Colorimetric method for PARP-1 detection based on preventing AuNRs from etching by molybdate

Yong Liu,Ensheng Xu,Chao Xu,Wei Wei

As a tumor-related enzyme, PARP-1 activity is closely related to the occurrence of cancer. Sensitive detection of PARP-1 activity is of significance in the early diagnosis and treatment of cancer. Therefore, the gold nanorods (AuNRs) based biological nanoprobe was constructed for the visual and label-free detection of PARP-1 activity. dsDNA which activate PARP-1 was modified on AuNPs ( [email protected] ). MoO 4 2− is added to catalyze H 2 O 2 and KI to accelerate the etching of AuNRs. In the presence of PARP-1, hyperbranched poly (ADP-ribose) polymers (PAR) formed on the surface of [email protected] ( [email protected] @PAR). A large number of PO 4 3- of the PAR reacted with MoO 4 2− to generate PMo 12 O 40 3- . Thus, PO 4 3- diminished MoO 4 2− concentration in solution, preventing the etching of AuNRs. Therefore, PARP-1 prevented the etching of AuNRs. With the increasing concentration of PARP-1, the color of the solution changed from light blue to brown obviously. The method was applied to assay actual samples with satisfactory results. The strategy has the advantages of rapid quantitative analysis of PARP-1 activity with naked eyes, simple operation and good application prospect.

A PNA-DNA2 Triple-Helix Molecular Switch-Based Colorimetric Sensor for Sensitive and Specific Detection of microRNAs from Cancer Cells

Mengjia Xu,Pan Fu,Shu Xing,Yang Zhao,Chao Zhao

Graphical Point-of-care cancer catching ? A novel PNA-DNA 2 triplex consisting of a homopurine PNA strand and two homopyrimidine DNA strands is reported for the first time. On the basis of this phenomenon, a simple, inexpensive colorimetric PNA-DNA 2 triple-helix molecular switch-based sensing platform for sensitive and selective detection of miRNAs from cancer cells is demonstrated. Peptide nucleic acids (PNAs), the synthetic DNA mimics that can bind to oligonucleotides to form duplexes, triplexes, and quadruplexes, could be advantageous as probes for nucleic acid sequences owing to their unique physicochemical and biochemical properties. We have found that a homopurine PNA strand could bind to two homopyrimidine DNA strands to form a PNA-DNA 2 triplex. Moreover, the cyanine dye DiSC 2 (5) could bind with high affinity to this triplex and cause a noticeable color change. On the basis of this phenomenon, we have designed a label-free colorimetric sensing platform for miRNAs from cancer cells by using a PNA-DNA 2 triple-helix molecular switch (THMS) and DiSC 2 (5). This sensing platform can detect miRNA-21 specifically with a detection limit of 0.18 nM, which is comparable to that of the THMS-mediated fluorescence sensing platform. Moreover, this colorimetric platform does not involve any chemical modification or enzymatic signal amplification, which boosts its applicability and availability at the point of care in resource-limited settings. The universality of this approach can be simply achieved by altering the sequences of the probe DNA for specific targets.

Label-free molecular probe based on G-quadruplex and strand displacement for sensitive and selective detection and naked eye discrimination of exon 2 deletion of AIMP2

Ruimin Zang,Xin Wang,Yanyan Zhu,Tianming Yao,Shuo Shi

Exon 2 deletion of aminoacyl tRNA synthetase complex-interacting multifunctional protein 2 (AIMP2) is a genetic deletion related to various cancers, for instance ovarian and lung cancers. It can be worked as an indicator of cancer for diagnosis of diseases. Here, we developed a label-free method based on the formation of split G-quadruplex in the presence of target DNA combined with strand displacement to detect exon 2 deletion of AIMP2 (DE2) sensitively and selectively. This method is easy-operating and cost-saving. Moreover, it has observed discrimination of gene deletion from wild-types by naked eyes. The results demonstrate that this strategy can be further used for the detection of different gene deletions to achieve early diagnosis of diseases and allow better prognosis.

PAMAM-Based DNA Fluorescence Nanoprobe for Rapid Whole Cellular APE1 Detection and Imaging

Huimin Li,Xinru Yuan,Mei Ren,Jiajia Liu,Yixin Zheng,Ziyi Lin,Zixin Chen,Zhao Yang,Xin Su,Heyun Shen

Apurinic/apyrimidinic endonuclease 1/redox effector factor 1 (APE1/ref-1, APE1), a vital protein for DNA repair and cellular redox regulation, is frequently overexpressed in tumor cells, underscoring the importance of developing sensitive detection methods for early cancer diagnosis. However, the rapid detection and visualization of nuclear APE1 in tumor cells are still challenging. In this study, we successfully developed a novel DNA fluorescent nanoprobe based on polyamidoamine (PAMAM) for the rapid detection of cytoplasmic and nuclear APE1. The PAMAM surface was modified with arginine (Arg), named PR, and its hydrophobic core encapsulated the 1,6,7,12-tetrachloroperylene tetracarboxylic acid dianhydride (TA) dye to construct fluorescent nanoparticles (TPR). Furthermore, an APE1-responsive dsDNA (SP) was linked on the TPR surface, containing apurinic/apyrimidinic sites (AP sites) and the black hole quencher 2 (BHQ2) and ensuring that fluorescence remains off in the absence of APE1. TPR-SP exhibited a detection range of 0.125–25 U mL–1 and a detection limit as low as 0.03 U mL–1. Compared with the Arg-free nanoprobes (TP-SP), TPR-SP significantly accelerated endocytosis and nuclear penetration, reducing the APE1 detection time to one-quarter (from 2 to 0.5 h). Notably, the APE1 signal in the whole nucleus can also be significantly detected. Thus, the TPR nanoprobe achieves the rapid enrichment and amplification of fluorescence signals, leading to highly sensitive whole cellular APE1 detection. This innovative and efficient detection method greatly expands the technological means for early cancer detection.

IR780-based diffuse fluorescence tomography for cancer detection

Zhuanxia He,Limin Zhang,Lingxiu Xing,Wenjing Sun,Xiujun Gao,Yanqi Zhang,Feng Gao

IR780 iodide is a commercially available targeted near-infrared contrast agent for in vivo imaging and cancer photodynamic or photothermal therapy, whereas the accumulation, dynamics, and retention of IR780 in biological tissue, especially in tumor is still under-explored. Diffuse fluorescence tomography (DFT) can be used for localization and quantification of the three-dimensional distribution of NIR fluorophores. Herein, a homemade DFT imaging system combined with tumor-targeted IR780 was utilized for cancer imaging and pharmacokinetic evaluation. The aim of this study is to comprehensively assess the biochemical and pharmacokinetic characteristics of IR780 with the aid of DFT imaging. The optimal IR780 concentration (20 μg/mL) was achieved first. Subsequently, the good biocompatibility and cellar uptake of IR780 was demonstrated through the mouse acute toxic test and cell assay. In vivo, DFT imaging effectively identified various subcutaneous tumors and revealed the long-term retention of IR780 in tumors and rapid metabolism in the liver. Ex vivo imaging indicated IR780 was mainly concentrated in tumor and lung with significantly different from the distribution in other organs. DFT imaging allowed sensitive tumor detection and pharmacokinetic rates analysis. Simultaneously, the kinetics of IR780 in tumors and liver provided more valuable information for application and development of IR780.

MoS2 quantum dots featured fluorescent biosensor for multiple detection of cancer

Yuhong Liu,Jinzha Zhang,Yang Shen,Jinduo Yan,Zaiying Hou,Chun Mao,Wenbo Zhao

Transition metal ions, such as those generated through MoS2 material, possess an intrinsic fluorescence quenching property towards organic dye molecules; thus, they can be used to construct biosensors as quenchers. However, we found that the conventional bulk MoS2 blocks the view of fluorescence imaging, and is incapable of tracing and visualizing mucin 1-overexpression cancer cells. Herein, a FAM fluorophore-labeled ssDNA fluorescent probe (P0-FAM) stacked on the surface of MoS2 quantum dots (QDs) was used to construct a MoS2 QDs–P0-FAM biosensor. MoS2 QDs exhibit a high fluorescence quenching ability towards fluorescent dyes, possess large specific surface area and a large number of active sites to adsorb and quench more fluorescent probes, promoting sensitivity between quenching and the recovery signal. In addition, the lighter color of unstack-MoS2 QDs is beneficial to define the location of cancer cells compared to MoS2 nanosheets. The novel MoS2 QDs-based biosensor demonstrates high sensitivity to MUC1 with a detection limit of 0.5 nM, and may become an important tool toward the detection of cancer cells.

NIR II Light-Response Au Nanoframes: Amplification of a Pressure- and Temperature-Sensing Strategy for Portable Detection and Photothermal Therapy of Cancer Cells

Sha Liu,Siyu Lu,Shihao Sun,Jun Hai,Genping Meng,Baodui Wang

Quantitative detection of cancer cells using portable devices is promising for the development of simple, fast, and point-of-care cancer diagnostic techniques. However, how to further amplify the detection signal to improve the sensitivity and accuracy of detecting cancer cells by portable devices remains a challenge. To solve the problem, we, for the first time, synthesized folic-acid-conjugated Au nanoframes (FA-Au NFs) with amplification of pressure and temperature signals for highly sensitive and accurate detection of cancer cells by portable pressure meters and thermometers. The resulting Au NFs exhibit excellent near-infrared (NIR) photothermal performance and catalase activity, which can promote the decomposition of NH4HCO3 and H2O2 to generate corresponding gases (CO2, NH3, and O2), thereby synergistically amplifying pressure signals in a closed reaction vessel. At the same time, Au NFs with excellent peroxidase-like activity can catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to produce TMB oxide (oxTMB) with a strong photothermal effect, thereby cooperating with Au NFs to amplify the photothermal signal. In the presence of cancer cells with overexpressing folate receptors (FRs), the molecular recognition signals between FA and FR can be converted into amplified pressure and temperature signals, which can be easily read by portable pressure meters and thermometers, respectively. The detection limits for cancer cells using pressure meters and thermometers are 6 and 5 cells/mL, respectively, which are better than other reported methods. Moreover, such Au NFs can improve tumor hypoxia by catalyzing the decomposition of H2O2 to produce O2 and perform photothermal therapy of cancer. Together, our work provides new insight into the application of Au NFs to develop a dual-signal sensing platform with amplification of pressure and temperature signals for portable and ultrasensitive detection of cancer cells as well as personalized cancer therapy.

A biomimetic design for a sialylated, glycan-specific smart polymer

Chen Zhonghui,Lv Ziyu,Wang Xue,Yang Hong,Qing Guangyan,Sun Taolei

Increasing evidence has indicated that glycans and their interactions with glycan-specific binding proteins have crucial roles in most physiological processes. One emerging topic in biomaterials is the construction of smart materials capable of recognizing and responding to these valuable glycans. However, because of the complicated compositions and structures as well as the low abundances and microheterogeneity of glycans, developing glycan-responsive materials with a high sensitivity and specificity is a long-term, challenging goal. Here, we report a biomimetic polymer capable of capturing and discriminating sialo-complex-type glycans with the core Neu5Ac-Gal-GlcNAc-Man sequence, which is one of the most effective biomarkers for cancer detection. As an optimized dipeptide, L-Asp-L-Phe has a strong but differential binding affinity for N -acetyl-neuraminic acid (Neu5Ac), N -acetyl-glucosamine (GlcNAc), galactose (Gal), and mannose (Man), which are the core construction units of sialylated glycans. In addition, the L-Asp-L-Phe-grafted polyethyleneimine film displays remarkable adsorption behavior for the model sialylated glycans and can discriminate their linkage isomers, which is accompanied by significant changes in the surface morphology and stiffness. These features facilitate the highly selective capture of sialylated glycopeptides from complex protein samples using PEI- g -L-Asp-L-Phe-modified mSiO 2 @SiO 2 @Fe 3 O 4 core-shell microspheres. The mechanism analysis demonstrates that the favorable polymeric spatial structures and multiple synergetic hydrogen-bonding interactions among the dipeptides and glycan are the main driving forces, indicating a clear direction for designing glycan-specific biomaterials.

Highly ordered graphene-isolated silver nanodot arrays as SERS substrate for detection of urinary nucleosides

Shicai Xu,Shouzhen Jiang,Guodong Hu,Jie Wei,Li Wang,Junye Zhang,Qiuju Li

An efficient surface enhanced Raman scattering (SERS) substrate has been developed based on highly ordered arrays of graphene-isolated Ag nanodot (G/AgND) arrays. By combining the plasmonic activity of AgND arrays and unique physical/chemical properties of graphene, the G/AgND arrays show high performance in terms of sensitivity, signal-to-noise ratio and reproducibility. The SERS signals of R6G are clearly detected even at very low concentration of 10−11M. The maximum deviations of SERS intensities from 20 positions of the SERS substrate are less than 5%. The G/AgND arrays were employed for detection of human urinary nucleosides. The diagnostic algorithms based on principal component analysis achieved a diagnostic sensitivity of 91.1% and specificity of 93.3% for separating cancer from normal samples. This work demonstrates that G/AgND arrays used in Raman spectroscopy could be developed as a smart and promising system for detection and screening of cancers.

High sensitivity surface plasmon resonance biosensor for detection of microRNA based on gold nanoparticles-decorated molybdenum sulfide

Wenyan Nie,Qing Wang,Xiaohai Yang,Hua Zhang,Zhiping Li,Lei Gao,Yan Zheng,Xiaofeng Liu,Kemin Wang

A high sensitive surface plasmon resonance (SPR) biosensor was proposed based on the gold nanoparticles-decorated molybdenum sulfide (AuNPs-MoS 2 ) for the first time. This SPR platform using the AuNPs-MoS 2 nanocomposites as signal labels for the sensitive and facile measurement of microRNA (miRNA) was executed in only two steps. At first, the thiol-modified DNA oligonucleotide probes , including a sequence complementary to the target miRNA-141, were fixed on the Au film to identify the segment sequence of target miRNA-141. Then, the assistant DNA-linked AuNPs-MoS 2 nanocomposites were used to combine with the other section of the miRNA-141. Performance of SPR biosensor was enhanced by taking advantage of the AuNPs-MoS 2 nanocomposites, thus this newly presented sensing assay exhibited high sensitivity toward miRNA with a detection limit of 0.5 fM. Furthermore, the method showed high specificity, resulting in distinguishing differences among miRNA-200 family members. Especially, the assay could also be used to detect human miRNA from cancer cells, and the results were in excellent accord with the ones obtained using qRT-PCR. What is more, this presented method could be feasible for determining miRNA in 10% human serum. This assay may provide a great potential as a miRNA quantification method in complex samples, and exert significant effect on biomedical research and clinical early diagnosis.

Periodic magnetic modulation enhanced electrochemical analysis for highly sensitive determination of genomic DNA methylation

Yitao Liang,Yu Jiang,Lu Fang,Zhen Dai,Shanshan Zhang,Yue Zhou,Yu Cai,Dong Wang,Zhaoyang Wang,Xuesong Ye,Bo Liang

DNA methylation aberrations have a strong correlation with cancer in early detection, diagnosis, and prognosis, which make them possible candidate biomarkers. Electrochemical biosensors offer rapid protocols for detecting DNA methylation status with minimal pretreatment of samples. However, the inevitable presence of background current in the time domain, including electrochemical noise and variations, limits the detection performance of these biosensors, especially for low concentration analytes. Here, we propose an ultrasensitive frequency-domain electrochemical analysis strategy to effectively separate the weak signals from background current. To achieve this, we employed periodic magnetic field modulation of magnetic beads (MBs) on and off the electrode surface to generate a periodic electrochemical signal for subsequent frequency-domain analysis. By capturing labeled MBs with as low as 0.5 pg of DNA, we successfully demonstrated a highly sensitive electrochemical method for determination of genome-wide DNA methylation levels. We also validated the effectiveness of this methodology using DNA samples extracted from three types of hepatocellular carcinoma (HCC) cell lines. The results revealed varying genomic methylation levels among different HCC cell lines, indicating the potential application of this approach for early-stage cancer detection in terms of DNA methylation status.

Finger-actuated Microfluidic Chip Integrated with Visual Immunoassay for Ultrasensitive Detection of PSA in Whole Blood

Chaoshan Zhao,Junju Wang,Liwei Hou,Hong He,Chuang Ge,Yuping Yang,Li Wang,Yi Xu,Shunbo Li

Cumbersome preprocessing and specialized manual operations in clinical blood samples remain a significant challenge for achieving high sensitivity and accurate quantification in point-of-care testing. In this paper, a finger-driven integrated microfluidic chip based on visualization of single nanoparticle scattering was proposed for the detection of prostate-specific antigen (PSA) in the whole blood. To control on-chip fluid, a finger-driven module based on a Tesla valve was designed to unidirectionally regulate fluid mixing and separation in the microchannel. In addition, a membrane separation unit was designed to efficiently separate blood cells and serum, reducing interference from blood cells in the detection process. For quantitative PSA concentration detection, a Multi-functional core-satellite magnetic probe was constructed by using the principle of complementary base pairing of ligands on the surface of gold nanoparticles and magnetic beads. In the presence of target PSA, the constructed core-satellite nanostructure was decomposed, producing a characteristic fluorescence signal and releasing gold nanoparticles with green scattering spots under dark-field microscopy. By correlating the concentration with the number of green scattering spots, cancer risk levels were displayed intuitively using a traffic light system. This biosensor achieves an ultra-low detection limit of 0.5 pg/mL for PSA. Due to the ultra-sensitive ability in detection, the monitoring of PSA concentrations for patients during treatment was also demonstrated. Compared with other methods, this proposed microfluidic assay technology has the advantages of small sample volume, minimal operation, high sensitivity and accuracy. Overall, this biosensor provides a new approach for cancer recurrence monitoring and early diagnosis.
