In this work, a novel ascorbic acid (AA) ratiometric biosensor was prepared by using the PCN-333 (Al) metal-organic frameworks (MOFs) (PCN stands for porous coordination network) to encapsulate Ketjen black (KB) as catalyst for catalyzing oxidation of AA and thionine (Thi) as an internal reference signal simultaneously. The encapsulation of KB and Thi in the pores of PCN-333 (Al) MOFs not only improved efficiency of KB and Thi greatly because PCN-333 (Al) MOFs could effectively avoid the cohesion or aggregation of KB and Thi on electrode surface but also enhanced the stability of biosensors because they were immobilized in the pore firmly. Furthermore, PCN-333 (Al) MOFs could also selectively accumulate target analytes into their pores to enhance the selectivity of biosensors. The oxidation peak current of AA catalyzed by KB at −0.05 V increased with the increasing concentration of AA, while the oxidation peak current of Thi at −0.24 V kept constant, which resulted in a novel ratiometric biosensor for AA detection. The ratiometric biosensor exhibited a wider linear range from 14.1 ± 0.2 to (5.5 ± 0.1) х10 3 μM (R 2 = 0.998) and a lower detection limit of 4.6 ± 0.1 μM with high accuracy, selectivity, reproducibility and sensitivity. The ratiometric electrochemical approach is not only a new method for AA detection but also opens a new way for sensitive detection of other analytes.

A sensitive immunosensor for quantifying prostate specific antigen (PSA) had been described. In the design, gold nanoparticles decorated thionine-functionalized carbon nanospheres (CNSs/Thi@AuNPs) for immobilization of primary antibody (Ab 1 ) as an electrochemical sensing platform. Silicon dioxide nanospheres (SiO 2 NSs), with large surface area and good biocompatible, were loaded with horseradish peroxidase (HRP) and horseradish peroxidase labeled secondary antibodies (HRP-Ab 2 ). In the presence of PSA, a sandwich type immunocomplex from Ab 1 -Ag-Ab 2 had been formed. Differential pulse voltmmetry (DPV) was employed to obtain the response signal of immunosensor. Under the suitable experimental conditions, the changes of current intensity were linear with the logarithm of PSA concentration from 0.1 pg·mL -1 to 100 ng·mL -1 , and the detection limit was 0.025 pg·mL -1 (S/N=3). Moreover, the immunosensor possessed satisfactory specificity, repeatability and stability, suggesting that it had potential application value in the detection of tumor markers.

Taking a composite of a nanomaterial and a signal molecule as a substrate material can provide a label-free electrochemical platform. Besides, the nanomaterial with a high catalytic activity towards the signal molecule can improve the sensitivity of the platform. Herein, a thionine functionalized Fe–N–C nanocomposite was employed as the substrate. Firstly, the electrocatalytic activity of Fe–N–C towards the electroreduction of thionine was explored. Then, an immobilization-free and label-free electrochemical platform for the determination of microRNA-21 based on Fe–N–C–thionine/Fe3O4@AuNPs was constructed. A magnetic glassy carbon electrode (MGCE) was used to keep the magnetic Fe–N–C–thionine/Fe3O4@AuNPs modified onto the surface of the MGCE. Fe–N–C and Fe3O4 nanoparticles can co-catalyze the electroreduction of thionine and a strong electrochemical reduction signal of thionine could be realized in the differential pulse voltammetry (DPV) test. Also, a catalytic hairpin assembly (CHA) reaction was utilized to enhance the sensitivity of the developed electrochemical biosensor. Besides, the developed biosensor shows excellent specificity and reproducibility in the test of human serum samples, indicating its wide application prospects in the early-stage diagnosis of tumors.

Protein kinase activity analysis is essential and important for elucidation of many fundamental biological processes, disease diagnosis, and drug discovery. Herein, a novel electrochemical biosensing method for protein kinase (PKA) activity was demonstrated by the use of a reduced graphene oxide-zirconium dioxide–thionine (rGO-ZrO2–Thi) nanocomposite, which interestingly served as an integral phosphopeptide-recognizing, signal amplifying and reporting platform. The ZrO2 nanoparticle-decorated reduced graphene oxide (rGO-ZrO2) was first prepared by a hydrothermal reaction route, and then the thionine was conjugated onto the rGO surface via π–π stacking as an excellent electrochemical probe. The prepared rGO-ZrO2–Thi nanocomposites were well-characterized by various techniques. With the full advantage of specific recognition of ZrO2 nanoparticles for the phosphate group, signal amplification, and transduction of abundant thionines onto the rGO surface, and excellent conductivity of rGO, the rGO-ZrO2–Thi nanocomposite endowed a label-free and one-step electrochemical analysis of kemptide phosphorylation catalyzed by PKA. The detection limit for PKA activity was experimentally achieved as 0.005 U/mL, which was evidently lower than most of the reported methods. The proposed sensing strategy could be also applied for an efficient inhibitor evaluation. Therefore, it offered an excellent pathway for a generic and sensitive electrochemical assay of PKA activity and inhibitor.

In order to effectively monitor the residue of kanamycin (KAN), a dual-signal-amplified electrochemiluminescence (ECL) aptasensor based on multi-walled carbon [email protected] dioxide/thionine ( [email protected] 2 /Thi) was proposed. [email protected] 2 with large specific surface area and favorable biocompatibility could accelerate charge transfer and enable high loading of luminol to enhance ECL response. As a perfect electronic mediator, Thi could also accelerate electron conductivity to further enhance ECL intensity. The ECL intensity of [email protected] 2 /Thi was enhanced for 3.6-fold compared with that of individual Thi because Thi could strongly interact with MWCNTs through π-π stacking force to enhance the electronic transmission. With the outstanding electron transfer property of [email protected] 2 and Thi, ECL intensity of the proposed aptasensor was obviously increased. Upon addition of KAN, the aptamer bound to its target, which caused that the ECL intensity decrease significantly. Therefore, KAN concentration could be monitored on the basis of signal intensity. Under optimal conditions, the constructed aptasensor exhibited a sensitive response towards KAN and a low detection limit of 0.049 ng mL −1 was obtained. It also possessed the excellent specificity, favorable stability and good reproducibility. Importantly, the application of proposed ECL aptasensor provides an efficient approach for highly sensitive detection of various small molecular contaminants.

A highly sensitive electrochemical adenosine aptasensor was fabricated by covalently immobilizing 3′-NH 2 -terminated capture probe (SSDNA1) and thionine (TH) on Au–GS modified glassy carbon electrode. 3′-SH-terminated adenosine aptamer (SSDNA2) was adsorbed onto palladium/copper alloyed supported on MWCNTs ( [email protected] )-conjugated multiple bienzymes, glucose oxidase (GOx), and horseradish peroxidase (HRP) (SSDNA2/ [email protected] /HRP/GOx). Then, it was immobilized onto the electrode surface through the hybridization between the adenosine aptamer and the capture probe. The signal was amplified based on the gradual electrocatalytic reduction of GOx-generated hydrogen peroxide by the multiple HRP through the mediating ability of the loaded multiple TH. However, the peak current of TH decreased in the presence of adenosine because the interaction between adenosine and its aptamer made SSDNA2/ [email protected] /HRP/GOx release from the modified electrode . Various experimental parameters have been optimized for the detection of adenosine and tests for selectivity , reproducibility and stability have also been performed. Under the optimal condition, the proposed aptasensor displayed a wide linear range (10–400 nM) with the low detection limit (2.5 nM), which has been applied in human serum samples with satisfactory results. Thus, the combination of Au–GS as a sensor platform and [email protected] /HRP/GOx as labels can be a promising amplification strategy for highly sensitive adenosine detection.

An ultrasensitive electrochemical aptasensor for the quantitative detection of kanamycin antibiotic was fabricated based on a novel signal amplification strategy. This aptasensor was developed using thionine functionalized graphene (GR-TH) and hierarchical nanoporous (HNP) PtCu alloy as biosensing substrates for the first time. HNP-PtCu alloy with controllable bimodal ligament/pore distributions was successfully prepared by two-step dealloying of a well-designed PtCuAl precursor alloy combined with an annealing operation. GR-TH composite was synthesized by one-step reduction of graphene oxide (GO) in TH solution. Greatly amplified sensitivity was achieved by using GR-TH/HNP-PtCu composite owing to its large specific surface and good electron-transfer ability. Under the optimized conditions, the proposed aptasensor exhibited a high sensitivity and a wider linearity to kanamycin in the range 5×10 −7 –5×10 −2 μg mL −1 with a low detection limit of 0.42 pg mL −1 . This aptasensor also displayed a satisfying electrochemical performance with good stability, selectivity and reproducibility. The as-prepared aptasensor was successfully used for the determination of kanamycin in animal derived food.

A label-free DNA-based electrochemical biosensor owning high sensitivity and selectivity has been established for detecting bisphenol A in a wide range of applications. Coupling the high electrochemical performance of graphene oxide-thionine-Au nanomaterial with the specific binding capacity of the aptamers to BPA, the monitoring of trace amount of BPA was realized, the detection limit was 3.3 pg ⋅ mL −1 with strong anti-interference. Besides, using molecular docking, it was found that BPA binds to the bases DC-49, DC-51, DG-52, DG-53 and DA-63 on the aptamer via hydrogen bonding and π-π stacking interactions. Finally, the biosensor had been successfully applied in different real samples.

In this paper, a sensing platform for the electrocatalytic oxidation of nicotinamide adenine dinucleotide (NADH) was proposed. It was prepared by the electropolymerization of poly(thionine-methylene blue) (PTH-MB) in ternary nature deep eutectic solvent (NADES) on a glassy carbon electrode (GCE) that was modified with electrochemically reduced graphene oxide (ERG). The modified electrode was denoted as PTH-MB NADES -ERG/GCE. Cyclic voltammetry (CV), chronocoulometry (CC), electrochemical impedance spectroscopy (EIS), scanning electron microscope (SEM), Raman spectroscopy, and Ultra Violet-Visible Spectroscopy (UV–Vis) were used to characterize the preparation processes. The electrochemical performances of the electrode in NADH oxidation were studied via CV and current-time methods. The NADH oxidation overpotential was lower on PTH-MB NADES -ERG/GCE compared to that on GCE, ERG/GCE, and PTH-MB NADES /GCE. The PTH-MB NADES -ERG/GCE showed excellent electrocatalytic activity for NADH oxidation with a linear range of 0.51–3333.33 µM (R 2 = 0.9534) and a detection limit of 0.159 nM. The sensor was further tested for NADH determination in artificial urine samples, showing promising biomedical applications.
