Noncovalent chitosan (CS)/hyaluronic acid (HA) hydrogel has been characterized by favorable cytocompatibility, biodegradability and non-toxicity. It bears a close resemblance to human skin and holds promising potential applications in medical engineering. Nevertheless, the CS/HA hydrogels have yet to achieve widespread application due to the relatively weak toughness. In this study, salts were utilized to regulate the mechanical properties of CS/HA hydrogel. The results indicated that copper nitrate was the most effective regulator, which transformed intramolecular hydrogen bonds and electrostatic interactions into intermolecular hydrogen bonds and cation chelations, respectively. The regular domains in the hydrogel were reduced while the crosslink was strengthened. Consequently, the toughness of the hydrogel was increased to 7.8 MJ/m3, 1253-fold that of covalent CS/HA hydrogel. The salt effect was replicated in the other two hydrogels, attesting to its generality. Hence, salt regulation proves to be an effective way to enhance mechanical properties of hydrogel. In addition, the copper nitrate-regulated hydrogel exhibited favorable drug delievring behavior and photothermal response. Under the exposure of near-infrared light, the release rate and amount of the drug loaded in the hydrogel were increased by 40% and 39% respectively within 20 minutes, manifesting a remarkable potential for application as a drug carrier.

DNA-functionalized gold nanoparticles (DNA–AuNPs) have shown great potential and exciting opportunities for constructing machine-like nanodevices. Nonthiolated DNA can be grafted onto gold surfaces via DNA bases, such as polyadenine (polyA)–DNA. The colloidal stability of polyA–DNA–AuNPs has a significant dependency on salt and pH that affects the assembly of AuNPs and their application in polyA–DNA molecular machines. High salt and low pH value contribute to the stabilization of polyA–DNA–AuNPs. In acid conditions, adenine can be protonated and becomes positively-charged, thus enhancing the adsorption of polyA–DNA onto the gold surface by electrostatic interactions; coordination of multiple interactions achieves a high DNA grafting density and colloidal stability. In addition, the length of adenine has an important effect on the efficiency of the DNA machine, while the length of thymine has little effect when the thymine length is less than or equal to seven. The assembly of AuNPs driven by dynamic polyA–DNA molecular machines was successfully accomplished with A5-DNA and A9-DNA. A moderate concentration of catalyst oligomer (50 nM) could improve the DNA hybridization efficiency. The A9-DNA based molecular machine is more efficient than the A5-DNA based one because of the larger amount of A9-DNA on the AuNPs, which increases the probability of collisions between complementary DNA strands. Therefore, polyA–DNA functionalized nanoparticles can be used as a basic unit to construct assembly-ordering structures and achieve dynamic molecular machines to be applied in the molecular diagnostics field.

Hydrogels are versatile functional materials with applications in biomedical devices, sensors, and energy systems. However, tailoring their mechanical properties remains a critical challenge for expanding their practical utility. Here, the mechanical properties of poly(vinyl alcohol) (PVA) hydrogels prepared via the freeze–thaw method were effectively tailored through the incorporation of various chloride salts. This study reveals the molecular mechanisms by which salt ions influence the water structure, polymer chain interactions, and freezing behavior to modulate hydrogel performance. Using Raman spectroscopy, X-ray diffraction (XRD), and Fourier-transform infrared spectroscopy (FTIR), we elucidate how cation-specific effects alter the crystallinity, cross-linking density, and hydrogen bond network in the hydrogels. Hydrogels containing K+ exhibited superior rigidity and toughness due to enhanced crystallinity and optimized water–polymer interactions, while hydrogels of Mg2+ and Ca2+ had much lower modulus and adhesion strength since the divalent ions reduced freezing points resulting in reduced polymer aggregation. These findings provide a molecular-level understanding of the salting effects on hydrogel properties, offering new insights into designing materials with tunable mechanical performance for diverse applications.
