Toxicity of Arsenic to Photobacterium phosphoreum, Daphnia magna, and Danio rerio at Different pH Levels

Qun He,Ruijuan Qu,Xinghao Wang,Zhongbo Wei,Ping Sun,Zunyao Wang

Although it has been widely known that arsenic poses great threat to organisms and arsenic species account for the toxic effect of the metalloid, relatively little attention has been given to the arsenic toxicity to different organisms at different pH values. Three kinds of aquatic organisms including Photobacterium phosphoreum , Daphnia magna , and Danio rerio were used to study the effects of pH on the toxicity of arsenic. Toxicity test results were analyzed by calculating EC 50 values and for arsenic species at five different pH values (5.0, 6.0, 7.0, 8.0, and 9.0). As a result, As(III) is more toxic to the tested organisms than As(V), and P. phosphoreum was more sensitive to the toxicity of arsenic than D. magna and D. rerio . The toxicity of As(III) species is different for different organisms: dihydrogen arsenite, H 2 AsO 3 – , is more toxic to P. phosphoreum and D. rerio than arsenous acid, H 3 AsO 3 , while H 3 AsO 3 is more toxic to D. magna . As for As(V), arsenate, HAsO 4 2− , is the most toxic to P. phosphoreum , whereas dihydrogen arsenate, H 2 AsO 4 – , may be responsible for the toxicity to D. magna and D. rerio . As(III) and As(V) are more toxic to P. phosphoreum in an alkaline environment, while for D. magna in an acidic environment and As(III) is more toxic to D. rerio in an alkaline environment and As(V) in an acidic environment.

Organosilicon enhances rice root suberization and antioxidant gene expression under cadmium/arsenic stress

Zhiheng Wang,Jia Wu,Shuxin Tu,Khurram Shehzad,Jingtao Hou,Shuanglian Xiong,Menghua Cao

Organosilicon exhibits unique physicochemical and biological properties with wide applicability across diverse fields, including agriculture and industry. Previous research has verified the effectiveness of organosilicon-modified fertilizers in inhibiting the uptake of cadmium (Cd) and arsenic (As) by plants. However, further investigations are necessary to elucidate the underlying mechanisms. In this study, we explored the potential of organosilicon to mitigate the toxic effects of Cd/As and lessen their uptake and accumulation during rice seed germination. Our results showed that under Cd/As stress, organosilicon treatments significantly increased suberin biosynthesis in rice roots. This was manifested as an increased level of root suberization and an enhanced apoplast barrier, as verified by observations made through fluorol yellow (FY) staining and transmission electron microscopy (TEM). Consequently, the uptake and translocation of Cd and As in rice seedlings were significantly reduced by 48.66% and 72.19% in shoots, and 43.23% and 68.93% in roots, respectively. Moreover, the application of organosilicon enhanced the activities of antioxidant enzymes in rice, This lead to an accelerated glutathione-oxidized glutathione (GSH-GSSG) cycle, up-regulated expression of the rice glutathione peroxidase gene ( OsGPX ), and increased GPX activity. These modifications effectively scavenged reactive oxygen species (ROS) generated by Cd/As stress and alleviated oxidative damage in rice. Overall, our study has unraveled the physiological and molecular mechanisms underlying the role of organosilicon in alleviating Cd/As toxicity in rice and has also provided new insights for the application of suberin in reducing heavy metal toxicity in plants.

Melatonin alleviates arsenite toxicity by decreasing the arsenic accumulation in cell protoplasts and increasing the antioxidant capacity in rice

Yan Li,Yutan Chu,Hongyu Sun,Qiongli Bao,Yizong Huang

Arsenic (As) is a common environmental pollutant that seriously interferes with the normal growth of organisms. There is an urgent need to take environment-safe and efficient strategies to mitigate As toxicity. Melatonin (MT) is a pleiotropic molecule that regulates plant growth and organ development and alleviates heavy metal stresses. The experiment aims to explore the mechanism of MT in reducing arsenite toxicity by hydroponic rice seedlings. The results showed that MT application reduced the As content in rice roots and shoots by 26.4% and 37.5%, respectively, and mainly decreased As content in the soluble fractions of the rice root cell. MT application also increased the As content of chelated-soluble pectin and alkali-soluble pectin in the cell wall by 14.7% and 74.4%, respectively. It promoted the generation of the functional group of the root cell walls by the FTIR analysis, indicating that MT may promote the fixation of As on the cell wall. Meanwhile, MT contributed to scavenging excess H 2 O 2 , reducing MDA content, and maintaining normal morphology of root cells by stimulating SOD, POD and CAT activities and increasing the level of GSH. The research deepens our understanding of how MT participates in maintaining redox homeostasis in rice cells, reducing As toxicity, and decreasing As concentration in rice seedlings, thereby providing more possibilities for reducing As accumulation in rice.
