Coculture of the Pacific white shrimp Litopenaeus vannamei and the macroalga Ulva linza enhances their growth rates and functional properties

Guang Gao,Lin Gao,Qianqian Fu,Xinshu Li,Juntian Xu

Integrated multi-trophic aquaculture (IMTA) has been proposed as a potential solution to supply aquatic food in an environmentally friendly way. However, little is known regarding the impacts of IMTA on the growth rates and food quality of both animals and plants, as well as the seasonal effects. In this study, we conducted field mesocosm experiments using monoculture and coculture systems with the shrimp Litopenaeus vannamei and the macroalga Ulva linza for four weeks in different seasons (autumn, spring and summer) to address the research gap. To evaluate the environmental pollution caused by shrimp culture, there was no water exchange for culture systems including shrimps. Compared to monoculture of L. vannamei , coculture with U. linza significantly reduced dissolved inorganic nitrogen (DIN, 98.5–99.0%) and dissolved inorganic phosphorus (DIP, 98.2–98.8%) but enhanced dissolved oxygen (DO, 56.2–68.7%) and pH (10.7–18.6%) by the end of culture. Compared to monoculture, coculture stimulated the growth rates of L. vannamei by 38.1–58.8% and U. linza by 241.9–290.4% in all seasons, and increased the lipid content of L. vannamei by 24.2% in autumn and by 37.9% in summer and the contents of protein and ash of U. linza by 23.8–29.0% and by 27.6–68.6%, respectively, in all seasons. Coculture enhanced the content of most amino acids in U. linza and the content of total fatty acids (FA) and polyunsaturated fatty acids in L. vannamei in all seasons in comparison with monoculture. In addition, coculture lifted swelling capacity by 28.9–40.5%, water holding capacity by 39.8–43.3% and oil holding capacity by 31.4–32.4% for U. linza in autumn and summer. Apart from relieving eutrophication , deoxygenation and acidification, IMTA increased growth rates of both shrimp and Ulva and improved functional properties of U. linza , suggesting a green and productive aquaculture mode.

Inactivation effect and kinetics of cathepsin L from white shrimp (Litopenaeus vannamei) by dense phase carbon dioxide

Ouyang Zheng,Xiaojie Cao,Yanqing Teng,Qinxiu Sun,Shucheng Liu

The effects of dense phase carbon dioxide (DPCD) pressure and temperature on the activity of cathepsin L in white shrimp ( Litopenaeus vannamei ) were studied. Meanwhile the effects of DPCD and heat treatment on the activity of cathepsin L were compared. The results of inactivation kinetics showed that under the same treatment temperature, the inactivation effect of DPCD on cathepsin L increased gradually with the increase of pressure and time. The effect of DPCD pressure on the activity of cathepsin L accorded with the first-order kinetic model. Under the same treatment pressure, with the increase of temperature, the inactivation effect of cathepsin L was significant at the initial stage of DPCD treatment (rapid inactivation period), and decreased with the extension of time (stable inactivation period). The effect of DPCD treatment temperature on cathepsin L activity accorded with the first-order kinetic model at 35 °C, and two-stage kinetic model at 40–60 °C. The difference of relative enzyme activity between the two treatments showed that the dependence of DPCD on temperature was greater than that of heat. E a, F and E a, S of DPCD were higher than that of heat, which indicated that cathepsin L was more easily inactivated under DPCD treatment.

A novel c-type lysozyme from Litopenaeus vannamei exhibits potent antimicrobial activity

Feng Hu,Yan Wang,Jingjie Hu,Zhenmin Bao,Mengqiang Wang

Lysozyme is relevant to the innate immune system as a vital protein for crustaceans. In the present study, we cloned and characterized a novel c-type lysozyme gene ( Lv LYZ) from the Pacific white shrimp ( Litopenaeus vannamei ). The obtained full-length cDNA of Lv LYZ was 990 bp and contained an open reading frame of 693 bp. Its deduced amino acid sequence consisted of 230 amino acids (aa) with a 17 aa signal peptide at the N-terminal and 130 aa functional domains. The multiple sequence alignment (MSA) indicated that the typical active sites in Lv LYZ were similarly conserved as c-type lysozymes from other species. The transcription of Lv LYZ appeared in all detected tissues and had relatively higher expression levels in hemocytes , hepatopancreas , gill and intestine. The mRNA expression profiles of Lv LYZ were up-regulated in hemocyte and hepatopancreas post the stimulation of Vibrio parahaemolyticus or white spot syndrome virus (WSSV), respectively. The recombinant protein of Lv LYZ (r Lv LYZ) exhibited antibacterial activities against various microbes, including Escherichia coli , Vibrio splendidus , Micrococcaus luteus , Vibrio parahaemolyticus and Staphylococcus aureus . These results indicated that Lv LYZ could cope with bacteria in L. vannamei and may play a significant role in immune response against invading pathogens.

Anesthetic effects of eugenol on adult ridgetail white prawn (Exopalaemon carinicauda) and its protective effects under simulated transportation

Chengsong Zhang,Fuhua Li

Anesthesia is a common practice used to improve animal welfare and reduce the negative impacts of stress factors in aquaculture settings. However, few studies have been conducted to investigate the application of anesthetics in ridgetail white prawn ( Exopalaemon carinicauda ). In the present study, the anesthetic efficacy and protective effects of eugenol in adults of this species were evaluated. When the prawns ( n  = 5) were immersed in different concentrations of eugenol in the range of 0–240 μL/L for 30 min, the results showed that 180 and 240 μL/L could lead to quick sedative and anesthesia effects with 100% mortality after 30 min exposure, while 30 μL/L failed to induce any sedative and anesthesia effects. Further tests showed that adult individuals were quickly anesthetized by eugenol at a concentration of 60–90 μL/L with a 30 min immersion, while half of the prawns were sedated in 30 μL/L eugenol after 4 h exposure, then recovered at 2 h without any mortality. Furthermore, we checked the water quality parameters after the adult prawns were immersed in four eugenol concentrations (0, 10, 20, 30 μL/L) for 8 h of simulated transportation followed by a 96 h recovery, we found that 30 μL/L eugenol treatment could significantly decrease the ammonia nitrogen concentration , and increased the survival rate at 96 h post recovery. The oxidative responses of the prawns during the 8 h transportation at 30 μL/L eugenol followed by 96 h recovery period were analyzed. The results showed that eugenol administration significantly decreased the malonaldehyde (MDA) concentration and the activities of most antioxidant-related enzymes immediately. The present data indicated that eugenol was an efficient anesthetic, which could be used for long-distance transportation of this species. It exhibited excellent protective effects to improve the animal's welfare by improving water quality and alleviating oxidative stress .

Shelf-life extension of Pacific white shrimp (Litopenaeus vannamei) using sodium alginate/chitosan incorporated with cell-free supernatant of Streptococcus thermophilus FUA 329 during cold storage

Shuangshuang Zhao,Yingying Bian,Gewen Zhang,Guang Yang,Xiaoyue Hou,Jiajin Gui,Shuting Mu,Shu Liu,Yaowei Fang

Seafood is highly perishable and has a short shelf-life. This study investigated the effect of chitosan and alginate (CH-SA) coating combined with the cell-free supernatant of Streptococcus thermophilus FUA329 (CFS) as a preservative on the quailty of white shrimp (Litopenaeus vannamei) refrigerated at 4° for 0, 3, 6, 9, 12, 15 days. Freshly shrimps were randomly divided into four groups: the CFS group (400 mL); the CH-SA group (1% chitosan/1% alginate); the CFS-CH-SA group (1% chitosan/1% alginate with 400 mL CFS) are treatment groups, and the control group (400 mL sterile water). The CFS–CH–SA coating effectively suppressed microbial growth total viable count and chemical accumulation (pH, total volatile basic nitrogen, thiobarbituric acid reactive substance) compared with the control. Additionally, the CFS–CH–SA coating improved the texture and sensory characteristics of shrimp during storage. The coated shrimp exhibited significantly reduced water loss ( p < 0.05). The combination of CH–SA coating with CFS treatment can extend the shelf life of shrimp. Practical Application Recently, edible films have received more consideration as a promising method to enhance the shelf life of seafood. The presence of Lactic acid bacteria metabolites in edible films reduces spoilage and improves consumer health. Our findings encourage the application of edible coating incorporated with cell-free supernatant of Streptococcus thermophilus FUA 329 to design multifubctional foods and preserve the qualities of shrimp.
