Polymeric micelles (PMs) have been applied widely to transport hydrophobic drugs to tumor sites for cancer treatment. However, the low load efficiency of the drug in the PMs significantly reduces the therapeutic efficiency. We report here that disulfide-linked camptothecin (CPT) as a kind of dimeric drug can be effectively embedded in the core of poly(ε-caprolactone)–poly(ethylene glycol)–poly(ε-caprolactone) (PCL–PEG–PCL) PMs for improving drug-loading efficiency, and PEG can be used as a hydrophilic shell. Moreover, the dimeric CPT-loaded PCL–PEG–PCL PMs exhibited excellent solubility in phosphate-buffered saline (PBS) media and significant cytotoxicity to cancer cells. Keywords:dimeric drug;camptothecin;micelles;cancer therapy

Concurrent induction of apoptosis and ferroptosis is promising in handling heterogenous cancers. We report a tailored polymeric micellar nanoplatform for the combinational anti-tumor therapy. Two stimuli-responsive amphiphlic block copolymers were synthesized, bearing three functional moieties, i.e. azobenzene, nitroimidazole and 3-fluorophenylboronic acid. Azobenzene could enhance the cellular uptake of micelles. Nitroimidazole and 3-fluorophenylboronic acid could deplete the reduced nicotinamide adenine dinucleotide phosphate (NADPH), glucose and adenosine triphosphate (ATP) under hypoxia, sensitizing ferroptotic and apoptotic cell death. The proof-of-concept was demonstrated in a triple-negative breast cancer cell line (MDA-MB-231). Irrespective of the free or encapsulated form, doxorubicin and auranofin showed a synergistic action, evidenced by a low combination index (< 1). The co-delivery micelles showed improved potency than the single drug-loaded micelles in terms of the biomarkers of apoptosis ( e.g. caspase 3/9, cytochrome c and ATP) and ferroptosis ( e.g. thioredoxin reductase, thioredoxin, glutathione, NADPH, malondialdehyde and lipid peroxides). The apoptosis and ferroptosis induction ability of cargo-free micelles was proved. The in vivo efficacy was verified in the MDA-MB 231 tumor-bearing nude mice model. The current work offers a promising strategy of combinational anti-tumor drug delivery for potent induction of ferroptosis and apoptosis via the sensitization effect of vehicle in the hypoxic tumor.

Photothermal therapy uses photothermal agents (PTAs) to convert light energy to heat energy under near-infrared light to kill local tumors in cancer patients or speed up wound healing in diabetic patients. However, it is difficult to achieve high photothermal conversion efficiency for most of PTAs. Herein, daptomycin (Dap) micelles-stabilized palladium nanoflowers (Dap-PdNFs) were prepared for the first time. The palladium nanoflowers (PdNFs) inside of the Dap-PdNFs were 106 nm. The temperature of the Dap-PdNFs solution quickly rose from 26.8 °C to 52.0 °C within 10 min under irradiation with high photothermal conversion efficiency up to 38%. In addition, the cell viability of HeLa cells and HT-29 cells of Dap-PdNFs exceeded 95% in the absence of near-infrared light, indicating that Dap-PdNFs had good biocompatibility. Meanwhile, the inhibition rate of Dap-PdNFs on HeLa cells was as high as 71.2% under irradiation of 808 nm near-infrared light. More importantly, Dap-PdNFs had a good healing effect on wounds of diabetic mice under irradiation of 808 nm near-infrared light. In short, this research provides a facile method for the application of Dap-PdNFs in safe and efficient tumor treatment and wound healing.

We present an intelligent photothermal therapy agents by functionalizing gold nanoparticles with specific nucleic acid sequences. Hairpin nucleic acids are modified to the nanoparticles, forming AuNPs-1 and AuNPs-2. Upon infiltrating cancer cells, these nanoparticles undergo catalytic hairpin assembly in the presence of target miRNA, leading to aggregation and subsequent photothermal conversion. Under near-infrared laser irradiation, aggregated gold nanoparticles exhibit efficient photothermal conversion, selectively damaging cancer cells. This approach offers heightened selectivity, as nanoparticles only aggregate in environments with cancer biomarkers present, sparing normal cells. Cytotoxicity assays confirm minimal toxicity to normal cells. In vivo studies on mice bearing solid tumors validate the system's efficacy in tumor regression. Overall, this study highlights the potential of nucleic acid-functionalized gold nanoparticles in intelligent and selective cancer photothermal therapy, offering insights for targeted diagnosis and treatment development.

Graphical Spot on : A light-inducible Lip-cRGDfk/ICG/Dox liposome simultaneously loads a water-soluble drug (Dox) and photothermal transduction agents (ICG), targets the tumor site, and locally heats it up under external laser irradiation. Due to the conformational transformation of CP lipids, the anticancer drug Dox is released at a specific time and in a specific space to kill tumor cells. This brings about a combination of chemotherapy and photothermal therapy for the noninvasive treatment of tumors. Choline phosphate lipids have been designed and developed as new-generation zwitterionic nanocarriers with excellent biocompatibility and bioorthogonality to provide a more programmable performance for cancer therapy. However, there is a lack of spatiotemporal and reversible control for drug release at target tumor cells, which can lead to severe adverse effects to normal tissue and discounted treatment outcome. Here, light-inducible Lip-cRGDfk/ICG/Dox liposomes were developed for synergistic cancer therapy. ICG can effectively convert light energy into selective heating in a local environment upon laser irradiation, thus inducing thermal ablation of tumor cells, and further reversibly trigger the spatiotemporal release of anticancer drugs (Dox) at tumor cells due to the conformation transformation of CP lipids to synergistically kill tumor cells. That Lip-cRGDfk/ICG/Dox exhibited a significant improvement for breast cancer therapy in vitro and in vivo is also demonstrated, thus it can serve as an efficient platform to noninvasively and spatiotemporally control the activation of cytotoxicity at tumor cells for precision cancer therapy.

Zwitterionic poly(2-methacryloyloxyethyl phosphorylcholine) (PMPC) is a promising alternative for poly(ethylene glycol) (PEG) as micelle shell-forming block. Three different PMPC-based polymeric micelles were designed and compared with PEGylated micelles. Cholesterol-conjugated and
disulfide-modified poly(D, L-lactide)-SS-poly(2-methacryloyloxyethyl phosphorylcholine) (CPLA-SS-PMPC) micelles, cholesterol-conjugated CPLA-PMPC micelles without disulfides, and PLA-PMPC micelles without cholesterol-conjugation were compared to PLA-PEG micelles. These PMPC-based micelles
showed better hydrophilicity and less protein adsorption. In cellular uptake evaluation, PMPC-based micelles exhibited much better cellular uptake than that of the PLA-PEG, which might reflect strong interaction between the biomimetic PMPC surface and the cellular membrane. In addition, although
there was no statistical difference in cellular internalization behavior among these three PMPC-based micelles, the anticancer activity of paclitaxel (PTX)-loaded CPLA-SS-PMPC micelles was significantly higher than those of the other two kinds of micelles. This superiority was attributed to
the rapid release of the anticancer drug under the redox environment inside cancer cells.

Doxorubicin (DOX) is a traditional broad-spectrum antitumor drug, which has a wide range of clinical applications, but has no tumor non-specificity. Nanoparticles have been explored as drug delivery agents to enhance the therapeutic efficacy and reduce toxic and side effects. Carbon dots (CDs), a carbon-based nanomaterial, has many unique advantages such as easy synthesis, good biocompatibility, and low toxicity. In this study, folic acid was used as raw material to prepare new CDs, and DOX was loaded on the surface of CDs through electrostatic interaction. The prepared nano-drugs CDs/DOX could effectively release DOX under mild acidic pH stimulation. Cell imaging showed that CDs/DOX could transport doxorubicin (DOX) to cancer cells and make them accumulated in nucleus freely. Flow cytometry tests and cellular toxicity assay together confirmed that CDs/DOX could target tumor cells with high expression of folate receptor and increase anti-tumor activity. The therapeutic effect on 4T1 tumor-bearing mice model indicated that CDs/DOX could alleviate DOX-induced toxicity, effectively inhibit tumor growth, and prolong the survival time. Hence, such a targeting nanocarrier is likely to be a candidate for cancer treatment.

Stimuli-responsive prodrug-based nanoplatform with synergistic antitumor activity is of central importance to the development of promising nanomedicines for cancer therapy. Here, we describe a polydopamine-drug conjugate nanocomposite (ZP-PDA-DOX) with targeted cancer photothermal-chemotherapy (PTT-CT), which constructed by a gradual copolymerization of dopamine (DA) and pH-sensitive dopamine-derived prodrug (DA-DOX) into the porous channels of zeolite imidazolate frameworks-8 (ZIF-8), followed by PEGylation with amino-terminated folic acid-polyethylene glycol (NH 2 -PEG-FA) to acquire the high biocompatibility, specificity, and excellent tumor-targeting property. The incorporation of polydopamine strengthened the stability and dispersion of ZIF-8, and also conferred photothermal conversion effect. In the tumor acidic microenvironment, the acid-labile hydrazone linker of DA-DOX and ZIF-8 promptly degraded to release activated DOX. Moreover, the generated hyperthermia due to the high photothermal conversion efficiency of PDA component could accelerate drug release, and simultaneously thermally ablate tumor tissue to maximize the DOX-induced CT, which could also assist PTT to eradicate tumor cells. This study provides a promising strategy for targeted cancer PTT-CT with synergistic anti-tumor effect.

Cancer chemotherapy has limitations such as nonselective distribution of drugs, detrimental side effects on normal tissues; here we reported a smart pH and magnetic sensitive drug delivery system (DDS) based on PEGylated Fe3O4 superparamagnetic nanoparticles. The
citric-coated Fe3O4 (CIO) nanoparticles were firstly synthesized and further functionalized by biocompatible poly(ethylene glycol) bis(carboxymethyl ether) (COOH-PEG-COOH), thus PEGylated CIO (GCIO) nanoparticles were obtained. Doxorubicin (DOX) was conjugated as a model
drug to the nanoparticles via hydrazone bond. The roughly sphere GCIO nanoparticles were comparatively strong magnetism with high drug loading capability (~89%) in relatively uniform size. Drug release study revealed that the GCIO-DOX performed pH-responsive drug-release properties. MTT assay
demonstrated that GCIO nanoparticles possessed low cytotoxicity and good physiological stability. Further, cell viability results indicated that the GCIO-DOX showed effective cytotoxicity only a bit lower than free DOX. All obtained data indicated that the combined pH-responsive and magnetic
multifunction drug delivery system can has excellent potential applications in cancer therapy.

Two-dimensional (2D) nanomaterials able to effectively absorb near-infrared (NIR) radiation have shown considerable potential as multifunctional platforms in the treatment of cancer. Here, we report a molybdenum dioxide (MoOx)-based system for synergistic chemo- and photothermal therapy of cancer. MoOx nanosheets were generated via a one-step hydrothermal route, grafted with poly(ethylene glycol) (PEG), and decorated with melanin (Mel), with successful functionalization confirmed by IR spectroscopy. The mean diameter and thickness of the MoOx particles were found to be 302 ± 34 nm and 19 ± 2 nm using atomic force microscopy. The hydrodynamic size of the MoOx nanosheets was 105 ± 17 nm, and the resulting final MoOx-PEG-Mel-DOX nanocomposite formulation had a uniform diameter of 161 ± 26 nm. The MoOx-PEG-Mel nanosheets efficiently convert NIR light to heat, possessing a photothermal conversion efficiency of 61.7%. They can also be loaded with doxorubicin hydrochloride (DOX), giving a high drug loading (325.6 mg DOX/g MoOx-PEG-Mel). DOX release from the formulation is influenced by both pH and NIR irradiation. Systematic in vitro and in vivo evaluations reveal that synergistic chemo- and photothermal therapy using MoOx-PEG-Mel-DOX can completely eradicate a tumor with no observable off-target cytotoxicity. This is the first report of the fabrication of enhanced photothermal agents by combining MoOx nanosheets and a natural product. This work proffers a strategy for efficiently treating cancer as well as potentially extending the photothermal applications of 2D nanomaterials by surface melanin functionalization.

It is of vital importance to engineer the surface structures of carbon dots (CDs) to satisfy their practical biomedical applications, including imaging and treatment. In this work, one type of hyaluronic acid-derived CD (HA-CD) was synthesized via a facile one-step hydrothermal method using cancer cell-targeted HA as a precursor. The as-prepared HA-CDs were targeted actively toward CD44 receptor-overexpressing cancer cells because a partial HA structure remained on the HA-CD surface. Beyond this, HA-CDs can act as a novel photosensitizer, because they can generate O2˙− under 650 nm laser irradiation, and they also exhibit excellent blue photoluminescence emission. The in vitro results revealed that HA-CDs imaged selectively CD44-overexpressing cancer cells and inhibited their growth under 650 nm laser irradiation. Thus, HA-CDs can serve as a promising self-targeted imaging-guided photodynamic therapy (PDT) agent for cancer. The present research provides a promising new method to simply construct multifunctional CD-based targeted phototheranostic systems.

Nanomedicine platforms that have the potential to simultaneously provide the function of molecular imaging and therapeutic treatment in one system are beneficial to address the challenges of cancer heterogeneity and adaptive resistance. In this study, Cyclic RGD peptide (cRGD), a less-expensive active tumor targeting tri-peptide, and doxorubicin (DOX), a widely used chemotherapeutic drug, were covalently attached to Ag2S quantum dots (QDs) to form the nano-conjugates Ag2S-DOX-cRGD. The optical characterization of Ag2S-DOX-cRGD manifested the maintenance of QDs fluorescence, which suggested the potential of Ag2S for monitoring intracellular and systemic drug distribution. The low biotoxicity of Ag2S QDs indicated that they are promisingly safe nanoparticles for bio-applications. Furthermore, the selective imaging and favorable tumor inhibition of the nanoconjugates were demonstrated at both cell and animal levels. These results indicated a promising future for the utilization of Ag2S QDs as a kind of multi-functional nano platform to achieve imaging-visible nano-therapeutics.
