Pilot-scale production of selenium-enriched Nostoc sphaeroides colonies and polysaccharides using two-phase cultivation strategy

Jingjing Hu,Peihua Liu,Qiangqiang Wang,Xuyuan Nie,Jianhua Tan,Junchao Shu,Junjian Mai,Yi Cao,Yongdong Zou,Zebo Huang

Nostoc sphaeroides , originally discovered in selenium-rich fields, is a photosynthetic, nitrogen-fixing cyanobacterium that forms edible spherical macrocolonies. However, prolonged selenium supplementation negatively impacts colony quality in culture. To address this, a two-phase cultivation strategy was developed to spatiotemporally separate biomass accumulation from selenium enrichment, resulting in high-quality selenium-enriched N. sphaeroides colonies. The first phase focused on colony growth in nitrogen-free BG-11 0 medium, while the second phase emphasized selenium enrichment in selenium-supplemented, phosphorus-reduced, and nitrogen-containing BG-11 medium (“selenium enrichment medium”). Scale-up to 300 L confirmed the robustness of this process, achieving desired colony hardness of ∼3 newtons, selenium content of ∼350 μg/g, and yield of ∼0.5 g/L (dry weight). Additionally, polysaccharides containing ∼110 μg/g selenium were isolated from the colonies, exhibiting biochemical properties similar to selenium-free polysaccharides from control colonies. These results provide valuable insight into the potential of N. sphaeroides as a cell factory chassis for producing selenium bioproducts.

Preparation and characterization of selenium enriched-Bifidobacterium longum DD98, and its repairing effects on antibiotic-induced intestinal dysbacteriosis in mice

Hui Zhu,Yan Zhou,Yan Qi,Rui Ji,Junliang Zhang,Zhixiang Qian,Chunzhen Wu,Jun Tan,Lei Shao,Daijie Chen

The aim of this study was to investigate the characteristics of a novel selenium-enriched Bifidobacterium longum DD98 (Se-B. longum DD98) supplement food and its repairing effects on the intestinal ecology of mammals. We assessed the growth, Se accumulation, and Se biotransformation of B. longum DD98 and its effects on antibiotic-induced intestinal dysbacteriosis in mice. The viable bacterial count at the end of fermentation was not significantly affected by the presence of Se. Bifidobacterium longum DD98 took up inorganic Se from the medium and biotransformed it into Se-containing proteins and selenoamino acids. The dominant Se species was selenomethionine (SeMet), which comprised 87% of the total Se in Se-B. longum DD98. Furthermore, Se-B. longum DD98 showed better regulation of the disrupted intestinal microbiota back to normal levels and repaired damaged colon tissues compared to the natural recovery and B. longum DD98 treatments. These findings suggest that B. longum DD98 efficiently biotransformed inorganic Se into more bioactive organic Se forms and may have therapeutic potential for the restoration of antibiotic-induced intestinal dysbacteriosis.

Isolation and identification of green alga Chlorella vulgaris HNUFU001 for environmental selenium enrichment in heterotrophic growth regime

Zhenyu Zhang,Li Wang,Yinqiang Wu,Chenshuo Li,Pengcheng Fu,Jing Liu

Microalgae , particularly Chlorella , have promising selenium (Se) enrichment capacity and great potential in wastewater treatment and the development of nutraceuticals. To acquire Chlorella strains with strong heterotrophic propensity and robust Se tolerance, this research utilized a heterotrophic isolation strategy along with a high Se concentration medium as selection factors. The combined sequence of the ribosomal small subunit (SSU) and internal transcribed spacer (ITS) was utilized for phylogenetic analysis , and comparative analysis of ITS secondary structures was also integrated to identify the desired strain. A novel subtropical microalgal strain, Chlorella vulgaris HNUFU001 with high Se tolerance, was isolated from water samples in a lake at Hainan University, China. Gaussian process regression was utilized to assess the strain's growth under varying Se concentrations. The intracellular accumulation and extracellular removal of Se were measured using Inductively Coupled Plasma-Mass Spectrometry (ICP-MS) and Inductively Coupled Plasma Optical Emission Spectrometer (ICP-OES), respectively. The maximum intracellular organic Se (92 % of total Se) was observed as 108.835 μg/g dry weight (DW) for 1 mg/L Se treatment. The highest Se removal rate was 40 %. The high Se tolerance and enrichment traits enable Chlorella vulgaris HNUFU001 to be a promising candidate for the heterotrophic production of organic Se-related nutraceuticals.

Transformation of W/O/W emulsions and O/W/O emulsions for co-loading selenium-enriched peptide and vitamin E: Design and characteristics

Fang Xie,Yuan Fang,Xin Liu,Xin Cong,Yangchao Luo,Jiaojiao Zhou,Zia-ud Din,Shuiyuan Cheng,Jie Cai

Double emulsions (e.g., O/W/O and W/O/W emulsions) have important applications in healthy food development for food industry. Herein, we prepared O/W/O and W/O/W emulsions by two-step emulsification method only using two food grade emulsifiers (Octenyl succinate starch ester (OSA starch) and polyglycerol polyricinoleate (PGPR)), respectively. The optimal process parameters (in external emulsion and internal emulsion) for the preparation of O/W/O and W/O/W emulsions were systematically studied. It is noteworthy that the facile conversion of the two O/W/O and W/O/W emulsions could be achieved by changing the order of emulsifier addition. The double emulsions can be used for co-loading of plant-derived selenium-enriched peptide (Se-peptide) and vitamin E (V E ) with efficient loading rates, and the emulsions of encapsulated Se-peptide and V E have good antioxidant properties. The results of fuzzy numerical sensory evaluation and electronic nose showed that both emulsion systems were effective in masking the odor of Se-peptide.

Anti-aging effect of Limosilactobacillus fermentum CGMCC 17434 in mice fed with fermented selenium-enriched yogurt

Jie Luo,Zhuhui Tian,Wenying Yuan,Xiaopeng Peng,Hui Zhou,Qingwu Shen,Yang Luo,Yuxing Guo,Zihang Shi,Xiaoxiao Jiang,Bo Chen,Daodong Pan,Xiankang Fan

Aging and Se deficiency can lead to numerous health problems in the organism, so there is a need to develop new technologies for anti-aging and daily selenium supplementation. In this study, efficient selenium-enriched lactic acid bacteria were screened and selenium-enriched yogurt was developed, and its effect on D-galactose-induced senescence in mice was further investigated. The results showed that Limosilactobacillus fermentum CGMCC 17434 could be efficiently enriched in selenium, and its organic selenium conversion rate was 88.12 %. Secondly, the content of organic selenium and resveratrol in peanut germination was 3.48 ± 0.16 μg/g d.w. and 20.36 ± 0.78 μg/g d.w., respectively, under the Na 2 SeO 3 concentration of 12 mg/L, soaking time of 10 h, soaking temperature of 30 °C, and germination temperature of 28 °C. The selenium-enriched yogurt developed on this basis contained 172.10 ± 0.56 μg/L of organic selenium and delayed the aging of D-galactose-induced senescence model mice. This might be due to the selenium-enriched Limosilactobacillus fermentum CGMCC 17434 and organic selenium in yogurt increased glutathione levels and superoxide dismutase activity, and decreased malondialdehyde, protein carbonyls and reactive oxygen species levels, as well as decreasing the release of oxidative stress-generated inflammatory factors such as IL-1β, IL-6 and TNF-α.

Biodegradable Liquid Slow-Release Mulch Film Based on Bamboo Residue for Selenium-Enriched Crop Cultivation

Chaoqi Chen,Zhaoshuang Li,Kuaile Yin,Lei Li,Zhen Zhang,Xu Xu,He Liu,Yan Qing,Xingong Li,Yiqiang Wu

The development of biodegradable mulch film is an effective means to address plastic pollution and promote modern green agriculture. In this work, with compounding sodium carboxymethyl cellulose (CMC) and quaternized lignin (QL), a biodegradable liquid mulch film (PVA@CMC/QL) was constructed by introducing polyvinyl alcohol (PVA) and a selenium-containing cross-linking agent through electrostatic interaction. The effect of sodium carboxymethyl cellulose and QL on different liquid mulch films was examined. PVA@CMC/QL had exceptional spray-film-forming properties of liquid mulch film and was capable of generating a dense mulch film above the soil/on top of the soil under natural conditions. PVA@CMC/QL exhibited excellent oxygen transmission rate (60.2 cm3·m−2·d−1·Pa−1) and water vapor transmission rate (753.4 g·m−2·d−1). Soil temperature and humidity increased by 0.4 to 2.1 °C and 0.5% to 2.8%, respectively, in the soil covered with PVA@CMC/QL compared to those in other controls, thereby confirming its exceptional moisture retention and insulation capabilities. PVA@CMC/QL combined remarkable weed suppression with only 13.3% weed germination under the mulch. Optimal rhizome growth of pak choi seedlings was observed under the PVA@CMC/QL cover, as demonstrated by the planting of both pak choi seedlings and weeds. Roots and stems increased by 3.8 ± 0.3 and 1.2 ± 0.3 cm, respectively. The weed suppression mechanism of PVA@CMC/QL was explained through the lens of density functional theory. In addition, the selenium content of pak choi seedlings under PVA@CMC/QL cover could reach 28.5 μg/kg, making the mulch film both degradable and highly reusable. This work not only improved the value-added utilization of bamboo residues but also gave new insight into the research on multifunctional bamboo–plastic mulch film.

Preparation, physicochemical characterization and identification of two novel mixed ACE-inhibiting peptides from two distinct tea alkali-soluble protein

Zhu Jiangxiong,Du Mengke,Wu Meirong,Yue Pengxiang,Yang Xiufang,Wei Xinlin,Wang Yuanfeng

In this study, two novel mixed ACE-inhibiting peptides, namely Se-TAPep 1 and TAPep 1, were prepared and purified from selenium-enriched and common alkali-soluble proteins using a series of process, and then its physicochemical properties and oligopeptide sequence were analyzed. The results showed that Se-TAPep 1 and TAPep 1 could be successfully separated, but their ACE inhibitory activities were different (IC50 values were 13.39 mg/mL and 23.56 mg/mL, respectively), this may be related to the difference in selenium content. Further analysis of physicochemical properties showed that the protein or peptide fractions contained their characteristic absorption peaks and contained aromatic amino acids, while the apparent morphology was different, the presence of selenium could change the apparent morphology of protein or polypeptide fractions, arranging of selenium-enriched fractions more irregular. The amino acid composition analysis showed that the Pro content of the selenium-enriched fractions was higher than that of the ordinary component, whereas the oligopeptide sequence analysis showed that both LQPSLGFP derived from Se-TAPep 1 and AETGEIKGHY derived from TAPep 1 contained some characteristics of highly active ACE-inhibiting peptides, but the characteristics of LQPSLGFP were more conspicuous, which may be related to its molecular weight and source of mixed peptides. These findings indicated that the selenium-enriched tea mixed peptides had better potential uses than ordinary tea mixed peptides in the preparation of functional food.

Gut Microbiota and Liver Metabolism Regulation Mediate the Protective Effects of Inactivated Selenium-Enriched Yeast Against Alcohol-Induced Liver Damage in Mice

Zihua Liang,Xiangchen Zhang,Shiwei Chen,Meiting Wang,Deying Men,Wangxin Liu,Xucong Lv

Inactivated Selenium-enriched yeast (YSe), as an organic source of selenium with multiple physiological activities, has attracted widespread attention. However, its potential to alleviate alcoholic liver injury (ALD) and its underlying mechanisms remain largely unexplored. This study explores the protective effects of inactivated YSe intervention on ALD in mice and clarifies its mechanism of action. The results indicated that, at the same selenium dose, inactivated YSe intervention was superior to inorganic selenium (sodium selenite) in alleviating ALD. Specifically, high-dose inactivated YSe significantly reduced the levels of serum ALT and AST in alcohol-exposed mice (38.69% and 24.67%, respectively), increased the level of HDL-C (16.83%), and effectively improved alcohol-induced lipid metabolism disorders and liver oxidative damage. At the same time, it significantly increased the concentration of short-chain fatty acids (SCFAs) in feces. 16S rRNA sequencing indicates that inactivated YSe intervention enhances the abundance of beneficial flora (such asBlautia,Oscillibacter,Anaerotruncus,Butyricicoccus, andRuminiclostridium) and simultaneously inhibits potentially harmful microbiota (such asxylanophium,Escherichia–Shigellaandoscilliumspirates) to restore the homeostasis of the intestinal microbiota in ALD mice. Liver metabolomics analysis revealed that inactivated YSe intervention significantly altered the liver metabolic profile. The core pathways that are regulated by YSe after alcohol disruption include glutathione metabolism, purine metabolism, riboflavin metabolism, etc. In conclusion, this study demonstrates that inactivated YSe can effectively alleviate ALD in mice by regulating the structure of the intestinal flora and restoring liver metabolic homeostasis, providing a scientific basis for its potential functional food component in the prevention and auxiliary management of ALD.

Antioxidant activity of selenium-enriched Chrysomyia megacephala (Fabricius) larvae powder and its impact on intestinal microflora in D-galactose induced aging mice

Xie Dandan,Jiang Liqin,Lin Yao,Liu Zhenwei

Background The purpose of this study was to assess the antioxidative activity of selenium-enriched Chrysomyia Megacephala (Fabricius) ( C. megacephala ) larvae powder (SCML) and its impact on the diversity and structure of intestinal microflora in a mouse model of D-galactose (D-gal)-induced oxidative damage. Methods Sixty male ICR mice were equally randomized to a normal control (NC) group, a model group, a positive group, a low-dose SCML (L-SCML) group, a mid-dose SCML (M-SCML) group, and a high-dose SCML (H-SCML) group. Animals in NC and model groups received water, animals in the positive group received 40 mg/Kg vitamin E (VE), and those in the three SCML groups received SCML which include 300, 1000 and 3000 μg/Kg selenium (Se) respectively. An oxidative damage model induced by subcutaneous injection of D-gal for 6 weeks via the neck was established. Serum oxidative stress levels and tissue appearance were evaluated. Tissues oxidative stress levels were detected by commercially available kit. Nuclear erythroid 2-related factor (Nrf2) and gut microbiota were determined by western blot and high throughput sequencing 16S rRNA gene respectively. Results An oxidative damage model was established successfully as represented by a significant elevation of malondialdehyde (MDA) and protein carbonylation, and inhibition of the antioxidants including superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), total antioxidant capacity (T-AOC) and glutathione (GSH). It was found that oxidative damage and histological alterations were attenuated, the expression of Kelch-like ECH-associated protein (Keap1) was decreased, and the expression of Nrf2 and hemeoxygenase-1 (HO-1) was increased after SCML treatment. In addition, significant changes were observed in the gut microbiota, including Proteobacteria and the ratio of Bacteroidetes to Firmicutes at the phylum level, as well as Helicobacter , Clostridium and Lactobacillus at the genus level. Conclusion SCML exerted an antioxidative effect in vivo, probably by increasing the antioxidant activity and reducing the production of oxidation products via the Nrf2 signaling pathway. SCML could also redress the intestinal flora imbalance induced by oxidative stress. All these findings suggest that SCML could serve as a functional food and natural drug additive to protect the human body against oxidative damage.

Molecular mechanisms of selenium-biofortified soybean protein and polyphenol conjugates in protecting mouse skin damaged by UV-B

Xing Zhang,Hui He,Tao Hou

Selenium-biofortified crops are a quality functional food resource because of their anti-tumor and anti-cancer properties. In the present study, the conjugates of selenium-biofortified soybean protein and polyphenols were prepared and evaluated by alkali-induced synthesis and in vitro antioxidant tests. Moreover, the antioxidant mechanisms of protecting mice skin damaged by UV-B were studied. The results showed that the antioxidant activity of the conjugate between 7S globulin from selenium-enriched soybean (Se-7S) and EGCG (Se-7S-EGCG) was significantly higher (P < 0.05) than that of Se-7S-GA. Structural characterizations implied that the polymerization of polyphenols with amino acid residues occurred. Se-7S-EGCG inhibited the apoptosis of epidermal cells induced by UV-B. The overexpression of phosphorylated proteins in the MAPK signaling pathway, the activation of related inflammatory factors, and the boost in the MMPs were reversed by Se-7S-EGCG. Overall, this research provides a theoretical and experimental basis for the application of protein and polyphenol conjugates in food and medicine fields.

Optimization of Bacillus cereus Fermentation Process for Selenium Enrichment as Organic Selenium Source

Chen Xujun,Li Shuyi,Cong Xin,Yu Tian,Zhu Zhenzhou,Barba Francisco J.,Marszalek Krystian,Puchalski Czesław,Cheng Shuiyuan

Selenium is an essential trace element and micronutrient for human health. Application of organic selenium in plants and microorganisms as trace element supplement is attracting more and more attention. In this study, Bacillus cereus, an important probiotic, was used for selenium enrichment with sodium selenite as selenium source. The growth curve of B. cereus was investigated, and 150 μg/ml was selected as the concentration of selenium for B. cereus fermentation. With application of response surface methodology, the optimal fermentation conditions were obtained as follows: inoculation quantity of 7%, culture temperature of 33°C, and shaking speed of 170 rpm, leading to the maximal selenium conversion ratio of 94.3 ± 0.2%. Field emission scanning electron microscope and energy dispersive spectrometry evidenced that inorganic selenium had been successfully transformed. This study may contribute to get a strain with high Se conversion ratio, so as to extract organic selenium in the form of selenoprotein to be used for further application.

Synthesis and Characterization of Chitosan Oligosaccharide Selenium Nanoparticles and Selenium Enrichment Evaluation and Mechanism on Soybean Sprouts

Xin Xu,Jinhang Wang,Huihui Wu,Xuedan Cui,Daibing Hou,Jun Cui,Rui Lu,Aijun Lin

Biofortification is an effective way to increase micronutrient levels in food crops. This study investigated selenium enrichment as a biofortification strategy in soybean sprouts. Chitosan oligosaccharide selenium nanoparticles (COS-Se NPs) were synthesized and used in soybean sprout culture to study their effects on the sprouts. The results showed that the enrichment factor value (EF) was higher when the concentration was 2.36–9.43 mg·L–1 compared with other concentrations, and the enrichment factor (EF) reached a maximum value of 0.9782 at a COS-Se NP concentration of 4.72 mg·L–1. Translocation factors (TFradicle-hypocotyl and TFhypocotyl-cotyledon) reached maximum values of 0.7866 and 0.9723 at 25.15 mg·L–1 and 15.09 mg·L–1, respectively, suggesting the relationship between enrichment of nanoselenium and concentration was not proportional. Most of the organic selenium was present mainly in the protein fraction, and albumin and glutelin were the main protein-bound seleniums. The contents of protein and biomacromolecule selenium reached higher values at a COS-Se NP concentration of 2.36–9.43 mg·L–1. Furthermore, the COS-Se NPs at 2.36–9.43 mg·L–1 concentration significantly affected the nutrient parameters in soybean sprouts. Different concentrations of COS-Se NPs were observed to have different effects on the mineral element content of the bean sprouts. In vitro digestion and dialysis experiments showed that the bioavailability of K, Mg, P, and Se was at a high level, while that of Fe and Ca was at a low level in soybean sprouts. Overall, no significant impact on the bioaccessibility of most elements was observed during the enrichment process compared to the control group. This work shows that the selenium enrichment strategy can be a potential soybean sprout production method and help solve the problem of nutritional deficiencies with selenium and reduce malnutrition.
