Skin defects, soft tissue damage, and fractures often occur simultaneously in severe trauma. Under current medical technology, fractures can be quickly fixed by internal or external repair techniques, and early functional exercises can be performed. However, skin defects heal over a long time and can even be difficult to heal. Functional exercise may cause cutting of fresh granulation to break and impair wound healing. Functional exercise and wound healing seem to contradict each other. In this study, an alginate hydrogel was developed. With self-healing characteristics, the hydrogel tightly adhered to the wound and could self-heal breaks in the gel caused by functional exercises. These characteristics enable this hydrogel to be used in complex clinical situations to solve sports rehabilitation and skin defect repair problems. In addition, this hydrogel can slowly release strontium ions, promote angiogenesis and collagen deposition in the wound, and quickly heal the wound.

Biological scaffolds prepared after decellularization are used for the restoration of damaged tissues. A number of chemicals are used for bioscaffold preparation, and some of them damage their composition and architecture. Herein, we investigated the Sapindus mukorossi fruit pericarp extract (SPE) (5%) for decellularization of the caprine dermis. The dermal samples were processed in 5% SPE over magnetic stirrer for 96h at room temperature. The decellularization efficiency of SPE was analyzed by histological examination, DAPI staining, scanning electron microscopy (SEM), quantification of DNA hydroxyproline and hemocompatibility determination. Further, these acellular caprine dermal scaffolds were transplanted on full thickness skin wounds of group III New Zealand white rabbits. The wounds were left open in group I (Sham) and reconstructed by autograft in group II (n = 6 in each group). Continuous agitation of native caprine dermal tissues in 5% SPE for 96 hours leads to complete decellularization without affecting the extracellular matrix architecture. Microscopic observation of decellularized samples did not show any nuclei. DNA quantity was reduced (p < .05) in decellularized samples and scaffolds were found to be hemocompatible. Complete healing was observed on day 28 in groups II and III. No significant difference was noted in IgG in all the groups. Quantitative assessment of MDA showed a significant increase in groups I and II. Our results suggested that the 5% SPE solution effectively decellularized the native caprine dermis and the scaffolds were well tolerated by the animals.

Natural hydrogels are growing in interest as a priority for wound healing. Plant polysaccharides have a variety of biological pharmacological activities, and chitosan hydrogels have proven strong antimicrobial effects, but hydrogels prepared with polysaccharides alone have certain deficiencies. Polysaccharides from flowers ofLonicera japonicaThunb. (LP) and the aerial parts ofMentha canadensisL. (MP) were extracted and oxidized by sodium periodate (NaIO4) and then cross-linked with oxidized-carboxymethylated chitosan (O-CCS) to develop oxidized plant- polysaccharides-chitosan hydrogels (OPHs). SEM observation showed that OPHs had porous interior structures with interconnecting pores. The OPHs showed good swelling, water-retention ability, blood coagulation, cytocompatibility properties, and low cytotoxicity (classed as grade 1 according to United States Pharmacopoeia), which met the requirements for wound dressings. Then the cutaneous wound-healing effect was evaluated in BALB/C mice model, after 7 days treatment, the wound-closure rate of OPHs groups were all greater than 50%, and after 14 days, all were greater than 90%, while the value of the control group was only 72.6%. Of them, OPH-2 and OPH-3 were more favorable to the wound-healing process, as the promotion was more significant. The plant polysaccharides and CS-based hydrogel should be a candidate for cutaneous wound dressings. Keywords:hydrogel;chitosan;Lonicera japonicaThunb. polysaccharides;Mentha canadensisL. polysaccharides;wound healing;skin defect model

In this paper, skin-inspired multilayer Janus composite nanofibrous membranes with superhydrophilicity and superhydrophobicity as a wound dressing were designed and prepared to manage and utilize the exudates. Wettability, morphology, in-vitro release, cytotoxicity, antimicrobial activity and healing effect were investigated. Animal experiment and histological analysis confirmed the composite membranes were more effective in improving the healing of full-thickness defect wound, the growth of collagen fibers and collagen deposition. The four-layer composite membranes include the drug-loading layer containing Ciprofloxacin (CIP) and Astaxanthin (ATX) with superhydrophilic surface for self-pumping and drugs backflow, the hydrophobic polyvinylidene fluoride layer for delaying the exudates loss, and the superhydrophobic coating with self-cleaning properties for avoiding adhesion of foreign microorganisms. The exudates were absorbed into the composite membrane and locked in the nanofiber network to avoid dehydration of the wound, meanwhile, CIP and ATX were delivered to the wound bed to effectively inhibit bacterial growth and stimulate tissue regeneration. These results demonstrate that skin-inspired Janus composite nanofibrous membrane is a promising wound dressing for promoting full-thickness skin defect wound healing, displaying its great potential applications in skin tissue engineering.

Long-term protection of skin-defect wounds is challenging for the common wound dressings cotton gauze due to its weak skin adhesion, limited drug-loading capacity and frequent replacement. Hydrogels are considered as the promising wound dressings benefiting by their good mechanical compatibility and biocompatibility. Nevertheless, the hydrogels always suffer from the drawbacks of fast water evaporation, lack of antibacterial ability and uncontrollable drug release, which greatly shorten their long-term wound protection duration. Herein, a novel and multifunctional phase change gel (PCG) combining flexibility of hydrogel and release-controllable ability of phase change materials (PCMs) was fabricated through a facile solvent-exchange strategy. Specifically, the bacterial cellulose (BC)/polyethylene glycol based phase change gels (BC/PEG PCGs) were prepared by direct impregnation of BC hydrogel into massive PEG melts. The solvent-exchange mechanism between BC hydrogel and PCM melts was firstly studied based on the impregnation experiments of BC gels into various PCMs. The as-prepared BC/PEG PCGs were then characterized in terms of structure, mechanical, phase change and wound dressing properties, subsequently. This work provides a comprehensive understanding of the structure relationship on their excellent flexibility, good adhesive, skin-cooling, antibacterial and controllable drug-release functions of BC/PEG PCGs, extending their potential in the long-term protection of skin-defect wounds.

Bacterial infection and free radical oxidative stress at the wound site could easily cause cascade inflammation and hinder the healing process of the wound. In this study, chitosan-cysteine-gallic acid (CCG) hydrogel with antibacterial and antioxidant properties was synthesized by chitosan (CS), cysteine (Cys), and gallic acid (GA) for a preliminary evaluation of its therapeutic efficacy in a mouse model of full-layer skin defect. In vitro analysis showed that the CCG hydrogel had good antibacterial activity and blood compatibility. In vivo , the CCG hydrogel wound dressings accelerated wound healing, stimulate angiogenesis, increase collagen deposition and anti-inflammatory factor expression. The CCG hydrogel wound dressing is designed to promote the regeneration of damaged skin tissue and is expected to become a potential candidate for clinical treatment.

Full-thickness skin wounds are have continued to be reconstructive challenges in dermal and skin appendage regeneration, and skin substitutes are promising tools for addressing these reconstructive procedures. Herein, the one-step fabrication of a cell sheet integrated with a biomimetic hydrogel as a tissue engineered skin for skin wound healing generated in one step is introduced. Briefly, cell sheets with rich extracellular matrix, high cell density, and good cell connections were integrated with biomimetic hydrogel to fabricate gel + human skin fibroblasts (HSFs) sheets and gel + human umbilical vein endothelial cells (HUVECs) sheets in one step for assembly as a cell sheet-laden hydrogel (CSH). The designed biomimetic hydrogel formed with UV crosslinking and ionic crosslinking exhibited unique properties due to the photo-generated aldehyde groups, which were suitable for integrating into the cell sheet, and ionic crosslinking reduced the adhesive force toward the substrate. These properties allowed the gel + cell sheet film to be easily released from the substrate. The cells in the harvested cell sheet maintained excellent viability, proliferation, and definite migration abilities inside the hydrogel. Moreover, the CSH was implanted into a full-thickness skin defects to construct a required dermal matrix and cell microenvironment. The wound closure rate reached 60.00 ± 6.26% on the 2nd day, accelerating mature granulation and dermis formation with skin appendages after 14 days. This project can provide distinct guidance and strategies for the complete repair and regeneration of full-thickness skin defects, and provides a material with great potential for tissue regeneration in clinical applications.

Effective wound healing is one of the most concerning health issues worldwide. Adding traditional Chinese herbal ingredients into functional hydrogel materials may be an efficient strategy to promote wound healing. In this study, Shikonin (SHK) was simply introduced into polyvinyl alcohol (PVA) hydrogel to construct SHK/PVA composite hydrogel through alkali crosslinking method. SHK/PVA hydrogel could not only improve cell adhesion and proliferation, but also significantly increase collagen deposition, enhance angiogenesis and reduce inflammatory response, and finally accelerate wound healing in rat skin defect models. Findings from this study indicated that the introduction of SHK would be a crucial candidate for wound healing. Therefore, SHK/PVA hydrogel is expected to provide a new choice for clinical repair and regeneration of acute and chronic wounds.

Background Most of the natural extracellular matrix (ECM) is a three-dimensional (3D) network structure of micro/nanofibers for cell adhesion and growth of 3D. Electrospun fibers distinctive mimicked 2D ECM, however, it is impossible to simulate 3D ECM because of longitudinal collapse of continuous micro/nanofibers. Herein, 3D electrospun micro/nano-fibrous sponge was fabricated via electrospinning, homogenization, shaping and thermal crosslinking for 3D tissue regeneration of cells and vascular. Results Fibrous sponge exhibited high porosity, water absorption and compression resilience and no chemical crosslinked agent was used in preparation process. In vitro studies showed that the 3D short fiber sponge provided an oxygen-rich environment for cell growth, which was conducive to the 3D proliferation and growth of HUVECs, stimulated the expression of VEGF, and well promoted the vascularization of HUVECs. In vivo studies showed that the 3D short fiber sponges had a good 3D adhesion to the chronic wound of diabetes in rats. Furthermore, 3D short fibrous sponges were better than 2D micro/nanofiber membranes in promoting the repair of diabetic full-thickness skin defects including wound healing, hair follicle regeneration, angiogenesis, collagen secretion. Conclusion Therefore, electrospun short fibrous sponges are special candidates for mimicking the 3D ECM and promoting 3D regeneration of tissue. Graphic Abstract

Purpose Wound healing, especially of infected wounds, remains a clinical challenge in plastic surgery. This study aimed to manufacture a novel and multifunctional wound dressing by combining graphene oxide/copper nanocomposites (GO/Cu) with chitosan/hyaluronic acid, providing significant opportunities for the therapy of wound repair in wounds with a high risk of bacterial infection. Methods In this study, GO/Cu-decorated chitosan/hyaluronic acid dressings (C/H/GO/Cu) were prepared using sodium trimetaphosphate (STMP) crosslinking and the vacuum freeze-drying method, and chitosan/hyaluronic acid dressings (C/H) and GO-incorporated chitosan/hyaluronic acid dressings (C/H/GO) served as controls. The surface characterization, in vitro degradation under various pH values, antimicrobial potential, cytocompatibility and in vivo therapeutic efficacy in a bacteria-infected full-thickness skin defect model were systematically evaluated. Results Our experimental results indicated that the acidic environment facilitated the release of copper (CuNPs and Cu2+) from the dressings, and prepared C/H/GO/Cu dressings exhibited significant in vitro antimicrobial activities against the two tested bacterial strains (ATCC35984 and ATCC25923). All three dressings showed satisfactory cytocompatibility with mouse fibroblasts (NIH/3T3-L1). Moreover, remarkably accelerated wound healing was found in the C/H/GO/Cu group, with controlled inflammatory infiltration and improved angiogenesis in granulation tissues. In addition, no pathological damage was noted in the tissue structures of the tested organs (heart, lung, liver and kidney) in any of the four groups. Conclusion Collectively, GO/Cu-incorporated chitosan/hyaluronic acid dressings suggested a synergistic antimicrobial efficacy and acceptable biocompatibility both in vitro and in vivo, as well as a significantly accelerated healing process of bacteria-infected wounds. Thus, the multifunctional C/H/GO/Cu composite is expected to be a potential alternative for wound dressings, especially for the management of intractable wounds caused by bacterial infection.

Clustered regularly interspaced short palindromic repeat activation (CRISPRa) technology has emerged as a precise genome editing tool for activating endogenous transgene expression. While it holds promise for precise cell modification, its translation into tissue engineering has been hampered by biosafety concerns and suboptimal delivery methods. To address these challenges, we have developed a CRISPRa non-viral gene delivery platform by immobilizing non-viral CRISPRa complexes into a biocompatible hydrogel/nanofiber (Gel/NF) composite scaffold. The Gel/NF scaffold facilitates the controlled and sustained release of CRISPRa complexes and also promotes cell recruitment to the scaffold for efficient and localized transfection. As a proof of concept, we employed this CRISPRa delivery platform to activate the vascular endothelial growth factor (VEGF) gene in a rat model with full-thickness skin defects. Our results demonstrate sustained upregulation of VEGF expression even at 21 days post-implantation, resulting in enhanced angiogenesis and improved skin regeneration. These findings underscore the potential of the Gel/NF scaffold-based CRISPRa delivery platform as an efficient and durable strategy for gene activation, offering promising prospects for tissue regeneration. Statement of significance Translation of clustered regularly interspaced short palindromic repeat activation (CRISPRa) therapy to tissue engineering is limited by biosafety concerns and unsatisfactory delivery strategy. To solve this issue, we have developed a CRISPRa non-viral gene delivery platform by immobilizing non-viral CRISPRa complexes into a biocompatible hydrogel/nanofiber (Gel/NF) composite scaffold. This scaffold enables controlled and sustained release of CRISPRa and can induce cell recruitment for localized transfection. As a proof of concept, we activated vascular endothelial growth factor (VEGF) in a rat model with full-thickness skin defects, leading to sustained upregulation of VEGF expression, enhanced angiogenesis and improved skin regeneration in vivo . These findings demonstrate the potential of this platform for gene activation, thereby offering promising prospects for tissue regeneration.

Mammalian skin is composed of hydrophobic polymers such as proteins, polysaccharide and oily matrices, serving as a crucial barrier to prevent adverse factors from rapidly infiltrating the body while also functioning as a key organ for material exchange. However, open wounds resulting from skin defects can disrupt the internal stability of the body's environment, leading to water loss and delaying the wound healing process. Mannan is believed to synergize with natural moisturizing factors (NMF). Inspired by the hydrophobic structure of skin, firstly, acetylated yeast mannans (AM) were synthetized as a hydrophobic and moisturizing component. Subsequently, we developed a composite electrospun nanofiber (PVCQAM), which integrates polyvinyl alcohol (PVA, a biocompatible component), AM, collagen (Col, extracellular matrix component), and quaternized chitosan (QCS, a hydrophobic and antibacterial component) to the mimic the architecture and functions involved in wound healing. AM enhanced the surface hydrophobicity compared to the mannan-containing nanofibers (PVCQM). Furthermore, as the degree of acetylation increased, nanofibers manifested improved hydrophobic properties. The nanofibers demonstrated thermal stability, and suitable stretchability that aligns with human tissue characteristics. The incorporation of QCS imparted inherent antibacterial effects to the biomimetic nanofibers against S. aureus and E. coli . Notably, excellent cytocompatibility and hemocompatibility in vitro have prompted us to evaluate the therapeutic efficacy in a cutaneous defect model. The electrospun nanofibers in this study displayed significant anti-inflammatory effects and promoted collagen deposition, resulting in an impressive wound closure rate of 95 %. The skin-inspired nanofiber dressing based on hydrophobic AM presents significant potential for moisturizing and facilitating benign tissue repair in defect management.
