JNK signaling pathway. Moreover, administration of the exosome secretion inhibitor GW4869 or a TNFα-neutralizing antibody alleviated UPEC-mediated tissue damage in mice with UPEC-induced cystitis by reducing the bacterial burden of the bladder and dampening the associated inflammatory response. Collectively, these findings suggest that MB49-U-Exo regulate macrophage function in a way that exacerbates UPEC-mediated tissue impairment. Thus, targeting exosomal -release or TNFα signaling during UPEC infection may represent promising non-antibiotic strategies for treating UTIs. Urinary tract infections, which are typically caused by uropathogenic Escherichia coli (UPEC), affect over 150 million people each year, worldwide. Because the incidence of antibiotic resistance among UPEC strains is on the rise, urinary tract infections are becoming increasingly challenging to treat using this standard treatment method. Moreover, although the host immune response against UPEC has been intensively studied, the mechanism of communication between bladder epithelial cells and macrophages during UPEC infection remains largely unexplored. Here, we characterized the exosomes released from bladder epithelial cells following UPEC infection and evaluated their impact on macrophages. We demonstrated that bladder tissue destruction following excessive innate immune cell activation during UPEC infection may be driven by exosomes secreted from bladder epithelial cells. We further showed that pharmacologically inhibiting exosomal release from epithelial cells limited macrophage-associated inflammation, innate immune cell infiltration, and histopathological injuries in a mouse model of UPEC-induced cystitis. Collectively, our findings suggest that modulating exosome release represents a promising non-antibiotic therapy for UTIs.
