Hyaluronic acid-liposomes hybridized with HucMSC exosomes for enhanced exosomes transdermal delivery and acute skin photodamage repair

Yuhui Yang,Yong He,Hui Xing,Ziyi Zhao,Jianjin Wang,Shanying Li,Xiaosong Xu,Dong Ma,Yunfeng Hu

Long-term exposure to ultraviolet (UV) radiation can damage human skin, resulting in photodamage. Repairing photodamaged skin has been a major focus of research in recent years. Extensive research has shown that human umbilical cord mesenchymal stem cell-derived exosomes (Exo) possess anti-inflammatory, pro-angiogenic, and wound healing properties, holding great potential for treating skin damage. However, due to the limitations of exosomes alone, such as poor transdermal penetration, instability, and low utilization, there is an urgent need for new delivery strategies. We designed a hybrid nanovesicle (HL@Exo) by combining ultrasonic incubation with membrane extrusion to fuse Exo with HL. HL@Exo capitalizes on the advantages of liposomal carriers and the permeation-enhancing properties of hyaluronic acid to effectively facilitate transdermal delivery of Exo. The successful fusion of HL@Exo and its skin penetration were verified by methods such as fluorescent labeling, western blotting, Transwell assays, and in vivo imaging. In vitro studies on photodamaged keratinocytes and endothelial cells demonstrated HL@Exo ability to promote cell proliferation, repair, angiogenesis, and reduce inflammation. In a laser-induced skin photodamage model, HL@Exo enhanced collagen regeneration, accelerated wound healing, and demonstrated significant anti-inflammatory effects, indicating its potential as a non-invasive treatment and offering a novel strategy for the clinical application of Exo.

Preparation and characterisation of a novel silk fibroin/hyaluronic acid/sodium alginate scaffold for skin repair

Wei Yang,Hongjie Xu,Yong Lan,Qiyu Zhu,Yu Liu,Shaoshan Huang,Shengjun Shi,Andrei Hancharou,Bing Tang,Rui Guo

To mimic the natural structure of tissue extracellular matrix, a novel silk fibroin (SF)/hyaluronic acid (HA)/sodium alginate (SA) composite scaffold (92% in porosity) was prepared by freeze-drying. Fourier-transform infrared spectroscopy and Raman spectra indicated interactions among SF, HA, and SA molecules. Scanning electron microscopy showed that the prepared SF/HA/SA scaffold had soft, elastic characteristics, with an average pore diameter of 93 μm. Mechanical property, thermogravimetric analyses and degradation results indicated that the SF/HA/SA scaffold had good physical stability in body fluid and mechanical movement-related environments. Cell proliferation, morphological, and live-dead analyses showed that NIH-3T3 fibroblast cells were better able to attach, grow, and proliferate on the SF/HA/SA scaffold compared with SF, SF/HA, and SF/SA scaffolds. We evaluated the wound healing effects in a rat full-thickness burn model. The hematoxylin-eosin (H&E) and Masson's trichrome staining results from SF/HA/SA scaffold showed that improved re-epithelialization, enhanced extracellular matrix remodeling. Our findings showed that the prepared SF/HA/SA scaffold can provide a potential way as a wound dressing for skin repair.

Fabrication of a MXene-based shape-memory hydrogel and its application in the wound repair of skin

Jingchuan Hu,Jun Xie,Tao Peng,Qingwen Shi,Chen Pan,Haiying Tan,Jiuxiao Sun

Wound dressings can generally complete hemostasis and provide temporary protection after skin damage. Herein, a MXene-based hydrogel was prepared from MXene, gelatin, poly(ethylene glycol)diacrylate (PEGDA) and N,N′-methylenebis(acrylamide) (HEAA) to prepare wound-dressing hydrogels for skin repair. HEAA and PEGDA crosslink polymerization formed the first layer of the network. Hydrogen bonds between MXene, PHEAA, and gelatin formed the second layer of the network. To make the hydrogel more suitable for skin repair, the mechanical properties of the hybrid hydrogel were adjusted. The MXene-based hydrogel could recover its original shape in 16 s upon immersion in water or for a few minutes under light irradiation. The obtained hydrogel showed good photothermal properties upon light irradiation (808 nm, 1 W cm−2) for 20 s, and its temperature on the surface could reach 86.4 °C. Due to its good photothermal properties, this MXene-based hydrogel was suitable for skin repair.

Photo-crosslinkable amniotic membrane hydrogel for skin defect healing

Qiang Zhang,Caiwang Chang,Chunyu Qian,Wanshu Xiao,Huajun Zhu,Jun Guo,Zhibing Meng,Wenguo Cui,Zili Ge

The human amniotic membrane (HAM) collagen matrix derived from human placenta can be decellularized (dHAM) to form a natural biocompatible material. dHAM has different bioactive substances and has been used widely in human tissue engineering research. However, dHAM has some disadvantages, e.g., poor mechanical properties , easy degradation and inconvenient operation and use, so it is not conducive to large-area or full-thickness skin defect healing. To overcome these limitations, for the first time, dHAM was grafted with methacrylic anhydride (MA) to form photocrosslinked dHAM methacrylate (dHAMMA); dHAMMA was then blended with methacrylated gelatin (GelMA), followed by the addition of a photosensitizer for photocrosslinking to obtain the fast-forming GelMA-dHAMMA composite hydrogel. Further, GelMA-dHAMMA was found to have the porous structure of a bicomponent polymer network and good physical and chemical properties. In vitro experiments, GelMA-dHAMMA was found to promote fibroblast proliferation and α-smooth muscle actin (α-SMA) expression. In vivo investigations also demonstrated that GelMA-dHAMMA promotes wound collagen deposition and angiogenesis , and accelerates tissue healing. GelMA-dHAMMA inherits the good mechanical properties of GelMA and maintains the biological activity of the amniotic membrane, promoting the reconstruction and regeneration of skin wounds. Thus, GelMA-dHAMMA can serve as a promising biomaterial in skin tissue engineering. Statement of significance Since the early 20th century, the human amniotic membrane (HAM) has been successfully used for trauma treatment and reconstruction purposes. dHAM has different bioactive substances and has been used widely in human tissue-engineering research. In this work, the dHAM and gelatin were grafted and modified by using methacrylic anhydride (MA) to form photocrosslinked dHAMMA and methacrylated gelatin (GelMA). Then, the dHAMMA and GelMA were blended with a photosensitizer to form the GelMA-dHAMMA composite hydrogel derived from gelatin-dHAM. GelMA-dHAMMA exhibits a bicomponent-network (BCN) interpenetrating structure. dHAM dydrogel has advantages, e.g., good mechanical properties, slow degradation and convenient operation, so it is conducive to large-area or full-thickness skin defect healing.

Design and evaluation of transdermal nano collagen dressings for enhanced healing of UV-damaged skin

Shuangni Shi,Jingting Zhang,Yi Yang,Biyang Ling,Linyan Yao,Jianxi Xiao

Excessive UV radiation is one of the most common factors contributing to skin damage, manifesting as desquamation, inflammation, and even the development of skin cancer. Collagen has been developed for the treatment of photodamaged skin; however, the risk of pathogen transmission and its limited ability to penetrate the skin barrier have significantly restricted its clinical applications. Transdermal nano collagen, a type of short synthetic collagen, has attracted growing attentions for its ability to mimic the structure and function of natural collagen, while its applications in skin repair dressings remain unexplored. Herein, we have for the first time developed a novel type of transdermal nano collagen dressing (TNCD) for accelerated healing of photodamaged skin. The TNCD was formulated using the transdermal nano collagen, GRR2, as the efficacious ingredient, exhibiting highly stable physical and chemical properties. DermaLab Combo evaluations revealed that, compared to commercial collagen dressings, TNCD significantly accelerated the recovery of skin density, erythema index, and transepidermal water loss (TEWL) in damaged skin to levels comparable with healthy skin. Histological analyses further confirmed that TNCD effectively repaired photodamaged skin by promoting the regeneration of collagen fibers. Transdermal nano collagen dressings present promising strategies for improving collagen absorption through the skin and promoting wound healing, with significant market potential in the field of advanced skincare products.

Green Agarose Hydrogel Implants With High Biocompatibility for Skin Repair and Regeneration in Photoaged Skin

Ying Guo,Bei Tian,Yi Xie,Xiuxia Sun,Jianxi Xiao

Skin aging is a multifaceted process marked by the destruction of skin structure and the diminishment of skin function, significantly impacting both physical and mental health. Injectable hydrogels are promising for skin repair, but chemical crosslinking in most hydrogels can cause cytotoxicity, whereas agarose hydrogels avoid crosslinking yet face injectability challenges. We have herein developed green agarose hydrogel implants with high biocompatibility for skin repair and regeneration in photoaged skin. The hydrogel is prepared by utilizing agarose's ability to dissolve at high temperatures and gel at low temperatures. This process yields implants with particle sizes predominantly ranging from 100 to 150 μm. The agarose hydrogel implant exhibits excellent injectability, with a steady injection force curve consistently around 4 N. The agarose hydrogel implant promotes the proliferation of human dermal fibroblasts and does not induce hemolysis or pyrogenic reactions. In a photoaging mouse model, the agarose hydrogel implant improves skin density and hydration, reduces transepidermal water loss, and stimulates collagen regeneration. This agarose hydrogel implant provides a novel approach to repairing aged skin and holds significant potential in the fields of skin health and tissue regeneration. Graphical

Identification of the Wound Healing Activity Peptidome of Edible Bird’s Nest Protein Hydrolysate and the In Silico Evaluation of Its Transport and Absorption Potential in Skin

Min Wang,Yinglu Qing,Qunyan Fan,Shan Li,Tao Yan,William Kwame Amakye,Yongzhao Xu,Xuncai Liu,Jiaoyan Ren

In this study, edible bird’s nest (EBN) was proven to be a suitable source of bioactive peptides via enzymatic hydrolysis. The ultrafiltration component of the EBN peptides (EBNPs, Mw < 3 000 Da) could be responsible for moderate moisture retention and filaggrin synthesis. It was found that EBNP had a great capacity to protect HaCaT keratinocytes from DNA damage caused by UVB-irradiation and enhance wound healing by increasing the migratory and proliferative potential of cells. Furthermore, the external application of EBNP could effectively repair high glycolic acid concentration-induced skin burns in mice. A total of 1 188 peptides, predominantly the hydrophobic amino acids (e.g., Leu, Val, Tyr, Phe), were identified in the EBNP by liquid chromatography with tandem mass spectrometry (LC-MS/MS). Molecular docking showed that hydrophobic tripeptides from EBNP had a good binding affinity to proton-dependent oligopeptide transporter PepT1. Our data indicated that the hydrophobic amino acid-rich EBNP plays an important role in skin wound healing.

High molecular weight hyaluronic acid-liposome delivery system for efficient transdermal treatment of acute and chronic skin photodamage

Hui Xing,Xiangjun Pan,Yihan Hu,Yuhui Yang,Ziyi Zhao,Huanqi Peng,Jianjin Wang,Shanying Li,Yunfeng Hu,Guowei Li,Dong Ma

Photodamage is one of the most common causes of skin injury. High molecular weight hyaluronic acid (HHA) has shown immense potential in the treatment of skin photodamage by virtue of its anti-inflammatory, reparative, and antioxidative properties. However, due to its large molecular structure of HHA, HHA solution could only form a protective film on the skin surface in conventional application, failing to effectively penetrate the skin, which necessitates the development of new delivery strategies. Liposomes , with a structure similar to biological membranes, have garnered extensive attention as transdermal drug delivery carriers because of their advantages in permeability, dermal compatibility, and biosafety. Herein, we have developed a HHA-liposome transdermal system (HHL) by embedding HHA into the liposome structure using reverse evaporation, high-speed homogenization, and micro-jet techniques. The effective penetration and long-term residence of HHA in skin tissue were multidimensionally verified, and the kinetics of HHA in the skin were extensively studied. Moreover, it was demonstrated that HHL significantly strengthened the activity of human keratinocytes and effectively inhibits photo-induced cellular aging in vitro . Furthermore, a murine model of acute skin injury induced by laser ablation was established, where the transdermal system showed significant anti-inflammatory and immunosuppressive properties, promoting skin proliferation and scar repair, thereby demonstrating immense potential in accelerating skin wound healing . Meanwhile, HHL significantly ameliorated skin barrier dysfunction caused by simulated sunlight exposure, inhibited skin erythema, inflammatory responses, and oxidative stress, and promoted collagen expression in a chronic photodamage skin model. Therefore, this transdermal delivery system with biocompatibility represents a promising new strategy for the non-invasive application of HHA in skin photodamage, revealing the significant potential for clinical translation and broad application prospects. Statement of significance The transdermal system utilizing hyaluronic acid-based liposomes enhances skin permeability and retains high molecular weight hyaluronic acid (HHL). In vitro experiments with human keratinocytes demonstrate significant skin repair effects of HHL and its effective inhibition of cellular aging. In an acute photodamage model, HHL exhibits stronger anti-inflammatory and immunosuppressive properties, promoting skin proliferation and scar repair. In a chronic photodamage model, HHL significantly improves skin barrier dysfunction, reduces oxidative stress induced by simulated sunlight, and enhances collagen expression.

Carboxymethyl Chitosan/Tannic Acid Hydrogel with Antibacterial, Hemostasis, and Antioxidant Properties Promoting Skin Wound Repair

Xujie Zhou,Qing Zhou,Qiang Chen,Yahao Ma,Zhenfang Wang,Lei Luo,Qiang Ding,Hang Li,Shunqing Tang

Local causes of slow wound healing include infection and wound hemorrhage. Using sodium bicarbonate as a neutralizer, a variety of carboxymethyl chitosan–tannic acid (CMC-TA) composite hydrogels solidify through hydrogen bonding in this study. The best-performing hydrogel was synthesized by altering the concentration of TA and exhibited remarkable mechanical properties and biocompatibility. Following in vitro characterization tests, the CMC-TA hydrogel exhibited remarkable antibacterial and antioxidant properties, as well as quick hemostasis capabilities. In the in vivo wound healing study, the results showed that the CMC-TA hydrogel could relieve inflammation and promote the recovery of skin incision, re-epithelialization, and collagen deposition. Overall, this multifunctional hydrogel could be an ideal wound dressing for the clinical therapy of full-thickness wounds.

Sustained release of cathepsin D/cathepsin K from injectable hydrogel microspheres in anti-photoaging

Wenyi Hou,Zhaoyi Lin,Xinling Chen,Yu Sun,Yao Lin,Mengran Peng,Feng Zhou,Huamin Jiang,Yan Li,Yue Zheng

There have been various interventions for photoaging skin, such as retinoic acid treatment and laser therapy. However, more precise, secure, and effective technologies and materials were still need to be explored. Cathepsin D (CTSD) repairs the epidermal barrier in chronic photodamaged skin through increasing TGase-1 expression and activity. Cathepsin K (CTSK) affects the metabolism of skin elastic fibers. Therefore, gelatin/alginate composite hydrogel microspheres loaded with CTSD or CTSK were fabricated. The microspheres with an average size of 79.8 ± 30.4 μm were stable in PBS (pH 7.4) for 5 days and then disintegrated in vitro . 0.1 mg/mL of microspheres continuously released 12-13 ng/mL CTSD or CTSK for the first three consecutive days. In vivo , the microspheres were almost completed degraded on day 7 after the subcutaneous injection. The biosafety and efficacy of CTSD- and CTSK- microspheres were confirmed in vitro and in vivo . They reduced ROS and inhibited the degradation of skin collagen and elastic fibers by upregulating Nrf2 and downregulating MMP-1 and MMP-3, which reversed photodamage of skin induced by UV. In conclusion, the gelatin/alginate composite microspheres loaded with CTSD or CTSK showed a great potential for chronic photodamaged skin.

Injectable Hybrid Poly(ε-caprolactone)-b-poly(ethylene glycol)-b-poly(ε-caprolactone) Porous Microspheres/Alginate Hydrogel Cross-linked by Calcium Gluconate Crystals Deposited in the Pores of Microspheres Improved Skin Wound Healing

JinFeng Liao,YanPeng Jia,BeiYu Wang,Kun Shi,ZhiYong Qian

In our study, a hybrid alginate hydrogel cross-linked by calcium gluconate crystals deposited in poly(ε-caprolactone)-b-poly(ethylene glycol)-b-poly(ε-caprolactone) (PCL-PEG-PCL, abbreviated as PCEC) porous microspheres was developed for skin engineering. The diameter of microspheres was ∼212 μm, and the pore size was ∼8 μm. The PCEC porous microspheres supplied different functions in the hydrogel: (1) Calcium gluconate crystals were loaded in the inner pores of the microspheres, which can induce alginate hydrogel to cross-link in a few minutes once they were mixed. (2) The porous structure of the microspheres provided more anchor points for fibroblast attachment and growth, resulting in the enhancement of cell growth in the hybrid hydrogel. The PCEC microspheres/Alg hydrogel (MPs/Alg hydrogel) possessed excellent compatibility, because cell viability remained around 100% even at a concentration of 500 μg/mL. Meanwhile, the morphology of 3T3 and L929 cells attached on both PCEC porous microspheres and MPs/Alg hydrogel were confirmed by confocal laser spectrometry (CLSM). What’s more, MPs/Alg hydrogel promoted wound regeneration in a full-thickness skin defect model of rats. The mild inflammation reaction existed at the early stage of wound repair and gradually disappeared. These findings suggested that MPs/Alg hydrogel may possess great potential in the application of skin tissue engineering.
