Skin damage caused by excessive UV exposure has gradually been one of the most common skin diseases, leading to desquamation, scab formation, inflammation and even skin cancer. Animal-derived hydrolyzed collagen peptides have been developed to treat UV-damaged skin; however, they have raised severe concerns such as potential viral transmission, random sequences and the lack of triple helix structure. Nano collagen, a novel type of short collagen, has attracted increasing attention in the mimicking of natural collagen, while their applications in UV-damaged skin treatment remains vacancies. Herein, we have created a series of nano collagens, and for the first time studied their capability to accelerating UV-damaged skin healing. Nano collagens, consisting of repetitive (GPO)n triplets and a GFOGER motif, display stable triple-helical structure, significantly promoting the fibroblasts adhesion, proliferation, and migration. The repair effects of nano collagens have been investigated using acute UV-damaged skin mouse model. Combo evaluations indicate nano collagen contributes to recovering the dermis density and erythema index of UV-damaged skin. Histological analysis further demonstrates their capability on promoting the healing of damaged skin by accelerating the re-epithelialization and collagen regeneration. These highly bioactive triple‑helical nano collagens present a novel strategy for the treatment of UV-damaged skin, providing promising applications in cosmetics and dermatology.

Soft tissue deficiencies profoundly impact the daily lives, and mental well-being of patients. Microspheres facilitate collagen synthesis by establishing a conducive environment for fibroblast growth. Existing synthetic polymer microspheres are typically prepared through emulsification and cross-linking at normal temperatures. However, residues of cross-linking agents can adversely affect biocompatibility, thereby limiting their biomedical applications. Agarose has garnered significant attention owing to its biodegradability and excellent biocompatibility. We have for the first time developed high-temperature emulsification coupled with low-temperature gelation for fabrication of agarose microsphere implants with well-controlled size for skin tissue enhancement. The agarose microspheres exhibited favorable sphericity and dispersion, possessing a uniform particle size with an average diameter of 37.24 μm. Furthermore, the microspheres demonstrated commendable injectability and biodegradability. Additionally, the implants displayed remarkable biocompatibility, effectively promoting the proliferation of human foreskin fibroblast-1 (HFF-1) cells. The microspheres exhibited no systemic toxicity and induced no hemolytic or thermogenic reactions. In photoaged mice skin models, the agarose microspheres augmented dermal density, and enhanced skin elasticity. The microspheres showed the capacity to stimulate the regeneration of collagen fibers. The agarose microspheres offer a novel avenue for soft tissue filling and hold significance in the field of tissue engineering and skin regeneration.

Excessive ultraviolet (UV) exposure can lead to sunburn, characterized by skin barrier damage, inflammation, pain, and an increased risk of skin cancer. Recombinant collagens have gradually attracted attention due to their high purity, low immunogenicity, batch-to-batch consistency, and excellent solubility. Additionally, the type of dressing significantly affects wound repair. Gels are ideal for sunburn treatment because they maintain a moist environment, adhere firmly, and do not need to be removed. Herein, we have created bioactive triple-helical recombinant collagen (THRC) gels for improved healing of sunburned skin. The THRC gels remained stable after a three-month stability test, displaying a rheological behavior characteristic of non-Newtonian pseudoplastic fluids. In vivo skin irritation tests conducted on New Zealand rabbits demonstrated that THRC gels were safe for use. A sunburned mice model was established to study the biological effects of THRC gels. Non-invasive combo evaluations indicated that THRC gels exhibited an exceptional capability of recovering dermis density, erythema index (EI), hydration, and transepidermal water loss (TEWL) of sunburned skin to a healthy state. Histological observations revealed that THRC gels significantly enhanced the repair of damaged skin by accelerating the recovery process, promoting collagen deposition and regeneration. Molecular biological characterizations further demonstrated their remarkable antioxidant properties, including the inhibition of lipoperoxidation and the enhancement in superoxide dismutase (SOD) and glutathione (GSH) activities. These safe and bioactive recombinant collagen gels provide a novel approach for sunburn repair and show potential for long-term cosmetic benefits.
