Microplastics and bisphenol A hamper gonadal development of whiteleg shrimp (Litopenaeus vannamei) by interfering with metabolism and disrupting hormone regulation

Yu Han,Wei Shi,Yu Tang,Weishang Zhou,Hongxiang Sun,Jiongming Zhang,Maocang Yan,Lihua Hu,Guangxu Liu

Gonadal development is a prerequisite for the reproductive success of an organism, and might be affected by environmental factors such as emergent pollutants. Although marine crustaceans are threatened by ubiquitous emergent pollutants such as microplastics (MPs) and bisphenol A (BPA) under realistic scenarios, studies about the impacts of these pollutants on the gonadal development of crustacean species are rare. In this study, the effects of MPs and BPA, alone or in combination, on gonadal development were investigated in whiteleg shrimp ( Litopenaeus vannamei ). The results obtained demonstrated that whiteleg shrimp exposed to MPs and BPA had significantly smaller gonad-somatic index (GSI) and an obvious delay in the gonad developmental stage. In addition, exposure of whiteleg shrimp to pollutants tested resulted in a reduction in the oxygen consumption rate, elevation in the ammonia excretion rate, decline in the O: N ratio, and downregulation in the expression of metabolism-related genes, indicating significant disruptions of shrimp metabolism by the pollutants. Furthermore, in addition to a few exceptions, both the in vivo contents of gonadal development-related hormones (GIH and MIH) and the expression of genes encoding regulatory hormones ( GIH , MIH , and CHH ) were upregulated by the exposure of whiteleg shrimp to the pollutants investigated, suggesting a significant obstruction of endocrine regulation. Moreover, MP-BPA coexposure was shown to be more toxic to whiteleg shrimp than the corresponding single exposures and significantly greater amount of BPA accumulated in the gonads (both testis and ovaries) of shrimp with the coexistence of MPs, which may be caused by the Trojan horse effect and summation of the toxic impacts on common targets. In general, the data obtained in this study demonstrated that MPs and BPA at environmentally realistic concentrations significantly inhibited the gonadal development of whiteleg shrimp probably by interfering with metabolism and disrupting endocrine regulation.

Characterization and interaction mechanism of apigenin-loaded composite hydrogels based on whiteleg shrimp (Penaeus vannamei) powder and κ-carrageenan

Yingrui Wang,Tianqi Zhou,Lin Li,Yuqiao Wang,Jianan Yan,Ce Wang,Bin Lai,Haitao Wu

The novel food composite hydrogel with delivery function consisting of shrimp powder (SP) and κ-carrageenan (κ-C) was constructed and used for loading apigenin (API). Binary mixtures of liquid SP and κ-C could form gels with solid behavior. Furthermore, the addition of API increased the initial modulus of elasticity of SP/κ-C by approximately 47.3%, as well as a 14.9% reduction in the relaxation time T 23 . Moreover, the O-H stretching band and the amide I band of SP/κ-C binary gel loaded with API were blue shifted, and the XRD characteristic peak of API disappeared in SP/κ-C/API. In addition, broken macropores and dispersed patch microstructures were presented in SP and κ-C, whereas the SP/κ-C and SP/κ-C/API hydrogels were homogeneously distributed with dense mesh and solid mesh walls. Hydrophobic interactions were the main forces in the SP/κ-C/API composite hydrogels, which were improved 1.3-fold. Furthermore, SP/κ-C/API was extended and released to more than 55% extent during simulated intestinal colonic digestion. These results suggested that a stable and dense binary gel was formed by SP and κ-C, which could seal and extended release effect of API to the colon.

Label-free based proteomics analysis of protein changes in frozen whiteleg shrimp (Litopenaeus vannamei) pre-soaked with sodium trimetaphosphate

Bin Zhang,Jun-long Mao,Hui Yao,Santiago P. Aubourg

Muscle proteins in peeled shrimp ( Litopenaeus vannamei ) are known to be unstable and prone to denaturation affected by freezing and frozen storage. In this study, label-free proteomics were performed to explore the stabilization of frozen (30 days at −18 °C) shrimp muscle proteins when a pre-soaking treatment with distilled water (DW)- or sodium trimetaphosphate (ST) was applied; comparison to fresh samples (FS) was carried out. In total, 163 differentially abundant proteins (DAPs) were down-regulated in DW batch when compared to FS, these including ribosomal proteins, actins, myosin, paramyosin, myosin heavy chains, and tropomyosin; interestingly, most of these DAPs (181 proteins) were up-regulated in ST batch when compared to DW shrimp, mainly due to the incorporation of ST into muscle tissues. The results revealed the decreased protein degradation resulting from the reduced damage from ice-crystal growth. Gene ontology (GO) analysis suggested that these DAPs were mainly involved in catalytic activity, binding, and metabolic processes. Kyoto encyclopedia of genes and genomes (KEGG) results indicated that many pathways, including phototransduction, metabolic, and ribosomal pathways that interacted with phosphoglycerate mutase, actins, and ribosomal proteins were altered. Additionally, Eukaryotic clusters of orthologous group (KOG) results confirmed that incorporated ST maintained the stability of these DAPs in shrimp muscle, especially for cytoskeleton proteins, and retarded the degradation of muscle proteins during frozen storage.
