Integrated network pharmacology and GC–MS-based metabolomics to investigate the chemical profile and efficacy of Anemarrhenae Rhizoma and its processed products

Wenyu Li,Jinhuan Wei,Xi Tian,Huiyi Zhang,Mengxin Yang,Yukun Niu,Qian Zhang,Nan Wang,Yiran Jin,Yingfeng Du

Rationale Anemarrhenae Rhizoma (AR) has been a frequently utilized traditional Chinese medicine (TCM) for an extended period, with its salt-processed variant being a prevalent application form. Contemporary pharmacological investigations have demonstrated that the salt-processed iteration exhibits a multitude of markedly augmented pharmacological properties. However, whether the pharmacodynamic material basis of this change is related to volatile substances remains unclear. The aim of this study was to develop a strategy to screen volatile pharmacodynamic substances in AR and salt-processed AR (SAR). Methods A comprehensive approach was developed to identify volatile pharmacodynamic compounds by integrating plant metabolomics, target network pharmacology, and molecular docking technology. Plant metabolomics using GC–MS analysis was conducted to identify volatile chemical markers distinguishing between AR and SAR. Subsequently, network pharmacology was utilized to investigate the correlation between chemical markers and associated diseases. Following this, molecular docking technology was utilized to explore the correlation between chemical markers and disease targets, resulting in the discovery of potential quality control markers. Results Fifty volatile compounds were isolated and identified in the salt of AR and SAR. The findings from plant metabolomics analysis demonstrated a distinct differentiation, revealing 13 volatile chemical markers that distinguish between AR and SAR. A target (PPARG) associated with diabetes was identified through target network pharmacology analysis. Thirteen volatile components were subsequently chosen as potential quality markers, taking into account their hypoglycemic activity. Conclusions The method developed provides a novel strategy for the identification of pharmacophores in AR and SAR, as well as establishing a foundation for the exploration of the volatile differential components and pharmacodynamics in various processed products of TCMs. Additionally, the findings of this study can serve as a theoretical framework for the development and utilization of volatile components in AR and its processed derivatives.

Relationship Between Metabolomics of T2DM Patients and the Anti-Diabetic Effects of Phellodendri Chinensis Cortex-Anemarrhenae Rhizoma Herb Pair in Mice

Jia-Lin Yu,Zhen-Yang Zhang,Sheng-Ping Liu,Hong-Ping Long,Ting-Ting Wang,Feng-Qing Huang,Jia Guo,Wei-Long Xu,Fei Li

Ethnopharmacological relevance Type 2 diabetes mellitus (T2DM) poses significant threats to public health. In Traditional Chinese Medicine (TCM), the Phellodendri Chinensis Cortex-Anemarrhenae Rhizoma (PCC/AR) herb pair has long been used for T2DM treatment, although its specific anti-diabetic mechanisms remain unclear. Aim of the study This study aimed to elucidate the relationship between metabolomics of T2DM patients and the anti-diabetic effects of PCC/AR herb pair in mice through clinical metabolomics and both in vitro and in vivo experiments. Materials and methods In this study, a T2DM mouse model was established via high-fat feeding (HFD) and streptozotocin (STZ) injection. The effects of PCC/AR on blood glucose, lipid metabolism, and inflammatory markers were evaluated. High-performance liquid chromatography-mass spectrometry (HPLC-MS) was performed for metabolomics analysis of T2DM patients. Results Serum metabolomics analysis identified significant alterations in metabolites linked to the biosynthesis of unsaturated fatty acids and purine metabolism in T2DM patients, with elevated 2-hydroxyvaleric acid (2HB) levels. In T2DM mice, PCC/AR intervention normalized FBG, GHbA1c, TC, TG, LDL-C, HDL-C, TNF-α and IL-1β levels, while improving insulin sensitivity and pancreatic β-cell function in T2DM mice. Notably, PCC/AR reduced key enzymes in gluconeogenesis and fatty acid synthesis, PEPCK and ACC1. Conclusion PCC/AR herb pair exerts an anti-diabetes effect in T2DM mice by regulating 2HB through ACC1 inhibition, thereby reducing FFA and TG synthesis. Additionally, PCC/AR may also exert its effects by modulating glucose and lipid metabolism and reducing inflammation. These results support further investigation into the PCC/AR herb pair as a complementary therapy for T2DM.

Identification, biological characteristics and fungicide sensitivity of Colletotrichum species that cause anthracnose on Anemarrhena asphodeloides in China

Ruofan Wei,Rong Wang,Yong Li,Moyi Yue,Wanlong Ding

Zhi mu ( Anemarrhena asphodeloides Bunge.) is an important medicinal plant in China, and its growth is greatly threatened by the fungal disease anthracnose, which can be caused by a number of species of Colletotrichum . Studying the phytopathogen and its sensitivities to fungicides will help to control anthracnose on zhi mu. Ten isolates of Colletotrichum were collected from zhi mu in Hebei, Anhui and Shanxi Provinces in China for this study. The isolates were identified as C . gloeosporioides , C. liriopes and C. spaethianum based on multi-locus phylogenetic analyses, including ACT , CHS - 1 , GADPH , HIS3 and ITS , micromorphological characteristics and the characteristics of colonies. Koch's postulates were confirmed by inoculating the leaves of zhi mu in vitro . The levels of virulence of 10 isolates varied significantly. Isolate YC-1 was the most virulent, while BZ-2 was the least virulent. The sensitivity of isolates YC-1, BY-1 and BY-2 ( C . spaethianum , C. gloeosporioides and C. liriopes respectively) to fungicides were examined. The EC 50 values of BY-1, BY-2, and YC-1 to tebuconazole, difenoconazole, prochloraz, fludioxonil, pyraclostrobin, carbendazim and azoxystrobin confirm that all of the isolates were markedly sensitive to the seven fungicides tested, strongly suggesting that these fungicides could be used to control anthracnose on zhi mu in the field.

A zinc-modified Anemarrhena asphodeloides polysaccharide complex enhances immune activity via the NF-κB and MAPK signaling pathways

Shaojie Zhang,Qian Zhang,Chong Li,Na Xing,Pengfei Zhou,Yukun Jiao

Anemarrhena asphodeloides polysaccharide (AAP70–1) was reported to have immunomodulatory effects in our previous report. To further improve the immunomodulatory effects of AAP70–1, an A. asphodeloides polysaccharide-zinc complex (AAP-Zn) was synthesized using a ZnCl 2 modification method, and the potential mechanisms by which AAP-Zn activates macrophages were investigated. The results showed that the structural features of AAP-Zn were similar to those of AAP70–1 with a Zn content of 0.2 %, confirming that Zn mainly interacted with AAP70–1 by forming Zn O coordination bonds and Zn…OH bonds. In addition, the administration of AAP70–1 and AAP-Zn effectively improved the immunomodulatory effects by enhancing phagocytosis and upregulating the mRNA expression of cytokines (TNF- α , IL-6, IL-1 β , and IL-18), as well as increasing the production levels of nitric oxide (NO) and reactive oxygen species (ROS) in zebrafish embryos. The intracellular mechanism by which AAP-Zn activates macrophages was found to involve activation of the NF- κ B and MAPK signaling pathways. Our findings suggest that AAP-Zn may be a potential immunopotentiator in the field of biomedicine or functional foods.

Anemarrhenasaponin I suppresses ovarian cancer progression via inhibition of SHH signaling pathway

Simin Deng,Yuan Xu,Binbin Gao,Tingting Yu,Lun Kuang,Bo’ang Han,Shaolun Feng,Haodong Chi,Qing Cao,Shen Yue,Chen Liu

Objectives The Sonic hedgehog (SHH) signaling is essential in animal development and tissue homeostasis. Aberrant activation of SHH pathway has been implicated in tumorigenesis and progression of several cancers, including ovarian cancer. Therefore, targeting SHH pathway may pave the way for successful ovarian cancer treatment. Methods To identify the potential SHH inhibitors from traditional Chinese medicines, we herein employed two in vitro cell models. In addition, western blotting and quantitative real-time PCR were performed to evaluate the inhibitory activity of Anemarrhenasaponin I (An-I) on SHH signaling in ovarian cancer cells. Cell proliferation assay and transwell assay were used to assess the effect of An-I on tumorigenicity. We also applied RNA-seq to examine the potential mechanism of An-I against ovarian cancer. Results Drug screening results showed that An-I drastically inhibited SHH signaling. More importantly, An-I effectively suppressed ovarian cancer cell proliferation and aggressiveness. RNA-seq-based transcriptome data showed that An-I affected ovarian cancer cells by suppressing SHH-WNT-Matrix metalloproteinases (MMPs) pathway. Conclusions An-I suppressed ovarian cancer progression by inhibiting SHH-WNT-MMP signaling transduction, providing a new treatment strategy for ovarian cancer.

Lipidomics of the erythrocyte membrane and network pharmacology to explore the mechanism of mangiferin from Anemarrhenae rhizoma in treating type 2 diabetes mellitus rats

Yanmei Zhong,Yingying Xu,Yongzhen Tan,Xuanxuan Zhang,Ruolun Wang,Danmin Chen,Zhaotao Wang,Xunlong Zhong

Mangiferin , a natural C-glucoside xanthone , is one of the major bioactive ingredients derived from the dry rhizome of Anemarrhenae rhizome , which has been reported to exhibit various pharmacological effects , including anti-oxidant, anti-inflammatory, anti-fatty liver, anti-metabolic syndrome, and anti-diabetic. However, the precise molecular mechanisms underlying its impact on phospholipid metabolism in the erythrocyte membrane of type 2 diabetes mellitus (T2DM) remain unclear. The present research aimed to evaluate the effects of mangiferin on glucose and lipid metabolism in T2DM model rats and discuss the relationship between lipid metabolites and potential targets involved in the hypoglycemic effects by integrating lipidomics and network pharmacology method. After 8 consecutive weeks of treatment with mangiferin, the T2DM model rats exhibited significant improvements in several biochemical indices and cytokines, including fasting blood glucose (FBG) levels after 12 h of fasting, fasting insulin level (FINS), total cholesterol (T-CHO), triacylglycerols (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), homeostasis model assessment of insulin resistance (HMOA-IR), TNF-α and IL-6. A total of 22 differential lipid metabolites were selected from erythrocyte membrane phospholipids, which were closely associated with the processes of T2DM. These metabolites mainly belonged to glycerophospholipid metabolism and sphingolipid metabolism. Based on network pharmacology analysis, 22 genes were recognized as the potential targets of mangiferin against diabetes. Moreover, molecular docking analysis revealed that the targets of TNF, CASP3, PTGS2, MMP9 , RELA, PLA2G2A, PPARA, and NOS3 could be involved in the modulation of inflammatory signaling pathways and arachidonic acid (AA) metabolism to improve IR and hyperglycemia . The combination of immunohistochemical staining and PCR showed that mangiferin could treat T2DM by regulating the expression of PPARγ protein and NF-κB mRNA expression to impact glycerophospholipids (GPs) and AA metabolism. The present study showed that mangiferin might alleviate IR and hyperglycemia of T2DM model rats via multiple targets and multiple pathways to adjust their phospholipid metabolism, which may be the underlying mechanism for mangiferin in the treatment of T2DM.

Timosaponin AIII inhibits gastric cancer by causing oxidative stress and blocking autophagic flux

Chunyang Zhu,Shuming Chen,Yangyang Lu,Jialin Song,Shasha Wang,Jing Guo,Xiaoxi Han,YuanYuan Fang,Siyi Zhang,Wensheng Qiu,Weiwei Qi

Background : Gastric cancer (GC) is a prevalent malignant tumor worldwide, with limited treatment targets. Timosaponin AIII (Tim AIII) is the naturally steroid saponin isolated from Anemarrhena, while this study initially confirms the anti-GC effect of Tim AIII. Methods : MTT assay, cell cycle analysis, and wound healing assay were used to evaluate the inhibitory effects of Tim AIII on GC cells (AGS and HGC27). Evaluate the oxidative stress (OS) by measuring reactive oxygen species (ROS) and malondialdehyde (MDA), as well as the Kelch-like ECH-associated protein 1 (Keap1) - Nuclear factor erythroid-derived 2-like 2 (Nrf2) pathway. RNA sequencing and proteomics analysis were utilized to investigate deeper molecular mechanisms. To track the autophagic flux using transmission electron microscope, detecting changes in autophagy-related pathway proteins, staining with LC3B and lysosome. Experiments related to cell viability, OS, and autophagy levels were performed on normal gastric mucosal epithelial cells (GES-1) as parallel controls. Finally, Nude mouse subcutaneous tumor model to evaluate the anti-GC ability in vivo. Results : Tim AIII inhibits the viability, proliferation, and migration of GC cells. Tim AIII causes OS in GC cells by the increasing intracellular ROS and MDA levels and inhibiting the Keap1-Nrf2 pathway. RNA sequencing and proteomics analysis mainly focused on the autophagy-associated pathways and lysosome in GC cells. Tim AIII activates autophagy, as indicated by an increase in the number of autophagosomes, inhibition of the PI3K-AKT pathway, and activation of the AMPK pathway in GC cells. However, Tim AIII inhibits autophagy-lysosome fusion and impairs lysosomal function, which results in autophagic flux blockage in GC cells. The Tim AIII concentration that significantly inhibited GC cells in this study was applied to GES-1 cells. The results showed that at this concentration, Tim AIII exhibited no significant cytotoxic effects on GES-1 cells, did not induce OS, and had no impact on autophagy. Finally, Tim AIII also has the ability to inhibit tumor growth in vivo. Significance : In summary, the results of our study indicate Tim AIII as a novel late-stage autophagy inhibitor, which may provide novel medical possibilities for GC.

Structural characteristics and antioxidant and hypoglycemic activities of a heteropolysaccharide from Anemarrhena asphodeloides Bunge

Juncheng Chen,Lin Li,Xia Zhang,Yuan Zhang,Qingsong Zheng,Meijuan Lan,Bing Li

In this study, an acid polysaccharide (AABP-1B) was extracted from the rhizome of Anemarrhena asphodeloides Bunge and purified using 60 % alcohol precipitation and DEAE-52 cellulose. The molecular weight of AABP-1B was 105 kDa, and it consisted of mannose (Man), rhamnose (Rha), galacturonic acid (GalA), glucose (Glc), galactose (Gal), and arabinose (Ara) in a ratio of 6.3:1.3:1.1:0.2:0.4:0.7. Methylation and NMR analyses revealed that the backbone of AABP-1 consists of 4)-β-D-Man p -(1 and 4)-2- O -acetyl-β-D-Man p -(1. In addition, the biological activity assays showed that AABP-1B not only displays potential antioxidant activity but also exhibits the α-glucosidase and α-amylase inhibitory effect. Moreover, AABP-1B enhanced glucose consumption and glycogen synthesis in insulin-resistant (IR) HepG2 cells. These results suggest that AABP-1B has potential hypoglycemic activity.

Anemarrhena asphodeloides Bunge total saponins lower lipid via modulating MAOA activity to enhance defense mechanisms in mice and C. elegans

Kexin Shi,Bentian Jing,Yifan Feng,Yong Yu

Ethnopharmacological relevance Within Anemarrhena asphodeloides Bunge (AAB), the pivotal bioactive constituents are identified as Anemarrhena asphodeloides Bunge total saponins (ABS). In traditional pharmacology, ABS has exhibited notable anti-inflammatory, hypoglycemic, and cardioprotective properties. Despite these observed effects, the specific protective mechanisms of ABS against metabolic diseases and improving the endocrine system remain largely uncharted. Aim to study This work intends to shed light on the effects and intrinsic mechanisms of ABS on metabolic diseases. Materials and methods The characterization of ABS components was achieved through High-Performance Liquid Chromatography/Mass Spectrometry (HPLC/MS). To evaluate ABS's anti-inflammatory efficacy, mouse macrophages underwent analysis using the Griess method. Induced differentiation of mouse fibroblasts was assessed through Oil Red O staining. In an obesity model with C57BL/6 N mice, ABS administration prompted measurements of glucose and insulin tolerance. Western blot analysis quantified lipolysis and anti-inflammatory protein expression. Nile red staining gauged body fat content in C. elegans post-ABS treatment. The mechanism of ABS action was elucidated through mRNA sequencing, further validated using RNA interference technology, and nematode mutants. Results ABS showcased the ability to diminish Nitric Oxide (NO) production in inflammatory macrophages and shrink adipocyte lipid droplets. In mice experiments, ABS was effective in alleviating fat accumulation and affecting serum lipid metabolism in diabetic mice. It enhanced oral glucose tolerance and insulin tolerance while increasing lipolysis-associated protein expression. ABS notably reduced fat content in C. elegans . Mechanistically, ABS downregulated NOD-like receptor thermal protein domain associated protein 3 (NLRP3) and monoamine oxidase A (MAOA) expression while enhancing UGT, ilys-2 , and ilys-3 . Lipolysis emerged as a pivotal pathway for ABS in the therapeutic intervention of metabolic diseases. Conclusions Our investigation has revealed that ABS exert a role in combating metabolic diseases by enhancing the body's defense mechanisms. ABS activate the NLRP3-neurotransmitter-visceral adipose pathway in mice, thereby bolstering resistance and diminishing fat accumulation. In C. elegans , ABS downregulated the expression of MAOA, bolstered resistance, and augmented glucuronidase activity, consequently leading to a reduction in fat content.
