Simultaneous determination of phenols in the four main original plants of the famous traditional Chinese medicine Shihu by pressurized capillary electrochromatography

Xiao-Quan Liu,Chao-Feng Qin,Nai-Dong Chen,Jing-Wen Hao,Shi-Tang Ma,Min Zhang,Yu Song,Yun-Jiang Min,Ya-Qin Bu,Sheng Li

A rapid pressurized capillary electrochromatography (pCEC) method has been established for the simultaneous analysis of 11 phenols in the four main original plants of the famous traditional Chinese medicine (TCM) Shihu. The effects of wavelength, mobile phase, flow rate, pH value, concentration of buffer, and applied voltage were systematically studied. The investigated 11 phenols could be isolated in 35 min on a reversed-phase EP-100-20/45-3-C18 capillary column using the established method. To apply the established pCEC method, all phenols except tristin (11) were detected in the four Dendrobium plants. A total of 10 components were detected in D. huoshanense, 6 components in D. nobile, 3 components in D. chrysotoxum, and 4 components in D. fimbriatum. The consistent evaluation revealed that the similarities among the four original plants of Shihu were 38.2–86.0% based on the 11 polyphenols and 92.5–97.7% based on the pCEC fingerprints. These further suggested that the components of the four original plants of TCM Shihu might be significantly different. Further investigation should be conducted to confirm and evaluate if the four species could be used as the same medicine with the same amount according to Chinese Pharmacopoeia (ChP).

DoRWA3 from Dendrobium officinale Plays an Essential Role in Acetylation of Polysaccharides

Can Si,Jaime A. Teixeira da Silva,Chunmei He,Zhenming Yu,Conghui Zhao,Haobin Wang,Mingze Zhang,Jun Duan

The acetylation or deacetylation of polysaccharides can influence their physical properties and biological activities. One main constituent of the edible medicinal orchid,Dendrobium officinale, is water-soluble polysaccharides (WSPs) with substitutedO-acetyl groups. BothO-acetyl groups and WSPs show a similar trend in different organs, but the genes coding for enzymes that transfer acetyl groups to WSPs have not been identified. In this study, we report that REDUCED WALL ACETYLATION (RWA) proteins may act as acetyltransferases. ThreeDoRWAgenes were identified, cloned, and sequenced. They were sensitive to abscisic acid (ABA), but there were no differences in germination rate and root length between wild type and35S::DoRWA3transgenic lines under ABA stress. Three DoRWA proteins were localized in the endoplasmic reticulum.DoRWA3had relatively stronger transcript levels in organs where acetyl groups accumulated thanDoRWA1andDoRWA2, was co-expressed with polysaccharides synthetic genes, so it was considered as a candidate acetyltransferase gene. The level of acetylation of polysaccharides increased significantly in the seeds, leaves and stems of three35S::DoRWA3transgenic lines compared to wild type plants. These results indicate that DoRWA3 can transfer acetyl groups to polysaccharides and is a candidate protein to improve the biological activity of other edible and medicinal plants. Keywords:acetyl groups;Dendrobium officinale;REDUCED WALL ACETYLATION;endoplasmic reticulum

Isolation, characterization, and activity of the polysaccharides in Bulbophyllum kwangtumgense Schltr

Liuyue Xiao,Jiaojiao Hou,Ping Chen,Xiao Tan,Xiangxiang Qin,Jing Nie,Hua Zhu,Shian Zhong

A microwave-assisted method was used to extract the polysaccharide from Bulbophyllum kwangtumgense Schltr, and the optimum extraction conditions were determined by orthogonal experiments. The purified polysaccharide component BKP-1 had an obvious polysaccharide characteristic structure on the infrared spectrum and was mainly composed of xylose and glucuronic acid at a ratio of 0.568:0.432. Its molecular weight distribution (polydispersity index was 0.58) was uniform with 1.92× 10 6  Da. The main structural skeleton of BKP-1 was →4)-β-D-Xylp-(1 → [4)-β-D-Xylp-(1] 4  → 4)-α-D-GlcAp-(1 → [4)-α-D-GlcAp-(1] 2 →. BKP-1 could clear certain free radicals, among which the hydroxyl free radical scavenging ability was the best, with a clearance rate of 61.46 %. Furthermore, BKP-1 significantly modulated the secretion of both pro-inflammatory and anti-inflammatory cytokines at a concentration of 125 μg/mL. Notably, the mechanism BKP-1 exhibiting inhibitory effects were related to regulating of the NF-κB signaling pathway.

Multi-fingerprint analysis for interpretation of the quality differences in polysaccharides during Dendrobium huoshanense traditional processing

Baojie Zhu,Cunwu Chen,Jiaxian He,Shaoping Li,Jing Zhao

The interpretation of the chemical quality differences during Dendrobium huoshanense ( D. huoshanense ) traditional processing into ‘Fengdou’ is critical for scientifically evaluating the medicinal value, optimizing the processing methods, and ensuring the stability and predictability of its efficacy. In this study, the quality differences of polysaccharides during the processing of D. huoshanense were conducted utilizing high performance size exclusion chromatography coupled with multi-angle laser light scattering and refractive index detector (HPSEC-MALLS-RID), nuclear magnetic resonance (NMR) and saccharide mapping based on polysaccharides analysis by using carbohydrate gel electrophoresis (PACE) and high-performance anion exchange chromatography-pulsed amperometric detection (HPAEC-PAD). The results showed that the concentration of non-starch D. huoshanense polysaccharides (NDHP) rose and exhibited greater stability during the processing. The molecular weight (Mw), molecular weight distribution, cumulative molar mass, radius of gyration (Rz), and monosaccharide composition molar ratio of NDHP did not have significant changes during the processing, and these parameters would be affected by the presence of starch. The NMR investigation indicated that the primary glycosidic linkages of β -1,4-Man p and β -1,4-Glc p , together with the acetyl group substitutions at O-2 and O-3 of 1,4-linked Man p , were unaltered. Additionally, the PACE and HPAEC-PAD profiles of acidic and enzymatic hydrolysates of NDHP were similar. These findings contribute to a deeper understanding of the changes in bioactive polysaccharides during the processing of D. huoshanense into ‘Fengdou’, providing theoretical support for improving its processing techniques.

Similarity Evaluation of Different Origins and Species of Dendrobiums by GC-MS and FTIR Analysis of Polysaccharides

Nai-Dong Chen,Nai-Fu Chen,Jun Li,Cai-Yun Cao,Jin-Mei Wang,He-Ping Huang

GC-MS method combined with FTIR techniques by the analysis of polysaccharide was applied to evaluate the similarity between wild (W) and tissue-cultured (TC) Dendrobium huoshanense (DHS), Dendrobium officinale (DO), and Dendrobium moniliforme (DM) as well as 3 wild Dendrobium spp.: Dendrobium henanense (DHN), Dendrobium loddigesii (DL), and Dendrobium crepidatum (DC). Eight monosaccharides involving xylose, arabinose, rhamnose, glucose, mannose, fructose, galactose, and galacturonic acid were identified in the polysaccharide from each Dendrobium sample while the contents of the monosugars varied remarkably across origins and species. Further similarity evaluation based on GC-MS data showed that the values of different origins of DHS, DO, and DM were 0.831, 0.865, and 0.884, respectively, while the values ranged from 0.475 to 0.837 across species. FTIR files of the polysaccharides revealed that the similarity coefficients between W and TC-DHS, DO, and DM were 88.7%, 86.8%, and 88.5%, respectively, in contrast to the similarity coefficients varying from 57.4% to 82.6% across species. These results suggested that the structures of polysaccharides between different origins of the investigated Dendrobiums might be higher than what we had supposed.

Rapid quantification of polysaccharide and the main onosaccharides in Dendrobium huoshanense by near-infrared attenuated total reflectance spectroscopy

Jing-Wen Hao,Nai-Dong Chen,Cun-Wu Chen,Fu-Cheng Zhu,De-Liang Qiao,Yong-Jun Zang,Jun Dai,Xiang-Wen Song,Han Chen

A rapid, green, low cost and nondestructive attenuated total reflection near infrared (ATR NIR) method was developed to quantify the total polysaccharide and the main monosaccharides mannose and glucose in Dendrobium huoshanense. Total 100 D. huoshanense samples from different places were analyzed using ATR NIR method. Potential outlying samples were initially removed from the collected NIR data using the PCA-Mahalanobis distance method. Spectral data preprocessing was studied in the construction of a partial least squares (PLS) model and six different signal pretreatment methods, including multiplicative scattering correction (MSC), standard normal transformation (SNV), first and second derivatives, the combination of MSC with the first derivative, and the combination of SNV with the first derivative, were compared. The results showed that the best signal pretreatment method was the spectral data pretreated by SNV combined with the first derivative due to it showed the lowest root-mean-square error of cross-validation (RMSECV), highest R 2 for both the polysaccharide and its main monosaccharides . In order to improve the performance of the model, the pretreated full spectrum was calculated by different wavelength selection method. The results showed that the optional wavelength selection model was the one simultaneously selecting the NIR wavelength ranges 7500–5750 cm −1 , 5250–4700 cm −1 , 4450–4300 cm −1 and 4200–4100 cm −1 because of the lowest RMSECV and the highest R 2 among the ten wavelength selection models. The external validation and the complete external validation confirmed the robustness and reliability of the developed NIR model. The contents of the total polysaccharide and the main monosaccharides are the essential quality assessment criterion for plant medicines while their traditional quantification methods involved sample destruction, tedious sample processing and non-environmentally friendly pretreatment, therefore, our study might provide an efficient technique tool for the rapid, green and nondestructive quantification of the total polysaccharide and the main monosaccharides for D. huoshanense and other rich-in-polysaccharide plant medicines.

Characterization and immunological activity of polysaccharides from two types of Dendrobium devonianum with different appearance

Wen Cao,Baojie Zhu,Xuan Zhang,Jurun Zhao,Shaoping Li,Jing Zhao

Dendrobium devonianum , a precious species of Dendrobium , is also called “ Zi-pi Shi-hu ” in Chinese due to its purple-skinned appearance. In this study, polysaccharides from green (GDP) and purple (PDP) skin D. devonianum were isolated by homogenate assisted extraction, ethanol precipitation and ultrafiltration. Their difference in physicochemical properties and immunological activities were investigated and compared. The results suggested that both GDP and PDP mainly contained glycosidic linkages of β -1,4-Man p and β -1,4-Glc p , but their molecular weights , 2.044 × 10 5 (GDP) vs 3.563 × 10 5 (PDP), and compositional monosaccharides of mannose to glucose ratio, 16.46: 1.00 (GDP) vs 7.61: 1.00 (PDP), were significantly different. Especially, GDP showed stronger immunological potentiation activity in vitro . Although the extraction yield of GDP was lower than that of PDP, considering the production of raw material and immunological activity of its polysaccharides, green skin D. devonianum should be a good alternative of purple one. These findings are helpful to provide evidence for improving the cultivation and production of D. devonianum .

Effect of Yeast Fermentation on the Physicochemical Properties and Bioactivities of Polysaccharides of Dendrobium officinale

Hang Chen,Xueqin Shi,Lanyan Cen,Lin Zhang,Yifeng Dai,Shuyi Qiu,Xiangyong Zeng,Chaoyang Wei

Fermentation is an effective method for enhancing the biological activity of polysaccharides, but research on its effect onDendrobium officinalpolysaccharides is rare. In this study, the effects of mono-fermentation (Saccharomyces cerevisiaeFBKL2.8022,Sc;Wickerhamomyces anomalousFBKL2.8023,Wa) and co-fermentation (Sc+Wa) on the physicochemical properties and bioactivity ofDendrobium officinalpolysaccharides were investigated. Meanwhile, the polysaccharide (DOP) obtained fromDendrobium officinalewas used as a control. Four homogeneous polysaccharides were obtained by isolation and purification and named DOSCP, DOWAP, DOSWP, and DOP. The results showed that DOSCP, DOWAP, DOSWP, and DOP consisted of mannose and glucose with ratios of 3.31:1, 5.56:1, 2.40:1, and 3.29:1, respectively. The molecular weights (Mws) of the four polysaccharides were 25.73 kDa, 15.01 kDa, 17.67 kDa, and 1268.21 kDa. The antioxidant activity of DOSCP, DOWAP, and DOSWP was better than that of DOP. Additionally, all four polysaccharides were able to reduce the inflammatory response of LPS-induced RAW 264.7 macrophages in the mice without a significant difference. Yeast fermentation significantly reduced the molecular weight and improved the antioxidant activity ofDendrobium officinalepolysaccharides, indicating a potential way to improve its antioxidant activity. Keywords:Dendrobium officinale;polysaccharides;fermentation;antioxidant activity;immune activity

Natural product osthole can significantly disrupt cell wall integrity and dynamic balance of Fusarium oxysporum

Ke Hu,Rongyu Li,Feixu Mo,Yi Ding,Aiai Zhou,Xue Guo,Ruotong Li,Min Li,Minggui Ou,Ming Li

Dendrobium officinale Kimura et Migo is a traditional Chinese herbal medicinal plant . However, the frequent occurrence of soft rot disease (SRD) is one of the most harmful diseases in D. officinale production in recent years, which can seriously affect its yield and quality. In this study, the major pathogenic fungus (SR-1) was isolated from D. officinale with typical symptoms of SRD, and was identified as Fusarium oxysporum through morphological and molecular identification. The biological activities of five natural products were determined against F. oxysporum using a mycelial growth inhibition assay. The results showed that osthole had the highest antifungal activity against F. oxysporum , with an EC 50 value of 6.40 mg/L. Scanning electron microscopy (SEM) showed that osthole caused F. oxysporum mycelia to shrink and deform. Transmission electron microscopy (TEM) showed that the organelles were blurred and the cell wall was thickened in the presence of osthole. The sensitivity of F. oxysporum to calcofluor white (CFW) staining was significantly enhanced by osthole. Relative conductivity measurements and propidium iodide (PI) observation revealed that osthole had no significant effect on the cell membrane. Further experiments showed that the activity of chitinase and β-1,3-glucanase were decreased, and expression levels of chitinase and β-1,3-glucanase related genes were significantly down-regulated after treatment with osthole. In conclusion, osthole disrupted the cell wall integrity and dynamic balance of F. oxysporum , thereby inhibiting normal mycelial growth.

Comparison of the Antioxidant Activities and Polysaccharide Characterization of Fresh and Dry Dendrobium officinale Kimura et Migo

Wang Zhang,Xinjie Liu,Xun Sun,Rongchun Han,Nianjun Yu,Juan Liang,An Zhou

It is generally believed that freshDendrobium officinale(FDO) has more significant pharmacological activity than driedDendrobium officinale(DDO); however, the difference has not been clearly shown. Our study compared their antioxidant properties both in vitro and in vivo, and the molecular weight arrangement and monosaccharide composition of the freshDendrobium officinalepolysaccharides (FDOPs) and the driedDendrobium officinalepolysaccharides (DDOPs) were analyzed by HPLC-GPC and GC-MS. The results showed that the FDO and its polysaccharides had more significant effects on scavenging DPPH, ABTS, and hydroxyl radicals than the DDO. In addition, both the FDO and DDO significantly reduced lipid peroxidation levels and increased the SOD, T-AOC, CAT, and GSH levels in mice with acute liver damage caused by CCl4, while the FDO and its polysaccharides were more effective. Histopathological analysis further verified the protective effect of the Dendrobium polysaccharides on CCl4-induced liver injury. The determination of the polysaccharides revealed that the polysaccharide and mannose contents of the FDO were significantly higher than their dried counterparts, and the homogeneous arrangement of the polysaccharides in the FDO was degraded into three polysaccharide fragments of different molecular weights in the DDO. Overall, our data identified differences in the antioxidant activities of the FDO and DDO, as well as the reasons for these differences. Keywords:Dendrobium officinale;antioxidant activity;polysaccharide;mannose;monosaccharide composition

Structure, anti-fatigue activity and regulation on gut microflora in vivo of ethanol-fractional polysaccharides from Dendrobium officinale

Ming Cai,Hua Zhu,Lei Xu,Jian Wang,Jing Xu,Zhenhao Li,Kai Yang,Jianyong Wu,Peilong Sun

This study was to investigate the antifatigue, prebiotic effects and their relationships to the structure properties of three ethanol precipitated polysaccharides from Dendrobium officinale (EPDO), as EPDO-40, EPDO-60 and EPDO-80. EPDOs with anti-fatigue activity were screened out by forced swimming test, and blood lactic acid (BLA), blood urea nitrogen (BUN), superoxide dismutase (SOD), liver glycogen, muscle glycogen, and intestinal microflora were investigated. Results showed that purified EPDO-60, 277.3 kDa, with a backbone consisted of 4-Man p and 4-Glc p . EPDO-60 had the best anti-fatigue activity, because it could significantly prolong the forced swimming time, as well as down-regulating the levels of BLA and BUN, increasing SOD. Proportions of Bacteroidetes and Firmicutes and abundance of Lactobacillus and Bifidobacterium in gut microflora increased after treated with EPDO-60. Accordingly, EPDO-60 could affect the community structure of gut microflora, leading to promote the balance of oxidation and antioxidation, and accelerated the fatigue metabolism in vivo .
