Treatment of hospital wastewater by electron beam technology: Removal of COD, pathogenic bacteria and viruses

Jianlong Wang,Shizong Wang,Chuanhong Chen,Jun Hu,Shijun He,Yuedong Zhou,Huanzheng Zhu,Xipo Wang,Dongming Hu,Jian Lin

The effective treatment of hospital sewage is crucial to human health and eco-environment, especially during the pandemic of COVID-19. In this study, a demonstration project of actual hospital sewage using electron beam technology was established as advanced treatment process during the outbreak of COVID-19 pandemic in Hubei, China in July 2020. The results indicated that electron beam radiation could effectively remove COD, pathogenic bacteria and viruses in hospital sewage. The continuous monitoring date showed that the effluent COD concentration after electron beam treatment was stably below 30 mg/L, and the concentration of fecal Escherichia coli was below 50 MPN/L, when the absorbed dose was 4 kGy. Electron beam radiation was also an effective method for inactivating viruses. Compared to the inactivation of fecal Escherichia coli , higher absorbed dose was required for the inactivation of virus. Absorbed dose had different effect on the removal of virus. When the absorbed dose ranged from 30 to 50 kGy, Hepatitis A virus (HAV) and Astrovirus (ASV) could be completely removed by electron beam treatment. For Rotavirus (RV) and Enterovirus (EV) virus, the removal efficiency firstly increased and then decreased. The maximum removal efficiency of RV and EV was 98.90% and 88.49%, respectively. For the Norovirus (NVLII) virus, the maximum removal efficiency was 81.58%. This study firstly reported the performance of electron beam in the removal of COD, fecal Escherichia coli and virus in the actual hospital sewage, which would provide useful information for the application of electron beam technology in the treatment of hospital sewage.

Dual-signal multiplex assays based on non-tandem enzymes for simultaneous detection of multiple pathogenic bacteria

Xiang Tianxin,Wen Jin,Fang Jianhua,Nie Chuang,Cheng Na,Ying Ying,Tan Hongliang

Enzymatic assays represent a promising alternative to conventional multiplex optical methods for multi-target detection due to their exceptional specificity and catalytic versatility. Nevertheless, most existing enzymatic systems are limited to single-target detection. In this work, we developed a dual-signal multiplex assay based on non-tandem enzymes for the simultaneous detection of multiple pathogenic bacteria. This assay utilizes the distinct catalytic activities of horseradish peroxidase (HRP) and invertase (INV) to produce two orthogonal signal outputs. Specifically, HRP catalyzes the oxidation of 3,3′,5,5′-tetramethylbenzidine to produce a blue chromogenic signal measurable by absorbance, while INV hydrolyzes sucrose to yield glucose, which is quantified with a personal glucose meter. To implement this assay, HRP and INV are individually encapsulated within zeolitic imidazolate framework-90, and Fe 3 O 4 nanoparticles functionalized with phenylboronic acid are employed for dual-recognition of target pathogens. By integrating spatiotemporal control of enzymatic reactions with distinct signal separation, the assay effectively minimizes crosstalk and enhances detection accuracy and reliability. This assay demonstrates excellent sensitivity and selectivity in milk samples, highlighting its potential for practical applications. We believe this work establishes a versatile and robust platform for multiplex pathogen detection, with significant implications for food safety, clinical diagnostics, and environmental monitoring. Inhibitory effects of fucoidan from Laminaria japonica against some pathogenic bacteria and SARS-CoV-2 depend on its large molecular weight

Xiaona Sun,Chunqing Ai,Chengrong Wen,Haoran Peng,Jingfeng Yang,Yuna Cui,Shuang Song

Fucoidan is a highly sulfated polysaccharide with a wide range of bioactivities, including anti-pathogenic activity. However, the relationship between structure and activity of fucoidan in inhibiting pathogen infections remains unclear. Here, different-molecular-weight fucoidans were prepared by photocatalytic degradation followed by membrane ultrafiltration, and their chemical structures and anti-pathogenic microbiota activity were compared. Results showed that photocatalytic degradation could effectively degrade fucoidan while its structure block and sulfate groups were not destroyed obviously. Fucoidan (90.8 kDa) of 5 mg/mL could inhibit the growth of S. aureus , S. typhimurium and E. coli , but its degradation products, Dfuc1 (19.2 kDa) and Dfuc2 (5.5 kDa), demonstrated lower inhibitory effect. In addition, compared to Dfuc1 and Dfuc2, fucoidan showed stronger capability to prevent the adhesion of S. aureus , L. monocytogenes , V. parahaemolyticus and S. typhimurium to HT-29 cells. Moreover, the inhibitory effect against SARS-CoV-2 and the binding activity to S protein were also positively correlated to molecular weight. These results indicate that natural fucoidan with higher molecular weight are more effective to inhibit these pathogenic bacteria and SARS-CoV-2, providing a better understanding of the relationship between structure and activity of fucoidan against pathogenic microbiota.

Effective photocatalytic antibacterial of Bi2WO6/Bi4O5I2 driven by visible light

Jingmei Li,Luyin Zhao,Chunjiao Hu,Ling Zhang

The escalation of medication resistance in pathogenic bacteria has resulted in significant damage to public health and safety, as bacterial and fungal illnesses proliferate. Additionally, pathogenic bacteria disseminate via water resources, leading to water contamination and adversely impacting the quality of the living environment. Consequently, the creation of an efficient antibacterial material has emerged as a focal point of study. Photocatalysis, as an innovative, environmentally friendly, and effective technique, has been extensively used in degrading and antibacterial domains. For the first time, Bi 2 WO 6 /Bi 4 O 5 I 2 composites with molar ratios of 1:1, 1:2, 1:3, and 1:4 were produced using the water bath-calcination technique including bismuth nitrate pentahydrate, sodium tungstate dihydrate, and potassium iodide, and their photocatalytic antibacterial characteristics were examined. Their characterization was conducted using XRD, XPS, SEM, FT-IR, and UV techniques. The antibacterial efficacy was evaluated using a low-power 35 W LED light irradiation dilution coated plate technique. The experimental findings demonstrated that, in the presence of 35w LED lighting, the antibacterial rate against E.coli , Bacillus subtilis , and P.aeruginosa reached 100 % in 10 min at a concentration of 1000 mg/L of BWOI-3 composite material, and the antibacterial efficiency against S. aureus reached 100 % in 5 min. Within 50 min, the antibacterial efficacy against Candida albicans was 100 %. The Bi 2 WO 6 /Bi 4 O 5 I 2 compound has shown exceptional broad-spectrum antibacterial efficacy.

Isolation, Identification, and Control of Pathogenic Endophytic Fungi in Nymphaea candida Presl Tissue Culture

Yuwei Xing,Cong Liu,Xumeng Cui,Haonan Lv,Jun Wang

Nymphaea candida Presl is a rare hardy water lily at risk of extinction and has been included on the ‘Red List’ of threatened species of the International Union for the Conservation of Nature. To protect germplasm resources and propagate N. candida seedlings, this study conducted tissue culture and found that pathogenic endophytic fungal infection was the main reason for failure of tissue culture. Compared with the stems and leaves, the roots of N. candida had the highest rates of fungal infection during tissue culture. Subsequently, three isolated endophytic fungi, Fusarium oxysporum , Phytopythium helicoides , and Alternaria sp., showed the highest frequency of occurrence in tissue culture. Furthermore, an antifungal formulation comprising 0.1 μg/mL pyrimidin suspension, 1 μg/mL mancozeb wettable powder, and 1 μg/mL carbendazim was constructed and could reduce the infection rates of root and stem tissues to 7.5% and 0%, respectively. Finally, the usefulness of this antifungal formulation for inhibition of endophytic fungi in tissue culture of N. candida was validated. This study not only provides important technical support for mass production of seedlings of N. candida , but also provides a scientific reference for the protection of endangered aquatic plant species.

Ultra-efficient amplification-free biosensor based on electric field and crowding agent-promoted hybridization for the detection of pathogenic bacteria

Hao Gong,Yang Li,Yaru Liu,Yuting Shan,Yijie Wang,Cuiping Ma,Chao Shi

Pathogenic bacteria represent a significant threat to public health and the global economy due to their high infectiousness and lethality. Consequently, the rapid and accurate identification of these bacteria remains a major challenge. Electrochemical amplification-free detection methods have shown several advantages, including rapidity, simplicity and high sensitivity. However, these methods also possess certain drawbacks, such as the inefficient solid-phase hybridization of long-chain nucleic acids on the electrode surface, it is challenging to directly detect long-chain nucleic acids. In response, we have established a novel sensor platform for efficient and rapid detection of long chain nucleic acids based on the principle of HRP signal amplification. This platform incorporates macromolecular crowding agents and electric field assistance to enhance pathogenic bacteria detection efficiency in clinical samples without amplification. The detection process begins by introducing the target into a strand displacement reaction system containing a crowding agent. The crowding effect of the crowding agent is such that the long chain target rapidly replaces the biotinylated probe. Subsequently, an electric field is applied to the surface of the screen-printed electrodes for rapid hybridization, which has been modified with a capture probe. This assay is straightforward and expeditious, reducing hybridization time from 150 min to 90 s. The sensor platform showed a detection linear range of 10 2 –10 7  CFU mL −1 with the limit of detection of 10 CFU mL −1 within 25min. This platform showcases the significant potential for on-site detection of pathogenic bacteria in clinical settings and is expected to be integrated into existing diagnostic equipment.

Insights into effects of algae on decay and distribution of bacterial pathogens in recreational water: Implications for microbial risk management

Tingting Fang,Zuotao Zhang,Hui Wang,Matt Rogers,Qijia Cui

The decay and distribution of bacterial pathogens in water is an important information for the health risk assessment to guide water safety management, and suspended algae might affect bacterial pathogens in water. This study established microcosms to investigate the effects of algae-related factors on the representative indicators and opportunistic pathogen species in water. We found that suspended algae increased the persistence of targeted species by 1-2 orders of magnitude of concentrations compared to microcosms without algae; and the effect of algae on microbial survival was affected by water nutrient levels (i.e., carbon, nitrogen and phosphorus), as the increased microbial persistence were correlated to the increased algae concentrations with more nutrient supplies. Moreover, decay and distribution profiles of representative species were determined. The three opportunistic pathogen species ( Pseudomonas aeruginosa, Aeromonas hydrophila and Staphylococcus aureus ) showed lower decay rates (0.82–0.98/day, 0.76–0.98/day, 0.63–0.87/day) largely affected by algae-related factors, while the enteric species ( Escherichia coli and Enterococcus faecalis ) had higher decay rates (0.94–1.31/day, 0.89–1.21/day) with little association with algae, indicating the propensity for attachment to algae is an important parameter in microbial fate. Together results suggest suspended algae played an evident role in the decay and distribution of bacterial pathogens, providing important implications regarding microbial safety in recreational water.

Visualizing germination of microbiota endospores in the mammalian gut

Ningning Xu,Liyuan Lin,Yahui Du,Huibin Lin,Jia Song,Chaoyong Yang,Wei Wang

Transmission of bacterial endospores between the environment and people and the following germination in vivo play critical roles in both the deadly infections of some bacterial pathogens and the stabilization of the commensal microbiotas in humans. Our knowledge about the germination process of different bacteria in the mammalian gut, however, is still very limited due to the lack of suitable tools to visually monitor this process. We proposed a two-step labeling strategy that can image and quantify the endospores’ germination in the recipient’s intestines. Endospores collected from donor’s gut microbiota were first labeled with fluorescein isothiocyanate and transplanted to mice via gavage. The recipient mice were then administered with Cyanine5-tagged D-amino acid to label all the viable bacteria, including the germinated endospores, in their intestines in situ. The germinated donor endospores could be distinguished by presenting two types of fluorescent signals simultaneously. The integrative use of cell-sorting, 16S rDNA sequencing, and fluorescence in situ hybridization (FISH) staining of the two-colored bacteria unveiled the taxonomic information of the donor endospores that germinated in the recipient’s gut. Using this strategy, we investigated effects of different germinants and pre-treatment interventions on their germination, and found that germination of different commensal bacterial genera was distinctly affected by various types of germinants. This two-color labeling strategy shows its potential as a versatile tool for visually monitoring endospore germination in the hosts and screening for new interventions to improve endospore-based therapeutics.

Rapid identification of pathogenic bacteria from clinical positive blood cultures via virus-like magnetic bead enrichment and MALDI-TOF MS profiling

Zhirou Zhang,Enyun Xing,Wenzhuo Zhao,Minghui Song,Cuiping Zhang,Hong Liu,Xiaomin Li,Hongxiu Yu

Reducing the time required for the detection of bacteria in blood samples is a critical area of investigation in the field of clinical diagnosis. Positive blood culture samples often require a plate culture stage due to the interference of blood cells and proteins, which can result in significant delays before the isolation of single colonies suitable for matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis. In this study, we developed a non-specific enrichment strategy based on SiO2-encapsulated Fe3O4 nanoparticles combined with MALDI-TOF MS for direct identification of bacteria from aqueous environments or positive blood culture samples. Three distinct types of Fe3O4@SiO2 magnetic nanoparticles (MNPs) with unique surface morphologies were developed: spherical MNPs with smooth surfaces (Fe3O4@SN), mesoporous silica coated MNPs (Fe3O4@MSN), and MNPs exhibiting a viral spiked structure (Fe3O4@VSN). These MNPs exhibited excellent binding affinity towards both Staphylococcus aureus and Klebsiella pneumoniae in PBS and artificial saliva solutions. Furthermore, the strategy of using Fe3O4@VSN, which involves non-specific interactions between bacterial cells and the virus-like surface, resulted in a dramatic reduction in the minimum detectable concentrations of target pathogens by up to 1000-fold compared to conventional methods. Our results demonstrate that the use of Fe3O4@VSN has the potential to significantly reduce the processing time required after blood culture and may be useful for enrichment and identification of microorganisms in complex clinical samples.

An ultrasensitive fluorescence detection template of pathogenic bacteria based on dual catalytic hairpin DNA Walker@Gold nanoparticles enzyme-free amplification

Yan Zhou,Zefeng Wang,Shengnan Zhang,Le Deng

Integrating the advantages of catalytic hairpin components and multi-foot DNA walker, we designed a 16S rRNA detection probe template for pathogen bacteria, which utilizes DNA ligation quencher and dual catalytic [email protected] walker to induce signal recovery. The dual catalytic [email protected] walker uses the walking position of the target on the AuNP as a foothold to promote the reaction, so that the biosensing of the low-abundance target sequence can induce signal recovery. During the entire experiment, no enzyme is required, which can avoid the limitation of enzyme degradation under unfavorable conditions and the inability to detect the target. Most importantly, the detection template has the advantages of high sensitivity, and its detection limit is significantly better than that of single hairpin DNA walker probe. As the detection system can sensitively and rapidly detect its targeted bacteria and not rely on any enzyme and sophisticated instrumentation, it has great potential for sensitive and specific pathogenic bacteria detection.

Antibacterial Properties of Silver Nanoparticles Synthesized Using Piper betle L. Leaf Extract

Tien Hieu Vu,Van Huan Bui,Ngoc Thang Nguyen

The green synthesis of silver nanoparticles (AgNPs) using herbal plants has gained much attention due to their potential widespread applications, especially in biomedical application to control pathogenic microbes. The aim of our study was to evaluate the antibacterial properties of synthesized AgNPs using aqueous leaf extract of Piper betle L., an important medicinal plant. The AgNPs were identified by UV-Visible spectrometry (UV-Vis), X-ray diffraction (XRD), transmission electron microscopy (TEM) and Fourier transform infrared spectrometry (FT-IR). The presence of surface plasmon band around 420-460 nm indicated AgNPs formation. Spherical nature, unique size-distribution and crystal structure of the AgNPs with diameter around 10-20 nm were affirmed by TEM and XRD analyses. The FTIR measurements showed the presence of bioactive compounds in the extract responsible for the efficient reduction of silver ions and stabilization of the AgNPs. The results from the antimicrobial assays suggested that the biosynthesized AgNPs were potent against pathogenic bacteria including Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus.

Detecting Bacteria with Ultralow Concentrations by Enzymatic Cascade Reaction–Amplifying Strategy

Qingzhi Peng,Huakun Wan,Zhenguo Yu,Shiyao Li,Hui Huang,Li Zhang,Yinli Guo,Dong Wang,Zhentan Lu

Bacteria can cause infectious diseases even at ultralow concentrations (<1 CFU/mL). It is important to rapidly identify bacterial contamination at ultralow concentrations. Herein, FITC-labeled gelatinase-sensitive nanoparticles (GNPs@FITCs) and NFM@GNP@FITCs are designed and fabricated as ultralow concentration bacteria detection platforms based on an enzymatic cascade reaction–amplifying strategy. Bacterial secretions could trigger the dissociation of GNPs@FITCs to release FITC, with gelatinase used as the model secretion. The detectable signal of ultralow concentration bacteria could be amplified effectively by the gelatinase-triggered cascade reaction. Bacterial concentration was evaluated by the change in fluorescence density. The results showed that the GNPs@FITCs and NFM@GNP@FITCs could be used for identifying bacterial contamination qualitatively, even when the bacterial contamination is lower than 1 CFU/mL. Moreover, the method has better timeliness and convenience, when compared with national standards. As solid films, NFM@GNP@FITCs have better long-term storage stability than GNPs@FITCs. The potential applications of GNPs@FITC and NFM@GNP@FITCs were proved by detecting pathogenic bacteria in food. All the results showed that the method has great potential for screening pathogenic bacterial contamination qualitatively.
