Chrysin Attenuates High Glucose-Induced BMSC Dysfunction via the Activation of the PI3K/AKT/Nrf2 Signaling Pathway

Yu Li,Ximei Wang

Purpose High glucose environment in diabetes mellitus induces the dysfunction of bone marrow-derived mesenchymal stromal cells (BMSCs) and impairs bone regeneration. Chrysin is a natural polyphenol with outstanding anti-inflammation and anti-oxidation ability. However, whether and how chrysin affects BMSCs in high glucose conditions remain poorly understood. The present study aimed to explore the effects and underlying mechanisms of chrysin on the BMSCs exposed to high glucose environment.Materials and Methods Cell viability was detected by cell counting kit 8 assay and 5-ethynyl-2’-deoxyuridine staining, while cell apoptosis was determined through flow cytometry using Annexin V-FITC/PI kit. The oxidative stress in BMSCs was evaluated by detecting the reactive oxygen species production, malondialdehyde content, and superoxide dismutase activity. Alkaline phosphatase staining, Alizarin Red staining, and quantitative real-time PCR were performed to determine the osteogenic differentiation. Western blot was used to examine the expression of the PI3K/ATK/Nrf2 signaling pathway. Furthermore, chrysin was injected into calvarial defects of type 1 diabetic SD rats to assess its in vivo bone formation capability. Results Chrysin reduced oxidative stress, increased cell viability, and promoted osteogenic differentiation in BMSCs exposed to high glucose. Blocking PI3K/ATK/Nrf2 signaling pathway weakened the beneficial effects of chrysin, indicating that chrysin at least partly worked through the PI3K/ATK/Nrf2 pathway. Conclusion Chrysin can protect BMSCs from high glucose-induced oxidative stress via the activation of the PI3K/AKT/Nrf2 pathway, and promote bone regeneration in type 1 diabetic rats.

Improvement of Baicalein and Wogonin Yields from Scutellaria baicalensis Georgi Roots Fermented by a Novel Endophytic Fungus Capable of Producing Cellulase and β-Glucosidase

Fu Jin-Xian,Jiao Jiao,Gai Qing-Yan,He Xiao-Jia,Fu Yu-Jie,Feng Xue,Gao Jie

Baicalein and wogonin, two health-promoting flavonoid aglycones in Scutellaria baicalensis Georgi roots, have better bioactivity and bioavailability than their glycoside precursors (baicalin and wogonoside). The search for novel microorganisms for the fermentation of S. baicalensis roots to improve the yields of baicalein and wogonin is of great interest in the food and nutraceutical industries. In this study, a novel endophytic fungus ( Penicillium rubens ) capable of producing cellulase (0.82 ± 0.12 U/mL) and β-glucosidase (4.55 ± 0.97 U/mL) was used to ferment S. baicalensis roots to obtain high yields of baicalein and wogonin. Semi-solid-state fermentation with homogeneous fungal cultures was found to be the appropriate strategy. Under the optimal fermentation parameters (temperature 30 °C, inoculation dose of fungal cultures 4 mL/g, and time 60 h), the yields of baicalein and wogonin (46.95 ± 4.98 mg/g DW and 31.41 ± 9.36 mg/g DW) from S. baicalensis roots were tremendously increased by 16.13-fold and 15.47-fold over control, respectively. Results of industrial enzyme bioprocessing and scanning electron microscope indicated that the remarkable increase in aglycone product yields could be due to the synergistic effects of cellulase and β-glucosidase produced by P. rubens on cell wall hydrolysis and glycoside deglycosylation. In addition, extracts of S. baicalensis roots fermented by P. rubens showed a significant increase in antioxidant and antimicrobial activities. Overall, the semi-solid-state fermentation of S. baicalensis roots using the novel P. rubens was an effective strategy that could yield high quantities of baicalein and wogonin, which had promising potential in the food and nutraceutical industries. Semi-Pilot Scale Extraction of Pinocembrin and Galangin from Populus alba L. × berolinensis K. Koch via Enzymatic Pretreatment and Ultrasonication

Ru Zhao,Xiaoli Li,Yazhou Bao,Wenjun Xu,Chen Xu,Rongrong Wang,Tianlan Xia,Tingli Liu,Ailing Ben

In this investigation, pinocembrin and galangin were efficiently extracted from the male inflorescence ofPopulus albaL. ×berolinensisK. Koch through an enzymatic pretreatment–ultrasonic-assisted strategy (EP-UAS), and the feasibility of their pilot-scale application was validated. The optimal parameters (ethanol volume fraction, cellulase dosage, incubation temperature, incubation time, pH, liquid‒solid ratio, ultrasonic irradiation power during incubation, duty cycle, ultrasonic irradiation power and time during extraction) affecting pinocembrin and galangin yields were systematically explored. The Box–Behnken design (BBD) results provided optimal parameters for the EP-UAS process. Under the optimal conditions, the actual yields of pinocembrin and galangin were 2158.33 ± 0.13 μg/g and 1257.96 ± 0.06 μg/g, respectively. Stability, recovery and reproducibility were determined under the above optimized conditions to evaluate the proposed EP-UAS method. Moreover, laboratory-scale experimental results revealed that the conditions selected via single-factor and response surface experiments were also applicable to pilot-scale production, facilitating industrialization.

Functional chitosan/HP-β-CD hydrogel for targeted co-delivery of Rhubarb-derived nanovesicles and kaempferol for alleviating ulcerative colitis

Xiao Tang,Kun Wang,Zihan Liu,Xu Luo,Ming Wu,Hui Ding,Gang Liu,Qian Du

Ulcerative colitis (UC) remains a major challenge in clinical treatment due to its multivariate pathology. Developing an oral formulation that encapsulates and delivers multiple active ingredients to target colon tissues by suppressing intestinal inflammation and restoring the intestinal barrier is crucial for effectively treating UC. Here, we developed rhubarb-derived nanovesicles (RNs) and a supramolecular hydrogel platform formed by furfural-functionalized chitosan-mannose polymer and synthesized 3-maleimide HP-β-CD, with kaempferol (Kae) integrated into the hydrophobic cavity. The hydrogel's cross-linking network effectively encapsulates RNs, forming the Kae/CMCHD@RNs system. Rheology, SEM, TGA, degradation behavior, in vitro drug release, and a macrophage-targeted permeability test were performed. The results indicate that the hydrogel utilizes pH/enzyme sensitivity to ensure sustained release in the colon, while also facilitating targeted delivery to macrophages. In vivo imaging further reveals a prolonged local drug retention time in the colon. Moreover, both in vitro and in vivo studies demonstrate RNs and Kae exhibit synergistic therapeutic effects for UC, including inflammation reduction, oxidative stress alleviation, M1-to-M2 macrophage repolarization, and restoration of the intestinal barrier. Consequently, this study underscores the potential of Kae/CMCHD@RNs as a promising therapeutic approach for managing UC.

Active colorimetric bilayer polycaprolactone-eucalyptus oil@silk fibroin-bayberry anthocyanins (PCL-EO@SF-BAs) membrane with directional water transport (DWT) for food packaging

Li Xia,Linlin Li,Yewen Xiao,Feng Xiao,Lanhua Liu,Xiangying Chen,Xingjiang Li,Hualin Wang

Currently, designing smart membranes with multifunctional effectiveness is crucial to food freshness monitoring and retention. Herein, an active colorimetric Janus bilayer membrane with directional water transport (DWT) performance is constructed by electrospinning, which comprises a hydrophilic layer of silk fibroin-bayberry anthocyanins (SF-BAs) and a hydrophobic layer of polycaprolactone-eucalyptus oil (PCL-EO). The entities of BAs and EO are well dispersed in the fiber matrix by hydrogen bonds and physical interactions, respectively. BAs endow the membrane colorimetric response and antioxidant activity, and EO contributes to the antibacterial activity while DWT performance is generated from the asymmetric wettability of the two layers. The bilayer membrane has an accumulative one-way transport index of 1077%, an overall moisture management capacity of 0.76 and a water evaporation rate of 0.48 g h −1 . Moreover, the release of BAs and EO was predominantly controlled by Fickian diffusion. As a pH-sensing indicator, PCL-EO@SF-BAs is highly sensitive to external pH stimuli and the response is reversible. In addition to freshness monitoring, PCL-EO@SF-BAs can extend the shelf-life of pork beyond 100% at 4 °C. Also, it can extend the shelf life of shrimp by approximately 70% at 25 °C with the synergistic effect of antibacterial activity and the DWT performance.

Evaluation of Anti-Fungal Activities of Environmentally Friendly Wood Preservative from Thermal-Induced Lignified Twigs

Xinqi Gao,Yafang Lei,Teng Sun,Yuanze Ma,Hao Guan,Li Yan

Enhancing the decay resistance ofPopulus tomentosawood through environmentally friendly methods is crucial for improving its durability and market competitiveness. Lignified twigs (LT), typically unsuitable as timber due to their small diameter, are rich in lignin, which degrades during thermal induction to produce antifungal organic compounds. In this context, the objective of this study was to develop a lignified twig preservative (LTP) by thermal induction from the LT ofActinidia chinensisvar.Jinyang. The antifungal activity of LTP under varying thermal conditions was analyzed, along with its chemical composition. Enzyme activity, cell membrane integrity, and respiratory metabolism in fungi treated with LTP were examined to elucidate antifungal mechanisms. Additionally, the decay resistance of LTP-treated wood was evaluated. Results revealed that LTP produced under N2at 220 °C exhibited robust antifungal activity againstTrametes versicolorandGloeophyllum trabeum, attributed to phenolic compounds such as syringaldehyde, syringone, vanillin, and vanillone. LTP inhibited fungal cellulases, hemicellulases, and ligninases by 30%–60%, disrupted cell membrane functionality, and suppressed respiratory metabolism. Poplar wood treated with LTP demonstrated significantly enhanced decay resistance (mass loss < 10%). This thermal-induced feedback pattern shows great potential for LT in wood preservation.

Hemicellulose-rich transparent wood: Microstructure and macroscopic properties

Yan Jiang,Mengyang Zhang,Mengling Weng,Xiuyu Liu,Xianjian Rong,Qin Huang,Guoning Chen,Shuangfei Wang,Lijun Wang

Hemicellulose plays a vital role in nature wood matrix. Here, we first demonstrate the significance of hemicellulose for improving the microstructure and macroscopic properties of transparent wood (i.e. wood/polymer composite), an emerging structural material. A delignified basswood templet with high hemicellulose content (~23 wt%) and well-preserved cellulose supramolecular structure was prepared via a peracetic acid-based process. This hemicellulose-rich delignified wood templet possesses nearly intact cell wall structure and sufficient space in the lumen and middle lamella for petroleum-based polymer space-filling. Consequently, the resultant hemicellulose-rich wood/PMMA composite exhibits high optical transmittance (~85 %), medium haze (~72 %), acceptable optical stability as well as excellent mechanical strength (tensile strength of 165.3 MPa and toughness of 1.65 MJ/m 3 ), benefitting from the absence of microstructural defects and heterogeneity in the matrix. It is anticipated that our study would provide new insights into the fabrication of advanced transparent wood material from the point-of-view of wood chemical composition .

Mitochondrial toxicity evaluation of traditional Chinese medicine injections with a dual in vitro approach

Shen Yunfu,Guo Kaiqiang,Ma Aijun,Huang Zhe,Du Jingjing,Chen Junhe,Lin Qianyu,Wei Chengming,Wang Zi,Zhang Fuming,Zhang Juan,Lin Wanjun,Feng Na,Ma Wenzhe

There are technical obstacles in the safety evaluation of traditional Chinese medicine (TCM) injections due to their complex chemical nature and the lack of rapid and accurate in vitro methods. Here, we established a dual in vitro mitochondrial toxicity approach combing the conventional “glucose/galactose” assay in HepG2 cells with the cytotoxic assay in mitochondrial respiration deficient cells. Using this dual in vitro approach, for the first time, we systematically assessed the mitochondrial toxicity of TCM injections. Four of the 35 TCM injections, including Xiyanping, Dengzhanhuasu, Shuanghuanglian, and Yinzhihuang, significantly reduced cellular ATP production in galactose medium in the first assay, and presented less cytotoxic in the respiration deficient cells in the second assay, indicating that they have mitochondrial toxicity. Furthermore, we identified scutellarin, rutin, phillyrin, and baicalin could be the potential mitochondrial toxic ingredients in the 4 TCM injections by combining molecular docking analysis with experimental validation. Collectively, the dual in vitro approach is worth applying to the safety evaluation of more TCM products, and mitochondrial toxic TCM injections and ingredients found in this study deserve more attention.

Berberrubine, a Main Metabolite of Berberine, Alleviates Non-Alcoholic Fatty Liver Disease via Modulating Glucose and Lipid Metabolism and Restoring Gut Microbiota

Sa Yang,Shijie Cao,Congyu Li,Jichao Zhang,Chang Liu,Feng Qiu,Ning Kang

Non-alcoholic fatty liver disease (NAFLD) is a major public health problem in many countries. Berberine (BBR) is an effective therapeutic agent in alleviating NAFLD. Berberrubine (BRB) is one of the main active metabolites of BBR, which shows significant anti-obesity and antihypoglycemic effects. However, whether BRB is responsible for the in vivo therapeutic effect and the underlying mechanism of BRB on NAFLD have not been elucidated. In this study, the ability of BRB to ameliorate NAFLD, together with its molecular mechanism, was investigated. The results showed that BRB treatments could significantly improve hepatic steatosis and insulin resistance in high-fat diet (HFD)-fed mice and oleic acid (OA)-treated HepG2 cells. Meanwhile, BBR and BRB treatment similarly prevented lipid accumulation by regulating the protein expression of ATGL, GK, PPARα, CPT-1, ACC1, FAS, and CD36. In addition, compared with BBR, BRB could maintain glucose homeostasis via GLUT2, GSK3β, and G6Pase in HFD-fed mice. Furthermore, the components of the gut microbiota in mice were analyzed by 16S rRNA gene sequencing. BBR and BRB treatment could greatly modify the structure and composition of gut microbiota. At the genus level, BBR and BRB treatment decreased Lactobacillus and Romboutsia , while BBR increased beneficial bacteria, such as Akkermansia and Bacteroides , and BRB increased beneficial bacteria, such as Ileibacterium and Mucispirillum . Altogether, both BRB and BBR were active in alleviating NAFLD in vivo and BRB might be used as a functional material to treat NAFLD clinically.
