The Two-Component System TtrRS Regulates the Expression of the 05890–05900 Operon that Promotes the Sulfur Metabolism of Vibrio parahaemolyticus

Bai Yameng,Liang Tianqi,Zhao Jie,Lin Yawen,Lu Qian,Zhou Xiujuan,Yang Menghua,Zhong Xiaojun

The marine bacterium Vibrio parahaemolyticus is responsible for the seafood-borne gastroenteritis in humans. Two-component system (TCS) TtrRS plays important roles in host colonization and works as an important regulator in V. parahaemolyticus . However, the mechanisms by which TtrRS regulates the downstream genes, as well as the function of their target genes, remain largely unknown. In this study, we performed a Find Individual Motif Occurrences (FIMO) analysis using the ttrR box to scan the V. parahaemolyticus genome and identified 68 potential binding sites harboring the ttrR box. Thereinto, a ttrR box exists in the promoter region of the 05890–05900 operon. We further demonstrated that TtrR directly regulates the expression of the 05890–05900 operon by binding to the ttrR box. In addition, another TCS, 05905/05910, could assist in the regulation of TtrR to the 05890 – 05900 operon. Subsequent study showed that the 05890–05900 operon promotes the sulfur metabolism of V. parahaemolyticus at the early growth stage. Our results suggested that TtrRS and their target genes generate a complex regulatory pathway to better control sulfur metabolism in V. parahaemolyticus .Please check and confirm the edit made in the article title and amend if necessary. It's OK.

Reduction of Pentavalent Vanadium via Enhanced Levels of Sulfhydryl by Elemental Sulfur Reducing Bacteria from a Stone Coal Mining Region

Zulv Huang,Xipeng Wei,Lijuan Zhang,Tao Chen,Yi Huang,Zhangwei Yang,Wanqing Zeng,Hongxing Chen,Lingtian Xie,Bo Yan

Microbial reduction of pentavalent vanadium [V(V)] to less toxic tetravalent vanadium [V(IV)] is associated with the biogeochemical cycling of sulfur. However, the capability of elemental sulfur-reducing bacteria (S0RB) to reduce V(V) in the presence of elemental sulfur (S0) remains largely unexplored. Herein, we systematically investigated the V(V) reduction capabilities and underlying mechanisms of Bacillus megaterium R01, a S0RB strain isolated from the stone coal mining region. Under sulfur-depleted conditions, B. megaterium R01 demonstrated limited reduction capacity (<54.3% removal efficiency for 200 mg/L V(V)). In contrast, S0 supplementation enabled near-complete V(V) reduction (97.6 ± 2.1%) even at elevated concentrations (400 mg/L), achieving a relatively low carbon consumption rate (TOC/V(V) of ∼0.6). Sulfhydryl (SH) compounds, reaching 424.2 ± 9.9 μmol/L within 5 days, were likely crucial for reducing V(V) to V(IV). This mechanism differs from that of other V-reducing bacteria. The transcriptome analysis revealed that the up-regulation of genes associated with sulfur metabolism, cysteine, and methionine metabolism in B. megaterium R01 is likely to contribute to its enhanced resistance toward V(V). Additionally, the biosynthesis of intracellular BSH in B. megaterium R01, and the extracellular SH groups (e.g., Trx and TrxR encode enzymes) play an irreplaceable role in reducing V(V) by B. megaterium R01. This study enhances our understanding of V(V) reduction in sulfur-rich environments and suggests a promising approach for remediating V(V)-contaminated groundwater, sediments, and soils.

Fumarate mitigates disruption induced by fenpropathrin in the silkworm Bombyx mori (Lepidoptera): A metabolomics study

Xue-Yang Wang,Zi-Qin Zhao,Cheng-Xian Song,Zhi-Hao Su,Mu-Wang Li,Yang-Chun Wu,Byung Rae Jin,Ming-Jie Deng

The silkworm Bombyx mori L. is a model organism of the order Lepidoptera. Understanding the mechanism of pesticide resistance in silkworms is valuable for Lepidopteran pest control. In this study, comparative metabolomics was used to analyze the metabolites of 2 silkworm strains with different pesticide resistance levels at 6, 12, and 24 h after feeding with fenpropathrin. Twenty-six of 27 metabolites showed significant differences after fenpropathrin treatment and were classified into 6 metabolic pathways: glycerophospholipid metabolism, sulfur metabolism, glycolysis, amino acid metabolism, the urea cycle, and the tricarboxylic acid (TCA) cycle. After analyzing the percentage changes in the metabolic pathways at the 3 time points, sulfur metabolism, glycolysis, and the TCA cycle showed significant responses to fenpropathrin. Confirmatory experiments were performed by feeding silkworms with key metabolites of the 3 pathways. The combination of iron(II) fumarate + folic acid (IF-FA) enhanced fenpropathrin resistance in silkworms 6.38 fold, indicating that the TCA cycle is the core pathway associated with resistance. Furthermore, the disruption of several energy-related metabolic pathways caused by fenpropathrin was shown to be recovered by IF-FA in vitro . Therefore, IF-FA may have a role in boosting silkworm pesticide resistance by modulating the equilibrium between the TCA cycle and its related metabolic pathways.

Mechanisms linking triclocarban biotransformation to functional response in sulfur-metabolism biofilm

Zhou-Yang Li,Min Li,Xue-Ning Zhang,Kun Zheng,Han-Bing Xiao,Ai-Jie Wang,Yi-Lu Sun

Triclocarban (TCC), a persistent antimicrobial compound widely present in municipal wastewater, poses potential risks to biological nitrogen removal processes. However, the potential risks posed by TCC to advanced treatment processes—particularly sulfur-metabolism biofilm reactor—remain poorly understood and require further elucidation. This study investigated the impact of TCC on sulfur-metabolism biofilm base on sulfur-metabolism biofilm reactor, focusing on nitrogen removal performance, TCC transformation pathways, and microbial community dynamics. The results showed that sulfur-metabolism biofilm maintained stable nitrogen removal efficiency under trace TCC (≤ 25 μg/L) stress with nitrate removal improved from 93 % to 98 %, whereas performance declined significantly at 100 μg/L, reducing nitrate removal to 81.8 ± 3.9 % and increasing effluent NO 2 ⁻-N and N 2 O-N by 6.0- and 28.0-fold, respectively. Functional predictions via FAPROTAX suggested TCC-induced alterations in sulfur and nitrogen metabolic pathways. In addition, TCC and its transformation intermediates (MCC, DCC, NCC, 3,4-DCA, 4-CA) accumulated in the biofilm. Network analysis revealed syntrophic interactions between sulfur-oxidizing bacteria (e.g., Sulfurisoma and Sulfuritalea ) and hydrogenase-rich TCC degraders (e.g., Unclassified Chloroflexi ). These findings highlight the potential of sulfur-metabolism biofilm to achieve simultaneous nitrogen removal and micropollutant transformation in low-carbon, sulfur-rich wastewater environments, offering a sustainable solution for advanced wastewater treatment systems.

Biofilm metagenomic characteristics behind high coulombic efficiency for propanethiol deodorization in two-phase partitioning microbial fuel cell

Jian Yu,Juping You,Piet N.L. Lens,Lichao Lu,Yaxue He,Zhenyi Ji,Jianmeng Chen,Zhuowei Cheng,Dongzhi Chen

ABSTRACT Hydrophobic volatile organic sulfur compounds (VOSCs) are frequently found during sewage treatment, and their effective management is crucial for reducing malodorous complaints. Microbial fuel cells (MFC) are effective for both VOSCs abatement and energy recovery. However, the performance of MFC on VOSCs remains limited by the mass transfer efficiency of MFC in aqueous media. Inspired by two-phase partitioning biotechnology, silicone oil was introduced for the first time into MFC as a non-aqueous phase (NAP) medium to construct two-phase partitioning microbial fuel cell (TPPMFC) and augment the mass transfer of target VOSCs of propanethiol (PT) in the liquid phase. The PT removal efficiency within 32 h increased by 11–20% compared with that of single-phase MFC, and the coulombic efficiency of TPPMFC (11.01%) was 4.32–2.68 times that of single-phase MFC owing to the fact that highly active desulfurization and thiol-degrading bacteria (e.g., Pseudomonas, Achromobacter ) were attached to the silicone oil surface, whereas sulfur-oxidizing bacteria (e.g., Thiobacillus, Commonas, Ottowia ) were dominant on the anodic biofilm. The outer membrane cytochrome-c content and NADH dehydrogenase activity improved by 4.15 and 3.36 times in the TPPMFC, respectively. The results of metagenomics by KEGG and COG confirmed that the metabolism of PT in TPPMFC was comprehensive, and that the addition of a NAP upregulates the expression of genes related to sulfur metabolism, energy generation, and amino acid synthesis. This finding indicates that the NAP assisted bioelectrochemical systems would be promising to solve mass-transfer restrictions in low solubility contaminates removal.

Bisphenol A Decelerates Sulfur Metabolism in Roseovarius nubinhibens at Environmentally Relevant Concentrations

Xin-Run Sun,Wan-Song Zong,Lingqiang Kong,Xiang Chen,Mei-Yan Liu,Su-Chun Wang,Peng-Fei Xia,Xian-Zheng Yuan,Li-Juan Feng

Bisphenol A (BPA) is a pervasive environmental contaminant known for its detrimental effects on human health. However, its impact on multiple generations of microorganisms and the biogeochemical cycles they mediate, particularly at environmentally relevant concentrations, remains poorly understood. This study explores the effects of BPA on Roseovarius nubinhibens, a crucial bacterium in the global sulfur cycle, under both short-term (F0 generation) exposure at the observed-effect concentration (26 mg/L) and long-term (F20 generation) exposure at an environmentally relevant concentration (0.06 mg/L). Short-term exposure to the observed-effect concentration significantly inhibited bacterial growth by 12.9%, while long-term exposure at the environmentally relevant concentration induced notable morphological changes without affecting growth. BPA at the observed-effect concentration also disrupted extracellular polymeric substance (EPS) production and protein structures, particularly in the soluble-EPS (S-EPS) fraction, leading to impaired bacterial flocculation. Additionally, BPA at both exposure levels decelerated the conversion of dimethylsulfoniopropionate (DMSP), with the observed-effect concentration particularly affecting the cleavage pathway, reducing dimethyl sulfide (DMS) production. This study provides the first direct experimental evidence that BPA disrupts the metabolic equilibrium of sulfur cycling. These findings underscore the need for deeper exploration of BPA’s environmental risks, especially at environmentally relevant concentrations, and its potential to interfere with microbial-driven sulfur biogeochemistry.

Bidirectionally Regulating Viral and Cellular Ferroptosis with Metastable Iron Sulfide Against Influenza Virus

Xinyu Miao,Yinyan Yin,Yulian Chen,Wenhui Bi,Yuncong Yin,Sujuan Chen,Daxin Peng,Lizeng Gao,Tao Qin,Xiufan Liu

Influenza virus with numerous subtypes and frequent variation limits the development of high-efficacy and broad-spectrum antiviral strategy. Here, a novel multi-antiviral metastable iron sulfides (mFeS) against various influenza A/B subtype viruses is developed. This work finds that mFeS induces high levels of lipid peroxidation and •OH free radicals in the conservative viral envelope, which depends on Fe 2+ . This phenomenon, termed as a viral ferroptosis, results in the loss of viral infectibility and pathogenicity in vitro and in vivo, respectively. Furthermore, the decoction of mFeS (Dc(mFeS)) inhibits cellular ferroptosis-dependent intracellular viral replication by correcting the virus-induced reprogrammed sulfur metabolism, a conserved cellular metabolism. Notably, personal protective equipment (PPE) that is loaded with mFeS provides good antiviral protection. Aerosol administration of mFeS combined with the decoction (mFeS&Dc) has a potential therapeutic effect against H1N1 lethal infection in mice. Collectively, mFeS represents an antiviral alternative with broad-spectrum activity against intracellular and extracellular influenza virus.

Simultaneous ammonium and sulfate biotransformation driven by aeration: Nitrogen/sulfur metabolism and metagenome-based microbial ecology

Haiguang Yuan,Jianqi Yuan,Yingying You,Biaojun Zhang,Yixiao Wu,Shaobin Huang,Yongqing Zhang

The present study aimed to clarify the effect of oxygen respiration on biotransformation of alternative electron acceptors (e.g., nitrate and sulfate) underlying the simultaneous removal of ammonium and sulfate in a single aerated sequencing batch reactor . Complete nitrification was achieved in feast condition, while denitrification was carried out in both feast and famine conditions when aeration intensity (AI) was higher than 0.22 L/(L·min). Reactors R1 [0.56 L/(L·min)], R2 [0.22 L/(L·min)], and R3 [0.08 L/(L·min)] achieved 72.39% sulfate removal efficiency in feast condition, but H 2 S release occurred in R3. Following exogenous substrate depletion, sulfate concentration increased again and exceeded the influent value in R1, indicating that sulfate transformation was affected by oxygen intrusion. Metagenomic analysis showed that a higher AI promoted sulfate reduction by switching from dissimilatory to assimilatory pathway. Lower AI-acclimated microorganisms (R3) produced H 2 S and ammonium, while higher AI-acclimated microorganisms (R1) accumulated nitrite, which confirmed that biotransformation of N and S was strongly regulated by redox imbalance driven by aeration. This implied that respiration control, a microbial self-regulation mechanism, was linked to the dynamic imbalance between electron donors and electron acceptors. Aerobic nitrate (sulfate) reduction, as one of the effects of respiration control, could be used as an alternative strategy to compensate for dynamic imbalance, when supported by efficient endogenous metabolism. Moderate aeration induced microorganisms to change their energy conservation and survival strategy through respiration control and inter-genus protection of respiratory activity among keystone taxa (including Azoarcus in R1, Thauera in R2, and Thiobacillus , Ottowia , and Geoalkalibacter in R3) to form an optimal niche in response to oxygen intrusion and achieve benign biotransformation of C, N, and S without toxic intermediate accumulation. This study clarified the biotransformation mechanism of ammonium and sulfate driven by aeration and provided theoretical guidance for optimizing existing aeration-based techniques.

A Comparative Transcriptome Analysis Unveils the Mechanisms of Response in Feather Degradation by Pseudomonas aeruginosa Gxun-7

Chaodong Song,Rui Liu,Doudou Yin,Chenjie Xie,Ying Liang,Dengfeng Yang,Mingguo Jiang,Hongyan Zhang,Naikun Shen

Microbial degradation of feathers offers potential for bioremediation, yet the microbial response mechanisms warrant additional investigation. In prior work,Pseudomonas aeruginosaGxun-7, which demonstrated robust degradation of feathers at elevated concentrations, was isolated. However, the molecular mechanism of this degradation remains only partially understood. To investigate this, we used RNA sequencing (RNA-seq) to examine the genes that were expressed differentially inP. aeruginosaGxun-7 when exposed to 25 g/L of feather substrate. The RNA-seq analysis identified 5571 differentially expressed genes; of these, 795 were upregulated and 603 were downregulated. Upregulated genes primarily participated in proteolysis, amino acid, and pyruvate metabolism. Genes encoding proteases, as well as those involved in sulfur metabolism, phenazine synthesis, and type VI secretion systems, were notably elevated, highlighting their crucial function in feather decomposition. Integration of Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) taxonomies, combined with a review of the literature, led us to propose that metabolic feather degradation involves environmental activation, reducing agent secretion, protease release, peptide/amino acid uptake, and metabolic processes. Sulfite has emerged as a critical activator of keratinase catalysis, while cysteine serves as a regulatory mediator. qRT–PCR assay results for 11 selected gene subset corroborated the RNA-seq findings. This study enhances our understanding of the transcriptomic responses ofP. aeruginosaGxun-7 to feather degradation and offers insights into potential degradation mechanisms, thereby aiding in the formulation of effective feather waste management strategies in poultry farming.

Heterogeneity of microbiome and flavor profiles of industrial-scale Laotan Suancai: Fermentation craft plays important roles

Shijin Xiong,Dandan Qiu,Xiaoyan Xu,Tonghao Du,Zhen Peng,Muyan Xiao,Tao Huang,Jinqing Huang,Hongbing Ren,Mingyong Xie,Tao Xiong

Laotan Suancai is a well-known Chinese traditional fermented vegetable with a unique flavor. However, there are significant differences in the quality of Laotan Suancai pickled by jars (LTSC-J) and pools (LTSC-P) on an industrial scale. To reveal the effect of fermentation craft on Laotan Suancai quality, the microbial community, flavor metabolites, and physicochemical properties of the two types of Laotan Suancai, as well as their interrelationship were investigated. LTSC-P had higher salinity, glucose content, and histidine content, while LTSC-J had higher TA, lactic acid content, and isobutyric acid content. Nine differential aroma-active compounds were found between the two types of Laotan Suancai: anethole , eucalyptol, diallyl sulfide, estragole, diallyl disulfide , eugenol, linalool, nonanal , and ethyl butyrate. LTSC-P showed a higher relative abundance of Weissella , Acinetobacter , Pseudomonas , Allorhizobium-Neorhizobium-Pararhizobium-Rhizobium , Inhella , and Leuconostoc compared to LTSC-J. Co-occurrence analysis indicated the distinct differences in the co-occurrence patterns of the microbial network across the two types of Laotan Suancai. RDA analysis indicated that glucose, salinity, TA, maltose , and nitrite were the main drivers for the differences in bacterial communities between the two types of Laotan Suancai. Spearman correlation analysis revealed strong correlations between 18 bacteria and 61 flavor compounds. Finally, bacterial metabolic function prediction indicated that sulfide and aromatic ester may be biosynthesized by sulfur metabolism , cysteine and methionine metabolism, pyruvate metabolism, and butanoate metabolism. Overall, these findings provide important guidance for improving the manufacturing and quality of Laotan Suancai.

The role of linalool in managing Alternaria alternata infection and delaying black mold rot in goji berry

Huaiyu Zhang,Yueli Zhou,Yue Yang,Lunaike Zhao,Hongbo Li,Lijie Long,Junjie Wang,Xuyan Wu,Zhao-Jun Wei

Alternaria alternata , a common fungal pathogen causes black mold in postharvest goji fruit. To explore the in vitro and in vivo impacts of linalool on the fungal infection, we treated A. alternata and infected goji fruit with varying linalool concentrations (0, 0.15, 0.45, and 1.35 mL L −1 ). Linalool repressed hypha growth, spore yield and germination, and germ tube length of A. alternata . Furthermore, we observed abnormal spore and mycelium morphology in the treated samples using optical, scanning electron, and transmission electron microscopy. Linalool reduced lesion diameter and disease incidence in goji fruit. Moreover, transcriptomic analysis of pathogen revealed that linalool inhibited nitrogen metabolism, altered chitin and β -1,3-glucan metabolisms, downregulated the key gene expression related to ergosterol synthesis and sulfur and glutathione metabolisms, regulated the enzymatic antioxidant system, and impacted the proportion of unsaturated fatty acids. Furthermore, linalool damaged the fungal cell wall integrity by inducing alkaline phosphatase and chitinase activities and β -1,3-glucan content and repressing β -1,3-glucanase and chitin synthase activities and chitin levels. Linalool elevated superoxide dismutase and catalase activities and declined peroxidase and glutathione reductase activities and glutathione content in A. alternata , leading to increased H 2 O 2 levels and ROS stress. Our data exhibited the promising antifungal effects of linalool with a future application in management of the postharvest rot of goji fruit.
