l-glutamate inhibits blue mould caused by Penicillium expansum in apple fruits by altering the primary nitrogen and carbon metabolisms

Jiali Yang,Tengfei Wang,Jianbing Di,Yaping Liu,Xiaoling Hao,Yu Wang

Summary Although several studies have shown the ability of l -glutamate (glutamate) to mitigate the stress imposed by pathogens, the underlying mechanism is still in its infancy. To gain further knowledge, this study focussed on the effect of glutamate on primary nitrogen and carbon metabolisms during apple- Penicillium expansum interaction. The obtained result showed that glutamate could effectively restrict blue mould rot development in apples, but had no direct impact on fungal growth in vitro . The application of glutamate increased the level of nitrogen in apples, resulting in a disrupted balance of carbon and nitrogen. Consistently, the key enzymes glutamine synthetase (GS) and glutamate synthase (GOGAT) involved in the GS/GOGAT cycle, and the deaminating activity of glutamate dehydrogenase (GDH), forming 2-oxoglutarate and ammonium from glutamate, were promptly stimulated by glutamate. Interestingly, glutamate led to considerable consumption of the tricarboxylic acid (TCA) cycle intermediates, such as isocitric acid and citric acid, accompanied by the enhancement of malate dehydrogenase and succinate dehydrogenase activities. Collectively, exogenous application of glutamate might confer blue mould resistance in apples, at least in part, by redirecting host’s primary nitrogen and carbon metabolisms, such as the activation of the GS/GOGAT cycle, deaminating activity of GDH and the TCA cycle.

Metabolic pathways of methylmercury in rotifer Brachionus plicatilis

Hangyu Lin,Xiaodong Mao,Yanlin Wei,Songzhang Li,Jianguang Qin,Song Zhu,Shengqi Su,Tao He

Methylmercury (MeHg) readily accumulates in aquatic organisms while transferring and amplifying in the aquatic food chains. This study firstly explores the in vivo accumulation sites and metabolic regulation of MeHg in the rotifer Brachionus plicatilis by aggregation-induced emission fluorogen (AIEgen) and metabolomics. Fluorescent image analysis by AIEgen showed that MeHg in B . plicatilis mainly occured in the ciliary corona, esophagus, mastax, stomach and intestine in the direct absorption group. In the other group, where B . plicatilis were indirectly supplied with MeHg via food intake, the accumulation of MeHg in the rotifer occurred in the ciliary corona, various digestive organs, and the pedal gland. However, the MeHg accumulated in the rotifer is difficult to metabolize outside the body. Metabolomics analysis showed that the significant enrichment of ABC transporters was induced by the direct exposure of rotifers to dissolved MeHg. In contrast, exposure of rotifers to MeHg via food intake appeared to influence carbon, galactose , alanine , aspartate and glutamate metabolisms. Besides, the disturbed biological pathways such as histidine metabolism, beta-alanine metabolism and pantothenate and CoA biosynthesis in rotifers may be associated with L-aspartic acid upregulation in the feeding group. The significant enrichment of ABC transporters and carbon metabolism in rotifers may be related to the accumulation of MeHg in the intestine of rotifers. In both pathways of MeHg exposure, the arginine biosynthesis and metabolism of rotifers were disturbed, which may support the hypothesis that rotifers produce more energy to resist MeHg toxicity. This study provides new insight into the accumulation and toxicity mechanisms of MeHg on marine invertebrates from the macro and micro perspectives.

Carbon fate and potential carbon metabolism effects during in-situ cyanobacterial inhibition by artemisinin sustained-release algaecides

Yushen Ma,Xiaoming Zhu,Lixiao Ni,Cunhao Du,Wenlu Sang,Chu Xu,Jiahui Shi,Yiping Li,Shiyin Li,Linyun Zhang

Carbon pools and microbial carbon metabolism can be significantly altered due to the diverse organic matter properties and low pH characteristics of artemisinin sustained-release algaecides (ASAs). These effects have still not been systematically studied, leading to uncertainty in the application of ASAs for cyanobacterial management in natural waters. This study assessed the effects of ASAs on carbon fate, carbon metabolism and potential ecological impacts during in-situ cyanobacterial inhibition. In the initial phase of ASAs-induced cyanobacterial inhibition (2–10 days), the carbon pool underwent significant changes due to the increased proportion of C4 plant-derived organic matter (97 %), humification level (FI =0.9–1.5), and inorganic carbon concentration (TIC = 80–110 mg/L). The cyanobacterial apoptosis triggered by ASAs produced particulate organic carbon, which provided a bioavailable carbon source for bacterial metabolism. ASAs enhanced the connection between bacteria and the carbon pool, as well as their carbon metabolism capabilities, by increasing the relative abundance of Proteobacteria (33.1 %−37.3 %), bacterial diversity, the proportion of Alphaproteobacteria and Betaproteobacteria (19.3 %−29.5 %) involved in carbon metabolism, and the complexity of the environment-bacteria co-occurrence network. These effects contributed to maintaining long-term ecosystem stability and resistance to cyanobacterial proliferation. Our findings elucidate the influence of bacterial carbon metabolism by ASAs as one of an important mechanism for achieving cyanobacterial inhibition in natural water, and emphasize the important role of bacteria in maintaining ecological stability during in situ cyanobacterial management.

A New Type of Quantum Fertilizer (Silicon Quantum Dots) Promotes the Growth and Enhances the Antioxidant Defense System in Rice Seedlings by Reprogramming the Nitrogen and Carbon Metabolism

Hao Jiang,Haodong Wang,Cancan Qian,Zhenlong Yang,Desong Yang,Jianghu Cui

To promote the growth and yield of crops, it is necessary to develop an effective silicon fertilizer. Herein, a new type of 2 nm silicon quantum dot (SiQD) was developed, and the phenotypic, biochemical, and metabolic responses of rice seedlings treated with SiQDs were investigated. The results indicated that the foliar application of SiQDs could significantly improve the growth of rice seedlings by increasing the uptake of nutrient elements and activating the antioxidative defense system. Furthermore, metabolomics revealed that the supply of SiQDs could significantly up-regulate several antioxidative metabolites (oxalic acid, maleic acid, glycine, lysine, and proline) by reprogramming the nitrogen- and carbon-related biological pathways. The findings provide a new strategy for developing an effective and promising quantum fertilizer in agriculture.

Unlocking the potential of Komagataeibacter europaeus FMES: A novel source of bacterial cellulose from Monascus vinegar biofilm

Siyu Liu,Jiaqi Luo,Ying Shi,Qihe Chen

Various bacterial species synthesize bacterial cellulose (BC), each exhibiting unique metabolic pathways and environmental adaptations. Herein, the microbial profile of Monascus vinegar biofilm was analyzed for the first time using metagenome sequencing, and a BC-producing bacteria, Komagataeibacter europaeus FMES, was isolated from Monascus vinegar biofilm and identified as a novel strain. The complete genome of strain FMES was analyzed using whole genome sequencing, revealing carbon metabolism and BC biosynthesis pathways in detail. The biosynthesis of BC is regulated by the bacterial cellulose synthase ( bcs ) operon, and three bcs operons were identified in the genome. Additionally, the BC production of strain FMES was evaluated under static conditions using different carbon sources. The highest BC yield of 5.89 ± 0.38 g/L was achieved using 5% glycerol as the only carbon source. The addition of 3% ethanol enhanced the biosynthesis of BC by K. europaeus FMES. This work brings valuable insights into the isolation of BC-producing strains from fermented foods and the metabolic pathways of BC synthesis, which provides useful information for further biological research on Komagataeibacter spp., and the potential use of BC in medical wound care and sustainable packaging.

Resistance characterization and transcriptomic analysis of imipenem-induced drug resistance in Escherichia coli

Chunyu Tong,Yimin Liang,Qi Liu,Honghao Yu,Wenzhi Feng,Bocui Song

Background Bacteria can develop resistance to various antibiotics under selective pressure, leading to multifaceted changes in resistance mechanisms. Transcriptomic sequencing allows for the observation of transcriptional level alterations in cells under antibiotic stress. Understanding the bacterial response to such stress is essential for deciphering their strategy against drug-resistant antibiotics and identifying potential targets for antibiotic development. Methods This study using wild-type (WT) Escherichia coli ( E. coli ) discovered that continuous in vitro induction screening for imipenem-resistant strains resulted in bacteria with enhanced biofilm-forming ability and mutations in antibiotic target sites. Transcriptomic sequencing of the resistant bacteria revealed significant changes in carbon and amino acid metabolism, nutrient assimilation, substance transport, nucleotide metabolism, protein biosynthesis, and cell wall biosynthesis. The up-regulated drug efflux genes were disrupted using gene knockout technology. Drug sensitivity tests indicated that drug efflux has a minimal effect on imipenem resistance. Results This suggests a strategy for E. coli drug resistance involving the reduction of unnecessary substance synthesis and metabolism, coupled with an increase in activities that aid in resisting foreign threats.

Targeting SHMT2-mediated membrane phospholipid remodeling for enhanced anti-GCSCs treatment

Liping Yang,Fangli Liao,Yanran Tong,Tong Huang,Yan-e Du,Siyang Wen,Linshan Jiang,Lanlang Peng,Hua Sun,Gaoli Zhang,Weixian Chen

Cancer stem cells exhibit flexible metabolic profiles. However, the underlying mechanisms for differential metabolic pathways affecting stemness maintenance in gastric cancer are poorly understood. Here, we reveal the role of serine hydroxymethyltransferase-2 (SHMT2)/serine-mediated crosstalk between one-carbon metabolism and lipid metabolism in the stemness maintenance of gastric cancer. Clinically, SHMT2 was significantly highly expressed in Gastric cancer cells (GCs) and gastric cancer stem cells, and was associated with clinical malignant features and poor prognosis in gastric cancer patients. Mechanistically, inhibition of SHMT2 expression resulted in diminished serine levels in one-carbon metabolism, which subsequently modified the composition and fluidity of membrane phospholipids, leading to a reduction in lipid rafts within cellular membranes. The remodeling of membrane phospholipids hindered the localization of γ-secretase to lipid rafts, thereby inhibiting the cleavage of CD44 and the subsequent production of CD44-ICD. Consequently, the transcriptional regulation of c-Myc and KLF4 by CD44-ICD was reduced, ultimately disrupting the maintenance of stemness in gastric cancer cells. Together, these results provide compelling evidence for the metabolic adaptability of cancer stem cells, and the SHMT2/serine/lipid rafts signaling axis holds promise as a potential biomarker for the diagnosis and prognosis of gastric cancer. Furthermore, we synthesized HA-Exo-si SHMT2 to investigate targeted therapy for GC, offering a novel approach for the clinical treatment of gastric cancer.

Distinct microbial metabolic activities of biofilms colonizing microplastics in three freshwater ecosystems

Lingzhan Miao,Yue Yu,Tanveer M. Adyel,Chengqian Wang,Zhilin Liu,Songqi Liu,Liuyan Huang,Guoxiang You,Meng Meng,Hao Qu,Jun Hou

Concerns are growing about the increasing amounts of microplastics (MPs) and their ecological impacts, especially the influences of “plastisphere” in the freshwater ecosystems. Although the microbial structure and composition of biofilms are investigated, knowledge of their microbial functions remains limited. Herein, we investigated the functional diversity of carbon metabolism in biofilms colonizing one inert (glass) and two MPs as polyvinyl chloride (PVC) and polyethylene terephthalate (PET) substrates incubated for 44 days in situ in the Niushoushan River, the Qinhuai River, and Donghu Lake. 2D confocal laser scanning microscopy images visualized distinct micro-structures and biofilm compositions on three substrates. BIOLOG ECO microplates indicated variation on carbon utilization capacities of biofilms of inert and MPs in three freshwater ecosystems. Biofilms on PET showed lower capacities and carbon metabolism rates than those on glass and PVC, indicating the presence of substrate-specific functional diversity. The Shannon-Wiener diversity, Simpson diversity and Shannon evenness indices for the Niushoushan River and Donghu Lake were ordered as glass > PVC > PET. Besides to MPs-specific factors, environmental factors including nutrient (i.e., TN and TP) and turbidity largely shaped biofilm carbon metabolism. Overall findings demonstrated that as specific niches, MPs influenced microbial-mediated carbon cycling in the freshwater ecosystems and MPs-promoted microbial communities posed ecological significance.

Responses of microbial carbon metabolism and function diversity induced by complex fungal enzymes in lignocellulosic waste composting

Zhuotong Zeng,Xueying Guo,Piao Xu,Rong Xiao,Danlian Huang,Xiaomin Gong,Min Cheng,Huan Yi,Tao Li,Guangming Zeng

Composting is an economic and effective technology for solid waste treatment , which is an essential method to promote the biogeochemical cycle of contaminants. However, the application of this technology was limited by the bio-degradative recalcitrance of lignin and other kind of phytotoxic substances release. The combination with microorganisms and enzymes is a popular and efficient way to enhanced composting. This study, referring to metabolic mechanisms, fungal molecular and biogeochemical cycles, was performed to investigate the effects of lignin degradation, carbon metabolic diversity, as well as the related changes induced by these two kinds of complex enzymes in composting. The biological diversity is important indicator in ecosystem, which concerns the environmental applicability of one technology. The carbon metabolism diversity reflected the biogeochemical cycles of organic matter, which was also an essential input to analyze the effects of composting. The changes on the diversity characteristics of carbon are essential to comprehensively understand the deep mechanisms of this process, and extended the application of complex enzymes in the field of enhanced composting. The analysis of Biolog revealed that the utilization of pyruvic acid methyl ester, α-Cyclodextrin, d -Mannitol, d -Galacturonic, Itaconic acid and l -asparagine were deeply promoted, and that of d , l -α-Glycerol-phosphate, l -Threonine, Glycyl- l -Glutamic acid and putrescine were depressed by adding the complex enzyme in composting. Moreover, according to the data, the addition of complex enzymes improved the degradation efficiency and the metabolic capacity of carbon in composting. These findings undoubtedly contribute to the development of enzyme-based technologies and the applications of complex enzymes in composting, which is of great benefit to eliminate the limitation and extend the application of composting.

Metabolomics analysis reveals that myrcene stimulates the spore germination of Penicillium digitatum via the upregulation of central carbon and energy metabolism

Jinxin Che,Yue Chen,Yalan Wu,Lu Li,Nengguo Tao

Tremendous economic losses in the citrus industry are caused by infestations of Penicillium digitatum . Previous research has shown that the terpene myrcene in citrus essential oils could stimulate the spore germination and mycelial growth of P. digitatum . However, the underlying mechanism is poorly known. Here, how myrcene induces the spore germination of P. digitatum was elucidated. The addition of myrcene significantly promoted the spore germination of P. digitatum and green mold in citrus fruit. As the incubation time increased, the intracellular and extracellular myrcene contents both significantly decreased in myrcene-treated samples, and no downstream products were detected through a Gas Chromatography-Mass Spectrometer. Metabolomics analysis revealed that the contents of metabolites involved in central carbon metabolism, such as glycolysis, pentose phosphate pathway, and citrate cycle, were significantly affected after the addition of myrcene. The adenosine triphosphate content and the intracellular energy charge were also drastically increased. Overall, our results suggest that the ability of myrcene to stimulate the spore germination of P. digitatum might be mediated by the upregulation of central carbon and energy metabolism.

Up-regulation of ribosomal and carbon metabolism proteins enhanced pyrene biodegradation in fulvic acid-induced biofilm system

Minfen Gu,WenWen Fang,Xiaoning Li,Weiben Yang,Michael Gatheru Waigi,Fredrick Orori Kengara,Shixi Wu,Cheng Han,Yinping Zhang

The polycyclic aromatic hydrocarbons (PAHs) that enter the aqueous phase usually coexist with fulvic acid (FA). Therefore, we initiated this investigation to explore the influences of FA on bacterial biofilm formation and its potential to biodegrade pyrene (PYR), using electron microscopic techniques and isobaric tags for relative and absolute quantification (iTRAQ). Our results revealed that FA stimulated biofilm formation and enhanced the biodegradation of PYR. First, FA favored the three-dimensional proliferation of bacteria, with an OD590/OD600 value of up to 14.78, and the extracellular surfaces covered by a layer of biomaterials. Distinctive intracellular morphologies of texture and organization were accompanied by reduced inter-bacterial distances of less than 0.31 μm. The biofilms formed displayed interactions between FA and surficial proteins, as noted by band shifts for the C–O and C O groups. Strikingly, FA triggered the upregulation of 130 proteins that were either operational in biofilm formation or in metabolic adjustments; with the changes supported by the increasing intensity of free amino acids and the newly generated N–O bonds. The results above revealed that the enhanced biodegradation was related to the up-regulation of the proteins functioned for ribosomal and carbon metabolism, and the ultra-structural changes in FA-induced biofilm system.

Acetylacetone Interferes with Carbon and Nitrogen Metabolism of Microcystis aeruginosa by Cutting Off the Electron Flow to Ferredoxin

Mihebai Yilimulati,Lang Zhou,Dmitry Shevela,Shujuan Zhang

The regulation of photosynthetic machinery with a nonoxidative approach is a powerful but challenging strategy for the selective inhibition of bloom-forming cyanobacteria. Acetylacetone (AA) was recently found to be a target-selective cyanocide for Microcystis aeruginosa, but the cause and effect in the studied system are still unclear. By recording of the chemical fingerprints of the cells at two treatment intervals (12 and 72 h with 0.1 mM AA) with omics assays, the molecular mechanism of AA in inactivating Microcystis aeruginosa was elucidated. The results clearly reveal the effect of AA on ferredoxin and the consequent effects on the physiological and biochemical processes of Microcystis aeruginosa. In addition to its role as an electron acceptor of photosystem I, ferredoxin plays pivotal roles in the assimilation of nitrogen in cyanobacterial cells. The effect of AA on ferredoxin and on nonheme iron of photosystem II first cut off the photosynthetic electron transfer flow and then interrupted the synthesis of adenosine triphosphate (ATP) and reduced nicotinamide adenine dinucleotide phosphate (NADPH), which ultimately might affect carbon fixation and nitrogen assimilation metabolisms. The results here provide missing pieces in the current knowledge on the selective inhibition of cyanobacteria, which should shed light on the better control of harmful blooms.
