As an important component of sunscreen products for sensitive skin, the potential damage mechanism of ZnO nanoparticles on skin surface with barrier structure or function defect caused by Cutibacterium acnes (C. acnes) has not been elucidated, which poses a serious challenge for reasonable selection of sunscreen products for acne-infected skin. In this work, we demonstrated for the first time that C. acnes induced significant changes in the membrane permeability and intracellular pH of fibroblasts through lipase up-regulation and lipid peroxidation, promoting endocytosis and ionization of ZnO NPs. High amounts of Zn2 + further delayed acne wound healing and aggravated scar hyperplasia by intervening matrix metalloproteinase-9 (MMP-9) and TGF-β1/Smad pathway. MMP9 was confirmed to be the key target of ZnO in delaying acne wound healing by the wound regulatory effects of MMP9 agonist and MMP9 inhibitor. In summary, this work clarified the interaction mechanism between ZnO NPs and acne skins, providing guideline for the application of physical sunscreens for special skins.Graphical abstract

Andrographis paniculatais mainly used to treat skin inflammations, wounds, and infections. In this study, Andrographis Herba, the aerial part of the plant, was proven to increase the viability of UVB-damaged HaCat cells and reduce reactive oxygen species levels. The chemical composition of Andrographis Herba extract (AHE) was analyzed using UPLC-Q-TOF-MS, and diterpene lactones were identified as its primary constituents. Then, the fraction of diterpene lactones was prepared and exhibited similar effects to AHE. AHE, its diterpene lactones component, and its representative constituent andrographolide all decreased the expression of IL-1β, IL-6, and CDKN1A. Furthermore, the protective effects of AHE and its active ingredients on UVB-damaged epidermal stem cells were investigated. Notably, the combined treatment with andrographolide and collagen XVII enhanced the viability of UVB-damaged epidermal stem cells, increased the expression of stemness markers integrin β1 and p63, and decreased the expression of the differentiation marker keratin 10. This combination demonstrated significant synergy in maintaining skin homeostasis, which provides evidences for the development of skin-protective products.

Sleep deprivation is a prevalent issue that disrupts the circadian rhythm of estrogen, particularly estradiol, thereby significantly affecting women’s skin health and appearance. These disruptions can impair skin barrier functionality and decrease dermal collagen synthesis. In this study, our results demonstrate that topical taurine supplementation promotes the expression of tight junction (TJ)-related proteins and enhances collagen production, effectively restoring skin homeostasis in sleep-deprived female mice. Mechanistically, taurine upregulates the expression ofTMEM38B, a gene encoding the TRIC-B trimeric cation channel, resulting in increased intracellular calcium ion levels. This, in turn, promotes the upregulation of TJ-related proteins, such as ZO-1, occludin, and claudin-11 in epidermal cells, while also enhancing the expression of type III collagen in fibroblasts, thus restoring skin homeostasis. These findings suggest that taurine may serve as an alternative to estradiol, effectively improving skin homeostasis disrupted by sleep deprivation while mitigating the potential risks associated with exogenous estrogen supplementation. Collectively, these results provide preliminary insights into the protective mechanisms of taurine against sleep deprivation-induced skin impairments and establish a foundation for its potential application in treating skin conditions related to estrogen imbalances, such as skin aging in menopausal women.

The skin serves as the primary defensive barrier of the human body against external stimuli and damage. Keratinocytes, which are the predominant cell type in the human epidermis, undergo a differentiation process that is crucial for the formation of the skin barrier. Myricetin, a dietary flavonoid present in various fruits and vegetables, is known to play a significant role in maintaining intestinal barrier function; however, its impact on the skin barrier remains inadequately understood. Consequently, this study investigates the effects of myricetin on the differentiation of epidermal keratinocytes and the integrity of the skin barrier. Differentiation of primary mouse keratinocytes was induced using 1.8 mM CaCl2. tudy demonstrated that myricetin effectively suppresses cell proliferation and induces both cell cycle arrest and calcium ion (Ca2+) influx, without influencing apoptosis. Concurrently, myricetin enhances the expression of differentiation markers, including K10, TGase1, Filaggrin, and Involucrin, and facilitates the formation of tight junctions. Upon examining the underlying mechanisms, we discovered that myricetin activates the TRPV4 channel, and the promotion of keratinocyte differentiation by myricetin is contingent upon the activation of this channel. In summary, these findings suggested that myricetin could promote keratinocytes differentiation and have well-established skin barrier protective function.

Conventional coupling agents (such as polyvinylpyrrolidone, methylcellulose, and polyurethane) are unable to efficiently transport drugs through the skin's dual barriers (the epidermal cuticle barrier and the basement membrane barrier between the epidermis and dermis) when exposed to ultrasound, hindering deep and noninvasive transdermal drug delivery. In this study, nanobubbles prepared by the double emulsification method and aminated hyaluronic acid are crosslinked with aldehyde-based hyaluronic acid by dynamic covalent bonding through the Schiff base reaction to produce an innovative ultrasound-nanobubble coupling agent. By amplifying the cavitation effect of ultrasound, drugs can be efficiently transferred through the double barrier of the skin and delivered to deep layers. In an in vitro model of isolated porcine skin, this agent achieves an effective penetration depth of 728 µm with the parameters of ultrasound set at 2 W, 650 kHz, and 50% duty cycle for 20 min. Consequently, drugs can be efficiently delivered to deeper layers noninvasively. In summary, this ultrasound nanobubble coupling agent efficiently achieves deep-layer drug delivery by amplifying the ultrasonic cavitation effect and penetrating the double barriers, heralding a new era for noninvasive drug delivery platforms and disease treatment.

Melanoma is characterized by high malignancy, ranking the third among skin malignancies, and is associated with a lack of specific treatment options and poor prognosis. Therefore, the development of effective therapies for melanoma is imperative. A critical challenge in addressing subcutaneous disease lies in overcoming the skin barrier. In this study, we engineered a microneedle (MN) system that integrates chemotherapy, photothermal therapy (PTT), and targeted therapy to enhance anti-tumor efficacy while effectively penetrating the skin barrier. In vitro studies have demonstrated that the MN drug delivery system (DDS) can effectively penetrate the stratum corneum of the skin, deliver therapeutics to subcutaneous tumor sites, and establish a drug reservoir at these locations to exert anti-tumor effects. Cellular experiments indicated that the engineered PTT chemotherapy-targeted MNs can be internalized by tumor cells, exhibiting enhanced cytotoxicity against them. In vivo pharmacological investigations revealed that the combination of PTT and chemotherapy delivered via this MN DDS produced synergistic anti-tumor effects, achieving a tumor inhibition rate of up to 98.15%. This in situ DDS minimizes involvement with other organs, significantly reducing chemotherapy-related side effects. In summary, the PTT chemotherapy-targeted MNs developed in this study demonstrate promising application potential by enhancing anti-tumor efficacy while minimizing adverse effects.

Chemical enhancers are widely used for facilitating drug penetration in transdermal drug delivery system (TDDS). However, there is a lack of knowledge about how the enhancement effect changes over time. In this study, on the basis of kinetic parameters of enhancement effect, molecular details of the dynamic enhancement process was described and a new hypothesis of the recovery mechanism of the skin barrier function was proposed. Using pretreated skin and flurbiprofen patch, the effects of Azone (AZ) and menthyl decanoate (MT-10) were evaluated with in vitro permeation experiment and further confirmed by confocal laser scanning microscopy (CLSM) and TEWL. The results showed that the enhancement ratio ( ER ) increased firstly, then reached a plateau and finally decreased. The enhancement effect of MT-10 was slower ( T onset, MT-10 > T onset, AZ ), weaker ( ER max, MT-10 < ER max, AZ ) and shorter ( T eff, MT-10 < T eff, AZ ) than that of AZ. According to the results of CLSM, ATR-FTIR and molecular dynamic simulation, the dynamic enhancement effect was caused by the variation of the diffusion coefficient of intercellular lipid in the stratum corneum (SC), which was dependent on the affinity between enhancers and lipid. Moreover, the skin barrier function recovered although a large amount of enhancers still existed in the SC. Additionally, according to the results of ATR-FTIR, molecular docking and skin retention study, the dynamic effect of AZ on the skin protein only induced skin irritation but showed no influence on drug penetration, so did the amount of the enhancer. In conclusion, dynamic enhancement effect was caused by the dynamic effect of the enhancer on the SC intercellular lipid, and the skin barrier function recovered by accepting the enhancer as a new component of the lipid bilayer .

Medicinal plasters have obvious value in promoting blood circulation and removing blood stasis, while their practical values are usually restricted by the skin barrier, resulting in lower local drug concentration and unsatisfactory therapeutic efficacy. Here, a novel non-invasive microneedle patch coated with ferulic acid Chinese herbal plasters (FA@MNs) is presented for the treatment of localised bruising and swelling. The FA@MNs consists of a pyramid-shaped blunt microneedle array core fabricated by poly(ethylene glycol) diacrylate (PEGDA) and traditional Chinese herbal plaster coating composed of ferulic acid and carbomer gel. This special design of microneedles can non-invasively open the skin barrier, enabling more efficient drug delivery while avoiding potential skin damage. Besides, the traditional Chinese herbal plaster coated in microneedle arrays provides further adhesion to the skin surface. It has been demonstrated that the FA@MNs could effectively reduce blood viscosity and inflammatory response in rats while ensuring skin tissue integrity. These results certify that the novel non-invasive microneedle patch has great potential in improving the effects of clinical medicinal plasters.

High-temperature-mediated transdermal delivery often relies on extreme heating to disrupt the skin barrier, but these approaches risk tissue damage and drug denaturation. Mild thermal stimulation promises safer uptake, yet achieving precise temperature control is inconvenient, and its transdermal efficacy and mechanisms remain unclear. A heat-responsive cream made from a stearic acid-lauric acid eutectic, polydopamine (PDA) nanoparticles, glycerin, and drugs is introduced. This cream maintains skin temperature ∼40 °C under simulated sunlight by absorbing excess heat through phase change, preventing overheating while preserving the effectiveness of the drug without requiring manual adjustments. Mild photothermal treatment boosted penetration of PDA nanoparticles (2.4–406.9 nm) by 5.5–7.1-fold, and achieved 28.9-, 24.6-, and 10.7-fold increase of fluorescent signals in dermis for rhodamine B (RB), RB-labeled 5 or 20 kDa dextran versus nonheated controls, respectively. Mechanistic studies revealed that the photothermal effect enhances transdermal delivery by inducing micropores on the skin and through the transappendageal route. Moreover, insulin delivered through this strategy reduced blood glucose by 75% in diabetic mice, and co-delivery of PDA NPs with methotrexate significantly improved psoriasis lesions. This self-regulating platform uniquely unites precise thermal control and dual-pathway enhancement, offering a generalizable route for enhancing transdermal delivery efficiency.

Skin barrier impairment is critical in the development of atopic dermatitis (AD), increasing vulnerability to external pathogens and disrupting cell metabolism, which leads to inflammatory stress and immune imbalance. In this study, a natural turmeric-derived nanovesicle (TDNV)-laden metal polyphenol hydrogel, termed Fe-HD@TDNV is proposed, to synergistically restore the compromised skin barrier in AD through a dual-repair strategy. The TDNV effectively regulates metabolic activity by upregulating the expression of skin barrier proteins, antioxidant enzymes, and antimicrobial peptides (AMPs) in keratinocytes, thereby reinforcing barrier integrity and combating pathogens. Simultaneously, the Fe-HD hydrogel, cross-linked by Fe 3 ⁺ ions and hyaluronic acid-graft-dopamine (HD), provides superior skin compatibility and establishes a low oxidative stress environment for potentiating the therapeutic efficacy of TDNV. By improving skin barrier conditions, the Fe-HD@TDNV hydrogel exhibited desirable performance in maintaining better skin hydration, reducing epidermal thickness, and decreasing abnormal immune responses in acute skin disruption models and AD models. This work is expected to offer insights into the cross-kingdom regulation between plant-derived nanovesicles and mammals, as well as the design of disease-specific dual-functional repair strategies.

Background and purpose Atopic dermatitis (AD) is a chronic inflammatory skin disease, characterized by pruritus and impaired skin barrier function. The pathology of AD involves in immune dysfunction and epidermal barrier disruption. Reactive oxygen species (ROS) are found to be associated with AD, and play a role in the immunological abnormalities and dysfunctional skin barrier. Nicotinamide mononucleotide (NMN) plays an important role in oxidative stress related diseases, but its role in AD is unclear. Methods KM mice were treated with DNFB to induce AD-like lesion and typical applied with NMN for two weeks. The dermatitis score, the degree of itching and TEWL were evaluated during modeling. Epidermal thickness of skin lesions and histopathological changes were detected. Further, inflammatory factors, epidermal differentiation-related genes, oxidative stress indicators and JAK2/STAT5 signaling pathway were evaluated. NHEK cells were stimulated by TNF-α/IFN-γ after pre-treatment with NMN, then ROS levels, inflammatory factors and JAK2/STAT5 signaling pathway were detected. Results NMN exhibited potent anti-atopic activities, shown by alleviated AD-like symptoms, inhibited the increased expression of inflammatory cytokines and restored proteins and mRNA level of skin barrier genes. In addition, NMN inhibited TNF-α/IFN-γ-stimulated elevation of inflammatory chemokines, which was associated with blocking the activation of ROS-mediated JAK2/STAT5 pathway. Conclusion NMN may have a positive effect on relieving symptoms of AD.

Psoriasis is a chronic, relapsing, and refractory immune-mediated skin disease with the etiology and pharmaceutical targets remaining unsatisfactorily addressed. Topical herbal-derived compounds, such as tryptanthrin (Tryp), have been considered as an alternative therapy for psoriasis due to their lower costs and fewer side effects compared to other therapies. However, the effectiveness of topically administered drugs is substantially limited by the thickened pathological skin barrier and the low bioavailability of drugs in the deeper layers of the lesion. Ethosomes, being a novel phospholipid-based vesicle system with high content of ethanol, have been implicated in enhancing topical drug absorption and restoring psoriatic lesions. In this study, taking advantages of ethosomes as a soft and malleable drug carrier, we constructed the Tryp-loaded ethosome (Tryp-ES) through a one-step microfluidics-based technique. The optimal formulation of Tryp-ES was achieved by adding amino-acid-derived surfactant sodium lauroyl glutamate, and Tryp-ES exhibited homogeneous particle size and favorable stability at room temperature. In vitro evaluations showed that Tryp of Tryp-ES could be easily internalized into cells and accumulated in cell nuclei, hence inhibited the abnormally proliferated keratinocytes by inducing apoptosis. In vivo and in vitro assessment using psoritic skin of mice revealed that Tryp-ES had preferred skin retention and permeation of loaded drugs within the initial 1 h of topical administration, which could be attributed to transient disintegrations of cell membranes by ethosomes, thus improved cellular fluidity and permeability. Notably, a synergistic effect of ethosomes and Tryp was found in psoriatic mice. Tryp-ES-treated mice showed substantially ameliorated symptoms of psoriasis and reduced pathological alterations due to hyperplasia, inflammation and angiogenesis, without detectable local or systemic toxicities. Interestingly, lipidomics analysis confirmed that the supplementation of phospholipids, as in the form of ethosome vehicles, was an alterantive strategy to relieve psoriatic pathologies. Taken together, this study provides a novel impact for ethosomal topical delivery of Tryp and underlines their potential as an effective therapy for the management of psoriasis.
