Background Duraplasty is one of the most critical issues in neurosurgical procedures because the defect of dura matter will cause many complications. Electrospinning can mimic the 3D structure of the natural extracellular matrix whose structure is similar to that of dura matter. Poly(L-lactic acid) (PLLA) has been used to fabricate dura matter substitutes and showed compatibility to dural tissue. However, the mechanical properties of the PLLA substitute cannot match the mechanical properties of the human dura mater. Methods and Results We prepared stereocomplex nanofiber membranes based on enantiomeric poly(lactic acid) and poly(D-lactic acid)-grafted tetracalcium phosphate via electrospinning. X-ray diffraction results showed the formation of stereocomplex crystallites (SC) in the composite nanofiber membranes. Scanning electron microscope observation images showed that composites nanofibers with higher SC formation can keep its original morphologies after heat treatment, suggesting the heat resistance of composite nanofiber membranes. Differential scanning calorimeter tests confirmed that the melting temperature of composite nanofiber membranes was approximately 222°C, higher than that of PLLA. Tensile testing indicated that the ultimate tensile strength and the elongation break of the stereocomplex nanofiber membranes were close to human dura matter. In vitro cytotoxicity studies proved that the stereocomplex nanofiber membranes were non-toxic. The neuron-like differentiation of marrow stem cells on the stereocomplex nanofiber membranes indicated its neuron compatibility. Conclusion The stereocomplex nanofiber membranes have the potential to serve as a dura mater substitute.

Dura mater defect and subsequent cerebrospinal fluid (CSF) leakage usually appear in trauma or neurosurgical procedures and are followed by a series of serious complications and even death. The use of a qualified dura mater substitute with multifunction of leakage blockade, adhesion prevention, and dura reconstruction is one of the promising treatment methods. However, even though some products have been used in the clinic, none of the substitutes achieved the required multifunction. In this study, we aimed to design and fabricate a dura repair composite with the ideal multifunction. By biomimicking the structure and component of natural dura, we applied poly(L-lactic acid) (PLLA), chitosan (CS), gelatin, and acellular small intestinal submucosa (SIS) powders to successfully prepare a triple-layered composite. Then, a series of specific devices and techniques were developed to investigate the performance. The results revealed that satisfactory structural stability could be realized under good synergistic interactions among the components. In addition, all the findings suggested that the bionic triple-layered composite showed satisfactory multifunction of leakage blockade, adhesion prevention, antibacterial property , and dura reconstruction potential, and thus, it might be a promising candidate for dura repair. Statement of significance Developing qualified dura mater substitutes with multifunction of leakage blockade, adhesion prevention, and dura reconstruction is crucial for treating dura mater defect and subsequent cerebrospinal fluid (CSF) leakage that appear in trauma or neurosurgical procedures. In this study, we designed and fabricated a triple-layered dura repair biocomposite with satisfactory structural stability and desired multifunction based on biomimicking of the structure and component of natural dura. Moreover, a series of specific devices and techniques were developed to investigate the relevant performance. Overall, the developed hydrogel electrospinning system exhibited excellent advantages in achieving multifunction and could be applied widely in the future to achieve multifunctional tissue repair materials.

The development and treatment of some diseases, such as large-area cerebral infarction, cerebral hemorrhage, brain tumor, and craniocerebral trauma, which may involve the injury of the dura mater, elicit the need to repair this membrane by dural grafts. However, common dural grafts tend to result in dural adhesions and scar tissue and have no further neuroprotective effects. In order to reduce or avoid the complications of dural repair, we used PLGA, tetramethylpyrazine, and chitosan as raw materials to prepare a nanofibrous dura mater (NDM) with excellent biocompatibility and adequate mechanical characteristics, which can play a neuroprotective role and have an antifibrotic effect. We fabricated PLGA NDM by electrospinning, and then chitosan was grafted on the nanofibrous dura mater by the EDC-NHS cross-linking method to obtain PLGA/CS NDM. Then, we also prepared PLGA/TMP/CS NDM by coaxial electrospinning. Our study shows that the PLGA/TMP/CS NDM can inhibit the excessive proliferation of fibroblasts, as well as provide a sustained protective effect on the SH-SY5Y cells treated with oxygen–glucose deprivation/reperfusion (OGD/R). In conclusion, our study may provide a new alternative to dural grafts in undesirable cases of dural injuries. Keywords:nanofibrous dura mater;antifibrosis;neuroprotection;PLGA;tetramethylpyrazine

This study presents a dual-layer artificial dura mater, a hierarchically structured fibrous membrane composed of poly(p-dioxanone) (PPDO) and bioactive glass (BG), fabricated using electrospinning and melt-casting techniques. Designed to address the challenges of dura mater repair, the membrane features a dense outer PPDO layer for mechanical resilience and an electrospun inner layer embedded with BG to enable controlled ion release, promoting tissue regeneration and angiogenesis. We evaluated the fibrous membrane’s surface morphology, mechanical properties, hydrophilicity, and in vitro degradation, demonstrating that increasing BG content enhances hydrophilicity, reduces crystallinity, and modulates degradation kinetics. In vitro assays using L929 fibroblasts and human umbilical vein endothelial cells reveal that the PPDO/BG membrane not only supports cell adhesion and proliferation but also fosters a pro-angiogenic environment through the controlled release of bioactive silicon ions. In vivo implantation in a rat dura mater defect model further validates its therapeutic potential, showing reduced adhesion, improved tissue integration, and enhanced vascularization, with the PBD-3 variant exhibiting superior performance due to its optimized BG composition. The synergistic effects of bioactive ion release, mechanical stability, and biocompatibility establish the PPDO/BG membrane as a highly promising dura mater substitute, offering a bioengineered solution for neurosurgical applications aimed at functional tissue regeneration.

Dural injuries often result in severe complications such as cerebrospinal fluid (CSF) leakage, intracranial infections, and brain herniation, which significantly impact patient recovery and quality of life. Conventional dural repair materials, which rely on suturing to peripheral tissues, fail to promote tissue regeneration and provide sufficient CSF leakage prevention, leading to suboptimal outcomes. To address these limitations, we developed a Janus adhesive bio-patch with both antibacterial and pro-angiogenic properties to enhance dura mater repair. This bio-patch consisted of a polyacrylic acid (PAA) adhesive gel layer loaded with vancomycin and magnesium carbonate (MgCO 3 ), integrated onto a small intestinal submucosa (SIS) extracellular matrix. It exhibited a burst strength of 20.50±2.89kPa, effectively sealing CSF leaks, while demonstrating excellent antibacterial efficacy (∼99%) and significant enhanced angiogenesis (3.47-fold higher than the control). In rat, rabbit, and dog dural injury models, the bio-patch adhered seamlessly to the injury site, successfully preventing leaks and promoting tissue regeneration. These results highlighted the Janus adhesive bio-patch as a promising solution for improving dural repair in neurosurgery, offering a safer and more effective alternative to conventional suturing techniques.

Dural closure after the neurosurgery can prevent postoperative complications. Although many types of dural substitute have been developed, most of them lack functional and structural characteristics compared with the natural dura mater. In this study, we used electrospinning method to fabricate a multilayer scaffold to promote dural repair. The inner layer of the scaffold that faces the brain tissue is composed of poly-lactic acid (PLA) to reduce tissue adhesion. The middle layer of the scaffold is composed of poly-ɛ-caprolactone and PLA, which provides a watertight seal. The outer layer of the scaffold contains a large amount of collagen to promote cell attachment and proliferation. The results from in vitro study and an animal model have shown that this multilayer fibrous scaffold has sufficient mechanic strength and biochemical properties to enhance dural repair. Therefore, fabrication of scaffold with multiple functional and structural layers may provide a novel approach for tissue engineering. © 2013 Wiley Periodicals, Inc. Cerebral lymphatic drainage is an important pathway for metabolic waste clearance in the brain, which plays a crucial role in the progression of central nervous system diseases. Recent studies have shown that norepinephrine (NE) is involved in the regulation of cerebral lymphatic drainage function, but the modulation mechanism remains unknown. In this study, we confirmed that NE rapidly reduced glymphatic influx and enhanced meningeal lymphatic clearance. Moreover, the transverse sinus (TS) was the vital region of cerebral lymphatic drainage regulation by NE. Further analysis revealed that NE inhibition could simultaneously enhance glymphatic drainage and dorsal meningeal lymphatic drainage, mainly acting on the TS region. This study demonstrated that the cerebral lymphatic drainage system can be regulated by NE, with the TS region serving as the primary modulating site. The findings provide a potential regulatory target for the amelioration of neurological diseases associated with cerebral lymphatic drainage function.

Management of dura tear during spine surgery remain challenging due to the complex focal environment. Current sealants could hardly independently repair dura tear while simultaneously regulate epidural inflammation and fibrosis induced by oxidative stress. A bioadhesive hydrogel sealant with active reactive oxygen species (ROS)-scavenging potential for suppression of epidural inflammation and fibrosis was designed to provide a versatile tool for spine surgeon. In order to achieve such goal, tannic acid-functionalized black phosphorus nanosheets with better ambient stability were incorporated and integrated into network of hydrogel sealant. Such integration was found to bring improved mechanical strength and sealing performance as evidenced by higher burst pressure and effective sealing of dura tear under in vivo environment. Moreover, instead of merely acting as a physical sealing barrier, the hydrogel sealant was also found with active ROS-scavenging capability. Effective inhibition on oxidative stress-mediated epidural inflammation and fibrosis was observed with employment of hydrogel sealant as revealed by both in vitro and in vivo studies. The proposed hydrogel sealant could provide a potential candidate for sealing of dura tear, and may hopefully provide a promising strategy for prevention of epidural fibrosis.

Cerebrospinal fluid (CSF) leakage caused by accidents or diseases resulting from traumatic brain injury, inflammation, tumor erosion and surgery can lead to many complications. In this study, a multifunctional composite double-layer hydrogel was designed by simulating the structure of native dura mater, which was composed of polyacrylic acid (PAA), polyethyleneimine (PEI), sodium alginate (SA), β-cyclodextrin (β-CD) and edaravone (Ed). The PAA/PEI layer had strong wet adhesion characteristics, while the PEI/SA@β-CD/Ed layer exhibited significant antioxidant, drug release and biocompatibility properties. By controlling the concentration of Ca2+, the gelation time can be adjusted rapidly within 95–215 s. Specifically, the final PAA/PEI/SA@β-CD/Ed composite hydrogel exhibited a porous network structure with high porosity and low swelling rate, improved tensile strength, sufficient biodegradability, favourable adhesion performance, enhanced DPPH and ABTS radicals scavenging abilities, and sustained Ed release capacity. In addition, the resulting hydrogel also showed excellent biocompatibility and protective effect on H2O2-induced oxidative damage in SH-SY5Y cells. These results preliminarily suggested that the PAA/PEI/SA@β-CD/Ed composite hydrogel would appear to be a promising candidate for dural repair.

The dura mater is the final barrier against cerebrospinal fluid leakage and plays a crucial role in protecting and supporting the brain and spinal cord. Head trauma, tumor resection and other traumas damage it, requiring artificial dura mater for repair. However, surgical tears are often unavoidable. To address these issues, the ideal artificial dura mater should have biocompatibility, anti-leakage, and self-healing properties. Herein, this work has used biocompatible polycaprolactone diol as the soft segment and introduced dynamic disulfide bonds into the hard segment, achieving a multifunctional polyurethane (LSPU-2), which integrated the above mentioned properties required in surgery. In particular, LSPU-2 matches the mechanical properties of the dura mater and the biocompatibility tests with neuronal cells demonstrate extremely low cytotoxicity and do not cause any negative skin lesions. In addition, the anti-leakage properties of the LSPU-2 are confirmed by the water permeability tester and the 900 mm H 2 O static pressure test with artificial cerebrospinal fluid. Due to the disulfide bond exchange and molecular chain mobility, LSPU-2 could be completely self-healed within 115 min at human body temperature. Thus, LSPU-2 comprises one of the most promising potential artificial dura materials, which is essential for the advancement of artificial dura mater and brain surgery.

Designing tough biopolymer-based hydrogels as structural biomaterials has both scientific and practical significances. We report a facile approach to prepare polysaccharide-based hydrogel films with remarkable mechanical performances and antiadhesion property. The hydrogel films with a thickness of 40–60 μm were prepared by mixing aqueous solutions of κ-carrageenan (κ-CG) and protonated chitosan (CS), evaporating the solvent, and then swelling the casted film in water to achieve the equilibrium state. The obtained κ-CG/CS gel films with a water content of 48–88 wt % possessed excellent mechanical properties with a breaking stress of 2–6.7 MPa and a breaking strain of 80–120%, superior to the most existing biopolymer-based hydrogels. The extraordinary mechanical properties of gel films obtained over a wide range of mass ratio of κ-CG to CS should be rooted in the synergistic effect of ionic and hydrogen bonds between the κ-CG and CS molecules. In addition, the tough gel films showed good self-recovery ability, biocompatibility, and cell antiadhesion property, making them promising as an artificial dura mater and diaphragm materials in the surgery. The design principle by incorporating multiple noncovalent bonds to toughen the biopolymer-based hydrogels should be applicable to other systems toward structural biomaterials with versatile properties.
