Structural elucidation of a glucan from Crataegus pinnatifida and its bioactivity on intestinal bacteria strains

Shihai Zhang,Chunqian Zhang,Meixia Li,Xia Chen,Kan Ding

Crataegus pinnatifida is widely used as functional food and traditional medicine in China for various applications. Polysaccharides may contribute to health benefits, such as modulating human gut microbiota (HGM). To investigate structure features of polysaccharides from this plant, fruits of C. pinnatifida were extracted with boiling water, yielding crude polysaccharides HAW. A novel polysaccharide, named HAW1-1, was pooled by DEAE Sepharose™ Fast Flow column and further purified by Sephacryl S-100 HR column. The molecular weight of HAW1-1 was 42.35 kDa. The carbohydrate content of HAW1-1 was 99.42% and no protein was detected. The monosaccharide analysis showed this polysaccharide was only composed of glucose. According to the partial acid hydrolysis, methylation analysis and NMR spectra, the backbone of HAW1-1 was consisted of 1,4-linked α-D-glucan and 1,4,6-linked α-D-glucan. The side chains were composed of 1,6-linked α-D-glucan and terminal-linked α-D-glucan. The bioactivity tests showed that HAW1-1 could promote the growth of three species of intestinal Bacteroides and produce short-chain fatty acids (SCFAs). The results suggested that HAW1-1 might be a potential prebiotic candidate to maintain intestinal homeostasis and improve human gut health.

Processing and Prebiotics Characteristics of β-Glucan Extract from Highland Barley

Yao Ren,Haoyu Xie,Li Liu,Dongying Jia,Kai Yao,Yuanlong Chi

β-glucan extract (GE) was obtained from highland barley bran using alkaline–acid–alcohol extraction method. The stability, solubility, foaming ability, and prebiotics characteristics of GE were assessed consecutively. GE demonstrated excellent heat stability (hardly degraded at 220 °C) and pH stability, especially at neutral or alkaline condition, and its solubility was significantly influenced by temperature instead of pH or NaCl, achieving 0.91 g/100 g at 100 °C. Good foaming ability and foam stability of GE were observed during low temperatures (≤40 °C), neutral or alkaline condition. GE indicated a strong anti-digestibility capacity of resisting the hydrolysis of α-amylase and simulated human gastric acid. Interestingly, GE could effectively promote the growth ofLactobacillus bulgaricusandBifidobacterium adolescentis, which was close to fructooligosaccharide. The results of this study could offer valuable information for the application of β-glucan from highland barley as prebiotics in promoting human intestinal health metabolism.Keywords:β-glucan;highland barley;processing characteristics;prebiotics

CAZymes-associated method to explore glycans that mitigate DSS-induced colitis via targeting Bacteroides cellulosilyticus

Xiaoxiang Gao,FuSheng Xu,Tangjun Li,Pan Huang,Leilei Yu,Fengwei Tian,Jianxin Zhao,Wei Chen,Qixiao Zhai

Improving Bacteroides cellulosilyticus abundance is a feasible approach to treating inflammatory bowel disease (IBD). Although B. cellulosilyticus is responsive to dietary components, untargeted manipulation cannot focus on target microbe and lead to an increase in harmful bacteria in the microbiota . Breakthroughs in methods for regulating specific microbes, but the protocols are expensive, time-consuming, and difficult to follow. Glycans based on microbial-carbohydrate-active enzymes (CAZymes) would provide a potential solution. We propose a method based on CAZymes to explore polysaccharides that target specific gut microbes and alleviate diseases. The designed polysaccharides (Arabinogalactan, AG) enrich the abundance of B. cellulosilyticus in single-strain co-cultures, fermentation in vitro , and mouse models in vivo . Supplementation with AG relieved mice from colitis and clinical symptoms. We reveal that AG directly alters B. cellulosilyticus level and cooperative microbes, resulting in remission of colitis. Our glycan design pipeline is a promising way to improve disease through the targeted enhancement of specific microbes.

Structural elucidation and functional characteristics of novel potential prebiotics produced from Limosilactobacillus reuteri N1 GtfB-treated maize starches

Jingjing Dong,Yanli Wang,Ying Chen,Qin Wang,Bo Zhang,Xiaoxiao Li,Zhengyu Jin,Yuxiang Bai

The recently characterized Limosilactobacillus reuteri N1 GtfB (LrN1 GtfB) from glycoside hydrolase family 70 is a novel 4,6-α-glucanotransferase acting on starch/maltooligosaccharides with high enzyme activity and soluble protein yield (in heterogenous system). In this study, the influence of the treatment by LrN1 GtfB on the fine structure and functional characteristics of three maize starches were furtherly investigated and elucidated. Due to the treatment of LrN1 GtfB, the starch molecules were transformed into reuterans containing linear and branched (α1 → 6) linkages with notably smaller molecular weight and shorter chain length. Moreover, the (α1 → 6) linkage ratios in the GtfB-modified high-amylose maize starch (GHMS)/normal maize starch (GNMS)/waxy maize starch (GWMS) increased by 18.3 %/12.6 %/9.0 % as compared to their corresponding controls. In vitro digestibility experiment revealed that the resistant starch content of GHMS, GNMS and GWMS increased by 16 %, 18 % and 25 % as compared to the starch substrates. Furthermore, the butyric acid yielded from GHMS, GNMS and GWMS in the in vitro fermentation experiments were 1.4, 1.5 and 1.4 times higher than those of commercial galactose oligosaccharides . These results indicated that the highly-branched short-clustered reuteran synthesized by LrN1 GtfB might serve as novel potential prebiotics , and provide insights for the synthesis of promising prebiotic dietary fiber from starch.

Hawthorn pectin mitigates high-fat diet induced hyperlipidemia in mice through promoting Dubosiella newyorkensis

Qianyun Zhao,Yubin Liu,Mingxing Li,Long Zhao,Tiangang Wang,Yaqin Xiao,Shulin Wei,Ke Wu,Jiayue Yang,Yaqiu Wang,Yueshui Zhao,Fukuan Du,Yu Chen,Shuai Deng,Jing Shen,Zhangang Xiao,Zhi Li,Shengpeng Wang,Xu Wu

Hawthorn fruit is rich in pectins; however, the gut microbiota-modulating effects of hawthorn pectin remain unclear. In this study, enzyme-extracted hawthorn pectin ( M w : 126.24 kDa; HG/RG-I ratio: 55.64 %/37.93 %; branching degree: 2.44) was fractionated into two components: D1 (spherical, 345.76 kDa, HG/RG-I 57.48 %/37.25 %, branching degree 2.16) and D2 (flexible, 17.20 kDa, HG/RG-I 78.24 %/21.76 %, branching degree 1.33). Hawthorn pectin directly stimulated Dubosiella newyorkensis and Bifidobacterium bifidum growth in vitro . In hyperlipidemic mice, it alleviated dyslipidemia and enriched Lactobacillus and Dubosiella . Parallel experiments confirmed that D. newyorkensis supplementation similarly reduced dyslipidemia and enhanced Lactobacillus abundance, mediated by bacterial acetate production. Only D1 mimicked the pectin's prebiotic activity, promoting D. newyorkensis and B. bifidum growth. The high-RG-I D1, with elevated branching and larger M w , was identified as the key driver of hawthorn pectin's prebiotic effect. These findings highlight that hawthorn pectin and D1 specifically target probiotic D. newyorkensis to synergize with Lactobacillus against hyperlipidemia.

Prebiotics and Postbiotics Synergistic Delivery Microcapsules from Microfluidics for Treating Colitis

Keli Yang,Xiaocheng Wang,Rongkang Huang,Hui Wang,Ping Lan,Yuanjin Zhao

Manipulation of gut microbiota by bacterial metabolites has shown protective effects against colitis; while the efficacy is strictly limited by the poor oral delivery efficiency and single drug usage. Here, a novel prebiotics and postbiotics synergistic delivery microcapsule composed of indole-3-propionic acid (IPA) postbiotic and three prebiotics including alginate sodium, resistant starch (RS), and chitosan via microfluidic electrospray for preventing and treating colitis are proposed. It is found that oral administration of IPA microcapsules (IPA@MC) to mice can exert significant protective effects to colitis, suggesting the therapeutic synergy between prebiotics and postbiotics. Furthermore, the mechanism of the IPA@MC is revealed in modulating the gut microbiota, that is by significantly increasing the overall richness and abundance of short-chain fatty acids (SCFA) producing bacteria such as Faecalibacterium and Roseburia . These results indicate that the prebiotics and postbiotics synergistic delivery microcapsules are ideal candidates for treating colitis.

Chlorella pyrenoidosa Polysaccharides as a Prebiotic to Modulate Gut Microbiota: Physicochemical Properties and Fermentation Characteristics In Vitro

Kunling Lv,Qingxia Yuan,Hong Li,Tingting Li,Haiqiong Ma,Chenghai Gao,Siyuan Zhang,Yonghong Liu,Longyan Zhao

This study was conducted to investigate the prebiotic potential ofChlorella pyrenoidosapolysaccharides to provide useful information for developingC. pyrenoidosaas a green healthy food.C. pyrenoidosapolysaccharides were prepared and their physicochemical characteristics were determined. The digestibility and fermentation characteristics ofC. pyrenoidosapolysaccharides were evaluated using in vitro models. The results revealed thatC. pyrenoidosapolysaccharides were composed of five non-starch polysaccharide fractions with monosaccharide compositions of Man, Rib, Rha, GlcA, Glc, Gal, Xyl and Ara.C. pyrenoidosapolysaccharides could not be degraded under saliva and the gastrointestinal conditions. However, the molecular weight and contents of residual carbohydrates and reducing sugars ofC. pyrenoidosapolysaccharides were significantly reduced after fecal fermentation at a moderate speed. Notably,C. pyrenoidosapolysaccharides could remarkably modulate gut microbiota, including the promotion of beneficial bacteria, inhibition of growth of harmful bacteria, and reduction of the ratio of Firmicutes to Bacteroidetes. Intriguingly,C. pyrenoidosapolysaccharides can promote growth ofParabacteroides distasonisand increase short-chain fatty acid contents, thereby probably contributing to the promotion of intestinal health and prevention of diseases. Thus, these results suggested thatC. pyrenoidosapolysaccharides had prebiotic functions with different fermentation characteristics compared with conventional prebiotics such as fructooligosaccharide, and they may be a new prebiotic for improving human health. Keywords:Chlorella pyrenoidosa;polysaccharides;fermentation;gut microbiota;prebiotic

Effects of simulated digestion and prebiotics properties of polysaccharides extracted from Imperatae Rhizoma based on different pilot processes

Mengge Sun,Haotian Huang,Haibao Tang,Jiajie Chen,Wei Chen,Dongsheng Yang

Recent studies have highlighted the prebiotic potential of natural plant polysaccharides, demonstrating their role in promoting beneficial gut microbiota and improving health. However, research on the digestive properties and prebiotic activities of Imperatae Rhizoma Polysaccharides (IRPs) remains limited. This study investigated fresh Imperatae Rhizoma as the research object. After processing, dry Imperatae Rhizoma and carbonized Imperatae Rhizoma were prepared. Three polysaccharides from the fresh, dry, and carbonized Imperatae Rhizoma were extracted with traditional hot water. And another polysaccharide was obtained by cold water extraction from fresh Imperatae Rhizoma. Total four IRPs were extracted and named: IRPs-F, IRPs-D, IRPs-C, and IRPs-J. This study evaluated the prebiotic activity of four polysaccharides derived from the roots of thatch, demonstrating their resistance to digestion, their ability to promote probiotic growth, and their enhancement of short-chain fatty acid (SCFA) production. The final results show that four IRPs exhibit strong resistance to digestion and IRPs-F ability to promote the growth of beneficial probiotics, making it a promising candidate for functional foods aimed at improving intestinal health, immune regulation, and metabolic benefits. This research is highly relevant to food microbiology and holds significant potential for application in the functional food and gut health sectors.

Chemical characterization of Cassia fistula polysaccharide (CFP) and its potential application as a prebiotic in synbiotic preparation

Dawood Hosni Dawood,Mohamed Samir Darwish,Asmaa A. El-Awady,Azza H. Mohamed,Ahmed A. Zaki,Mohamed A. Taher

Prebiotics are non-digestible food ingredients that are selectively fermented by probiotics. The aim of this study was to investigate the chemical properties of a polysaccharide extracted from Cassia fistula mature fruit pulp and to evaluate its effects on probiotic strains: L. casei, L. rhamnosus, E. coli Nissle 1917 (EcN), and E. faecalis. These strains were compared for their growth behavior in culture media supplemented with different Cassia fistula polysaccharide (CFP) concentrations. The molecular weight of CFP was approximately 8.707 × 105 Da. The recovered polysaccharide contained a low percentage of crude protein (4.4%). Aspartic acid, glutamic acid, and proline were the most abundant amino acids. Glucose and mannose were the predominant sugars followed by arabinose and rhamnose. L. casei grew faster at high CFP concentrations (2%) compared with the lower concentrations of CFP. The highest values for the prebiotic index and prebiotic activity score were observed for L. casei treated with 2% CFP, and it may be considered a prebiotic due to its high resistance against α-amylase and acidic conditions. CFP provides two ways to adjust nitric oxide (NO) synthesis in macrophages. Finally, the use of 1.5 and 2% CFP for cultured milk production significantly shortened the fermentation period from 210 min to 180 min and 150 min, respectively.

Fermented Soybean Dregs by Neurospora crassa: a Traditional Prebiotic Food

Zhou Ruolin,Ren Zhiqing,Ye Jun,Fan Yawei,Liu Xiaoru,Yang Jianyuan,Deng Ze-Yuan,Li Jing

Soybean dregs fermented by Neurospora crassa is a typical traditional food in Gannan district of China. In this study, in vitro imitated gut fermentation was carried out to evaluate whether the oligosaccharides from this fermented soybean dregs had potential prebiotic properties. 11.91% of oligosaccharides were extracted from the fermented soybean dregs at the optimized condition which of 1:25 for ratio of soybean dregs (g) to 50% ethanol (ml), 90 min of extracted duration at 70 °C for twice. The soybean dreg oligosaccharides (SBOS) were progressively purified with Sevag method and on columns filled with AB-8 macroporous resin, and then identified as cellobiose by HPLC-ESI-MS and FT-IR. Oligosaccharides of soybean dregs with 800 mg/L significantly decreased pH value ( p  < 0.05) and ammonia N concentration ( p  < 0.05), and increased short chain fatty acid (SCFA) level ( p  < 0.05) in imitated gut fermentation compared with control group. It was shown that this fermented soybean dregs could be a potential prebiotic food.

Bifidogenic effects of Cordyceps sinensis fungal exopolysaccharide and konjac glucomannan after ultrasound and acid degradation

Ang-Xin Song,Yu-Heng Mao,Ka-Chai Siu,Jian-Yong Wu

The bifidogenic effects of exopolysaccharide (EPS) of a medicinal fungus ( Cordyceps sinensis ) and a well-known food polysaccharide konjac glucomannan (KGM) with different molecular weight (MW) ranges were evaluated through in vitro experiments in liquid cultures of Bifidobacteria . Native EPS and KGM were partially degraded with power ultrasound (US) to improve the water solubility, and further hydrolysed with trifluoroacetic acid to much lower MW. The acid-hydrolysed fractions (EPS-AH and KGM-AH) supported the growth of all five tested bifidobacterial species, while the US-degraded high MW fractions, EPS-US and KGM-US, could only slightly support the growth of some species. All EPS fractions increased the acetic acid production of most bifidobacterial species. Most remarkably, the high MW EPS-US, EPS-AH and KGM-US fractions significantly enhanced the cell viability with much higher colony forming unit (CFU) counts, suggesting a protective effect of these high MW polysaccharides for the bacterial survival . The results have shown that MW was a significant factor on the bifidogenic properties of partially degraded EPS and KGM.

Fractionation, preliminary structural characterization and prebiotic activity of polysaccharide from the thin stillage of distilled alcoholic beverage

Wu Sun,Jingjing Xu,Zhongwei Yin,Huan Li,Juan Li,Li Zhu,Zhitao Li,Xiaobei Zhan

Thin stillage is the distillery stillage from distilled alcoholic beverage, which has great economic potential. In this study, a novel polysaccharide from the thin stillage of distilled alcoholic beverage (CRWP1) was purified with a molecular weight of 8554 Da. CRWP1 was mainly composed of glucose, galactose , mannose , and glucuronic acid at approximate percentages of 91.56, 2.13, 1.61 and 1.84, respectively. The structural chain of CRWP1 consisted of (1→4)-linked -Glcp, (1→)-linked-Glcp, (1→3,6)-linked-Manp, (1→6)-linked-Galp, (1→6)-linked-Glcp and contained with α and β glycosidic bonds by fourier transform infrared spectroscopy (FTIR), methylation and nuclear magnetic resonance (NMR) analysis. Following 48 h of fecal fermentation in a bionic gastrointestinal reactor, CRWP1 could promote the relative abundance of Bacteroides , Bifidobacterium , and Collinsella at the genus level, as well as the production of short-chain fatty acids (SCFAs). Furthermore, CRWP1 fermentation increased production of organic acids (especially butyric and oleic acids) and polyols (such as 1,3-propanediol). This is the first report on the prebiotic potential of CRWP1 and it provides a foundation for the potential application in economical food industry.
