Recently, Pickering emulsions stabilized by edible particles have attracted significant attention from the scientific community and food industry owing to their surfactant-free character. However, those edible particles are mostly used for stabilizing oil-in-water emulsions, whereas those for water-in-oil emulsions are very limited. In this article, stable water-in-oil Pickering emulsions were prepared through dispersing phytosterol particles in oil phase, and the effects of antisolvent treatment, the type of oil, particle concentration, and water fraction on the stability, type, and morphology of these emulsions were investigated. In addition, the release profile of salt as a model aqueous compound from these emulsions has also been studied. Results showed that due to its higher water content, the antisolvent pretreatment of phytosterol in the ethanol/water system facilitated the dispersion of dried phytosterol particles into oil phase as microcrystals. Water-in-oil Pickering emulsions with droplet sizes of 80–100 μm were fabricated at phytosterol concentrations of 1.5–3% w/v and water fractions of 0.2–0.6. The dissolved phytosterol molecules in oil phase could help in emulsion stabilization through interfacial crystallization during emulsification, evidenced by polar microscopic observations. Moreover, the salt release from phytosterol-stabilized Pickering emulsions showed a temperature-dependent profile which could have potential application in a controlled-release system. The current study provided important information for fabrication of stable water-in-oil emulsion using natural particles.

Nanoscaled light-triggered nitric oxide (NO) delivery vehicles with the ability of near-infrared (NIR) fluorescence imaging was presented, which consisted of chitosan (CS)-based S-nitrosothiols (SNO) and encapsulated silver sulfide quantum dots (Ag2S QDs). CS-SNO compounds that bore NO-storing functional groups were prepared via amino modification of chitosan. Water-soluble Ag2S QDs were synthesized and conjugated with the CS-SNO compounds with the aid of ethylenediaminetetraacetic acid (EDTA). The biocompatible Ag2S-CS-SNO nanospheres, with dimension of ∼117 nm, exhibited bright NIR fluorescence and satisfactory photostability under NIR irradiation. The Ag2S-CS-SNO nanospheres could release NO under irradiation of UV or visible light at physiological pH and temperature yet would hardly release NO if NIR irradiation was applied. Cell imaging was successfully performed, demonstrating that the Ag2S-CS-SNO nanospheres could emit readily observable NIR fluorescence and release NO in living cells. The NIR fluorescence imaging of the Ag2S-CS-SNO nanospheres did not interfere with the light-triggered NO release from them, which would provide new perspectives for the application of multifunctional nanostructured materials in diagnostics and imaging.

Keratin-based biomaterials have emerged as potential candidates for various biomedical and biotechnological applications due to their intrinsic biocompatibility, biodegradability, mechanical durability, and natural abundance. The objective of our study is to explore the feasibility and potential applications of keratin for promoting nitric oxide (NO) release. Herein, keratin was first extracted from human hair and then modified with iodoacetic acid to afford S-carboxymethyl keratin. The amino acid residues, isoelectric point, and molecular weight of keratin were quantified with amino acid analyzer, zeta potential measurement, and SDS-PAGE method. Interestingly, human hair keratin was proved to be capable of promoting NO release from endogenous donor of S-nitrosoglutathione (GSNO) in the presence of glutathione.

The aim of this study was to develop an EHD printing method to fabricate graphene-loaded polycaprolactone (PCL)/polyethylene oxide (PEO) dual-core matrices. Graphene was incorporated in shell PCL components, while gelatin and dopamine hydrochloride (DAH) were encapsulated in two PEO cores to enhance biocompatibility of graphene-loaded matrices. Furthermore, the effect of PEO concentration on dual-core fiber formation was evaluated. The influence of process parameters (applied voltage, inner flow rate, outer flow rate and X-Y-Z collector stage speed) on dual-core fiber morphology was evaluated. Our findings show graphene-loaded structures to possess two inner cores and increasing graphene content yields matrices with smoother surfaces, causing a slight reduction in their contact angle behavior. Furthermore, the addition of graphene to matrices results in reduced elasticity. DAH release from matrices comprising various graphene concentrations showed no significant difference and drug release mechanism was diffusion based. In vitro biological tests indicate resulting graphene-loaded dual-core matrices exhibit good biocompatibility and also improve PC12 cell migration. The findings suggest matrices to have potential applications in nerve restoration and regeneration.

Conventional hydrogel drug delivery systems are limited in recapitulating the natural spatiotemporal progression of bone regeneration due to their passive release mechanisms. Here, an enzyme-responsive deoxyribonucleic acid (DNA)-polyethylene glycol (PEG) hydrogel is developed to actively coordinate the sequential processes of angiogenesis, osteogenesis, and mineralization through rational material design. The hydrogel integrates matrix metalloproteinase (MMP)-cleavable peptide-crosslinked PEG networks conjugated with actin-stabilized vascular endothelial growth factor (VEGF)-binding DNA strands, enabling spatiotemporally controlled therapeutic release. Upon implantation, MMPs trigger the hydrogel degradation, releasing VEGF to induce angiogenesis while simultaneously promoting osteogenic differentiation. The actin-stabilized DNA framework maintains structural integrity during this stage, preventing premature phosphate release. Subsequent matrix remodeling liberates nuclease that catalyzes DNA to generate phosphate ions, which synergize with peptides in hydrogels and endogenous calcium to drive mineralization. Molecular dynamics simulations reveal the underlying mechanism of hydrogel-mediated mineralization, demonstrating enhanced calcium phosphate formation. Such temporally controlled cascade significantly improves vascular density, osteogenic marker expression, and mineral deposition compared to controls. This work establishes a bioresponsive platform that dynamically interacts with the biological microenvironment to orchestrate multi-phase bone regeneration, offering new possibilities for complex tissue repair.

Osteogenic–osteoclastic microenvironment imbalance impairs osseointegration between joint prostheses and the marrow cavity, which is a crucial contributor to the high incidence of complications such as implant loosening and periprosthetic fractures in osteoporosis (OPs). Previously, we employed hydrogel-based bisphosphonate (BPs) coatings on 3D-printed porous titanium alloy implants (pTi) to modulate the OPs microenvironment through controlled release. However, challenges persisted, including the limited duration of BPs release and associated organic material degradation-induced toxicity. To address these issues, we modified pTi surfaces by introducing chemically stable zeolites and BPs. In this study, we applied Ca 2+ -functionalized hierarchical zeolite coating on pTi for the first time. By combining mesoporous adsorption and Ca 2+ chelation, we achieved efficient encapsulation of zoledronate (ZOL), creating a bioactive interface characterized by a spatial gradient structure with a “microporous-mesoporous-macroporous” morphology. The coating achieves the release of ZOL through the mesoporous structure and facilitates the gradual release of chelated ZOL via ion exchange, resulting in hierarchical drug delivery. Moreover, the spatial gradient structure of the coatings significantly influenced cell extension. Simultaneously, we constructed a bioactive coating with a drug concentration gradient, achieving graded drug release within the microenvironment, which facilitated the accurate regulation of osteogenesis and bone resorption. This coating has the potential to substantially enhance the management of postoperative complications in OPs patients undergoing prosthetic replacement surgery by achieving a balanced osteogenic-osteoclastic response.
