An Experimental Evaluation of Toxicity Effects of Sodium Chloride on Oviposition, Hatching and Larval Development of Aedes albopictus

Xiang Guo,Siyun Zhou,Jing Wu,Xiaoqing Zhang,Yuji Wang,Zixuan Li,Xiao-Guang Chen,Xiaohong Zhou

Dengue virus, one of the most important mosquito-borne viruses, has shown a sharp upward trend, spreading around the world in recent years. Control of vectorsAedes aegyptiandAe. albopictusremains crucial for blocking dengue transmission. The lethal ovitrap (LO) is one of the cost-effective traps based on the classic “lure and kill” strategy, and finding a proper long-lasting effective toxin is key to achieving the desired effect. The concentration of inorganic salts of habitat environment plays a strong role in affecting oviposition, hatching, and development of mosquitoes, but the potential insecticide activity of Sodium Chloride (NaCl) in habitat water as well as LO still lacks research. In this study, we carried out laboratory experiments to systematically explore the effects of different concentrations of NaCl solutions on oviposition, egg hatching, and larval development ofAe. albopictus. Consequently,Ae. albopictuswas found to prefer freshwater to lay eggs; whereas 48.8 ± 2.6% eggs were laid in freshwater and 20% in ≥1.0% brackish water, few eggs were laid in 3.0% NaCl solution. Compared with egg hatching, larval development ofAe. albopictuspresented a higher sensibility to NaCl concentration. The mortality of the 3rd–4th larvae in 1.0% NaCl solution was 83.8 ± 8.7%, while in 3.0% it reached 100%. Considering the cumulative effect of NaCl, when NaCl concentration was ≥1.0%, no eggs could successfully develop into adults. These data suggested that NaCl solutions with a concentration ≥1.0% can be used as an effective cheap insecticide forAe. albopictusin subtropical inland aquatic habitats, and also as the “kill” toxin in LOs. Meanwhile, the concentration range from 0 to 2.0% of NaCl solution has the potential to be used as the “lure” in LOs. The technological processes of how to use NaCl as insecticide or in LOs still needs further in-depth exploration. Keywords:Aedes albopictus;habitat;Sodium Chloride (NaCl);dengue;50% lethal concentration

Detection of Dengue Fever Nonstructural Protein 1 Antigen by Proteolytic Peptide Imprinting Technology and UHPLC–MS/MS

Jinyi Wu,Shuyi Huang,Lei Tan,Yuling Li,Xiaotong Wu,Yong Liang

Dengue fever is caused by mosquito-transmitted dengue virus infection and continues to increase worldwide, threatening public health in tropical and subtropical regions. The primary difficulties in preventing a reduction of the medical burden of dengue fever lies in the lack of effective mosquito control, preventive dengue vaccines, and clinically effective antiviral drugs to treat dengue infections. Rapid and accurate diagnosis is crucial for proper patient care and effective control of epidemics. The present work proposes an alternative strategy for detecting the dengue virus nonstructural protein 1 (NS1) antigen in clinical serum samples by using ultrahigh-performance liquid chromatography tandem mass spectrometry (UHPLC–MS/MS) in combination with the molecularly imprinted polymers. Rather than the whole protein, the NS1 signature peptide is selected as a template for molecular imprinting and quantified as a stoichiometric readout of NS1. Three functional monomers with hydrophobic, positively charged, and negatively charged groups were synthesized by click reactions in terms of the signature peptide. These three functional monomers provide abundant recognition sites for the peptide, allowing the peptide template to be effectively imprinted during polymerization. The imprinting conditions were optimized, and the molecularly imprinted polymers were characterized and used for enriching the signature peptide from digested serum samples by solid-phase extraction and then detected by UHPLC–MS/MS. The proposed method is used to detect the dengue virus NS1 in clinical samples and holds significant promise for early confirmation of dengue virus infection.

Application of a graphene-oxide-modified surface plasmon resonance biosensor in dengue E-protein detection

Zhang Liang,Zhang Ran,Cui Lili,Li Tao,Gu Dayong,He Jian’an,Zhang Sixiang

The sensitization effect of graphene oxide (GO) on surface plasmon resonance (SPR) chip sensors has been widely reported. However, the absence of a fixed value for selecting the GO solution concentration is a significant limitation for reproducing the experiments reported in the literature. In this study, linear functionalized polyethylene glycol (SH-PEG-NH 2 ) is used to modify the surfaces of bare gold chips, using flow and immersion methods to produce GO-film-covered SPR sensor chips. Deionized water and phosphate buffered saline are applied as refractive index variants to compare the changes in sensor performance following chip modification, and to determine the optimal concentrations of GO solution under the respective modification methods. Using a sensor chip modified with the optimal GO concentration enhances the ability of a 10-year-old device to detect the dengue virus (DENV) E-protein by 357% and enables the detection of antibody protein concentrations as low as 62.5 ng/mL. Satisfactory results were also obtained using a DENV-positive serum to verify the chip usage. The results of this study are of significance for future efforts involving the rapid preparation of accurate and low-cost GO-SPR biosensor chips.

d-Tetramethrin causes zebrafish hepatotoxicity by inducing oxidative stress and inhibiting cell proliferation

Yang Li,Mijia Li,Shiyi Duan,Sijie Zhang,Huiqiang Lu,Xinchun Guo,Keyuan Zhong

d -Tetramethrin is one of the main components of mosquito control products, and is widely used for the control of dengue fever and insecticide production. Due to its widespread use, d -tetramethrin is a ubiquitous environmental pollutant and poses potential risks to human health. However, the effects of d -tetramethrin on liver morphology and function are not clearly established. In this study, we used zebrafish as an animal model to analyze the acute and chronic effects of d -tetramethrin exposure on the liver. We exposed zebrafish larvae and adults to different concentrations of d -tetramethrin and examined the impact of d -tetramethrin on lipid and glycogen metabolism, cellular properties, oxidative stress , cell proliferation , and apoptosis in the liver. We also analyzed transcriptional changes in genes related to apoptosis, inflammation, and cell proliferation using qPCR . Zebrafish exposed to d -tetramethrin exhibited severe liver damage, as evidenced by the presence of vacuoles and nuclear distortion in liver cells. The liver area in zebrafish larvae of the treatment group was significantly smaller than that of the control group. Significant lipid accumulation and decreased glycogen levels were observed in the livers of both zebrafish larvae and adults exposed to d -tetramethrin. Furthermore, d -tetramethrin exposure induced apoptosis and inflammation in zebrafish embryos. Additionally, d -tetramethrin caused liver damage, metabolic dysfunction, and impaired liver function. These results suggest that d -tetramethrin induces liver toxicity in zebrafish, by inducing oxidative stress and inhibiting cell proliferation.
