Self-healing, antioxidant, and antibacterial Bletilla striata polysaccharide-tannic acid dual dynamic crosslinked hydrogels for tissue adhesion and rapid hemostasis

Xiaojia Zhang,Yingying Mu,Lijie Zhao,Yanlong Hong,Lan Shen

Biomaterials capable of achieving effective sealing and hemostasis at moist wounds are in high demand in the clinical management of acute hemorrhage . Bletilla striata polysaccharide (BSP), a natural polysaccharide renowned for its hemostatic properties, holds promising applications in biomedical fields. In this study, a dual-dynamic-bonds crosslinked hydrogel was synthesized via a facile one-pot method utilizing poly(vinyl alcohol) (PVA)-borax as a matrix system, followed by the incorporation of BSP and tannic acid (TA). Chemical borate ester bonds formed around borax , coupled with multiple physical hydrogen bonds between BSP and other components, enhanced the mechanical properties and rapid self-healing capabilities. The catechol moieties in TA endowed the hydrogel with excellent adhesive strength of 30.2 kPa on the surface of wet tissues and facilitated easy removal without residue. Benefiting from the synergistic effect of TA and the preservation of the intrinsic properties of BSP, the hydrogel exhibited outstanding biocompatibility , antibacterial, and antioxidant properties. Moreover, it effectively halted acute bleeding within 31.3 s, resulting in blood loss of 15.6 % of that of the untreated group. As a superior hemostatic adhesive, the hydrogel in this study is poised to offer a novel solution for addressing future acute hemorrhage, wound healing , and other biomedical applications.

Jiahao Zhang,Yunxue Yin,Shuo Hu,Guangyu Wang,Yuxin Tong,Mingyang Zen,Zhile Liang,Minghao Li,Ran Yan,Yue Wang

The silver nanoparticle is a good antibacterial material being used as a broad-spectrum fungicide, including against some multidrug-resistant strains. Compared with the normal chemical and physical preparation methods, green synthesis has attracted wide attention, because of the pharmaceutical activities of the natural product, mild reaction conditions, and environmentally friendly, etc. In this study, the synthesis of silver nanoparticles (Ag NPs) was prepared fromBletilla striatapolysaccharide (BSP) and characterized by UV-vis spectroscopy and Dynamic Light Scattering (DLS). Scanning electron microscopy (SEM) and transmission electron microscopy (TEM) indicated the morphology of Ag NPs was subspherical with an average size of 20–35 nm.Bletilla striatapolysaccharide not only can be used as a natural reducing agent, but also has good repairing ability. Moreover, the antibacterial experimental results showed its great antimicrobial activity against Gram-positive bacteria (Staphylococcus aureus), Gram-negative bacteria (Escherichia coli) andCandida albicans. Keywords:silver nanoparticles;green synthesis;Bletilla striatapolysaccharide;antibacterial

Oxidized Bletilla rhizome polysaccharide-based aerogel with synergistic antibiosis and hemostasis for wound healing

Quan Yan,Xiaoyan Long,Puxiang Zhang,Wei Lei,Dequn Sun,Xiaoying Ye

Uncontrolled bleeding and infection following trauma remain a clinical challenge. Here, we developed a synergistic hemostatic and antibacterial aerogel dressing based on oxidized Bletilla rhizome polysaccharide Schiff Base (ORBPS) and polyvinyl alcohol (PVA) by using a combined freeze-drying/cross-linking process. Bletilla rhizome polysaccharide (RBP) was extracted by water extraction and alcohol precipitation. RBP contained mannose and glucose in the molar ratio of 2:1 and its molecular weight was 8.2 × 10 5  g/mol. ORBPS was prepared via Schiff base reaction between silver sulfadiazine and oxidized RBP (ORBP), and verified by FTIR and NMR spectra . The resultant ORBPS/PVA aerogel exhibited excellent antibacterial and hemostatic capabilities. The aerogel also showed good liquid absorption capacity and biocompatibility. Full-thickness skin defect experiments indicated the aerogel enhanced wound healing by reducing inflammation, promoting angiogenesis , and accelerating epithelialization . Therefore, ORBPS/PVA aerogel may be a potential hemostasis and anti-infection wound dressing.

The utilization of chitosan/Bletilla striata hydrogels to elevate anti-adhesion, anti-inflammatory and pro-angiogenesis properties of polypropylene mesh in abdominal wall repair

Di Yuntao,Wang Lu,He Wei,Liu Shuyan,He Yuqi,Liao Jie,Zhang Ruihong,Yin Lan,Xu Zhiwei,Li Xiaoming

Polypropylene (PP) mesh is commonly used in abdominal wall repair due to its ability to reduce the risk of organ damage, infections and other complications. However, the PP mesh often leads to adhesion formation and does not promote functional tissue repair. In this study, we synthesized one kind of aldehyde Bletilla striata polysaccharide (BSPA) modified chitosan (CS) hydrogel based on Schiff base reaction. The hydrogel exhibited a porous network structure, a highly hydrophilic surface and good biocompatibility. We wrapped the PP mesh inside the hydrogel and evaluated the performance of the resulting composites in a bilateral 1 × 1.5 cm abdominal wall defect model in rats. The results of gross observation, histological staining and immunohistochemical staining demonstrated the positive impact of the CS hydrogel on anti-adhesion and wound healing effects. Notably, the addition of BSPA to the CS hydrogel further improved the performance of the composites in vivo, promoting wound healing by enhancing collagen deposition and capillary rearrangement. This study suggested that the BSPA-modified CS hydrogel significantly promoted the anti-adhesion, anti-inflammatory and pro-angiogenesis properties of PP meshes during the healing process. Overall, this work offers a novel approach to the design of abdominal wall repair patches.

Preparation and characterization of photocrosslinked bletilla striata polysaccharide/hyaluronic acid injectable hydrogels

Yilong Wei,Yanan Jiang,Doudou Ma,Li Luo,Jiandong Ma,Xiaoliang Zhao,Yonggang Wang,Jing Zhang,Cunjin Wang,Weijie Zhang

Photocrosslinkable injectable hydrogels dressings prepared based on natural polymer materials not only have good biocompatibility and bioactivity, but also possess the ability to fill tissue defects of irregular shapes and the characteristics of non - invasive drug delivery, showing great research value in the biomedical field. However, it remains a challenge to prepare photocrosslinkable injectable hydrogels dressings with better performance by using natural materials. Therefore, the effects of both radical photo polymerization and photo-click chemistry method on the properties of hydrogels were explored in depth in this paper. A series of radical photo polymerization hydrogels and photo - click chemistry hydrogels were prepared under visible light, with natural Bletilla striata polysaccharide and hyaluronic acid as base material. Comparison of the two types of hydrogels revealed that photo - click chemistry hydrogels have more uniform and dense pore sizes, more superior mechanical properties, better cell compatibility, stronger antioxidant properties, as well as good injectability, drug - sustained - release performance and degradability. The results show that photocrosslinked hydrogels dressings prepared by photo - click chemistry method have better properties than radical photo polymerization method, and exhibit greater potential for application in biomedical fields such as wound healing.

Calcium alginate/silk fibroin peptide/Bletilla striata polysaccharide blended microspheres loaded with tannic acid for rapid wound healing

Nan Wang,Xiaoyong Tian,Baijie Cheng,Shanyi Guang,Hongyao Xu

Biodegradable natural polymers are receiving increasing attention as potential candidates for wound dressing. In the present study, composite microspheres (mCSB) based on calcium alginate (CA), silk fibroin peptide (SP), and Bletilla striata polysaccharide (BSP) were prepared by the reverse emulsion method. The excellent swelling properties of microspheres enable them to rapidly promote thrombosis. Microspheres can increase the platelet aggregation index to 1.5 and the aggregation rate of red blood cells to as high as 80 %. Furthermore, tannic acid (TA)-loaded microspheres demonstrate a slow-release effect on TA; this allows the microspheres to exhibit good long-lasting antibacterial properties . Due to the synergistic effects of SP and TA, the cell senescence was delayed, with a 126.69 % survival rate of fibroblasts after 3 days of incubation. In addition, TA led to a rapid reduction in inflammation levels, with a wound closure rate of >92.80 % within 7 days. The multifunctional TA-loaded mCSB has great application potential for rapid wound healing and the treatment of wound hemostasis .

Yue Zhongqun,Zhu Yue,Chen Teng,Feng Tingting,Zhou Ying,Zhang Jiaojiao,Zhang Ning,Yang Jing,Luo Gang,Wang Zuhua

Background: Vitamin E, which is also known as tocopherol, is a compound with a polyphenol structure. Its esterified derivative, Vitamin E succinate (VES), exhibits unique anticancer and healthcare functions as well as immunomodulatory effects. Natural polysaccharides are proved to be a promising material for nano-drug delivery systems, which show excellent biodegradability and biocompatibility. In this study, we employed a novel bletilla striata polysaccharide-vitamin E succinate polymer (BSP-VES) micelles to enhance the tumor targeting and anti-colon cancer effect of andrographolide (AG).Methods: BSP-VES polymer was synthesized through esterification and its structure was confirmed using 1H NMR. AG@BSP-VES was prepared via the dialysis method and the drug loading, entrapment efficiency, stability, and safety were assessed. Furthermore, the tumor targeting ability of AG@BSP-VES was evaluated through targeted cell uptake and in vivo imaging. The antitumor activity of AG@BSP-VES was measured in vitro using MTT assay, Live&Dead cell staining, and cell scratch test. Results: In this study, we successfully loaded AG into BSP-VES micelles (AG@BSP-VES), which exhibited good stability, biosafety and sustained release effect. In addition, AG@BSP-VES also showed excellent internalization capability into CT26 cells compared with NCM460 cells in vitro. Meanwhile, the specific delivery of AG@BSP-VES micelles into subcutaneous and in-situ colon tumors was observed compared with normal colon tissues in vivo during the whole experiment process (1–24 h). What’s more, AG@BSP-VES micelles exhibited significant antitumor activities than BSP-VES micelles and free AG.Conclusion: The study provides a meaningful new idea and method for application in drug delivery system and targeted treatment of colon cancer based on natural polysaccharides.

Mitigating gastrointestinal side effects of risedronate sodium: A study on Bletilla striata polysaccharide microneedle patches

Cong Wang,Wenting Liu,Yong Huang,Lin Zheng,Lili Wang,Ruixi Li,Chang Yang

Risedronate sodium (RIS) is a primary treatment for postmenopausal osteoporosis, but oral administration requires patients to remain upright to prevent side effects like esophagitis, gastritis, and ulcers, which can be difficult for those with fatigue or bone pain. This study developed RIS-loaded Bletilla striata polysaccharide microneedles (RIS-BMNs) with sufficient mechanical strength for effective skin penetration and drug delivery. Franz diffusion cell experiments showed that RIS-BMNs achieved 3 times higher in vitro transdermal absorption than the RIS solution and 2.6 times more than RIS patches. The stimulation test found that RIS-BMNs caused minimal skin irritation, which resolved within 12 h. RIS-BMNs also promoted cell proliferation and wound healing, as shown by in vitro cell viability and migration tests, and improved bone trabeculae and density in osteoporotic rats, according to micro-CT imaging and H&E staining. Biochemical markers (BGP, IL-6, CTX-I, TRAP, OPG, and PINP) indicated that RIS-BMNs had anti-osteoporotic effects comparable to oral RIS but without gastrointestinal side effects, as confirmed by H&E staining. Our new transdermal method for RIS could improve adherence in treating postmenopausal osteoporosis.

Guofeng Zhong,Mengyu Qiu,Junbo Zhang,Fuchen Jiang,Xuan Yue,Chi Huang,Shiyi Zhao,Rui Zeng,Chen Zhang,Yan Qu

In this study, a novel nanofiber material with Polylactic acid (PLA), natural plant polysaccharides- Bletilla striata polysaccharide (BSP) and Rosmarinic acid (RA) as the raw materials to facilitate wound healing was well prepared through coaxial electrospinning. The morphology of RA-BSP-PVA@PLA nanofibers was characterized through scanning electron microscopy (SEM), and the successful formation of core-shell structure was verified under confocal laser microscopy (CLSM) and Fourier transform infrared spectroscopy (FTIR). RA-BSP-PVA@PLA exhibited suitable air permeability for wound healing, as indicated by the result of the water vapor permeability (WVTR) study. The results of tension test results indicated the RA-BSP-PVA@PLA nanofiber exhibited excellent flexibility and better accommodates wounds. Moreover, the biocompatibility of RA-BSP-PVA@PLA was examined through MTT assay. Lastly, RA-BSP-PVA@PLA nanofibers can induce wound tissue growth, as verified by the rat dorsal skin wound models and tissue sections. Furthermore, RA-BSP-PVA@PLA can facilitate the proliferation and transformation of early wound macrophages, and down-regulate MPO + expression of on the wound, thus facilitating wound healing, as confirmed by the result of immunohistochemical. Thus, RA-BSP-PVA@PLA nanofibers show great potential as wound dressings in wound healing.

In vitro digestion and human gut microbiota fermentation of Bletilla striata polysaccharides and oligosaccharides

Wang Qiqi,Chen Huimin,Yin Mingzhu,Cheng Xue,Xia Hui,Hu Haiming,Zheng Junping,Zhang Zhigang,Liu Hongtao

Background: Bletilla striata is one of the commonly used traditional Chinese medicine. B. striata polysaccharides (BP) and oligosaccharides (BO) are one of the main components of B. striata, which have been proved to have a variety of biological activities. However, the digestion and fermentation characteristics of BP and BO are still unclear. Methods: The study evaluated different prebiotic effects of BP and BO by in vitro simulating digestion and gut microbiota fermentation. Results: The results show that the simulating saliva partly degraded BP, but had no effect on BO. The molecular weights of BP and BO remained basically unchanged in gastric and intestinal digestion. In addition, BP and BO could be rapidly degraded and utilized by gut microbiota. During in vitro fermentation, the growth rates of the BP and BO groups were higher than that of the Control group and the pH value and total carbohydrate content in BP group and BO group decreased significantly. Although the reducing sugar level in the BO group decreased rapidly, it remained at a low level in the BP group. Both BP and BO improved the composition and structure of gut microbiota, indicative of the upregulated abundances of Streptococcus and Veillonella, and the downregulated populations of Escherichia and Bacteroides. There were differences in the SCFA production by gut microbiota and antioxidant activities between the BP and BO groups. The fermentation broth of the BP group displayed a stronger suppression of O2-, but a higher scavenging effect on DPPH for the BO group. Conclusions: BP and BO displayed different digestion and fermentation characteristics in vitro due to their distinct polymerization degrees. The study point towards the potential of BP and BO as prebiotics in the application to human diseases by selectively regulating gut microbiota in the future.

Bletilla striata polysaccharide/ethanol extract composite hydrogel for accelerated wound healing

Hu Zhengbo,Zhao Kai,Zhou Fangmei,Ye Xiaoqing,Chen Yuchi,Jin Lixia,Ruan Xinming,Ding Zhishan,Zhu Bingqi

The purpose of this study is to utilize the active ingredients of Bletilla striata ( B. striata ) to prepare the hydrogel dressing for accelerated wound healing. To this end, a series of composite hydrogels were prepared by blending different proportions of B. striata polysaccharide (BSP) and ethanol extract of B. striata (EEB) with Carbomer 940 (CBM940). These hydrogels showed porous three-dimensional structures with good water retention properties. The cytotoxicity test demonstrated the high cytocompatibility of the EEB/BSP hydrogels. Additionally, the inclusion of EEB enhanced the antibacterial and anti-inflammatory properties of EEB/BSP hydrogel. The animal experiment illustrated the accelerated wound healing effect using the hydrogels and the highest healing rate was obtained in EEB1/BSP7 (m EEB :m BSP  = 1:7) group with the 82.00 ± 4.25% and 93.30 ± 0.97% on 7th day and 10th day, respectively. Histological observations further confirmed the excellent healing effect of EEB/BSP hydrogel. Therefore, EEB/BSP hydrogel had potential to becoming a suitable wound dressing. Graphical abstract

Multifunctional structural color Chinese herb hydrogel patches for wound management

Xinyue Cao,Yu Wang,Xiangyi Wu,Jinglin Wang,Haozhen Ren,Yuanjin Zhao

Chinese herb hydrogel has outstanding values in wound treatment. Tremendous effort is dedicated to developing functional and intelligent Chinese herb hydrogels for accelerating healing process. Herein, a novel multifunctional bilayer Chinese herb hydrogel patch for wound healing is presented. The patches are composed of a Ginsenoside compound K (GCK)-integrated chitosan inverse opal scaffold layer and a Bletilla striata polysaccharide (BP) hydrogel filler layer. As the photothermal conversion capacity of BP can trigger faster degradation of its hydrogel layer under near-infrared (NIR) light, the encapsulated BP can be controllably released to wound area and exhibit photothermal antibacterial and antioxidant effects. Besides, the inverse opal scaffold layer shows great mechanical property and moisturizing performance, and its released GCK can promote angiogenesis for long term therapeutic effects. Attractively, due to responsive structural color, the patch is endowed with self-reporting performance, which can real-time visually monitor the drug release degree. Taking advantages of these properties, the patches have been demonstrated to achieve satisfying therapeutic effects for in vivo wound treatment. Therefore, we believe that the proposed Chinese herb hydrogel patches are valuable as multifunctional patches for clinical treatment.
