Evaluation of Tacrolimus’ Adverse Effects on Zebrafish in Larval and Adult Stages by Using Multiple Physiological and Behavioral Endpoints

Wen-Wei Feng,Hsiu-Chao Chen,Gilbert Audira,Michael Edbert Suryanto,Ferry Saputra,Kevin Adi Kurnia,Ross D. Vasquez,Franelyne P. Casuga,Yu-Heng Lai,Chung-Der Hsiao,Chih-Hsin Hung

Simple SummaryDespite the clinical usefulness of tacrolimus in preventing organ transplant rejection, numerous studies have demonstrated its adverse effects in humans and animals. However, to date, no study has comprehensively addressed its toxicity toward zebrafish. Using various tests to assess the cardiovascular performance, various behaviors, and cognitive performance of the fish, we found that tacrolimus in relatively low concentrations altered the behaviors, social cohesion, and short-term memory of zebrafish in the larval and adult stages after acute and chronic exposures, respectively, with some alterations occurring in a dose-dependent manner. We hypothesize that tacrolimus may possess anxiotropic properties, which make it capable of modifying anxiety in the fish by affecting their nervous system. Nevertheless, considering the potential toxicity of tacrolimus, its usage, including in clinical applications, should be taken into account and monitored.AbstractTacrolimus (FK506) is a common immunosuppressant that is used in organ transplantation. However, despite its importance in medical applications, it is prone to adverse side effects. While some studies have demonstrated its toxicities to humans and various animal models, very few studies have addressed this issue in aquatic organisms, especially zebrafish. Here, we assessed the adverse effects of acute and chronic exposure to tacrolimus in relatively low doses in zebrafish in both larval and adult stages, respectively. Based on the results, although tacrolimus did not cause any cardiotoxicity and respiratory toxicity toward zebrafish larvae, it affected their locomotor activity performance in light–dark locomotion tests. Meanwhile, tacrolimus was also found to slightly affect the behavior performance, shoaling formation, circadian rhythm locomotor activity, and color preference of adult zebrafish in a dose-dependent manner. In addition, alterations in the cognitive performance of the fish were also displayed by the treated fish, indicated by a loss of short-term memory. To help elucidate the toxicity mechanism of tacrolimus, molecular docking was conducted to calculate the strength of the binding interaction between tacrolimus to human FKBP12. The results showed a relatively normal binding affinity, indicating that this interaction might only partly contribute to the observed alterations. Nevertheless, the current research could help clinicians and researchers to further understand the toxicology of tacrolimus, especially to zebrafish, thus highlighting the importance of considering the toxicity of tacrolimus prior to its usage.

Extracting Tissue Optical Properties and Detecting Bruised Tissue in Pears Quickly and Accurately Based on Spatial Frequency Domain Imaging and Machine Learning

Shengqiang Xing,Jiaming Zhang,Yifeng Luo,Yang Yang,Xiaping Fu

Recently, Spatial Frequency Domain Imaging (SFDI) has gradually become an alternative method to extract tissue optical properties (OPs), as it provides a wide-field, no-contact acquisition. SFDI extracts OPs by least-square fitting (LSF) based on the diffuse approximation equation, but there are shortcomings in the speed and accuracy of extracting OPs. This study proposed a Long Short-term Memory Regressor (LSTMR) solution to extract tissue OPs. This method allows for fast and accurate extraction of tissue OPs. Firstly, the imaging system was developed, which is more compact and portable than conventional SFDI systems. Next, numerical simulation was performed using the Monte Carlo forward model to obtain the dataset, and then the mapping model was established using the dataset. Finally, the model was applied to detect the bruised tissue of ‘crown’ pears. The results show that the mean absolute errors of the absorption coefficient and the reduced scattering coefficient are no more than 0.32% and 0.21%, and the bruised tissue of ‘crown’ pears can be highlighted by the change of OPs. Compared with the LSF, the speed of extracting tissue OPs is improved by two orders of magnitude, and the accuracy is greatly improved. The study contributes to the rapid and accurate extraction of tissue OPs based on SFDI and has great potential in food safety assessment. Keywords:spatial frequency domain imaging (SFDI);optical properties;absorption;reduced scattering;long short-term memory (LSTM)

CsPbBr3 QDs Employed as Gate to Fabricate an Artificial Photosynapse

Yubo Peng,Xudong Jin,Pingjun Zeng,Min Zhao,Zhipeng Gao,Jian-long Ji,Xiaona Li,Weiyi Chen

Recently, artificial photosynapses have attracted much attention from researchers because they have the ability to perceive, process, and remember information about the external environment, which can greatly improve the efficiency of visual information processing. This work prepared the artificial optical synaptic transistors based on organic electrochemical transistors (OECTs) and CsPbBr3 quantum dots (QDs) synthesized at room temperature. The fabricated device here could successfully imitate the visual functions by mimicking the important synaptic features such as excitatory postsynaptic currents, paired-pulse facilitation, spiking-rate-dependent plasticity, spiking-number-dependent plasticity, and short-term memory to long-term memory transition. Employing the device, we also simulated the capabilities of the human visual system of light adaptation, image recognition, and preprocessing. The investigations in this work indicated that CsPbBr3 QDs have a high potential application in fabricating artificial photosynapses of the type of OECTs to imitate human vision due to the simpler fabrication process.

Direct extraction of respiratory information from pulse waves using a finger-inspired flexible pressure sensor system

Zhang Xikuan,Chai Jin,Xu Lingxiao,Mei Shixuan,Wang Xin,Zhao Yunlong,Xue Chenyang,Wang Yongjun,Cui Danfeng,Zhang Zengxing,Zhang Haiyan,Gao Libo

The long-term monitoring of respiratory status is crucial for the prevention and diagnosis of respiratory diseases. However, existing continuous respiratory monitoring devices are typically bulky and require either chest strapping or proximity to the nasal area, which compromises user comfort and may disrupt the monitoring process. To overcome these challenges, we have developed a flexible, attachable, lightweight, and miniaturized system designed for extended wear on the wrist. This system incorporates signal acquisition circuitry, a mobile client, and a deep neural network, facilitating long-term respiratory monitoring. Specifically, we fabricated a highly sensitive (11,847.24 kPa −1 ) flexible pressure sensor using a screen printing process, which is capable of functioning beyond 70,000 cycles. Additionally, we engineered a bidirectional long short-term memory (BiLSTM) neural network, enhanced with a residual module, to classify various respiratory states including slow, normal, fast, and simulated breathing. The system achieved a dataset classification accuracy exceeding 99.5%. We have successfully demonstrated a stable, cost-effective, and durable respiratory sensor system that can quantitatively collect and store respiratory data for individuals and groups. This system holds potential for everyday monitoring of physiological signals and healthcare applications.

A flexible dual-gate hetero-synaptic transistor for spatiotemporal information processing

Xuerong Liu,Cui Sun,Zhecheng Guo,Yuejun Zhang,Zheng Zhang,Jie Shang,Zhicheng Zhong,Xiaojian Zhu,Xue Yu,Run-Wei Li

Artificial synapses based on electrolyte gated transistors with conductance modulation characteristics have demonstrated their great potential in emulating the memory functions in the human brain for neuromorphic computing. While previous studies are mostly focused on the emulation of the basic memory functions of homo-synapses using single-gate transistors, multi-gate transistors offer opportunities for the mimicry of more complex and advanced memory formation behaviors in biological hetero-synapses. In this work, we demonstrate an artificial hetero-synapse based on a dual-gate electrolyte transistor that can implement in situ spatiotemporal information integration and storage. We show that electric pulses applied on a single gate or unsynchronized electric pulses applied on dual gates only induce volatile conductance modulation for short-term memory emulation. In contrast, the device integrates the electric pulses coincidently applied on the dual gates in a supralinear manner and exhibits nonvolatile conductance modulation, enabling long-term memory emulation. Further studies prove that artificial neural networks based on such hetero-synaptic transistors can autonomously filter the random noise signals in the dual-gate inputs during spatiotemporal integration, facilitating the formation of accurate and stable memory. Compared to the single-gate synaptic transistor, the classification accuracy of MNIST handwritten digits using the hetero-synaptic transistor is improved from 89.3% to 99.0%. These findings demonstrate the great potential of multi-gate hetero-synaptic transistors in simulating complex spatiotemporal information processing functions and provide new platforms for the design of advanced neuromorphic computing systems.

Copper nanoclusters based short-term memory “eraser”

Kaigeng Hu,Yu Liu,Qingqing Wang,Yan Xiong,Zirun Guo,Zhenzhen Weng,Yu Liu,Yini Zhang,Hongguang Wu,Fanrong Ai,Xiaolei Wang

Short-term memory refers to processing information during short-term storage and manipulation which is affected by a variety of factors. If there exists a biocompatible material that can effectively enter the brain, remove some short-term memories (such as car accident, murder, etc.), and be excreted from the body automatically, it will be expected to significantly reduce level of depression, anxiety and even the odds of suicide. Herein, we synthesize the bovine serum albumin (BSA)-templated copper nanoclusters (BSA-CuNCs) and BSA-templated silver nanoclusters (BSA-AgNCs) respectively then discuss their potential as short-term memory “erasers”. In vivo experiments show that both CuNCs and AgNCs can effectively penetrate the blood–brain barrier (BBB) and decrease the recognition memory of mice by inhibiting neuronal cell conduction within 24 h. Interestingly, after 3 weeks, the discrimination index (DI) of the BSA-CuNCs-treated mice increases again and the memory can be recovered. In contrast, mice of the BSA-AgNCs group do not regain their memory. In addition, BSA-CuNCs exhibit good biocompatibility and can be metabolically cleared by the organism, which makes BSA-CuNCs intriguing potential for short-term memory erasure.

Optically Readable Electrochromic-Based Microfiber Synaptic Device for Photonic Neuromorphic Systems

Hui Shi,Banghu Wang,Xiaohong Wang,Longzhen Qiu,Lei Zheng

Optical synaptic devices possess great potential in both artificial intelligence and neuromorphic photonics. In this work, an optically readable electrochromic-based microfiber synaptic device was designed by the combination of a multimode fiber and an electrochromic device and using an external voltage to control the transmission of light in the fiber. The proposed synaptic device has the ability to imitate various basic functions of the biological synapses, such as synaptic plasticity, and paired-pulse facilitation (PPF), as well as the transition from short-term memory to long-term memory. Moreover, the proposed device decodes the output optical signal with the international Morse code to express the signal “HFUT” in two ways, and a 3 × 3 array composed of this device can simulate the perceptual learning process. The device can be easily prepared for a wide range of applications, and the incorporated microfibers can be replaced by planar optical waveguides, making it easy to be integrated into a more complex and versatile photonic neuromorphic system.

Quercetin inclusion complex gels ameliorate radiation-induced brain injury by regulating gut microbiota

Jinglu Hu,Wencheng Jiao,Ziyan Tang,Chunqing Wang,Qi Li,Meng Wei,Shiyong Song,Lina Du,Yiguang Jin

Radiation-induced brain injury (RIBI) is a serious adverse effect of radiotherapy. RIBI has garnered considerable clinical attention owing to its powerful effects on brain function and cognition; however, no effective treatment is available. The microbiota–gut–brain axis theory is a novel concept of treating RIBI by regulating gut microbiota. Quercetin, a particularly common flavonoid compound, has a wide range of biological activities and can regulate gut microbiota; however, it has poor solubility and dispersibility. In the present study, oral gels of inclusion complex comprising quercetin and HP-β-CD were prepared, which increased quercetin dispersion and extended its release time in the intestinal tract. First, the relative abundance and diversity of gut microbiota in RIBI mice changed after oral administration of quercetin inclusion complex gels (QICG). Second, the spontaneous activity behavior and short-term memory ability as well as anxiety level were improved. Third, changes in physical symptoms were observed, including a decrease in TNF-α and IL-6 levels. H&E staining revealed that gut epithelial injury and intestinal inflammation as well as hippocampal inflammation were ameliorated. Antibiotics treatment (Abx) mice were developed to disrupt the mice's original gut microbiota composition. No significant improvement was observed in behavior or histopathology after oral administration of QICG in Abx mice of RIBI, indicating that the effect of QICG on improving RIBI was regulated by intestinal microbiota. Finally, the QICG preparation is efficient, exerting a protective effect on RIBI by regulating gut microbiota via the microbiota–gut–brain axis, which provides a novel idea for RIBI treatment.

Machine learning combined with multi-source data fusion for rapid quality assessment of yellow rice wine with different aging years

Zhi-Tong Zhang,Yu Li,Lei Bai,Pan Chen,Yue Jiang,Yali Qi,Huanhuan Guan,Yaxuan Liang,Dongping Yuan,Tulin Lu,Guojun Yan

Aging is a crucial factor for high-quality alcoholic beverages, which improves the flavor of alcoholic beverages such as yellow rice wine (YRW). However, traditional analytical methods for evaluating the quality of YRW are very tedious, and there is still a lack of rapid quality assessment methods for YRW with different aging times. In this study, taking Jimo rice wine (JRW, a representative YRW in northern China) with different aging years as the object, traditional analytical methods and various antioxidant experiments were used to characterize their quality differences, a rapid quality evaluation method was established by machine learning combining with the data obtained from Flash GC e-nose and near-infrared. The results showed multiple physicochemical parameters coupled with multivariate statistical analysis could distinguish JRW with diverse aging years. JRW with different aging years also showed various antioxidant capacities and generally increased with the aged years. A total of 24 major aroma components were identified in JRW, of which five varied regularly with aging time. Then, the deep learning algorithm (long short-term memory, LSTM) showed excellent classification performance (100% accuracy) in JRW with different aging years, and a multi-source information fusion strategy can achieve 100% classification accuracy even when combined with traditional algorithms. Finally, the fused data improved the accuracy of the LSTM regression model in predicting the content of the main physicochemical parameters of JRW, with higher R 2 and lower RMSE compared to data from a single source. Overall, this study clarified the quality differences of JRW with diverse aging years, and a rapid and precise method combining multi-source data fusion and machine learning was developed to assess the quality of JRW, which could also apply to other beverages or foods.

Prediction of the postharvest quality of Boletus wild mushrooms stored with mesoporous silica nanoparticles antibacterial film using Long Short-Term Memory model combined with the Northern Goshawk Optimization (NGO-LSTM)

Yudi Liu,Kai Jiang,Yuyue Qin,Margaret Brennan,Charles Brennan,Jianxin Cao,Zhengxuan Wang,Thanapop Soteyome

This study aimed to address the challenge of extending the shelf life of Boletus wild mushrooms, which are prone to environmental and microbial contamination. An antibacterial film composed of polylactic acid (PLA) and mesoporous silica nanoparticles loaded with citral (CMP film) was developed for this purpose. Fifteen quality indices were assessed, and the data were integrated using AHP and TOPSIS to evaluate the film's efficacy. The CMP film effectively maintained the quality of mushroom over time. Additionally, a Nonlinear Global Optimization-Long Short-Term Memory (NGO-LSTM) model was employed to predict storage quality, using seven highly correlated quality indicators. The model achieved a high predictive accuracy, with the R 2 exceeding 0.999. This study presents a novel packaging solution and a predictive model that together enhance the storage and quality control of Boletus wild mushrooms.

Enabling high-accuracy lithium-ion battery status prediction via machine learning-integrated perovskite sensors

Jiaxin Sun,Xianjie Xu,Zhefu Mu,Zijun Huang,Guo Chen,Xinkai Qi,Hongwei Liu,Lei Zhu,Xiuquan Gu,Xinjian He,Sheng Huang

Monitoring the safety and reliability of lithium-ion batteries (LIBs) under extreme operating conditions faces significant challenges, due to the difficulty of conventional battery management systems (BMS) in capturing internal dynamic changes in real-time. To address this challenge, this paper proposes an implantable flexible sensor based on ZnO-passivated CsPbBr 3 quantum dots encapsulated in EPDM, which realizes real-time monitoring inside lithium batteries. Experiments demonstrate that the sensor resistance exhibits high synchronization with battery voltage changes (response time < 5 s), and the capacity fade rate difference after sensor implantation is only 14.2 % over 15 days. Theoretical calculations and in situ X-ray diffraction (XRD) reveal the energy band modulation mechanism of Li + adsorption - induced. During LIBs charging/discharging, changes in Li + concentration around the sensor alter the band structure and density of states (DOS) of the sensing material, leading to changes in its electrical signal. Thus, the working state of the LIBs can be monitored and predicted by tracking changes in the sensor's electrical signal. Further integration of an LSTM-multitask learning model enables advanced prediction of the LIBs operating state based on sensor signals (5000 s ahead), with a mean absolute error (MAE) of 6.10 %, and charge/discharge rate recognition with 96.7 % accuracy.
