High-Resolution Imaging of Biological Soft and Hard Tissues Using NIR Scintillators

Xinran Wang,Heng Dai,Yichun Liu,Linglu Kuang,Hao Song,Zhaowei Teng,Dawei Wang,Ting Wang,Xuhui Xu

Near-infrared (NIR) luminescence has emerged as a powerful tool in biological imaging due to its nondestructive nature, high spatiotemporal resolution, and deep tissue penetration. However, traditional NIR imaging often fails to provide detailed information about hard tissues such as bones. In this study, an approach for simultaneous imaging of soft and hard tissues using Lu 1.5 Mg 1.5 Al 3.5 Si 1.5 O 12 : Yb 3+ as an NIR scintillator is presented. Under X-ray irradiation, Lu 1.5 Mg 1.5 Al 3.5 Si 1.5 O 12 : Yb 3+ scintillator emits NIR emission with a peak at 970 nm with thermal quenching resistance, which is strongly absorbed by soft tissues, enabling soft tissue imaging. Additionally, the scintillator provides a high signal-to-noise ratio for X-ray imaging, allowing for high-resolution imaging of hard tissues, unaffected by ambient light, with a spatial resolution of up to 203 µm. By leveraging X-ray as the sole excitation source, this method provides a comprehensive view of internal biological structures, including vascular and skeletal tissues, with high resolution and minimal interference. This innovation holds great promise for advancing medical imaging technologies and enhancing diagnostic capabilities.

Assembled collagen films modified using polyacrylic acid with improved mechanical properties via mineralization

Xiaohui Chen,Zhilin Huang,Shuyun Zhang,Hong Li

The imperative task of enforcing collagen materials holds paramount significance in the field of hard tissue repair. We hereby present mineralized collagen fiber films via mineralization with improved mechanical properties. Self-extracted collagen was assembled into an array with an aligned fibrous pattern and then modified with polyacrylic acid (PAA) followed by mineralization in cationic polyacrylamide (CPAM)–SBF. Biomineralization occurred at the inner and outer surface of the assembled collagen fiber films. A tensile strength of up to 40.38 ± 3.08 MPa of mineralized collagen was obtained, for the first time, which may be attributed to the synergistic effect of polyanion and polycation on the mineralization process of assembled intrafibrillar collagen fibers. It was argued that PAA may facilitate the intra-fiber interaction of collagen, which extends the elongation at break of collagen fibers. This study introduces a pioneering approach for the preparation of mineralized collagen materials with superior mechanical properties, which would be beneficial for hard tissue repair.

Investigation of nature of starting materials on the construction of hydroxyapatite 1D/3D morphologies

Yin-chuan Wang,Wei-li Xu,Yu-peng Lu,Wen-hua Xu,Han Yin,Gui-yong Xiao

With the increasing requirement of bone repair materials, hydroxyapatite (HA) has been paid widely attention to investigation because of its good bioactivity and osteoconductivity. The structure of HA is a vital factor to expand its application in the field of hard tissue therapy. Thus, many strategies have been utilized in fabricating one-dimensional (1D) and three-dimensional (3D) nanostructured HA. In this paper, we successful synthesize HA with 1D nanofibers and 3D nanostructured microspheres using stearic acid as a template and different phosphates as phosphorus sources under the same synthetic system. The morphology of HA changes from nanofibers with high flexibility to nanostructured microspheres with good sphericity under the synergistic effect of stearic acid and various phosphates. The HA nanofibers and microspheres are promising for applications in biomedical fields. Base on characterization results, the formation mechanisms of HA nanofibers and HA microspheres self-assembled by nanorods are proposed. Furthermore, the HA morphology transition from nanofibers to nanostructured microspheres may be attributed to the formation of polyphosphate-induced water-in-oil microemulsion system in the synthesis process. The finding may provide a new direction to control HA morphology from 1D nanofibers to 3D microspheres based on previous strategies.

A versatile three-dimensional foam fabrication strategy for soft and hard tissue engineering

Changlu Xu,Yanjie Bai,Shaofeng Yang,Huilin Yang,David A Stout,Phong A Tran,Lei Yang

The fabrication strategies of three-dimensional porous biomaterials have been extensively studied and well established in the past few decades, yet the biocompatibility and versatility of porous architecture preparation is still lacking. Herewith, we present a novel and green 3D porous foam fabrication technique for both soft and hard engineering. By utilizing the gelatinization and retrogradation properties of starches, stabilized porous constructs made of various building blocks, from living cells to ceramic particles, were created for the first time. In soft tissue engineering applications, 3D cultured tissue foam (CTF) with controlled cell release properties was developed, and foams constituting osteoblasts, fibroblasts and vascular endothelial cells all exhibited high mechanical stability and preservation of cell viability or functions. More importantly, the CTF achieved sustained self-release of cells controlled by serum concentration (containing amylase) and the released cells also maintained high viability and functions. In the context of hard tissue engineering applications, ceramic/bioglass (BG) foam scaffolds were developed by a similar starch-assisted foaming strategy where the resultant bone scaffolds of hydroxyapatite (HA)/BG and Si3N4/BG possessed >70% porosity with interconnected macropores (sizes 200 ∼ 400 μm), fine pores (sizes 1 ∼ 10 μm) and superior mechanical properties despite the high porosity. Additionally, in vitro and in vivo evaluations of the biological properties revealed that porous HA/BG foam exhibits the desired biocompatibility and osteogenesis. The in vivo study indicated new bone ingrowth after 1 week and significant increases in new bone volume after 2 weeks. In conclusion, the presented foaming strategy provides opportunities for biofabricating CTF with different cells for different target soft tissues and preparing porous ceramic/BG foams with different material components and high strengths, showing great versatility in soft and hard tissue engineering.

Biomimetic Intrafibrillar Mineralization of Native Tendon for Soft–Hard Interface Integration by Infiltration of Amorphous Calcium Phosphate Precursors

Yangwu Chen,Yuxiang Zhang,Xiaoyi Chen,Jiayun Huang,Bo Zhou,Tao Zhang,Wei Yin,Cailian Fang,Zi Yin,Haihua Pan,Xiongfeng Li,Weiliang Shen,Xiao Chen

Soft and hard tissues possess distinct biological properties. Integrating the soft-hard interface is difficult due to the inherent non-osteogenesis of soft tissue, especially of anterior cruciate ligament and rotator cuff reconstruction. This property makes it difficult for tendons to be mineralized and integrated with bone in vivo. To overcome this challenge, a biomimetic mineralization strategy is employed to engineer mineralized tendons. The strategy involved infiltrating amorphous calcium phosphate precursors into collagen fibrils, resulting in hydroxyapatite deposition along the c -axis. The mineralized tendon presented characteristics similar to bone tissue and induced osteogenic differentiation of mesenchymal stem cells. Additionally, the interface between the newly formed bone and tendon is serrated, suggesting a superb integration between the two tissues. This strategy allows for biomineralization of tendon collagen and replicating the hallmarks of the bone matrix and extracellular niche, including nanostructure and inherent osteoinductive properties, ultimately facilitating the integration of soft and hard tissues.

Facile synthesis and in vitro bioactivity of radial mesoporous bioactive glasses

Yudong Wang,Tianshun Liao,Miao Shi,Cong Liu,Xiaofeng Chen

This paper reports a facile method for fabricating radial mesoporous bioactive glasses (RMBGs) using Hexadecyl trimethyl ammonium bromide (CTAB) micelle as the reaction center. The radial structure was formed with the extension of precursor dissolved in the hydrophobic solvent (cyclohexane) which gathered in the internal part of the CTAB micelle. The morphology, structure and in vitro bioactivity of RMBGs were investigated by various methods. The results indicate that the CTAB micelle could restrict the precursor-dissolved cyclohexane and was in favor of preparing the RMBGs with radial mesoporous structure, high specific surface area and good apatite-forming ability. The obtained RMBGs with the novel structure and properties may have potential application in drug delivery and hard tissue regeneration.

Microenvironment-Regulated Dual-Layer Microneedle Patch for Promoting Periodontal Soft and Hard Tissue Regeneration in Diabetic Periodontitis

Yaning Qu,Huajing Zeng,Lei Wang,Zhenlin Ge,Bin Liu,Zengjie Fan

Chronic periodontitis in individuals with diabetes can exacerbate the destruction of local periodontal soft tissues and accelerate the resorption of hard tissue. Currently, effective strategies to simultaneously restore both soft and hard periodontal tissues remain insufficient. To address this challenge, a multifunctional dual-layer microneedles (d-MNs) design is proposed to regenerate both periodontal soft and hard tissues in diabetic patients. The d-MNs substrate is composed of gelatin methacryloyl (GelMA) infused with nano-hydroxyapatite (nHA), which facilitates the differentiation of osteogenic cells into osteoblasts, thereby promoting alveolar bone regeneration. The tips of the d-MNs, on the other hand, are primarily made of hyaluronic acid (HA) combined with a magnesium-based metal-organic framework (Mg-MOF) loaded with glucose oxidase (GOX). This composition creates a hypoglycemic, angiogenic, and anti-inflammatory microenvironment, which supports soft tissue repair. When implanted at the site of periodontitis, the synergistic interaction between the d-MNs substrate and tips effectively promotes the regeneration of both soft and hard tissues, as demonstrated in diabetic rat models of periodontitis. These innovative d-MNs have the potential to revolutionize traditional approaches to treating diabetic periodontitis and can see broad application in dental clinics.

Bioinspired mineral-in-shell nanoarchitectonics: Functional empowerment of mineral precursors for guiding intradentinal mineralization

Zheng Xiaoran,Liu Yang,Li Mingjing,Li Yuyan,Gao Wanshan,Qiu Rongmin,Xing Jiaqi,Yang Jiaojiao,Chen Yantao,Xu Xinyuan,Ding Mingming,Luo Jun,Li Jianshu

Effective mineralization of biological structures poses a significant challenge in hard tissue engineering as it necessitates overcoming geometric complexities and multistep biomineralization processes. In this regard, we propose “mineral-in-shell nanoarchitectonics”, inspired by the nanostructure of matrix vesicles, which can influence multiple mineralization pathways. Our nanostructural design empowers mineral precursors with tailorable properties through encapsulating amorphous calcium phosphate within a multifunctional tannic acid (TA) and silk fibroin (SF) nanoshell. The bioinspired nanosystem facilitates efficient recruitment of mineral precursors throughout the dentin structures, followed by large-scale intradentinal mineralization both in vitro and in vivo , which provides persistent protection against external stimuli. Theoretical simulations combined with experimental studies attribute the success of intradentinal mineralization to the TA-SF nanoshell, which exhibits a strong affinity for the dentin structure, stabilizing amorphous precursors and thereby facilitating concomitant mineral formation. Overall, this bioinspired mineral-in-shell nanoarchitectonics shows a promising prospect for hard tissue repair and serves as a blueprint for next-generation biomineralization-associated materials.

An Interlocked Bone/Fiber Construct Recapitulating the Native Soft‒Hard Tissue Interfaces for Periodontal Complex Regeneration in Pigs

Yi Tian,Xuan Li,Yu-Zhe Chen,Dian Gan,Dao-Kun Deng,Lin Jing,Fang Li,Wen-Jie Zhang,Rui-Xin Wu,Fen Liu,Jia Wang,Bei-Min Tian,Fa-Ming Chen,Xiao-Tao He

The repair of periodontal complexes poses a significant challenge due to the lack of functional material constructs that can recapitulate the hierarchically interlocked structures of the soft–hard tissue interfaces within periodontium. Here, a bioinspired construct is fabricated by engineering interlocked bone–fiber interfaces within its hierarchical architectures to mimic the multiscale features found in native periodontal ligament (PDL)‒bone enthesis. The 3D-printed bone module and electrospun fiber module of the bioinspired constructs offered suitable architecture, biomechanics, and cytokine landscapes to recruit reparative cells and promote their osteogenic and fibrogenic differentiation. At the interfaces of the dual-module construct, the electrospun nanofibers of the fiber module are inserted into the bone module owing to various intermolecular interactions, which facilitate strong conjunction between the dual modules and provided biophysical gradients along the inserted nanofibers, similar to the microstructure and micromechanics of PDL‒bone insertions. When the constructs are placed in periodontal defects in pigs for 12 weeks, the alveolar bone is restored to its native level, and newly formed mature collagen fibers are inserted into the regenerated alveolar bone, culminating in superb integration between the soft and hard tissue interfaces. The findings demonstrate that bioinspired constructs with interlocked bone–fiber interfaces can regenerate complex soft‒hard tissue interfaces in pigs.

Biomineralization-inspired mineralized hydrogel promotes the repair and regeneration of dentin/bone hard tissue

Wen Bo,Dai Yuguo,Han Xue,Huo Fangjun,Xie Li,Yu Mei,Wang Yuru,An Ning,Li Zhonghan,Guo Weihua

Maxillofacial hard tissue defects caused by trauma or infection often affect craniofacial function. Taking the natural hard tissue structure as a template, constructing an engineered tissue repair module is an important scheme to realize the functional regeneration and repair of maxillofacial hard tissue. Here, inspired by the biomineralization process, we constructed a composite mineral matrix hydrogel PAA-CMC-TDM containing amorphous calcium phosphates (ACPs), polyacrylic acid (PAA), carboxymethyl chitosan (CMC) and dentin matrix (TDM). The dynamic network composed of Ca 2+ ·COO − coordination and ACPs made the hydrogel loaded with TDM, and exhibited self-repairing ability and injectability. The mechanical properties of PAA-CMC-TDM can be regulated, but the functional activity of TDM remains unaffected. Cytological studies and animal models of hard tissue defects show that the hydrogel can promote the odontogenesis or osteogenic differentiation of mesenchymal stem cells, adapt to irregular hard tissue defects, and promote in situ regeneration of defective tooth and bone tissues. In summary, this paper shows that the injectable TDM hydrogel based on biomimetic mineralization theory can induce hard tissue formation and promote dentin/bone regeneration.

Glycerol solutions of highly concentrated biomineral counter-ions towards water-responsive mineralization: Demonstration on bacterial cellulose and its application in hard tissue repair

Zhao Yunfei,Liu Xiaohao,Zhou Zhi,Feng Chaobo,Luo Nan,Yan Jiajun,Tan Shuo,Lu Yang,Chen Feng,Lu Bing-Qiang,He Shisheng

Mineralization has found widespread use in the fabrication of composite biomaterials for hard tissue regeneration. The current mineralization processes are mainly carried out in neutral aqueous solutions of biomineral counter-ions (a pair of cation and anion that form the corresponding minerals at certain conditions), which are stable only at very low concentrations. This typically results in inefficient mineralization and weak control over biomineral formation. Here, we find that, in the organic solvent glycerol, a variety of biomineral counter-ions (e.g., Ca/PO 4 , Ca/CO 3 , Ca/SO 4 , Mg/PO 4 , or Fe/OH) corresponding to distinct biominerals at significantly high concentrations (up to hundreds-fold greater than those of simulated body fluid (SBF)) are able to form translucent and stable solutions (mineralizing solution of highly concentrated counter-ions (MSCIs)), and mineralization can be triggered upon them with external solvents (e.g., water or ethanol). Furthermore, with pristine bacterial cellulose (BC) membrane as a model, we demonstrate an effective and controllable mineralization performance of MSCIs on organic substrates. This approach not only forms the homogeneous biominerals on the BC fibers and in the interspaces, but also provides regulations over mineralization rate, mineral content, phase, and dopants. The resulting mineralized BC membranes (MBCs) exhibit high cytocompatibility and favor the proliferation of rat bone marrow mesenchymal stem cells (rBMSC). Following this, we prepare a mineralized bone suture (MBS) from MBC for non-weight bearing bone fixation, which then is tested on a rabbit median sternotomy model. It shows firm fixation of the rabbit sternum without causing discernible toxicity or inflammatory response. This study, by extending the mineralization to the organic solution system of highly concentrated counter-ions, develops a promising strategy to design and build targeted mineral-based composites.

Rough surface of copper-bearing titanium alloy with multifunctions of osteogenic ability and antibacterial activity

Hui Liu,Rui Liu,Ihsan Ullah,Shuyuan Zhang,Ziqing Sun,Ling Ren,Ke Yang

Implant-related infection and early bone integration are the main risk factors of implants for long-term service, to overcome these difficulties, SLA-TiCu surface was prepared by sandblasting and large-grits etching (SLA) treatment on a novel antibacterial titanium-copper alloy (TiCu), which is the most prevalent surface treatment with micro/submicron hierarchical structures to titanium-based implants. Effects of SLA-TiCu surface on the adhesion, proliferation, apoptosis and differentiation of MC3T3-E1 cells as well as the antibacterial activity against a common orthopedic pathogen ( Staphylococcus aureus ) were studied. Compared to the following surfaces: sandblasting and large-grits etched pure titanium (SLA-Ti), mechanically ground pure titanium and TiCu alloy (M-Ti and M-TiCu), these results indicated that SLA-TiCu surface obviously enhanced the bone-related gene expressions (alkaline phosphates (ALP), collagen type I (COL I), Runt-related transcription factor 2 (RUN × 2), and osteopontin (OPN)). Moreover, SLA-TiCu surface could maintain a sustainable release of Cu 2+ ions and effectively inhibited the viability of bacteria. This study demonstrated that SLA-TiCu surface possessed multifunctional characteristics of improved osteogenic ability and antibacterial activity, making it promising as a novel implant material for hard tissue repairs such as orthopedics and dental implants .
