Adsorption of non-systemic pesticide chlorothalonil on grape cuticles and dewaxed grape cuticles: elucidating the role of wax

Huihui Wang,Yuhang Du,Aying Wen,Shaofeng Yuan,Hang Yu,Weirong Yao

BACKGROUND Cuticular wax is vital for preventing pesticide migration into fruit tissue, yet interactions between pesticides and wax components remain unclear. This study investigated the adsorption of non-systemic pesticide chlorothalonil on grape cuticles and dewaxed grape cuticles to quantify wax contributions and elucidate the interaction between chlorothalonil and wax components. RESULTS The wax was distributed on the surface of the grape cuticles with higher lipophilicity. The distinct chlorothalonil adsorption behaviors on grape cuticles (featuring heterogeneous and multilayer chemical adsorption) and dewaxed grape cuticles (featuring homogeneous and monolayer physical adsorption) were attributed to the presence of wax. Wax accounted for 40.36% of the total grape cuticle content and contributed 67.47% to the chlorothalonil adsorption capacity, indicating that wax was the main site for chlorothalonil adsorption. Through wax analysis and molecular docking, hexadecanoic acid and octadecanoic acid were considered as the major contributors, primarily engaging in hydrophobic interactions with chlorothalonil. Furthermore, gradual fluorescence quenching of chlorothalonil was observed with increasing concentrations of these acids (both below saturation concentrations, r = 0.99), confirming the intermolecular interaction between chlorothalonil and these acids. CONCLUSION The study clarified the residue behavior in fruit cuticles and the interaction between pesticides and cuticular wax. The findings provide important insights for optimizing pesticide application strategies to reduce pesticide residues in fruits and improve pest management efficiency. © 2025 Society of Chemical Industry.

Development of leaf-adhesive polyurethane microcapsules loaded with prochloraz for enhanced fungicide efficiency

Peng Xu,Dongmei Yu,Weishan Shi,Gang Xing,Ao Wang,Mi Tang,Jiawei Bao,Zhaogang Teng,Dejun Hao

Leaf-adhesive pesticide microcapsules represent a promising approach to improving pesticide efficacy while minimizing environmental pollution. In this study, we developed a leaf-adhesive polyurethane microcapsule loaded with prochloraz (Pro@CS-OP-10) using a reactive emulsifier (OP-10) via interfacial polymerization. Physicochemical analysis revealed microcapsules with smooth surfaces and uniform distribution, averaging 1.97 μm in size, with an encapsulation efficiency of 96.38 %. Comparative assessments demonstrated superior wetting and adhesion properties of Pro@CS-OP-10 on wheat leaves compared to other commercial formulations. Moreover, the polyurethane shell effectively enhanced the photostability of encapsulated prochloraz and hindered its rapid release. Fungal growth rate experiments indicated prolonged inhibitory effects of Pro@CS-OP-10 on Fusarium graminearum , with median inhibitory concentrations (EC 50 ) on the second and eighth day being 0.018 mg/L and 0.045 mg/L. Therefore, the leaf-adhesive Pro@CS-OP-10 shows great potential as an environmentally friendly fungicidal formulation.

Selective extraction of fungicides from fruit samples with defective UiO-66 as solid-phase microextraction fiber coating

Yang Li,Xiaohuan Zang,Yahuan Li,Shuaihua Zhang,Chun Wang,Zhi Wang

A defective UiO-66 with rich defect sites was synthesized by a modulator-induced defect formation approach and used as the solid-phase microextraction (SPME) fiber coating material. The extraction experimental results showed that the defective UiO-66 coating can selectively extract the fungicides chlorothalonil, vinclozolin, dimethachlon, procymidone and iprodione with a high adsorption capability. The superior adsorption capability can be attributed to hydrophobic, Lewis acid-base and hydrogen bonding interactions. After optimization, a SPME with the defective UiO-66 as the fiber coating combined with GC–MS detection was established for the sensitive and selective determination of the five fungicides in fruit samples. Under the optimized experimental conditions, the quantitative linear response was in the range of 0.83–300 ng g −1 , with the coefficients of determination in the range from 0.9970 to 0.9986 and the limits of detection between 0.25 and 9.0 ng g −1 for the analytes. The method was successfully applied for the determination of the fungicides in different fruit samples. Satisfactory method recoveries (82.7%-117.7%) and good precisions with the relative standard deviations being from 3.2% to 12% were achieved.

Density-adjusted liquid-phase microextraction with smartphone digital image colorimetry to determine parathion-methyl in water, fruit juice, vinegar, and fermented liquor

Xinyuan Bi,Haijuan Jiang,Xingle Guo,Min Wang,Yu Niu,Liyan Jia,Xu Jing

To achieve rapid and convenient on-site pretreatment and determination of parathion-methyl, a density-adjusted liquid-phase microextraction with smartphone digital image colorimetry was established to detect parathion-methyl in food samples. In this study, the environmentally friendly biomass-derived solvent guaiacol was used as the extractant. Salt and water, as density regulators, realized the two movements (floating–sinking) of the extractant and full contact between the extractant and the sample solution to establish an environmentally friendly, fast, and efficient pretreatment method. Under strong alkaline conditions, parathion-methyl generated a yellow product; then, a smartphone was used to obtain the image of the yellow product for intensity analysis. Parathion-methyl has a good linear relationship in the range of 0.01–1 mg L−1, and the limits of detection and quantification are 0.003 and 0.01 mg L−1, respectively. This method has been successfully applied to the determination of parathion-methyl in spiked water, fruit juice, vinegar, and fermented liquor with a recovery of 91.6–106.5% and a relative standard deviation of 0.6–6.0%. The established density-adjusted liquid phase microextraction with smartphone digital image colorimetry is rapid, convenient, and environmentally friendly for the determination of parathion-methyl in food samples.

Zhenpeng Men,Ragini Singh,Qi Zhang,Huibo Han,Sandeep Boddu,Jyothsna Vaadaala,Yesudasu Vasimalla,Bingyuan Zhang,Santosh Kumar

In this study, an ultrasensitive, and glyceraldehyde-3-phosphate dehydrogenase (GTP)-modified gold nanoparticles (AuNPs)-based localized surface plasmon resonance (LSPR) sensor for detection of chlorothalonil (CTL) in environmental and food safety applications. As a widely used fungicide, CTL plays an important role in agricultural production. However, its residues pose a potential threat to environmental ecology and human health. Therefore, it is particularly urgent and significant to develop an efficient and sensitive detection method for CTL. In this study, an innovative detection method of CTL is proposed based on LSPR sensor. A highly sensitive LSPR sensor was successfully constructed by carefully synthesizing AuNPs and modifying them with GTP. The sensor demonstrates high sensitivity, evidenced by its low limit of detection (LoD) for CTL and exhibits excellent selectivity and stability. More importantly, the recovery rate of the detection method in actual samples reached more than 90 %, which fully proved its broad application prospects in the field of environmental monitoring and food safety. The linear range of the sensor is 0-1000 μM, and the LoD is 75.41 μM. The sensitivity is as high as 0.0037 nm/(μM), which verifies that the sensor provides an indispensable technology and a cost-effective and portable solution for CTL-specific detection.

Genistein: A promising botanical fungicide candidate for enhancing tomato yield and quality by controlling Alternaria solani

Ling Zhang,Yu Long,Long-Fei Li,Xiang-Wei Xin,Xiao-Di Chen,Xiang Zhou,Li-Wei Liu,Zhi-Bing Wu,Song Yang

The overreliance on traditional chemical fungicides, combined with the emergence of resistance, poses significant challenges for food safety. Early blight, caused by the fungal pathogen Alternaria solani ( A. solani ), is among the most significant contributors to pre- and postharvest yield losses in tomato cultivation. To address this challenge, through evaluation of the antifungal activity of 52 commercially available natural products against A. solani , we revealed for the first time that the naturally occurring compound genistein exhibited potent fungicidal activity against A. solani , with an EC 50 value of 6.53 μg/mL, surpassing that of chlorothalonil (EC 50  = 8.35 μg/mL). In vivo assays demonstrated that genistein exhibited strong protective (48.1 %) and therapeutic (42.5 %) efficacy, effectively safeguarding tomatoes from fungal infection. Mechanistic investigations, including transcriptomic analysis, scanning electron microscopy, fluorescence imaging, and measurements of malondialdehyde and ATP levels, collectively indicated that genistein disrupted energy metabolism, leading to reactive oxygen species (ROS) accumulation. This oxidative stress cascade triggered both apoptotic-like responses and membrane lipid peroxidation, ultimately inhibiting the growth of A. solani . Interestingly, genistein showed enhanced anti- A. solani activity when used in combination with either chlorothalonil or tebuconazole. Additionally, genistein displayed low phytotoxicity and favorable drug-likeness properties. Taken together, these findings elucidate the mechanisms underlying genistein's antifungal activity against A. solani , underscoring its promise as a natural antifungal agent for enhancing food safety and improving tomato yield and quality. Furthermore, this study provides valuable insights for the rational design of flavonoid-based antifungal agents.

Captan exposure disrupts ovarian homeostasis and affects oocytes quality via mitochondrial dysfunction induced apoptosis

Quan-Kuo He,Chang-Long Xu,Yan-Ping Li,Zhi-Ran Xu,Yu-Shen Luo,Si-Cheng Zhao,Hai-Long Wang,Zhong-Quan Qi,Yu Liu

Captan is a non-systematic fungicide widely used in agricultural production, and its residues have been found in the environment and daily diet. Previous studies confirmed that captan exerts several toxic effects on tissues, but its effect on the mammalian female reproductive system is unclear. In current study, we reported that captan affected mouse ovarian homeostasis and disrupted female hormone receptor expression, leading to impaired follicular development. Ovarian follicles from the captan exposure group showed an increased level of inflammation, endoplasmic reticulum stress and apoptosis. In addition, captan exposure disrupted oocyte development. Transcriptomic analysis indicated that captan changed multiple genes expression in oocytes, including autophagy and apoptosis. Further molecular testing showed that captan induced oxidative stress and mitochondrial dysfunction, as indicated by the increased level of reactive oxygen species , disrupted mitochondrial structure and distribution, and depolarized membrane potential. Furthermore, captan triggered DNA damage, autophagy and early apoptosis, as shown by the enhanced levels of γ-H2AX, LC3, and Annexin-V and increased expression of related genes. Taken together, these results indicated that captan exposure impairs ovarian homeostasis and subsequently affects oocyte development.

Electrochemical Sensing of Organophosphorus Pesticides Based on Plant Esterase-Triggered Silver Deposition with CuNi-DAC Signal Amplification

Longying Zhao,Jiayu Chen,Donglin Xiang,Mierban Dawuti,Laping He,Yuangen Wu,Han Tao

Residual organophosphorus pesticides (OPs) in food pose a severe threat to public health, necessitating simple and sensitive on-site analytical methods. In this study, a signal-off electrochemical sensing platform for OPs was constructed based on the OP-induced inhibition of silver deposition triggered by red kidney bean esterase (RKBE), coupled with the signal amplification of a CuNi dual-atom catalyst (CuNi-DAC). RKBE catalyzes the hydrolysis of α-naphthyl acetate to produce α-naphthol, which reduces Ag⁺ to Ag 0 deposited on the electrode. The electroactive Ag 0 undergoes oxidation to generate a prominent oxidation current, providing a sensitive sensing signal for detection. CuNi-DAC modification of the electrode accelerates charge transfer, provides more active sites, and effectively amplifies the sensing signal. Upon exposure to OPs, the activity of RKBE is inhibited, thereby suppressing the production of α-naphthol. This leads to a diminished Ag 0 generation and subsequent a weakened oxidation current. The decrease degree of current signal shows a positive correlation with pesticide concentration. Under optimized conditions, the fabricated sensing platform exhibits a good linear response range of 10 pmol/L-1000 pmol/L for fenthion with a low detection limit of 5.4 pmol/L. Spiked recoveries in cabbage and apple samples ranged from 93.3% to 108.5%, validating the sensing platform's practicality and reliability. This work offers a facile and cost-effective electrochemical sensing approach for on-site screening of OPs.

Effects of silver nanoparticles and nitrile fungicides on soil microbial community structure, metabolic functions, and resistance genes

Yiwang Wang,Ting Gao,Jiajia Liu,Shaoheng Bao,Fuli Wang,Ge Yao,Xiaoyan Lin,Shijie Jiang,Jinlong Lai,Penggang Han,Jiang Hui

Nitrile fungicides and silver nanoparticles (AgNPs) are increasingly applied in agriculture, yet their synergistic toxicity to soil ecosystems remains unclear. This study investigated the individual and combined effects of AgNPs and three nitrile fungicides—azoxystrobin, flufenacet, and chlorothalonil—on soil microbial communities, metabolic activities, antibiotic resistance genes, metal resistance genes, and plant physiology. The results showed that the synergistic toxicity of AgNPs with the three nitrile fungicides induced oxidative stress in the maize to varying degrees. Nitrile fungicides alone increased sucrase activity, while AgNPs alone suppressed urease, sucrase, and acid phosphatase. Both treatments promoted dominance of Pseudomonadota , Bacteroidota , and Sphingopyxis . AgNPs specifically enriched Proteobacteria and Myxococcus , causing carbon/nitrogen utilization impediments partially mitigated by nitrile biodegradation. Synergistic exposure redirected nitrile catabolism from the benzoyl-CoA to catechol pathway. AgNPs alone upregulated silver resistance genes and activated antibiotic efflux pumps; synergistic exposure with fungicides suppressed Ag + efflux genes while exacerbating antibiotic efflux activation and weakening links between environmental factors (NH₄ + -N, NO₃ − -N, Ag + ) and resistance genes. These findings reveal the responses of plants and soil microorganisms to co-exposure of nitrile fungicides and AgNPs, and provide insights for assessing the ecological risks of combined application of nanomaterials and pesticides.

An electrochemical sensing strategy for organophosphorus pesticides based on dual-enzyme synergistic catalysis of plant esterase and diatomic FeCo–NC nanozyme

Donglin Xiang,Xiao Wang,Chun Ji,Laping He,Yuangen Wu,Han Tao

This study presents a novel electrochemical strategy for detecting organophosphorus pesticides based on the synergistic catalytic of red kidney bean esterase (RKBE) and iron–cobalt dual-atom catalyst embedded in N-doped carbon carrier (FeCo–NC). RKBE was extracted from the widely cultivated British red kidney beans ( Phaseolus vulgaris L.) . FeCo–NC exhibited excellent electrocatalytic and peroxidase-like activity, catalyzing o -phenylenediamine oxidation to produce electroactive 2,3-diaminophenazine, which served as signal probe generating a sensitive cathodic current. Notably, RKBE product 1-naphthol inhibited FeCo–NC activity, while organophosphorus pesticides counteracted this inhibition by suppressing RKBE activity, restoring the electrochemical signal of 2,3-diaminophenazine. Leveraging this signal-regulation mechanism, a dichlorvos sensor was developed, with wide linear range (50–50,000 pM), low detection limit (14 pM), and excellent real-sample recovery (93.33%–107.08%), demonstrating practical application potential. This study provides an affordable and easy-to-operate approach for large-scale on-site monitoring of organophosphorus pesticide residues in foods.

A design of thermal desorption unit for ultra-sensitive detection of compounds by ion mobility spectrometry

Leila Mohammadipour,Mohammad R. Rezayat,Peng Li,Mohammad T. Jafari,Lingfeng Li,Lihui Huang

A solvent-free, low-dead-volume design of a thermal desorption unit (TDU) was developed as an introduction system to significantly improve the performance of handheld ion mobility spectrometry (IMS). The TDU configuration eliminates solvent interference using solvent-free desorption, enhancing the sensitivity of IMS analysis and preventing signal overlap between the analyte and the solvents. Furthermore, the dead volume of the introduced TDU was decreased to diminish the peak broadening to a few seconds. The major advantage is the ability to maintain a uniform temperature within the TDU to avoid analyte condensation toward IMS. Furthermore, the designed platform was validated to analyze compounds with diverse physicochemical properties, including pesticides, drugs, and explosives. To demonstrate the potential of the TDU in real sample analysis, chlorothalonil (CTL) was chosen and determined as a test compound using the thin film microextraction method followed by IMS. Metal organic framework-808@5-nitroisophthalic acid (NIP) was synthesized by a solvothermal procedure, and electrospinning was applied to prepare the thin film. Several experimental factors influencing the extraction efficiency and signal of CTL were identified and optimized. The linearity, limit of detection, and limit of quantification were obtained to be 0.3-10 ng mL −1 , 0.09 ng mL −1 , and 0.3 ng mL −1 , respectively. The intra- and inter-day repeatabilities were calculated as 2% and 6% for 0.5 ng mL −1 , and as 3% and 7% for 5.0 ng mL −1 . Several real samples, consisting of tomato, cucumber, and lake water, were chosen to assess the performance of the suggested approach. The spiking recovery values ranged from 98% to 107%. Additionally, the accuracy of the proposed approach was evaluated using the EPA 508 reference method.

Construction of CuNCs/AgNPs based fluorescent platform for specific enzyme-free detection of trichlorfon

Songrui Li,Fubin Yang,Junying Li,Yongcun Zou,Qiong Jia

Background As a common organophosphorus pesticide, trichlorfon (TC) is being extensively used in agriculture due to its high efficiency in plant diseases and insect pest control. However, TC should be monitored in a timely manner due to its severe threat to public health. Currently reported fluorescence methods for TC detection largely depend on enzyme inhibition effect, which suffers from their intrinsic defects such as poor selectivity, complexity, or high cost. Hence, it is of essential value to develop a facile and low-cost fluorescence approach to achieve specific enzyme-free detection of TC. Results In this work, L-cysteine protected copper nanoclusters (CuNCs) with good stability were prepared as the fluorescence sensing probe. Under alkaline conditions, TC tends to be hydrolyzed to produce dichloroacetaldehyde, which was directly employed as an effective reducing agent to in-situ synthesize silver nanoparticles (AgNPs). The fluorescence of L-cys-CuNCs was quenched by AgNPs through the inner filter effect (IFE), realizing specific enzyme-free determination of TC. Besides, portable and visual detection of TC was achieved depending on a smartphone sensing platform. This method was further employed to monitor TC in cherry tomatoes, potatoes, and cultivated soil samples with satisfying results, illustrating its high potential in monitoring TC in real samples. Overall, this method possesses the attractive superiorities of simplicity, cost efficiency, and specificity for the detection of TC. Significance The strategy has the following advantages. (i) Specific enzyme-free detection of TC was achieved based on CuNCs/AgNPs fluorescent platform. (ii) AgNPs, as CuNCs fluorescence quencher, was directly in-situ synthesized using the hydrolysis products of TC, which avoids adding extra reducing agent and provides a possibility for the waste utilization of TC. (iii) A portable and low-cost smartphone sensing platform was constructed for visually specific determination of TC.
