Inflammation-reducing thermosensitive hydrogel with photothermal conversion for skin cancer therapy

Mengqi Jia,Ruilin Lu,Pengfei Li,Xiaoming Liao,Yanfei Tan,Shiyong Zhang

Photothermal therapy (PTT) has widely been utilized for postoperative treatment of skin cancer, while high temperature, usually >50 °C, would induce damage to healthy tissue and increased wound inflammation. Herein, we developed an “all in one” hydrogel to enhance mild PTT for postoperative skin cancer treatment while circumventing photothermo-induced inflammation by loading quercetin (Que)-coated tannin‑iron (TA-Fe) nanoparticles with poly (N-acrylylglycine) amine (PNAGA) hydrogel (Que@TA-Fe@PNAGA). Exposure to near-infrared light, Que.@TA-Fe@PNAGA occurred a mild temperature increase (∼47 °C), which induces local mild PTT and disrupts the hydrogen bonds within the hydrogel, triggering a gel-to-sol phase transition and the release of Que.@TA-Fe nanoparticles. These released nanoparticles inhibit the expression of heat shock proteins in tumor cells by producing reactive oxygen species and enter inflammatory cells to release TA and Que. via acid hydrolysis, reducing tumor necrosis factor-α expression by 66.6 % and promoting M1-to-M2 macrophage conversion. Based on this integrated functionality, Que.@TA-Fe@PNAGA hydrogel achieves over 99.4 % tumor inhibition rate, effectively avoids photothermo-induced damage in normal tissue and inflammation, and thus represents a new approach for postoperative photothermal therapy in skin cancer treatment.

Molecular spectroscopic behaviors of beta-arbutin in anti-skin cancer

Yanbin Su,Xuwei Sun,Ruixue Wu,Xing Zhang,Yunzhou Tu

The purpose of this study was to explore the molecular spectroscopic behaviors of the beta-arbutin on the toxic aldehyde of trans-crotonaldehyde in anti-skin cancer. Results revealed that both hydroxymethyl and phenolic hydroxyl of beta-arbutin led to the shift of the maximum ultraviolet peak of trans-crotonaldehyde from a very significant absorption peak at 300 nm to a very weak absorption peak at 310 nm and the significant shift of the aldehyde Raman band at 1689/cm of the toxic trans-crotonaldehyde in mitochondria and aldehyde dehydrogenase, respectively. The beta-arbutin acting on the toxic trans-crotonaldehyde highlighted the potential of anti-skin cancer and provided a valuable clue for the new application of the beta-arbutin.

Tea polyphenols nanoparticles integrated with microneedles multifunctionally boost 5-aminolevulinic acid photodynamic therapy for skin cancer

Yixuan Wang,Shijia Fu,Yao Zeng,Siwen Jiao,Guihong Chai,Yuehong Xu

5-aminolevulinic acid photodynamic therapy (ALA-PDT) is an emerging therapeutic strategy for skin cancer due to its noninvasiveness and high spatiotemporal selectivity. However, poor skin penetration, poor intratumoral delivery, the instability of aqueous ALA, and the tumor’s inherent hypoxia microenvironment are major hurdles hindering the efficacy of ALA-PDT. Herein, we aim to address these challenges by using microneedles (MNs) to assist in delivering nanoparticles based on natural polymeric tea polyphenols (TP NPs) to self-assemble and load ALA (ALA@TP NPs). The TP NPs specifically increase cellular uptake of ALA by A375 and A431 cells and reduce mitochondrial membrane potential. Subsequently, the photosensitizer protoporphyrin IX derived from ALA accumulates in the tumor cells in a dose-dependent manner with TP NPs, generating reactive oxygen species to promote apoptosis and necrosis of A375 and A431 cells. Interestingly, TP NPs can ameliorate the tumor’s inherent hypoxia microenvironment and rapid oxygen consumption during PDT by inhibiting hypoxia inducible factor-1α, thereby boosting reactive oxygen species (ROS) generation and enhancing ALA-PDT efficacy through a positive feedback loop. After ALA@TP NPs are loaded into MNs to fabricate ALA@TP NPs@MNs, the MNs enhance skin penetration and storage stability of ALA. Importantly, they exhibit remarkable antitumor efficacy in A375-induced melanoma and A431-induced squamous cell carcinoma with a reduced dose of ALA and reverse hypoxia in vivo . This study provides a facile and novel strategy that integrates MNs and green NPs of TP for addressing the bottlenecks of ALA-PDT and enhancing the ALA-PDT efficacy against skin cancers for future clinical translation.

Hybrid Nanogels-Based Printable Tattoos Inspired by Lady Costume from Tang Dynasty for Monitoring Solar UV Radiation

Neng Hu,Di Gao,Xin Cai,Lin Lei,Weijia Wang,Huiqing Fan,Peter Müller-Buschbaum,Qi Zhong

Inspired by the lady costume from Tang Dynasty, printable tattoos are prepared from hybrid nanogels to monitor ultraviolet (UV) radiation from sunshine. The hybrid poly(di(ethyleneglycol) methylether methacrylate- co -poly(ethylene glycol) methylethermethacrylate) nanogels containing graphitic carbon nitride (g-C 3 N 4 ) nanosheets, abbreviated as hybrid P(MA- co -MA 300 )/g-C 3 N 4 nanogels, are prepared by precipitation polymerization. Due to the excellent photocatalysis of the g-C 3 N 4 nanosheets, the dyed hybrid nanogels can be rapidly photodegraded under sunshine. Based on the linear correlation between the discoloration of dyed hybrid nanogels and the dose of solar radiation, the UV radiation from sunshine can be simply monitored by checking the color in real-time. By applying different types of dyes, the hybrid nanogels even can be used to trace the dose for the synthesis of vitamin D, induction of erythema and risk of skin cancer simultaneously. Favored by the stickiness of acrylate-based polymers, the hybrid nanogels can be printed to the body as tattoos to realize decoration and UV detection. By simply switching the template for printing, the tattoo patterns can be easily changed according to personal interests and applied position on the human body. Therefore, the present hybrid nanogels can be a promising candidate to monitor the solar radiation dose and decorate the human body.

In Situ Implantation of Chitosan Oligosaccharide-Doped Lipoic Acid Hydrogel Breaks the “Vicious Cycle” of Inflammation and Residual Tumor Cell for Postoperative Skin Cancer Therapy

Mengqi Jia,Ruilin Lu,Caikun Liu,Xiaodong Zhou,Pengfei Li,Shiyong Zhang

Surgical excision is the main treatment for skin cancer, but the tumor recurrence caused by the “vicious cycle” between residual tumor cells and postoperative inflammation remains a challenge. Herein, a new material, which can break the “vicious cycle”, was developed by incorporating chitosan oligosaccharides into lipoic acid hydrogel (COS@LA-hydrogel). When implanted at the resection site, the COS@LA-hydrogel would have a sustained release of LA and COS, which could not only kill residual tumor cells by synergistically reducing AKT phosphorylation but also decrease inflammation by inhibiting the expression of tumor necrosis factor-α (TNF-α) and inhibiting bacterial infection, respectively. As a proof of concept, in the postoperative melanoma resection model, the COS@LA-hydrogel reduced the expression of pro-inflammatory factors TNF-α and interleukin-6 (IL-6) by up to 78 and 80%, respectively, and they showed almost no tumors and the median survival of the mice was 2.5 times longer than that of the control group. The hydrogel with the function of “vicious cycle” breaking holds clinical potential.

Isolicoflavonol alleviates UVB-induced photodamage via protecting mitochondria and blocking the activation of NLRP3 inflammasome

Xing-Jie Zhang,Peng-Yun Yang,Ling Ding,Jun Wang,Xiao-Li Li,Wei-Lie Xiao

Photodamage, a type of skin inflammation caused by excessive exposure to solar radiation, leads to skin redness, inflammation, and even the development of skin cancer, posing a severe threat to individuals living at high altitudes. UVB radiation is considered the primary factor contributing to photodamage. It stimulates macrophages within the epidermis, triggers inflammasome activation, and increases the inflammatory cytokine interleukin-1β (IL-1β) production. This study examined the protective effects of the compound isolicoflavonol (ILF) and its mechanism against UVB-induced photodamage. We irradiated UVB to create a photodamage model in mice and macrophages. Next, we assessed ILF's ability to protect the skin and cells from UVB photodamage and its inhibitory effects on UVB-mediated NLRP3 inflammasome. Our findings indicated that ILF reduced UVB-induced skin injury and inflammation in mouse skin, decreased cell death, NLRP3 inflammasome activation, ROS production, and mitochondrial dysfunction. These results suggest that ILF may be a potent agent for protecting the skin against UVB-induced photodamage.

Confinement of Reactive Oxygen Species in an Artificial-Enzyme-Based Hollow Structure To Eliminate Adverse Effects of Photocatalysis on UV Filters

Enguo Ju,Kai Dong,Zhenzhen Wang,Yan Zhang,Fangfang Cao,Zhaowei Chen,Fang Pu,Jinsong Ren,Xiaogang Qu

Graphical Confine and eliminate : The integration of ZnO nanoparticles and CeO x nanoparticles into hollow microspheres (ZnO/CeO x HMS) could provide broad-spectrum UV protection and scavenge reactive oxygen species (ROS) generated under UV irradiation (see figure). Benefiting from the cooperative effect of the hollow structure and the antioxidative enzyme mimicking activity of CeO x , the generated ROS could be confined to the limited space and effectively scavenged. Skin cancers caused by UV irradiation have been a major public health problem. One simple and effective way to avoid the above detrimental effects is the use of UV-protective sunscreens. However, there has been considerable concern with the issue of the production of reactive oxygen species (ROS) through the photodegradation of commercial UV filters. Herein, for the first time, it is reported that the integration of ZnO nanoparticles and CeO x nanoparticles into hollow microspheres (ZnO/CeO x HMS) could provide broad-spectrum UV protection and scavenge generated ROS under UV irradiation. Benefiting from the cooperative effect of the hollow structure and the antioxidative activity of CeO x , ROS generated under UV irradiation could be confined to a limited space and effectively conversion into nontoxic molecules is catalyzed as a consequence of increased collision frequency. Therefore, both primary, direct UV-induced damage and secondary ROS toxicity could be greatly reduced.

A biocompatible and magnetic nanocarrier with a safe UV-initiated docetaxel release and cancer secretion removal properties increases therapeutic potential for skin cancer

Fei Kong,Xiao Huang,Danyang Yue,Jun Pan

Cancer is a leading fatal disease worldwide. To increase its therapeutic efficiency, more effective with less side effect and patient acceptable administration approach is expected. Moreover, modification of tumor microenvironment is proved to be operative recently. In this paper, a nanocarrier named LDEDDS was developed for intelligent tropical administration of skin cancer, along with removal of hydrophobic cancerous secretion to change tumor microenvironment. It was made by coating of amphipathic polymer P(BA- co -HBA) on docetaxel (TXT, a model hydrophobic anticancer drug) loaded Fe 3 O 4 @ZnO. Results showed that an optimal loading rate of TXT in Fe 3 O 4 @ZnO was 89.75   ±   0.15%, corresponding to loading capacity of 17.95   ±   2.97% when the mass ratio of Fe 3 O 4 @ZnO to TXT was 1:20. The LDEDDS had a narrow distribution size of 115.8   nm in average and was superparamagnetic. Without UV radiation, it had low TXT release (<   7% in 48   h) and cytotoxicity (<   14% in 96   h) to both the normal and carcinoma skin cells. While under a UV with a dose much lower than physiological dose of normal sunlight, LDEDDS released around 60% and 90% of TXT in 1 and 48   h. 1   h UV treated LDEDDS removed up to 62% of cancer secreted epidermal growth factor (EGF), a model hydrophobic secretion in 96   h. Consequently, 1   h UV treated LDEDDS inhibited up to 60% of the growth of skin cancer cells in 96   h, overriding those effects of the same concentration of TXT in in vitro cellular experiments. This is the first study to change tumor microenvironment by removal of cancerous secretion and is proved to be effective. Along with the superparamagnetic property, which provides potential for concentrating, increasing penetration and internalization into cancerated cells as well as removing from body under an external magnetic field, we predict LDEDDS will have potential applications in clinic skin cancer therapy.

Microwave assisted drug delivery of titanium dioxide/rose Bengal conjugated chitosan nanoparticles for micro-photodynamic skin cancer treatment in vitro and in vivo

Abd El-Kaream Samir Ali,Hassan Nasser Ali Mohamed,Saleh Hassan Saleh Abdullatif,Albahloul Mohamed Albahloul Salem,Khedr Abdalla Mohamed,El-Kholey Sohier Mahmoud

Background Micro-photodynamic therapy (MWPDT) combines photo-dynamic (PDT) and microwave-dynamic (MWDT) therapies with sensitizers, offers new avenues for cancer treatment. Despite the fact that novel sensitizers for MWPDT have been successfully synthesized, only a few are being employed effectively. The low tumor-targeting specificity, inability to transport sensitizer's deeper intratumorally, and deteriorating tumor microenvironment all restrict their anti-tumor efficacy. The current work was done aiming at microwave assisted drug delivery of titanium dioxide / rose Bengal conjugated chitosan nanoparticles (TiO 2 /RB@CSNP) for micro- photo-dynamic skin cancer (SKCA) treatment in vitro and in vivo as activated cancer treatment up-to-date modality. Materials and methods The study was conducted in vitro on human SKCA cells (A-375) and the study protocol application groups in vivo on Swiss albino mice treated with 7,12-dimethylbenz[a]anthracene (DMBA)/croton oil only and were not received any treatment for inducing SKCA, and only after SKCA induction the study treatment protocol began, treatment was daily with TiO 2 /RB@CSNP as MWPDT sensitizer with or without exposure to laser (IRL) or microwave (MW) or a combination of them for 3 min for two weeks. Results Revealed that CSNP can be employed as effective TiO 2 /RB delivery system that directly targets SKCA cells. Additionally TiO 2 /RB@CSNP is a promising MWPS for and when combined with MWPDT can be very effective in treatment of SKCA-A-375 in vitro (cell viability decreased in a dose-dependent basis, the cell cycle progression in G0/G1 was slowed down, and cell death was induced as evidenced by an increase in the population of Pre-G cells, an increase in early and late apoptosis and necrosis, and an increase in autophagic cell death) and DMBA/croton oil SKCA-induce mice in vivo (induced antiproliferative genes (caspase 3,9, p53, Bax, TNFalpha), suppressed antiapoptotic and antiangiogenic genes (Bcl2,VEGF respectively) effectively reducing the tumors growth and leading to cancer cell death as well as decreased oxidative stress (MDA), and ameliorated enzymatic and non-enzymatic antioxidants (SOD, GR, GPx, GST, CAT, GSH, TAC) as well as renal (urea, creatinine) and hepatic (ALT, AST) functions. This process could be attributed to MWPDT; microwave and/or photo-chemical TiO 2 /RB activation mechanism and antioxidant potential of non activated TiO 2 /RB as well. Conclusion The results indicate that TiO 2 /RB@CSNP has great promise as an innovative, effective delivery system for selective localized treatment of skin cancer that is activated by MWPDT.

Simulated Sunlight-Mediated Photodynamic Therapy for Melanoma Skin Cancer by Titanium-Dioxide-Nanoparticle–Gold-Nanocluster–Graphene Heterogeneous Nanocomposites

Yan Cheng,Yun Chang,Yanlin Feng,Ning Liu,Xiujuan Sun,Yuqing Feng,Xi Li,Haiyuan Zhang

Simulated sunlight has promise as a light source able to alleviate the severe pain associated with patients during photodynamic therapy (PDT); however, low sunlight utilization efficiency of traditional photosensitizers dramatically limits its application. Titanium-dioxide-nanoparticle–gold-nanocluster–graphene (TAG) heterogeneous nanocomposites are designed to efficiently utilize simulated sunlight for melanoma skin cancer PDT. The narrow band gap in gold nanoclusters (Au NCs), and staggered energy bands between Au NCs, titanium dioxide nanoparticles (TiO 2 NPs), and graphene can result in efficient utilization of simulated sunlight and separation of electron–hole pairs, facilitating the production of abundant hydroxyl and superoxide radicals. Under irradiation of simulated sunlight, TAG nanocomposites can trigger a series of toxicological responses in mouse B16F1 melanoma cells, such as intracellular reactive oxygen species production, glutathione depletion, heme oxygenase-1 expression, and mitochondrial dysfunctions, resulting in severe cell death. Furthermore, intravenous or intratumoral administration of biocompatible TAG nanocomposites in B16F1-tumor-xenograft-bearing mice can significantly inhibit tumor growth and cause severe pathological tumor tissue changes. All of these results demonstrate prominent simulated sunlight-mediated PDT effects.

Development of biodegradable nanogels for lipase accelerated drug release of 5-aminolevulinic acid

Xiao Liu,Yuan Zhang,Peng Zhang,Kang Ge,Ruzhi Zhang,Yixin Sun,Yang Sheng,Mark Bradley,Rong Zhang

Photodynamic therapy (PDT) using 5-aminolevulinic acid (5-ALA) is an important approach for the treatment of some skin diseases and cancers. A major defect of this approach is that it is difficult for 5-ALA to accumulate around lesions in deeper regions of tissue, resulting in poor conversion to the active fluorophore and photodynamic efficiencies. Because of their targeting and controlled release abilities, nanogel carriers could solve this problem. In this paper, nanogels were prepared by using micro-emulsion polymerization with various biodegradable polyester crosslinkers ( L -lactide and ε-caprolactone). The swelling and degradation properties and entrapment efficiency, drug loading and drug release ability of the nanogels were investigated. Nanogels co-cultured with skin cancer cells (A2058) allowed the efficiency of the PDT in vitro to be demonstrated. The results showed that the swelling rate of hydrogels reduced with increasing crosslinker levels, which caused a slow-down in the release of 5-ALA, but lipase accelerated degradation of nanogels increased 5-ALA concentrations in tumor cells and leading to higher PDT efficiency. It was proved by in vivo experiment indicating that the development of skin cancer tissues were efficiently inhibited by the 5-ALA loaded nanogels.
