Yields of three acids during simulated fermentation of inulin and xylo-oligosaccharides enhanced by six exogenous strains

Kong Lu,Zhao Xin-Huai

Three acids (acetic, propionic and butyric acids) yielded from simulated colonic fermentation of inulin and xylo-oligosaccharides (XOS) in the absence and presence of six exogenous strains were investigated in this study. Both inulin and XOS were fermented by fecal extract (FE) prepared from healthy adults and by an exogenous strain for 48 h, and three acids generated in fermentation products at five time periods were detected by gas chromatography. Inulin and XOS during fermentation were capable of producing more propionic and butyric acids, respectively, whilst inoculation using one strain culture into fermentation systems mostly increased yields of three acids. In inulin-containing fermentation systems, these strains increased yields of propionic and butyric acids by 6–34 and 206–250%, respectively. In XOS-containing fermentation systems, these strains did not enhance yield of propionic acid, but increased yield of butyric acid by 50–96%. In general, Lactobacillus brevis, Lactobacillus plantarum, Lactobacillus rhamnosus, Enterococcus faecalis and Enterococcus faecium were more efficient than Lactobacillus acidophilus to increase yields of propionic and butyric acids. It is thus concluded that the six strains in total displayed beneficial effects on simulated fermentation of inulin and XOS through enhancing yields of three acids especially propionic and butyric acids with healthcare functions.

Hypocholesterolaemic action of Lactobacillus casei F0822 in rats fed a cholesterol-enriched diet

C.F. Guo,J.Y. Li

Elevated serum cholesterol is a major risk factor for coronary artery disease. Probiotics may manage elevated cholesterol. The hypocholesterolaemic effects of Lactobacillus casei F0822 and its functional mechanisms were investigated in rats . The serum total cholesterol, low-density lipoprotein cholesterol and liver total cholesterol levels significantly decreased in rats fed a high-cholesterol diet plus drinking water supplemented with viable cells of strain F0822; however, there was no significant difference in the serum high-density lipoprotein cholesterol levels among the treatment groups. The hypocholesterolaemic mechanisms of strain F0822 were attributed to its ability to suppress the reabsorption of bile acids into the enterohepatic circulation though hydrolysis of conjugated bile acids in the small intestine, binding of deoxycholic acid and hyodeoxycholic acid in the large intestine, and increase in propionate:acetate ratio in the large intestine in rats. Strain F0822 may be a promising probiotic culture with potential hypocholesterolaemic action in human.

Macro/Microgel-Encapsulated, Biofilm-Armored Living Probiotic Platform for Regenerating Bacteria-Infected Diabetic Wounds

Huilong Xin,Zhe Cai,Jiahui Hao,Jing An,Yi Li,Min Wen,Zhaojun Jia

Infectious diabetic wounds pose an arduous threat to contemporary healthcare. The combination of refractory biofilms, persistent inflammation, and retarded angiogenesis can procure non-unions and life-threatening complications, calling for advanced therapeutics potent to orchestrate anti-infective effectiveness, benign biocompatibility, pro-reparative immunomodulation, and angiogenic regeneration. Herein, embracing the emergent “living bacterial therapy” paradigm, a designer probiotic-in-hydrogel wound dressing platform is demonstrated. The platform is constructed employing a “macrogel/microgel/biofilm” hierarchical encapsulation strategy, with Lactobacillus casei as the model probiotic. Alginate gels, in both macro and micro forms, along with self-produced probiotic biofilms, served as encapsulating agents. Specifically, live probiotics are enclosed within alginate microspheres, embedded into an alginate bulk matrix, and cultivated to facilitate biofilm self-encasing. This multiscale confinement protected the probiotics and averted their inadvertent escape, while enabling sustained secretion, proper reservation, and localized delivery of therapeutically active probiotic metabolites, such as lactic acid. The resulting biosystem, as validated in vitro/ovo/vivo, elicited well-balanced antibacterial activities and biological compatibility, alongside prominent pro-healing, vasculogenic and anti-inflammatory potencies, thus accelerating the regeneration of infected full-thickness excisional wounds in diabetic mice. Such multiple encapsulation-engineered “all-in-one” probiotic delivery tactic may shed new light on the safe and efficient adoption of live bacteria for treating chronic infectious diseases.

In vitro assessment of the effect of four polysaccharides on intestinal bacteria of mice with colitis

Shanshan Zhang,Yonggan Sun,Qixing Nie,Jielun Hu,Wenwen Su,Zheyu Guo,Yanli Zhang,Shaoping Nie

Polysaccharides are typically resistant to digestion in the gastrointestinal tract and are instead degraded by gut microbiota in the colon. As such, they are commonly employed as carriers for colon-targeted drugs, with the potential to regulate gut microbiota. Pectin, carrageenan, guar gum, and xanthan gum are commonly used polysaccharide carriers, but their degradation in the gut microbiota under colitis conditions as well as their effects on gut microbiota remains unclear. In this study, we performed the in vitro fermentation of these four polysaccharides using colonic content microbiota from mice with colitis and evaluated the degree of polysaccharide degradation and the effects on pH, short-chain fatty acids, and gut microbiota. Our findings indicate that pectin and guar gum had a greater degree of degradation and promoted the production of butyrate, inhibited the proliferation of harmful bacteria (e.g., Enterobacteriaceae ), and increased beneficial bacteria (e.g., Bifidobacterium ). In contrast, carrageenan and xanthan gum promote the proliferation of Enterobacteriaceae . These results provide theoretical guidance for the selection of drug delivery carriers for inflammatory bowel disease treatment and provide information on the relationship between polysaccharides and gut microbiota.

Aqueous extracts of Elsholtzia ciliata and Hovenia dulcis ameliorate loperamide-induced constipation in mice by promoting intestinal peristalsis and barrier function and the abundance of intestinal beneficial bacteria

Junnan Wu,Qilei Xin,Shuo Wang,Xu Zhang,Chunping Jiang

Objective: The aims of the present study were to determine the efficacy of edible traditional Chinese medicines (ETCMs) in treating constipation, verify their laxative effects, and conduct preliminary investigations into their mechanisms of action. Methods: ICR mice were treated with loperamide to induce constipation, and various fecal parameters, including fecal volume, water content, and intestinal transport function, were measured in these constipation model mice to screen for ETCMs with laxative properties. The mechanism of action was preliminarily explored by examining changes in the intestinal mucosal structure, protein expression levels, and alterations in intestinal flora composition. Results: In ICR mice with loperamide-induced constipation, Elsholtzia ciliata aqueous extract (ECAE) and Hovenia dulcis aqueous extract (HDAE) significantly ameliorated constipation symptoms, mitigated colonic pathological tissue damage, significantly increased the expression levels of proteins associated with the promotion of intestinal peristalsis [Stem Cell Factor Receptor (c-Kit) and Stem Cell Factor (SCF)] and the maintenance of the intestinal barrier [Zonula Occludens-1 (ZO-l), Occludin and Claudin-l], and promoted beneficial intestinal bacterial colonization. Conclusion: ECAE and HDAE ameliorated constipation in mice, and their mechanism of action may be related to the increased abundance of intestinal bacteria such as Turicibacter, Olsenella, and Odoribacter, which contribute to higher butyrate production. This increase in butyric acid reduces inflammation, improves intestinal barrier function, and increases the abundance of beneficial intestinal bacteria.

Lactiplantibacillus plantarum S1 as a Novel Dual-Functional Probiotic Strain for High-Efficiency Organoselenium Biotransformation in Functional Food Development

Lin Yuan,Jianfeng Yuan,Chen Gao,Haoming Zhao,Chengye Wu,Zhong-Hua Yang

The microbial conversion of inorganic Se into bioactive organoselenium compounds represents a cutting-edge strategy for developing functional foods with enhanced nutritional value. This study introduces Lactiplantibacillus plantarum S1, a novel Se-enriched probiotic strain isolated from traditional Chinese sauerkraut, and systematically optimizes its capacity for selenite biotransformation. Critical fermentation parameters—including sodium selenite supplementation timing (2 μg/mL added at mid-log phase, 7 h post-inoculation), pH (5.0), and anaerobic cultivation duration (12 h)—were identified as key determinants of conversion efficiency. The optimized protocol achieved a 72.3% organoselenium conversion yield, producing 626.6 μg/g cellular organoselenium while maintaining probiotic viability (2.28 × 10 9 CFU/mL). Se speciation analysis demonstrated that 78.51% of intracellular Se existed in organic forms, with protein-bound Se constituting the predominant fraction (85.33%), followed by polysaccharide-associated (6.42%) and nucleic acid-linked (3.38%) species. The strain’s dual functionality as both an efficient Se bioconverter and a resilient probiotic carrier highlights its potential for nutraceutical applications. These findings not only establish a robust bioprocess for Se-enriched probiotic production but also reveal mechanistic insights into preferential Se incorporation into protein matrices. This work bridges microbial Se metabolism research with scalable functional food innovation, offering a sustainable platform for developing Se-fortified products with dual health benefits.

Lactobacillus reuteri ZJ617 attenuates metabolic syndrome via microbiota-derived spermidine

Ma Yanfei,Zhong Yifan,Tang Wenjie,Valencak Teresa G.,Liu Jingliang,Deng Zhaoxi,Mao Jiangdi,Liu Daren,Wang Shanshan,Wang Yuhao,Wang Haifeng

Metabolic syndrome (MetS) is a difficult-to-manage disease that poses a significant risk to human health. Here, we show that the supplementation of Lactobacillus reuteri ZJ617 ameliorates symptoms of MetS in mice induced by the high-fat diet. L. reuteri ZJ617 modulates host metabolism by interacting with the microbiome, resulting in the production of spermidine synthesized by the microbiota. L. reuteri ZJ617 serves as a source of substrates for the microbiota to synthesize spermidine, hence preventing the decline of bacteria responsible for spermidine production. Spermidine treatment mimics the metabolic effects of L. reuteri ZJ617, whereas pharmacological inhibition of spermidine biosynthesis in mice abolishes these benefits. Our findings reveal the mechanism by which L. reuteri ZJ617 alleviates MetS symptoms and provide support for its potential use as a probiotic for promoting metabolic health.

Vibrio sp. 33 a potential bacterial antagonist of Vibrio splendidus pathogenic to sea cucumber (Apostichopus japonicus)

Ningning Liu,Shanshan Zhang,Weiwei Zhang,Chenghua Li

Recently, the use of probiotics as an alternative strategy to control bacterial diseases in aquaculture has garnered much attention due to the merits of environmental friendliness. In this study, a bacterium identified as Vibrio sp. V33 was isolated from healthy sepia and was determined to have strong antagonistic activity towards the pathogenic isolate Vibrio splendidus Vs using the disk diffusion method. The antagonistic substance was secreted into the supernatant and was thermo-stable. The antagonistic activity of Vibrio sp. V33 may be attributed to its higher ability to compete for iron than that of V . splendidus Vs, as deduced from siderophore quantification using the CAS method. Number of colony counts from seawater and Apostichopus japonicus infection experiments showed that Vibrio sp. V33 significantly decreased the quantity of V . splendidus Vs in natural seawater and promoted the survival rate of A . japonicus infected by V . splendidus , with a relative percentage survival of 43% under the current conditions. Our findings suggest that V33 could be potentially used as potential bacterial antagonist for controlling V . splendidus infections in sea cucumber aquaculture. Statement of relevance It might protect A . japonicus from V . splendidus infection.

Probiotics-fermented blueberry juices as potential antidiabetic product: antioxidant, antimicrobial and antidiabetic potentials

Hao Zhong,Abdullah,Minjie Zhao,Jun Tang,Lingli Deng,Fengqin Feng

BACKGROUND Fermentation is a traditional food-preserving technique. It is an effective process, widely used to enrich the nutrients diversity and bioactivity of the fermented foods since ancient times. This study aimed at investigating the effects of various fermentation starters on the physicochemical, antioxidant, antimicrobial, and antidiabetic properties of blueberry juices. The blueberry juices were fermented by natural fermentation (NFBJ), self-made starters fermentation (SFBJ), and commercial starters fermentation (CFBJ); fresh blueberry juice (BBJ) was processed without fermentation for comparison. RESULTS Probiotics-fermented blueberry juices (SFBJ and CFBJ) showed less total and reducing sugars, higher titratable acidity, and a wider variety and higher amounts of organic acids than non-fermented blueberry juice (BBJ) did. All the fermented blueberry juices (NFBJ, SFBJ, and CFBJ) showed significantly ( P < 0.05) higher antioxidant potentials than that of BBJ measured by 2,2′-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid, cupric-reducing antioxidant capacity, and ferric-reducing ability power assays. The SFBJ exhibited the highest antibacterial activities against Escherichia coli , Staphylococcus aureus , and Salmonella Typhimurium, with inhibition zone diameters of 38.84 ± 1.74 mm, 34.91 ± 1.53 mm, and 36.18 ± 3.16 mm respectively. Compared with BBJ, the α -glucosidase inhibitory activity of the SFBJ and CFBJ increased by two‑to threefold. The α -amylase inhibitory activity of the SFBJ and CFBJ increased by 600%, whereas the spontaneous fermentation showed no improvement. The SFBJ and CFBJ promoted glucose consumption of HepG2 cell lines, indicating the promising potential for a higher glucose bio-utilization. CONCLUSIONS The SFBJ and CFBJ showed remarkable improvements in the antioxidant, antimicrobial, and antidiabetic activities compared with non-fermented and spontaneous fermented juices, indicating their promising potentials as an antihyperglycemic agent. © 2021 Society of Chemical Industry

Orally Administered Functional Polyphenol-Nanozyme-Armored Probiotics for Enhanced Amelioration of Intestinal Inflammation and Microbiota Dysbiosis

Yong Zhu,Ziqu Fang,Jie Bai,Longhui Wang,Jiaqing Chen,Zehua Zhang,Qiang Wang,Weiwei Sheng,Xueyin Pan,Zhenyuan Gao,Dengqiu Xu,Pengkai Wu,Beicheng Sun

Maintaining microbiota balance and enhancing the antioxidant performance of nanozyme-based probiotic systems are crucial for effective inflammatory bowel disease (IBD) therapy. Despite significant advancements, developing a green and safe coating technology that functionalizes probiotics with nanozymes while preserving the activity of both components remains a challenge. To address this, chitosan-modified epigallocatechin gallate (EGCG-CS, EC)is synthesized, leveraging the intrinsic adhesive and coordination properties of polyphenols to capture gold nanozymes (AuNPs), forming ECA complexes that enhance nanozyme activity. When coated onto Escherichia coli Nissle 1917 (EcN), the resulting ECA@EcN system effectively scavenged reactive oxygen species (ROS), improving probiotic viability and promoting colon accumulation. Mechanistically, ECA protected EcN by suppressing the activation of the Flagellar Assembly and Branched-Chain Amino Acid Synthesis pathways, ultimately alleviating inflammation and modulating intestinal microbial communities to relieve IBD symptoms. Given the biocompatibility of its components and the environmentally friendly assembly approach, this polyphenol-nanozyme-armored probiotic system represents a promising platform for IBD treatment.

Metal-phenolic networks enhanced the protection of excipients for probiotics during freeze-drying

Tong Zhang,Chen Wang,Shengpeng Su,Axiang Sun,Ting Du,Jianlong Wang,Julong Liu,Wentao Zhang

Probiotic powder using a single protective method during freeze-drying is insufficient vitality because it lacks adequate protection. Here we developed a protection strategy through biointerfacial phenolic self-assembly to enhance the protection of excipients for probiotics to address existing challenges during freeze-drying. This strategy could strengthen the connections of excipients and phenolic protective layers containing hydroxyl groups with water molecules, improving the hydration layer's preservation and shielding bacteria from damage. The results indicated that, compared with origin probiotics, protected probiotics maintained higher viability at approximately 91 % and higher ATPase activity and exhibited a better survival rate in various environmental challenges after freeze-drying. The broad applicability of this protection strategy was confirmed across other LAB strains. Additionally, the protected probiotics demonstrated superior shelf life during 30 days of storage, indicating promising prospects for preparing bacterial powder via freeze-drying.

Harnessing Akkermansia muciniphila Membrane Coating for Probiotic Therapy in Inflammatory Bowel Disease

Zhenzhen Su,Weizheng Li,Shang Li,Ruiqi Wang,Dashuang Sheng,Guo-ping Zhao,Hongqian Cao,Lei Zhang

Inflammatory bowel diseases (IBDs) are characterized by dysregulated inflammatory responses, increased reactive oxygen species (ROS), and compromised intestinal barrier function. Current clinical treatments often yield suboptimal responses and significant side effects. Given the great potential of Akkermansia muciniphila (AKK) bacterial membrane components in IBD treatment and the possible pathogenicity of live bacteria, to essentially address these challenges, we develop a natural membrane-based single-cell coating, incorporating a self-assembled AKK bacterial membrane (AKM) and astaxanthin (AST), forming a protective biological barrier on probiotics. Utilizing Escherichia coli Nissle 1917 (EcN) for the oral delivery system, we engineer AKM-AST@EcN nanocoated probiotics, demonstrating exceptional gastrointestinal (GI) tract tolerance and robust bioactivity. Critically, our findings highlight the potential of AKM-AST@EcN as a promising therapeutic approach for IBD treatment while also presenting a distinctive strategy for probiotic oral delivery systems.
